Electrical resistance and ion diffusion through mesothelium.

(1) Since phospholipids (PHL) added on the luminal side of specimens of parietal pericardium of rabbits decrease diffusional permeability (P) to Na+, but not to Cl-, P to Rb+, a cation with hydrated radius similar to that of Cl- was measured. P(Rb+) was 13.1 (+/-1.1, S.E.)x10(-5) cm/sec and it was not decreased by PHL. This suggests that PHL decrease size of intercellular "pores" of mesothelium, and restrict diffusion of solutes with radius>0.2 nm. (2) Electrical resistance (Re) of pericardium specimens was measured without PHL, with PHL, and after mesothelium was scraped away, to obtain Re of

Intestinal ion transport in NKCC1-deficient mice.

The Na(+)-K(+)-2Cl(-) cotransporter (NKCC1) located on the basolateral membrane of intestinal epithelia has been postulated to be the major basolateral Cl(-) entry pathway. With targeted mutagenesis, mice deficient in the NKCC1 protein were generated. The basal short-circuit current did not differ between normal and NKCC1 -/- jejuna. In the -/- jejuna, the forskolin response (22 microA/cm(2); bumetanide insensitive) was significantly attenuated compared with the bumetanide-sensitive response (52 microA/cm(2)) in normal tissue. Ion-replacement studies demonstrated that the forskolin response in

Properties of cytotoxic peptide-formed ion channels.

Cytotoxic peptides are relatively small cationic molecules such as those found 1) in venoms, e.g., melittin in bee, scorpion toxins in scorpion, pilosulin 1 in jumper ant, and lycotoxin I and II in wolf spider; 2) in skin secretions (e.g., magainin I and II from Xenopus laevis, dermaseptin from frog, antimicrobials from carp) and cells of the immune system (e.g., insect, scorpion, and mammalian defensins and cryptdins); 3) as autocytotoxicity peptides, e.g., amylin cytotoxic to pancreatic beta-cells, prion peptide fragment 106-126 [PrP-(106-126)], and amyloid beta-protein (AbetaP) cytotoxic to…

Ion transporter mutations in Gitelman's and Bartter's syndromes.

The application of modern techniques in molecular genetics to classic diseases in clinical nephrology is highlighted by the recent description of the molecular basis of Bartter's and Gitelman's syndromes. A series of detailed studies are described that have resulted in the identification of specific mutations in four different genes, each of which causes hypokalemic alkalosis, salt wasting and hypotension. The importance of these genetic studies in understanding renal physiology and the regulation of blood pressure, and in developing new therapeutic strategies is discussed.

Chromosomal localization of 15 ion channel genes.

Several human Mendelian diseases, including the long-QT syndrome, malignant hyperthermia, and episodic ataxia/myokymia syndrome, have recently been demonstrated to be due to mutations in ion channel genes. Systematic mapping of ion channel genes may therefore reveal candidates for other heritable disorders. In this study, the GenBank and dbEST databases were used to identify members of several ion channel families (voltage-gated calcium and sodium, cardiac chloride, and all classes of potassium channels). Genes and ESTs without prior map localization were identified based on GDB and OWL databa…

Ion channels in human THP-1 monocytes.

The THP-1 human monocytic leukemia cell line is a useful model of macrophage differentiation. Patch clamp methods were used to identify five types of ion channels in undifferentiated THP-1 monocytes. (i) Delayed rectifier K+ current, IDR, was activated by depolarization to potentials positive to -50 mV, inactivated with a time constant of several hundred msec, and recovered from inactivation with a time constant approximately21 sec. IDR was inhibited by 4-aminopyridine (4-AP), tetraethylammonium (TEA+), and potently by charybdotoxin (ChTX). (ii) Ca-activated K+ current (ISK) dominated whole-ce

Erythrocyte ion transport alterations in hypertriglyceridaemic rats.

1. Hypertension appears to be associated with both abnormalities in ion transport and disturbances in lipid metabolism. The aim of the present study was to search for alterations in erythrocyte ion transport in hereditary hypertriglyceridaemic rats. This strain is characterized by a pronounced elevation of blood pressure which correlates with the plasma triacylglycerol level. 2. Erythrocyte Na+ content was elevated in hypertriglyceridaemic rats due to an increased ouabain-resistant Na+ net uptake. This was caused not only by the acceleration of bumetanide-sensitive Na+ inward co-transport, but…

gp120-mediated alterations in astrocyte ion transport.

The pathogenesis of the acquired immunodeficiency syndrome (AIDS) dementia complex (ADC) is unknown. However, recent work indicates that neurons and astrocytes are functionally compromised by exposure to viral components or cellular factors released from HIV-1-infected macrophages/microglia. We show that exposure of primary cultured rat astrocytes to the major HIV envelope glycoprotein gp120 results in alterations of ion and solute transport that may contribute to neuronal cell injury.

Negative ion formation in electrospray mass spectrometry.

Analytical and Chemical Sciences, Research Triangle Institute, Research Triangle Park, North Carolina, USA Negative ion electrospray (ES) operating on a single quadrupole mass spectrometer for the detection of low-molecular-weight molecules is discussed. The ES interface was operated at a positive cylindrical electrode potential to produce negative ions, and the results obtained were compared to the positive ion mode. As in the case of operation in the more common positive mode, negative ions with varying degrees of solvation and structurally relevant fragments can be obtained from a variety o…

[Ion pair-HPLC of a pharmaceutical multicomponent system].

A multicomponent system of non-ionic (methyl- and propylhydroxybenzoate) and ionic (theophylline, phenobarbital, atropine methobromide, demelverine hydrochloride) analytes, which are applied in therapy as bronchospasmolytically effective drug (Aminophyllin compositum tablets), was investigated by means of ion-pair-HPLC-technique on reversed phases. As ion-interaction-reagents (IIR) were tried alkylsulfonates and sodium cyclamate. The influence of some factors on retention was described. The use of "mixed" IIR for improvement of the separation was investigated.

Yeast metallothionein function in metal ion detoxification.

A genetic approach was taken to test the function of yeast metallothionein in metal ion detoxification. A yeast strain was constructed in wh … o mercury, zinc, silver, cobalt, nickel, gold, platinum, lanthanum, uranium, or tin. Two mutant versions of the metallothionein gene were constructed and tested for their ability to confer metal resistance in the cup1 delta background. The first had a deletion of a highly conserved amino acid sequence (Lys-Lys-Ser-Cys-Cys-Ser). The second was a hybrid …

Adrenaline and ion flux in isolated human leucocytes.

Hypokalaemia may be produced in man by intravenous adrenaline infusion, or as a result of pathological disturbances which have led to a high plasma adrenaline concentration. With isolated human leucocytes used as a cellular model, adrenaline at concentrations at and above 9 nmol/l increases the influx of rubidium (a model for potassium flux) into cells, with a simultaneous efflux of sodium. There is no effect on Na+,K+-ATPase activity in lysed leucocytes. Use of the adrenoceptor blockers timolol and atenolol shows that the demonstrated effect of adrenaline on coupled active transport of ions i…

The effect of somatostatin and enkephalin on ion transport in the intestine.

SRIF and enkephalin stimulate net Na and C1 absorption in the rabbit ileum and appear to do so primarily by stimulating the coupled influx of Na and C1 across the brush border membrane. The stimulatory effect of enkephalin on ion transport is blocked by verapamil and Ca-free solution, indirectly indicating that enkephalin may work as a Ca-channel blocker. The resultant lowering in cytosolic calcium level stimulates Na and C1 absorption. SRIF was able to block the effect of cyclic AMP-dependent and cyclic AMP-independent secretogogues in the rat colon without affecting cyclic AMP levels suggest
