Chronic administration of branched-chain amino acids impairs spatial memory and increases brain-derived neurotrophic factor in a rat model.

Maple syrup urine disease (MSUD) is a neurometabolic disorder that leads to the accumulation of branched-chain amino acids (BCAAs) and their α-keto branched-chain by-products. Because the neurotoxic mechanisms of MSUD are poorly understood, this study aimed to evaluate the effects of chronic administration of a BCAA pool (leucine, isoleucine and valine). This study examined the effects of BCAA administration on spatial memory and the levels of brain-derived neurotrophic factor (BNDF). We examined both pro-BDNF and bdnf mRNA expression levels after administration of BCAAs. Furthermore, this stu

The alternative oxidase, a tool for compensating cytochrome c oxidase deficiency in human cells.

In plants as well as in a number of micro-organisms, and in several animal phyla, but not in mammals, the alternative oxidase (AOX) possibly by-passes the cytochrome segment of the respiratory chain. The AOX is located at the inner surface of the inner mitochondrial membrane, being activated by over-reduction of the quinone pool and accumulation of keto-acids such as pyruvate. Since these conditions are frequently encountered in patients with mitochondrial diseases, we hypothesized that the expression of the ascidian Ciona intestinalis AOX might alleviate the consequences of a blockade of the

[Acute and persistent antiproteinuric effect of a low-protein diet in chronic kidney disease].

This study aimed to evaluate the anti-proteinuric effect of a very-low-protein diet supplemented with essential amino acids and keto analogs in patients with moderate to advanced chronic kidney disease and proteinuria already treated with both ACE inhibitors and angiotensin-receptor blockers. The study was a prospective randomized controlled cross-over trial comparing a very-low-protein diet (VLpD) and a low-protein diet (LpD). We enrolled 32 consecutive patients between June 2000 and June 2005. They were randomized to receive a VpLD (group A) or an LpD (group B) for 6 months; thereafter, pati

Purification and characterization of a novel recombinant highly enantioselective short-chain NAD(H)-dependent alcohol dehydrogenase from Thermus thermophilus.

The gene encoding a novel alcohol dehydrogenase (ADH) that belongs to the short-chain dehydrogenase/reductase (SDR) superfamily was identified in the extremely thermophilic, halotolerant gram-negative eubacterium Thermus thermophilus HB27. The T. thermophilus ADH gene (adh(Tt)) was heterologously overexpressed in Escherichia coli, and the protein (ADH(Tt)) was purified to homogeneity and characterized. ADH(Tt) is a tetrameric enzyme consisting of identical 26,961-Da subunits composed of 256 amino acids. The enzyme has remarkable thermophilicity and thermal stability, displaying activity at tem

Expression and Localization of Branched-Chain Ketoacid Dehydrogenase E1 Subunits and LAT1 Transporter in Rat Retinal and Ocular Tissues.

Branched-chain amino acids (BCAAs) are essential for various metabolic and physiological functions. The enzymes responsible for breaking down and transporting them are organized in a tissue-specific manner, playing a crucial role in moving metabolites between cells but often being overlooked. This is particularly true for the rate-limiting enzyme, branched-chain keto-acid dehydrogenase (BCKD). To address this gap, our study investigated the expression and distribution of the BCKDE1 subunits and the BCAA transporter LAT1 in normal rat eye tissues, with a focus on the retina, by applying immunoh

Crystal structures of the mycolic acid methyl transferase 1 (MmaA1) from Mycobacterium tuberculosis in the apo-form and in complex with different cofactors reveal unique features for substrate binding.

Mycolic acid methyl transferase 1 (MmaA1) protein from Mycobacterium tuberculosis plays a crucial role in the biosynthesis of cell wall mycolic acids that aid in survival of the bacteria under adverse conditions. The enzyme converts a cis to a trans olefin and adds a methyl group at the proximal position of both methoxy and keto-mycolic acid chains. Here we report the crystal structures of apo-MmaA1 and complexes with the cofactor S-adenosylmethionine (SAM), the end-product of methylation reactions - S-adenosylhomocysteine (SAH), and the nucleoside analog Sinefungin (SFG) at 1.4-1.9 Å resoluti

Expanding the genetic and phenotypic spectrum of branched-chain amino acid transferase 2 deficiency.

The first step in branched-chain amino acid (BCAA) catabolism is catalyzed by the two BCAA transferase isoenzymes, cytoplasmic branched-chai … biochemical profile with raised plasma BCAAs and, in contrast with MSUD, low-normal branched-chain keto acids (BCKAs) with undetectable l-allo-isoleucine. Interestingly, unlike in MSUD, none of the individuals with BCAT2 deficiency developed acute encephalopathy even with exceptionally high BCAA levels. We observed wide-ranging clinical phenotypes in indivi …

Free radical tissue damages in the anterior segment of the eye in experimental autoimmune uveitis.

PURPOSE: To investigate increased free radical activity and the accumulation and localization of lipid peroxidation in the anterior segment of the eye with uveitis. METHODS: Experimental autoimmune uveitis (EAU) was induced in rats using human S-antigen peptide. Conjugated dienes (CD) and keto-dienes (KD) were then extracted from the cornea, iris-ciliary body and lens of the EAU eyes. The quantity of CD and KD were determined by measuring ultraviolet absorption and estimating by means of a molar extinction coefficient. Frozen sections of EAU eyes were reacted with 3-hydroxy-2-naphthoic acid h

Glucose stimulation of beta-cell DNA replication in the intact rat and in pancreatic islets in suspension culture. Effects of alpha-ketoisocaproic acid, dibutyryl cyclic AMP, and 3-isobutyl-1-methylxanthine in the in vitro system.

DNA replication in pancreatic beta-cells was compared in intact rats maintained hyperglycemic by continuous glucose infusion up to 10 days and in rat islets maintained in suspension culture in RPMI 1640 medium up to 12 days. Replicative activity was evaluated by counting the proportion of labeled beta-cell nuclei after injection or addition of [3H]thymidine. In both experimental systems, DNA replication initially was markedly stimulated by high glucose; then it subsided, even though high levels of glucose were maintained. Culture of pancreatic islets in suspension permitted the study of variou

Pediatrics: office management of diabetes mellitus in children.

For children with diabetes and their families, education should start immediately upon diagnosis of the disease or recovery from the initial episode of ketoacidosis and should be directed specifically toward childhood diabetes. The physician or nurse should be accessible by phone at all times in case of severe hypoglycemia or ketoacidosis. Nonemergency problems are handled during routine office visits, which should be scheduled regularly every three months. Measures important to good control include accurate daily testing of urine, proper choice and careful regulation of insulin, and maintenan

Group B streptococci (GBS) injure lung endothelium in vitro: GBS invasion and GBS-induced eicosanoid production is greater with microvascular than with pulmonary artery cells.

Neonatal group B streptococcal (GBS) sepsis and pneumonia cause lung endothelial cell injury. GBS invasion of the lung endothelium may be a mechanism for injury and the release of vasoactive eicosanoids. Pulmonary artery endothelial cells (PAEC) and lung microvascular endothelial cells (LMvEC) were isolated from neonatal piglets and were characterized as endothelial on the basis of morphology, uptake of acyl low-density lipoprotein, factor VIII staining, and formation of tube-like structures on Matrigel. PAEC and LMvEC monolayers were infected with COH-1 (parent GBS strain), isogenic mutants o

Specific reactivity of recombinant human PDC-E1 alpha in primary biliary cirrhosis.

The mitochondrial autoantigens recognized by autoantibodies in patients with primary biliary cirrhosis have been identified as components of related multi-enzyme complexes, including acyltransferases of the pyruvate dehydrogenase complex (PDC), the branched-chain alpha-keto acid dehydrogenase complex (BCODH), the alpha-ketoglutarate dehydrogenase complex (OGDC), protein X and pyruvate dehydrogenase (PDC) E1 alpha and E1 beta. The major autoantigens, PDC-E2, BCODH-E2 and OGDC-E2, share some sequence homology; the epitopes on these antigens appear to be close to, or identical with, the lipoic ac

Metabolic impact of infant formulas in young infants. An outlook from the urine metabolome.

INTRODUCTION: Although breast milk is the ideal food source for newborns during the first six months of life, a high percentage of children receive infant formulas. There is evidence that specific diet habits may influence individual metabolic profile. Therefore, in newborns, such profile can be influenced by the use of infantile formulas given the composition differences that display compared to human milk. Up to now, there are no reports in the literature that address this issue. OBJECTIVES: this work aims to compare the metabolic profile of full-term newborns that were feed with either bre

Sodium-Glucose Linked Transporter Inhibitors as a Cause of Euglycemic Diabetic Ketoacidosis on a Background of Starvation.

We present a case of a male diabetic patient with one of the most well-known major complications of diabetes mellitus (DM), i.e., diabetic ketoacidosis (DKA). The finding of euglycemic DKA, or DKA with blood glucose levels of less than 200 mg/dL, is a rare occurrence especially in patients with type II DM. He presented with hypotension and increased anion gap metabolic acidosis on a background of keto diet for weight loss and recent use of sodium-glucose linked transporter inhibitors. He was successfully managed with dextrose water, insulin infusion, and potassium replacement. A ketogenic diet

Naja atra SVPLA2 Aggravates Acute Kidney Injury Through Metabolic Reprogramming-Dependent Macrophage Polarization and Defective Efferocytosis.

Snakebite envenoming remains a major global health challenge. Naja atra (N. atra) envenomation induces severe acute kidney injury (AKI), largely driven by snake venom phospholipase A2 (SVPLA2). Increasing evidence suggests that immune dysregulation, in addition to direct cytotoxicity, contributes to delayed renal injury. Here, we investigated whether N. atra SVPLA2 exposure is associated with macrophage immunometabolic remodeling and functional changes relevant to AKI progression. In vivo, AKI was induced in C57BL/6J mice by intraperitoneal administration of N. atra venom, followed by treatmen

Identification of six novel mutations in Iranian patients with maple syrup urine disease and their in silico analysis.

Maple syrup urine disease (MSUD) is a rare inborn error of branched-chain amino acid metabolism. The disease prevalence is higher in populations with elevated rate of consanguineous marriages such as Iran. Different types of disease causing mutations have been previously reported in BCKDHA, BCKDHB, DBT and DLD genes known to be responsible for MSUD phenotype. In this study, two sets of multiplex polymorphic STR (Short Tandem Repeat) markers linked to the above genes were used to aid in homozygosity mapping in order to find probable pathogenic change(s) in the studied families. The families who
