[The reconstruction of ion channels created by protein neurotoxins in bilayer lipid membranes as a research tool for exocytosis processes].

The direct transport of inorganic ions and much highly organized biomolecules across native membranes by the specific membrane proteins--ionic channels provides the functioning of a number of important biochemical processes like depolarization, Ca(2+)-release and exocytosis in nerve or muscle cells at the synaptic transmission. Therefore, the investigation of the structure and ion-conducting properties for neurotoxin channels and different factors affecting channels themselves and neuro-muscular transmission plays an important role in the research of pathologies that appear in intoxicated nerv

Nonsteroid anti-inflammatory drugs inhibit both the activity and the inflammation-induced expression of acid-sensing ion channels in nociceptors.

Nonsteroid anti-inflammatory drugs (NSAIDs) are major drugs against inflammation and pain. They are well known inhibitors of cyclooxygenases (COXs). However, many studies indicate that they may also act on other targets. Acidosis is observed in inflammatory conditions such as chronic joint inflammation, in tumors and after ischemia, and greatly contributes to pain and hyperalgesia. Administration of NSAIDs reduces low-pH-induced pain. The acid sensitivity of nociceptors is associated with activation of H(+)-gated ion channels. Several of these, cloned recently, correspond to the acid-sensing i…

Multiscale ion transport channels based on chitosan and nanocellulose: Synergistic enhancement of mechanical properties and ionic conductivity, and multifunctional integration.

Eutectogels are promising in flexible wearable electronics, but the inherent trade-off between mechanical strength and ionic conductivity limits their development. Here, we introduced polyethyleneimine-modified cellulose nanocrystals (PEI@CNC) and chitosan (CS) into a polyvinyl alcohol (PVA) matrix, and constructed a PVA composite eutectogel (PCxSy) with an aligned lamellar porous structure via directional freezing. The anisotropic low-tortuosity channels, combined with the incorporation of PEI@CNC and CS that provide additional ion-conducting pathways, endow the material with an ionic conduct

Transient receptor potential ankyrin 1 ion channels may control stress adaptation and conditioned fear responses in male mice following footshock.

INTRODUCTION: Stress maladaptation and dysregulated conditioned fear are hallmarks of trauma- and stressor-related disorders. The centrally projecting Edinger-Westphal nucleus (EWcp) is involved in stress adaptation. As the urocortin 1/cocaine- and amphetamine-regulated transcript (UCN1/CART)-positive neurons of mouse and human EWcp express transient receptor potential ankyrin 1 (TRPA1) mRNA, which reacts to stress, we hypothesized their role in stress adaptation and conditioned fear responses. METHODS: Male Trpa1 wild-type (WT) and knockout (KO) mice underwent footshock-induced fear conditio

Superficial grey layer of superior colliculus integrates visual cue-evoked learning and memory in rats: importance of TRPV3 ion channels.

The superficial grey layer (SuG) of the superior colliculus (SC) receives direct visual sensory inputs from the retina and V1 cortex to drive the motor command. Processing of information in this layer is known to mainly involve glutamatergic, GABAergic and cholinergic signalling systems. In addition, the occurrence of transient receptor potential vanilloid 3 (TRPV3) cationic channels has been detected in the superficial SC, but their functional significance has not been clarified. In our previous study, we have shown that SuG may play an important role in processing visual cues encoding reward

The Complex Interplay between Toxic Hallmark Proteins, Calmodulin-Binding Proteins, Ion Channels, and Receptors Involved in Calcium Dyshomeostasis in Neurodegeneration.

Calcium dyshomeostasis is an early critical event in neurodegeneration as exemplified by Alzheimer's (AD), Huntington's (HD) and Parkinson's (PD) diseases. Neuronal calcium homeostasis is maintained by a diversity of ion channels, buffers, calcium-binding protein effectors, and intracellular storage in the endoplasmic reticulum, mitochondria, and lysosomes. The function of these components and compartments is impacted by the toxic hallmark proteins of AD (amyloid beta and Tau), HD (huntingtin) and PD (alpha-synuclein) as well as by interactions with downstream calcium-binding proteins, especia…

A historical biophysical dogma vs. an understanding of the structure and function of voltage-gated tetrameric ion channels. A review.

The outstanding work of several eminent biophysicists has allowed the functional features of voltage-gated tetrameric ion channels to be disclosed using ingenious and sophisticated electrophysiological techniques. However, the kinetics and mechanism underlying these functions have been heavily conditioned by an arbitrary interpretation of the groundbreaking results obtained by Hodgkin and Huxley (HH) in their investigation of sodium and potassium currents using the voltage clamp technique. Thus, the heavy parametrization of their results was considered to indicate that any proposed sequence of…

Transcriptome-based screening of ion channels and transporters in a migratory chondroprogenitor cell line isolated from late-stage osteoarthritic cartilage.

Chondrogenic progenitor cells (CPCs) may be used as an alternative source of cells with potentially superior chondrogenic potential compared to mesenchymal stem cells (MSCs), and could be exploited for future regenerative therapies targeting articular cartilage in degenerative diseases such as osteoarthritis (OA). In this study, we hypothesised that CPCs derived from OA cartilage may be characterised by a distinct channelome. First, a global transcriptomic analysis using Affymetrix microarrays was performed. We studied the profiles of those ion channels and transporter families that may be rel…

Kv2 Ion Channels Determine the Expression and Localization of the Associated AMIGO-1 Cell Adhesion Molecule in Adult Brain Neurons.

Voltage-gated K+ (Kv) channels play important roles in regulating neuronal excitability. Kv channels comprise four principal α subunits, and transmembrane and/or cytoplasmic auxiliary subunits that modify diverse aspects of channel function. AMIGO-1, which mediates homophilic cell adhesion underlying neurite outgrowth and fasciculation during development, has recently been shown to be an auxiliary subunit of adult brain Kv2.1-containing Kv channels. We show that AMIGO-1 is extensively colocalized with both Kv2.1 and its paralog Kv2.2 in brain neurons across diverse mammals, and that in adult b

Ion permeation and block of the gating pore in the voltage sensor of NaV1.4 channels with hypokalemic periodic paralysis mutations.

Hypokalemic periodic paralysis and normokalemic periodic paralysis are caused by mutations of the gating charge-carrying arginine residues in skeletal muscle Na(V)1.4 channels, which induce gating pore current through the mutant voltage sensor domains. Inward sodium currents through the gating pore of mutant R666G are only approximately 1% of central pore current, but substitution of guanidine for sodium in the extracellular solution increases their size by 13- +/- 2-fold. Ethylguanidine is permeant through the R666G gating pore at physiological membrane potentials but blocks the gating pore a

[Recent insights into the implication of ion channels in familial forms of epilepsies associated or non associated to febrile convulsions].

The etiology of stroke in young patients is often unknown. Although systemic infections as well as specific infection agents, like herpes zoster virus or cysticercus, are often considered as risk factors, there are no indications that herpes simplex type 1 plays a role in the pathogenesis of stroke. We present the case of a young patient who suffered a stroke during a meningoencephalitis due to herpes simplex 1 and we review the relevant literature for a possible relation between the two entities.

Transduction without tip links in cochlear hair cells is mediated by ion channels with permeation properties distinct from those of the mechano-electrical transducer channel.

Tip links between adjacent stereocilia are believed to gate mechano-electrical transducer (MET) channels and mediate the electrical responses of sensory hair cells. We found that mouse auditory hair cells that lack tip links due to genetic mutations or exposure to the Ca(2+) chelator BAPTA can, however, still respond to mechanical stimuli. These MET currents have unusual properties and are predominantly of the opposite polarity relative to those measured when tip links are present. There are other striking differences, for example, the channels are usually all closed when the hair cell is not

Inhibitory effects of sulfur dioxide within the nucleus tractus solitarii of rats: involvement of Calcium Ion channels, Adenine nucleoside triphosphate-sensitive potassium channels, and the nitric oxide/cyclic Guanine trinucleotide phosphate pathway.

This study was designed to investigate the cardiovascular effects of sulfur dioxide within the nucleus tractus solitarii. Sulfur dioxide or artificial cerebrospinal fluid was unilaterally applied into the nucleus tractus solitarii of rats, and the effects on blood pressure, heart rate, and arterial baroreflex sensitivity (ABR) were determined. To explore the mechanisms of the effects of intra-nucleus tractus solitarii sulfur dioxide, various inhibitors were applied prior to sulfur dioxide treatment. Unilateral microinjection of sulfur dioxide produced a dose-dependent decrease in blood pressur

Mechanistic insights into PFAS-induced toxicity: from lipid membrane perturbation to ion channel and transporter dysregulation.

Per- and polyfluoroalkyl substances (PFAS) are persistent environmental contaminants associated with a wide range of adverse health outcomes, including metabolic, developmental, and neurodevelopmental disorders. Recent evidence suggests that PFAS can act as membrane-active compounds, directly interacting with lipid bilayers. Their amphiphilic properties allow incorporation into cellular membranes, altering membrane properties. These physicochemical changes can impact the structural dynamics and functional properties of membrane proteins, particularly ion channels. A growing body of research de…

The stiffness of three-dimensional biomimetic matrices regulates insulin secretion via the Piezo1 mechanosensitive ion channel.

INTRODUCTION: The extracellular matrix (ECM) within the pancreatic microenvironment plays an integral role in the pathogenesis of diabetes. However, how matrix stiffness regulates islet β-cell function through mechanosensitive ion channels remains poorly understood. METHODS: A three-dimensional biomimetic islet culture system with graded matrix stiffness was established using hyaluronic acid methacrylate (HAMA) hydrogels with different crosslinking times. MIN6 insulin-secreting cells were encapsulated in soft and stiff HAMA matrices, and cell viability, Piezo1 expression, intracellular Ca2+ l…

Structural Modifications Redirect Phenylpyrazole Ectoparasiticides from the Ion Channel Pore to an Allosteric Site on GABA Receptors.

Phenylpyrazole ectoparasiticides, exemplified by fipronil, act as noncompetitive antagonists of arthropod GABA receptors by blocking the ion channel pore. Here, we investigated the molecular mechanism of the newer phenylpyrazole nicofluprole using Drosophila melanogaster. CRISPR/Cas9-mediated mutagenesis and pharmacological assays showed that A301 substitutions in the pore-forming TM2 helix conferred strong resistance to fipronil but had little effect on nicofluprole sensitivity. In contrast, G335 mutations in TM3 markedly reduced nicofluprole activity. Molecular docking supported these findin

Pathophysiological significance of the TRPM2 ion channel as a potential target in cancer, neurological disorders, and ischemia/reperfusion injury.

The transient receptor potential melastatin 2 (TRPM2) ion channel is a redox-sensitive, non-specific cation channel that plays a vital role in the regulation of Ca2+ homeostasis and cellular functions in response to oxidative stress. However, aberrant expression of TRPM2 is associated with various pathological conditions. Overexpression of TRPM2 promotes cell survival in multiple malignancies, including neuroblastoma, lung, prostate, stomach, and pancreatic cancers. TRPM2 also mediates different neurological disorders, such as Alzheimer's disease (AD), Parkinson's disease (PD), and epilepsy, a

Verification of the expression of Piezo1 and Piezo2 ion channels in the mouse lens during the embryonic, postnatal and adult growth.

It has been proposed that in the absence of a blood supply and any direct innervation, the lens utilizes a variety of mechanosensitive ion channels to transduce changes to its internal and external environments into the activation of signaling pathways that alter fiber cell structure and hence overall lens function. One such mechanosensitive channel is Piezo1, the activation of which has been shown to phosphorylate Myosin light chain kinase (MLCK) and increase the expression of Transglutaminase 2 in fiber cells. To complement these functional studies, we have conducted a comprehensive mapping …

Leukotriene B4 potentiates acid-sensing ion channel currents in mouse dorsal root ganglion neurons.

Leukotriene B4 (LTB4) is a lipid mediator generated from arachidonic acid. Studies have shown that LTB4 plays a role in the pain process. However, little is known about the underlying molecular and cellular mechanisms. The present data demonstrated a functional link between LTB4 and acid-sensing ion channels (ASICs) in mouse dorsal root ganglion (DRG) neurons. LTB4 potentiated acid-activated ASIC currents in a concentration-dependent and pH-independent manner. The potentiating effect of LTB4 on ASIC currents was mediated by BLT1 receptors but not by BLT2 receptors. The LTB4-induced potentiatio…
