The expression of alpha 5 beta 1 (alpha 5 beta 1) integrin and its extracellular ligand fibronectin was studied immunohistochemically in 23 cases of Dupuytren's disease using an immunoperoxidase method for light microscopic visualization. All cases consisted of multiple nodules showing a variable degree of cellularity and fibrosis. Depending on the histological appearance of these nodules, each case was assigned to the three following phases: proliferative, involutional and residual. Alpha 5 beta 1 integrin was detected in the highly cellular areas of both proliferative and involutional phases

The integrins are a large family of heterodimeric cell-surface glycoproteins that play key roles in the adherence of cells to other cells and to extracellular matrix proteins. We have previously reported the identification of a novel integrin beta subunit partial cDNA from leukocytes. We have now determined the complete sequence of this subunit, designated as beta 7, from overlapping clones obtained from a PEER T leukemia cell library and a peripheral T cell library. The beta 7 cDNA contains a single large open reading frame predicted to encode a 798-amino acid protein precursor (signal peptid…

Microtubules are dynamic cytoskeletal filaments that have important organizing roles in all eukaryotic cells. Early in vitro studies on the effect of pH on microtubule stability focused on microtubules isolated from whole cell lysates, which can only replicate changes in intracellular pH. However, how extracellular pH can affect microtubule dynamics remains unclear. Here, we report that acidosis activates β1 integrin by increasing its affinity for RGD-containing ligands through the displacement of divalent ions in the metal-ion-binding sites of β1 integrin extracellular domain via protonation

OBJECTIVES: Anti-integrin αvβ6 Immunoglobulin G (anti-αvβ6 IgG) has been identified as a biomarker for ulcerative colitis (UC) in adults, but they were also detected in pediatric Crohn's disease (CD). We hypothesized that their less specificity in this population could reflect colonic inflammation rather than disease type and may be influenced by HLA-DQA1*05 carriage.
METHODS: We conducted a monocentric retrospective study using serum and clinical data from 213 pediatric inflammatory bowel disease patients and 110 controls, analyzing the association between anti-αvβ6 IgG and disease location,

The interactions between viral ligands and cellular receptors, along with the subsequent entry of Human cytomegalovirus (HCMV) into host cells, represent an important focus of research in virus-host relationships. This study investigated the function and mechanistic role of α2,6-sialylation of Integrin β3 during the early stages of infection in fibroblasts. Analysis of the human Integrin β3 glycomics database revealed that the surface glycans of Integrin β3 are extensively modified with terminal sialic acid residues and occur at relatively high abundance. To evaluate their functional relevance

Prostate cancer, particularly its metastatic castration-resistant form, presents major challenges due to tumor heterogeneity and limited treatment efficacy. The inherent limitations of single-target radiopharmaceuticals, such as variable antigen expression, underscore the need for innovative strategies. To address this, we developed a novel dual-targeting radioligand, 177Lu-PSMA-3-RGD, via solid-phase synthesis and click chemistry, designed to simultaneously bind prostate-specific membrane antigen (PSMA) and integrin αvβ3. The ligand demonstrated high radiochemical purity, excellent radiostabi

Integrins play a central role in a wide range of biological processes, including development, tissue homeostasis, immune responses, and disease progression. Their activation is traditionally explained by two major signaling modes: inside-out signaling, which converts integrins to ligand-competent and high-affinity states through intracellular activators, including the Rap1-talin-kindlin axis, and outside-in signaling, which transduces ligand engagement into cytoskeletal coupling and downstream signaling. Recent findings indicate that this division is incomplete. Ligand-engaged integrins can al…

BACKGROUND: Colorectal cancer (CRC) is the third most common cancer in the world. Bone metastasis, which occurs in advanced cancer, causes severe damage and accelerates patient death. TROP2, encoded by the TACSTD2 gene, is highly expressed in multiple types of cancer and is associated with tumor proliferation, invasion and metastasis. However, the role of TROP2 in CRC is unclear.
METHODS: TROP2 expression was detected in tumor tissues of CRC patients with or without bone metastases, as well as CRC cell lines. The functional role of TROP2 on CRC cells was validated by in vitro cell culture mod

Integrins, a family of transmembrane receptors that mediate cell-extracellular (ECM) interactions, play a pivotal role in the heart to maintain cardiac function and tissue integrity. As they sense extracellular signals, their expression changes in a microenvironment-dependent manner. Especially following myocardial infarction (MI), several alpha integrins are upregulated. Integrin α3 (ITGA3) expression has been found upregulated after MI, yet little is known about the function of ITGA3 in cardiac tissue. To evaluate the role of ITGA3 in vivo. we generated a mouse line with cardiomyocytes-speci…

BACKGROUND: Pancreatic ductal adenocarcinoma (PDAC) is one of the most lethal malignancies and is characterised by aggressive biological behaviour, marked therapeutic resistance and a dense desmoplastic tumour microenvironment. Integrin α5β1, the principal fibronectin receptor, has emerged as an important mediator of tumour-stroma interactions through its roles in cell adhesion, mechanotransduction, migration, survival and extracellular matrix (ECM) remodelling.
MAIN BODY: Increasing evidence indicates that integrin alpha 5 (ITGA5)/integrin α5β1 is upregulated in PDAC cells and stromal compar

The integrin heterodimer CD49d/CD29 (a.k.a. Very Late Antigen-4, VLA-4) mediates cell-cell and cell-matrix interaction through binding to its specific ligands. VLA-4 can be present on the cell surface at different conformation states that affect the binding affinity for the ligands. In chronic lymphocytic leukemia (CLL), higher VLA-4 levels have been demonstrated to be associated with a worse prognosis both in the chemo-immunotherapy era and in the BCR inhibitor setting, in keeping with the role of VLA-4 as a key molecule favoring CLL cell localization in protective niches of bone marrow and l

Curcumin has attracted attention for its nontoxic chemopreventive effects; however, the pathways underlying these effects in colon cancer remain unclear. We investigated the potential interplay between curcumin and integrin β1 in colon cancer. Human colon cancer (HCT) 116 cell line and fibroblast cells were treated with curcumin, and cell proliferation was assessed using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay and western blotting. Using western blotting and confocal microscopy in curcumin-treated HCT116 cells, we analyzed integrin β1 expression and the roles of tali

The blood-brain barrier (BBB) is critical for maintaining cerebral homeostasis, and its deterioration with age is an important pathogenic factor in the etiology of vascular dementia. Extracellular matrix-integrin interactions play a central role in regulating vascular stability. The αvβ3 integrin is not expressed by brain endothelial cells under stable conditions but is strongly induced by hypoxia. However, it is currently unclear whether αvβ3 integrin exerts a destructive or protective influence on BBB integrity. In young (8 to 10 wk) and aged (20 mo) mice, we examined the impact of a functio

Neutrophils are the first responders among peripheral immune cells to infiltrate the central nervous system following a traumatic brain injury (TBI), triggering neuroinflammation that can exacerbate secondary tissue damage. The precise molecular controls that dictate the inflammatory behavior of neutrophils post-TBI, however, remain largely elusive. Our comprehensive analysis of the molecular landscape surrounding the trauma in TBI mice has revealed a significant alteration in the abundance of β2 integrin (ITGB2), predominantly expressed by neutrophils and closely associated with immune respon
