Multigenerational endometriosis : consequence of fetal exposure to diethylstilbestrol ?

BACKGROUND: Endometriosis, which affects 10-15 % of women of reproductive age, is an estrogen-driven condition influenced by environmental and genetic factors. Exposition to estrogen-like endocrine-disrupting chemicals (EDCs) has been reported to contribute to the fetal origin of this disease. CASE PRESENTATION: We report here an informative family in which all prenatally DES-exposed daughters and subsequent granddaughters presented endometriosis, whereas the unexposed first daughter and her progeny presented no gynecological disorders. Moreover, the only post-pubertal great-granddaughter, wh

[Factors influencing conception rate after synchronization of ovulation and timed artificial insemination--a review].

This review describes factors that affect conception rate after synchronization of ovulation and timed artificial insemination. Intervals of 7 days between GnRH and PGF2alpha, 48 hours to the second GnRH treatment and a further 16 to 20 hours to the timed insemination have been proven to be most effective. Conception rates (CR) increase as lactation progresses up to 100 days in milk. Primiparous cows have higher CR than older cows. Anovular cows at the start of the synchronization protocols have poor CR. These are highest for cows started in early dioestrus. While poor body condition and some

Lactational high weight loss impairs follicular development by causing mitochondrial dysfunction of ovarian cells in sows and mitigated by butyrate supplement.

INTRODUCTION: In modern sows, lactational high weight loss (HWL), caused by the large litter size and inadequate feed intake, has a negative effect on follicular development after weaning, resulting in poor reproductive performance in the subsequent parity. However, the underlying mechanism remains unclear. OBJECTIVES: This research aimed to explore the mechanism that sows HWL during lactation damages follicular development and attempt to improve the reproductive function by treating with butyrate. METHOD: Four multiparous sister sows were chosen to build a HWL model for lactating sows throu

Dysphoric Milk Ejection Reflex: Report of Two Cases and Postulated Mechanisms and Treatment.

Background: The symptoms of Dysphoric Milk Ejection Reflex (D-MER) occur just before milk release, last no more than a few minutes, and then subside spontaneously, which are characterized by a sudden and transient feeling of dysphoria, depression, sadness, or other negative emotions. These emotions may adversely affect the mother's lactation behavior and mental health, negatively influence the mother-child relationship, and even result in self-harm or suicidal tendencies in lactating women. Case Presentation: We reported two cases of breastfeeding mothers with D-MER who experienced unpleasant

Histone H4 lysine 5 lactylation: A key regulator of immune metabolism in microglia during ischemic stroke.

Histone lactylation is associated with neurological disorders and the state of reactive microglia. However, the impact of elevated lactate levels, generated through glycolysis under hypoxic conditions, on the status and functionality of reactive microglia in the context of ischemic stroke (IS) remains inadequately understood. Immunofluorescence, Western blot and co-immunoprecipitation were performed to identify the histone lactylation modification sites in microglia after IS. CUT&Tag and RNA sequencing data were used to clarify the target genes of H4K5la in microglia after cerebral ischemia. T

Early-Life High-Fat-Diet Exposure Induced Pre-Puberty Obesity-Related MASLD via Autophagy-Mediated Ferroptosis in Male C57BL/6J Mice.

OBJECTIVES: Exposure to high-fat diets in early life plays an important role in metabolic dysfunction-associated steatotic liver disease (MASLD); however, the mechanism remains unclear. In this study, we explore the role of autophagy and ferroptosis in pre-puberty obesity-related MASLD caused by high-fat diets in early life. METHODS: Twenty-four male C57BL/6J mice were fed over a 6-week period, and were divided into three groups: control, lactation HFD, and lactation + post-weaning HFD group. AML12 cells were treated with 0.5 mM free fatty acids (palmitic acid:oleic acid = 1:2) for 24 h to es

Metabolic-Epigenetic-Immune axis in Ischemic Stroke: Lactate-derived Lactylation as a Key Regulator.

Ischemic stroke (IS) induces an energy metabolism crisis, driving ischemic core and penumbra tissues toward anaerobic glycolysis and massive lactate accumulation. Initially identified as a metabolic waste, lactate is now recognized as a crucial signaling molecule. Recent studies have demonstrated that this metabolic byproduct is not merely a toxic waste product but plays a complex spatiotemporal dual role in disease progression: it may exacerbate acidosis and inflammatory damage in the early stages, while serving as an alternative energy substrate or signaling molecule with neuroprotective eff

Lactylation-driven microglial GCH1-BH4 axis suppresses neuroinflammation and underlies the preventive effects of treadmill running against CSDS-induced depression-like behaviors in mice.

Exercise is a novel non-pharmacological intervention for depression, yet the molecular mechanisms linking exercise-induced metabolic signals to epigenetic regulation of neuroinflammation remain largely undefined. We hypothesized that treadmill running ameliorates depressive-like behaviors via suppressing neuroinflammation by lactate-mediated histone lactylation and activation of the GCH1-BH4 axis in the microglia. A chronic social defeat stress (CSDS) mouse model, combined with treadmill running, BH4 treatment, and C646 infusion, along with BV2 microglial cell experiments, were used to assess

Early-Life Antibiotic Cocktail Intervention Alters Cecal Microbiota Composition and Metabolic Profiles in Suckling Rats.

The early-life lactational window is critical for gut microbial colonization, intestinal maturation, and long-term host metabolic and immune programming. Although antibiotic-induced microbiota perturbation models are widely applied to investigate microbiota function, the overall microbiota perturbation efficiency and comprehensive metabolic alterations induced by broad-spectrum antibiotics during the lactational stage remain poorly characterized. In this study, neonatal SD rat pups were continuously administered a broad-spectrum antibiotic cocktail to establish a lactational antibiotic-perturb

Breastfeeding in panhypopituitarism: a case report, review of the literature, and management checklist.

BACKGROUND: Panhypopituitarism is a rare endocrine disorder characterized by deficiency of multiple pituitary hormones. Because prolactin and oxytocin are essential for lactation, breastfeeding is often presumed to be impossible in affected women, and guidance on lactation in this population is scarce. METHODS: We conducted a literature search to identify reports describing breastfeeding outcomes in women with hypopituitarism or panhypopituitarism and examined clinical, obstetric, and treatment-related factors (including prior pituitary surgery and radiation therapy) potentially associated wi

Identification of Hypoxia-Ischemia by Chemometrics Considering Systemic Changes of Physiology.

Perinatal asphyxia represents a major medical disorder and is related to around a fourth of all neonatal deaths worldwide. Specific thresholds for lactate or pH levels define the gold standard for detecting hypoxic-ischemic events as physiological abnormalities. In contrast to current gold standard, we analyze the systemic picture, represented by the whole set of biochemical parameters from blood gas analysis, by multiparametric machine learning algorithms. In a swine model with 22 objects, we investigate the impact of neonatal hypoxic-ischemic encephalopathy on 18 individual physiological par

Mogroside V Improves Follicular Development and Ovulation in Young-Adult PCOS Rats Induced by Letrozole and High-Fat Diet Through Promoting Glycolysis.

Polycystic ovary syndrome (PCOS) is a heterogeneous endocrine disorder characterized by hyperandrogenism, ovulatory dysfunction, and polycystic ovaries. In this study, we induced a young-adult PCOS rat model by oral administration of letrozole combined with a high-fat diet and then treated with mogroside V (MV) to evaluate the protective effects of MV on endocrine and follicle development in young-adult PCOS rats. MV (600 mg/kg/day) administration not only significantly reduced the body weight and ovary weight, but also attenuated the disrupted estrous cycle and decreased the level of testoste

Incidence and risk factors for ketosis in grazing dairy cattle in the Cundi-Boyacencian Andean plateau, Colombia.

Ketosis is a metabolic disorder that has an important impact on health and productivity in dairy cattle during early lactation. In Colombia, the incidence and risk factors for ketosis development have not been studied. The objectives of the present study were to determine the incidence of subclinical (SCK) and clinical ketosis (CK) and to determine the risk factors associated with ketosis within the first 6 weeks of lactation in dairy cattle in Colombia. A prospective cohort study was carried out. A convenience sample of 150 lactating dairy cows from ten commercial dairy farms in the Cundi-Boy

Effects of experimental tooth clenching on pain and intramuscular release of 5-HT and glutamate in patients with myofascial TMD.

OBJECTIVES: It has been suggested that tooth clenching may be associated with local metabolic changes, and is a risk factor for myofascial temporomandibular disorders (M-TMD). This study investigated the effects of experimental tooth clenching on the levels of 5-HT, glutamate, pyruvate, and lactate, as well as on blood flow and pain intensity, in the masseter muscles of M-TMD patients. METHODS: Fifteen patients with M-TMD and 15 pain-free controls participated. Intramuscular microdialysis was performed to collect 5-HT, glutamate, pyruvate, and lactate and to assess blood flow. Two hours after

Measurement of the urinary lactate:creatinine ratio for the early identification of newborn infants at risk for hypoxic-ischemic encephalopathy.

BACKGROUND: Newborn infants with perinatal asphyxia are prone to the development of hypoxic-ischemic encephalopathy. There are no reliable methods for identifying infants at risk for this disorder. METHODS: We measured the ratio of lactate to creatinine in urine by proton nuclear magnetic resonance spectroscopy within 6 hours and again 48 to 72 hours after birth in 58 normal infants and 40 infants with asphyxia. The results were correlated with the subsequent presence or absence of hypoxic-ischemic encephalopathy. RESULTS: Hypoxic-ischemic encephalopathy did not develop in any of the normal

Disrupted glial metabolism and myelin vulnerability in a mouse model of glucose transporter type 1 deficiency syndrome.

Glucose transporter type 1 deficiency syndrome (Glut1DS) is a neurodevelopmental disorder caused by impaired cerebral glucose transport and clinically characterized by absence-type seizures, neurodevelopmental delays, movement disorders, and cognitive deficits. The consequences of chronic cerebral glucose deficiency for glial metabolism and myelin homeostasis remain poorly understood. In this study, we combined metabolic profiling, diffusion MRI, transcriptomic and lipidomic analyses to delineate the metabolic, molecular, and microstructural alterations induced by Glut1 haploinsufficiency in a

Microbial and Metabolic Dysbiosis in Ruminal Acidosis: Mechanisms, Host Responses, and Microbiota-Targeted Mitigation Strategies.

Ruminal acidosis, particularly subacute ruminal acidosis, remains a major metabolic disorder that compromises animal health, production efficiency, and the sustainability of intensive ruminant systems. Although traditionally defined by reduced ruminal pH, it is increasingly recognized as a multidimensional disorder involving microbial ecological destabilization, disrupted metabolic cross-feeding, impaired epithelial barrier function, and dysregulated host inflammatory responses. This review synthesizes current knowledge of the microbial and metabolic mechanisms underlying acute and subacute ru

[Brain damage in sleep-disordered breathing: the role of glia].

Sleep-disordered breathing (SDB) is one of the most prevalent sleep-wake disorders and is associated with brain damage. In this review, we describe the role of astroglia, microglia and oligodendroglia as the main cellular mediators of brain damage in SDB based on the results of experimental studies. Specifically, we describe the role of the molecules that are expressed by glia and mediate oxidative stress (NADPH-oxidase), inflammation (hypoxia-inducible factor-1, inducible nitric oxide synthase, pro- and anti-inflammatory cytokines) and sympathetic hyperactivation (ATP, lactate).
