Live-Cell Imaging of Telocyte-like Cells in Primary Liver Cell Cultures.

Telocytes are small mesenchymal cells with unknown hepatic functions. In primary cultures of rat hepatocytes and non-parenchymal liver cells … s beneath cells. Their fast motility apart, hTCLs share structural and behavioral similarities with Kupffer cells/macrophages in NPLC cultures, suggesting a relationship between both cell types that needs to be addressed by further investigation. In EGF/insulin-treated hepatocyte cultures, hTCLs' motility declines, and the cells adopt an extremely small cell bo …

Treatment of multiple sclerosis with peptide-coupled red blood cells induces antigen-specific T regulatory cells.

Inducing antigen-specific immune tolerance is a long-standing aim in organ-specific autoimmune diseases including multiple sclerosis (MS). W … cells. Biodistribution studies in mice and humans demonstrate that pcRBCs are rapidly processed by Kupffer cells in the liver and by marginal zone macrophages in the spleen. In summary, pcRBCs are a promising autologous cell therapy for achieving antigen-specific immune tolerance in MS.

Neutrophil extracellular traps mediate pathophysiology of hepatic cells during liver injury.

Neutrophil extracellular traps (NETs) are web-like, bactericidal structures produced by neutrophils and are composed principally of extracel … onic liver diseases. The liver parenchyma contains hepatocytes, liver sinusoidal endothelial cells, Kupffer cells, and hepatic stellate cells. Each of these cells possesses unique structures and functions, and their interactions with NETs result in pathophysiological changes contributing to liver injury. This review updates the findings related to the modes of …

Lack of effect of calcitonin gene-related peptide and amylin on major markers of glucose metabolism in hepatocytes.

Effects of amylin and calcitonin gene-related peptide on several processes involved in carbohydrate metabolism were investigated in rat hepatocytes, non-parenchymal cells (Kupffer, Ito and endothelial cells) and alveolar macrophages. In hepatocytes, cAMP levels were increased 25-fold by glucagon (10 nM), less than 2-fold by calcitonin gene-related peptide (100 nM) and not at all by amylin (100 nM). In non-parenchymal cells and cultured alveolar macrophages, calcitonin gene-related peptide potently, and amylin weakly, stimulated cAMP levels. In hepatocytes neither amylin nor calcitonin gene-rel

Engineering a biomimetic system for hepatocyte-specific RNAi treatment of non-alcoholic fatty liver disease.

RNA interference (RNAi) presents great potential against intractable liver diseases. However, the establishment of specific, efficient, and safe delivery systems targeting hepatocytes remains a great challenge. Herein, we described a promising hepatocytes-targeting system through integrating triantennary N-acetylgalactosamine (GalNAc)-engineered cell membrane with biodegradable mesoporous silica nanoparticles, which efficiently and safely delivered siRNA to hepatocytes and silenced the target PCSK9 gene expression for the treatment of non-alcoholic fatty liver disease. Having optimized the Gal

Histological studies into rat liver following long-term application of aminophenazone, phenazone, and propyphenazone.

Morphological studies by means of light microscopy are important for toxicological drug testing. More complete information on potential damage can be obtained only by combination of pharmacological, biochemical, hepatofunctional, and morphological tests and studies. No systematic tests had been applied to rats, in the past. Therefore, aminophenazone, phenazone, and propyphenazone were tested for periods up to 16 weeks. Long-term application of the three pyrazolone derivatives resulted in the following dose-dependent and time-dependent alterations: hepatocyte enlargement, fatty degeneration and

Natural medicines for treating liver fibrosis by modulating post-translational modifications.

Hepatic fibrosis is a multifactorial process driven by hepatic stellate cell (HSCs) activation, participation of Kupffer cells and infiltrating immune cells, and profibrotic cytokine signaling (notably TGF-β), culminating in excessive extracellular matrix (ECM) and collagen deposition. Post-translational modifications (PTMs)-covalent changes added after protein synthesis-govern protein stability, localization, interactions, and activity. Common PTMs include phosphorylation, acetylation, ubiquitination, glycosylation, nitration, and methylation; collectively, they modulate fibrogenic pathways a…

Histopathology and immunocytochemical study of type 3 and type 4 complement receptors in the liver and spleen of dogs naturally and experimentally infected with Leishmania (Leishmania) chagasi.

The objective of this study was to compare the histopathological changes and expression of CR3 and CR4 in the liver and spleen of dogs naturally and experimentally infected with L. chagasi. The basic histopathological lesions observed mainly in naturally infected dogs were: epithelioid hepatic granulomas, hyperplasia and hypertrophy of Kupffer cells, Malpigui follicles and mononucleated cells of the red pulp of the spleen. Sections from the liver and spleen by immunocytochemistry technique showed the presence of CD11b, c/CD 18 antigens in the control and infected animals and no qualitative or …

A protein toxin from Pasteurella multocida type D causes acute and chronic hepatic toxicity in rats.

Pasteurella toxin given subcutaneously to rats caused severe liver damage and growth suppression in doses as low as 15.6 ng. Toxin was lethal at and above 31.25 ng. Survival times were dose-dependent, and lesions differed with time of survival after toxin. Rats dead of acute toxicity had focal hepatic necrosis. Liver lesions were associated with diffuse endothelial damage, intravascular trapping of leukocytes, and degeneration of hepatocytes (characterized by glycogen depletion, development of vacuoles, and eosinophilic cytoplasmic inclusions). Endothelial cells, Kupffer cells, and macrophages…

Involvement of natural killer cells in PolyI:C-induced liver injury.

BACKGROUND/AIMS: The roles of T cells, natural killer T cells (NKT) and macrophages in autoimmune hepatitis have been well documented. Howev … ly attenuated PolyI:C-induced liver injury. Neutralization of endogenous Interleukin-12 produced by Kupffer cells abrogated the accumulation of NK cells in the liver and subsequent liver injury. Circadian disruption aggravates non-alcoholic fatty liver disease by activating RIPK1-RIPK3-MLKL axis in mice.

Bidirectional Regulation of Sodium Acetate on Macrophage Activity and Its Role in Lipid Metabolism of Hepatocytes.

Short-chain fatty acids (SCFAs) are important metabolites of the intestinal flora that are closely related to the development of non-alcoholic fatty liver disease (NAFLD). Moreover, studies have shown that macrophages have an important role in the progression of NAFLD and that a dose effect of sodium acetate (NaA) on the regulation of macrophage activity alleviates NAFLD; however, the exact mechanism of action remains unclear. This study aimed to assess the effect and mechanism of NaA on regulating the activity of macrophages. RAW264.7 and Kupffer cells cell lines were treated with LPS and dif…
