Detection of circulating Trichinella spiralis larvae by polymerase chain reaction.

Several investigators have successfully applied the polymerase chain reaction to the amplification of DNA from Trichinella spiralis muscle-stage larvae. We show herein that specific DNA can be amplified from T. spiralis migratory larvae in the blood of experimentally infected mice. The polymerase chain reaction detected the presence of migratory larvae in mouse blood from day 5 to day 14 of infection. The technique may be applied to human trichinosis, but its diagnostic value will depend on the severity and stage of the infection.

[Experimental infestation of goats with first instar larvae of Oestrus ovis].

A protocol of experimental infection with OEstrus ovis larvae L1 was applied to kids in order to study their susceptibility to OEstrus ovis. Two-month-old kids, born and raised indoors during winter, were infected with single or repeated administrations of OEstrus ovis L1. Two or three months later, the kids were slaughtered and larvae harvested from their nasal cavities. Following a single administration of 30 L1, 8.3 and 3.3% of the larvae survived after two and three months respectively. After eight repetitive infections in two months with the same number of larvae (30 L1), only 2.2% surviv…

Vaccination of sheep against larvae of the sheep blowfly (Lucilia cuprina).

Four first stage larval antigens from the sheep blowfly were identified using supernatants from cultures of antibody secreting cells. These partially purified larval antigens, when added to Montanide ISA-25 containing recombinant ovine IL-1 beta (rovIL-1 beta) were used to successfully vaccinate sheep against larvae of the sheep blowfly. Significantly less strikes were recorded on vaccinated sheep compared to controls (P < 0.033) with surviving larvae from vaccinated sheep up to 85% smaller than larvae from control sheep. RovIL-1 beta was found to be an important component of the vaccine. Vacc…

Genes expressed in Brugia malayi infective third stage larvae.

We have used a tag sequencing approach to survey genes expressed in the third stage infective larvae of the human filarial nematode parasite Brugia malayi. RNA was isolated from late vector-stage L3 larvae after days 9 or 10 of infection in mosquitos, and converted to cDNA by reverse transcriptase. Double-stranded cDNA was produced by either conventional methods (non-SL cDNA library) or by PCR using the nematode spliced leader (SLI) and oligo(dT) primers (SL cDNA library). Two clone libraries (one from SL and one from non-SL cDNAs) were constructed in lambda ZapII. A set of these full-length c…

The significance of head movements in distance discrimination in praying mantis larvae.

1. When larvae of the praying mantis Polyspilota sp. and Tenodera sinensis want to leave an exposed position and can choose to move between stationary objects at different distances, they usually choose the nearest. Their ability to select the nearest object is greatest when the background has horizontal stripes and is least when it has vertical stripes. Object preference is based on a successive distance comparison, which may involve content-related memory processes. 2. Mantid larvae can determine the absolute distance to a stationary object. Vertical contrasting borders play an important rol…

Inhibition of mouse parenchymal cells proliferation by mosquito larvae extract.

As it is known, some cell populations of mosquito Aedes aegypti larvae do not proliferate. This fact suggests the existence of some mitosis inhibiting factor. We have assayed the effect of mosquito larvae crude extract on 85 C3HS young male mice (25 +/- 1 days old) and the effect of the dialyzed fraction of the same extract on 47 partially hepatectomized C3HS adult male mice. The injection was performed at 16:00 hour (before the raising of the DNAS curve) and the controls were made at 08:00/16, 12:00/20 and 16:00/24 (Time of Day/Time Post Injection). Colchicine arrested metaphases of nephrocyt…

On the control of ecdysone biosynthesis by the central nervous system of blowfly larvae.

Ecdysone was found to be the major secreted steroid of ring glands dissected from blowfly larvae and incubated in vitro. Other secretory products such as 3-dehydroecdysone and 20-deoxy-makisterone A could not be detected when the glands were labelled with tritiated cholesterol. Ecdysone synthesis and secretion were found to be tightly coupled. The highest rate of secretion was observed a few hours before pupariation. In vitro, the rate of ecdysone secretion by ring glands was affected significantly by coincubation with the central nervous system (CNS). Modulating effects from the CNS to the gl…

Efficacy of doramectin against warble fly larvae (Hypoderma bovis).

The efficacy of doramectin in the treatment of cattle harbouring naturally acquired infestations of first, second and third instar larvae of Hypoderma bovis was determined in two studies carried out in the Burgundy region of France. In the first study, cattle on six farms with a history of H. bovis infestations were treated during October 1989 with either doramectin at a dose of 200 micrograms kg-1 liveweight (186 animals) or with an equivalent volume of saline (157 animals). During the following March and April, all animals were examined for the presence of warbles. In the second study, cattl…

Distinction of human hookworm larvae based on lectin-binding characteristics.

A comparative study of lectin binding to ensheathed (EnL3) and exsheathed (ExL3) L3 larvae of Necator americanus, Ancylostoma duodenale and Ancylostoma ceylanicum revealed a number of differences between these hookworm species. These differences could provide a novel approach to distinguish infective L3 larvae in field conditions. For example, binding of Ulex europaeus agglutinin (UEA) and Ricinus communis agglutinin (RCA120) to N. americanus EnL3 distinguished them from those of A. duodenale and A. ceylanicum. Furthermore, UEA and RCA120 negative EnL3 could be separated into the two Ancylosto…

Inoculation of ferrets with ten third-stage larvae of Dracunculus insignis.

Infection with Dracunculus insignis was established in 7 of 10 ferrets experimentally inoculated intraperitoneally with 10 third-stage larvae (L3's). Worm recovery and infection rate were comparable to animals inoculated with 50 L3's. This study demonstrates that once infective larvae traverse the gut and pass into the peritoneal cavity, very few larvae are required to establish infection.

Identification of the surface polypeptide antigens of the infective larvae of Onchocerca volvulus.

The surface polypeptide antigens of third-stage infective larvae (L3) and adult males of Onchocerca volvulus have been compared after iodogen-catalysed radioiodination, separation by sodium dodecyl sulphate-polyacrylamide gel electrophoresis in beta-mercaptoethanol followed by autoradiography. L3 surfaces contained polypeptides with apparent molecular weights of 14, 18.5, 26, 34, 51, 68 and 130 kDa, whereas the adults contained 14, 19, 26, 34, 37.5 and 68 kDa components. By immunoprecipitation with patient's sera, only the 14 and 18.5 kDa components were shown to be antigenic on the L3, the ot…

The effect of Strongylus vulgaris larvae on equine intestinal myoelectrical activity.

The myoelectrical activity of the ileum, caecum and large colon was monitored from Ag-AgCl bipolar recording electrodes in four conscious 'parasite-naive' weanling foals. All foals were inoculated with 1000 infective 3rd-stage Strongylus vulgaris larvae and alterations to the myoelectrical activity observed. The frequencies of caecal and colonic spike bursts increased significantly in all post infection periods coinciding with assumed larval penetration into the intestinal mucosa and migration through the vasculature. Peaks in caecal and colonic activity occurred at Days 1 to 5 post infection.…

Toxocara canis: proteolytic enzymes secreted by the infective larvae in vitro.

Second-stage larvae of the dog nematode Toxocara canis are infective to man and cause the syndromes of visceral larva migrans and ocular toxocariasis. Larvae cultured in vitro secrete proteases which degrade components of a model of extracellular matrix and basement membranes. These enzymes have been characterized using a variety of techniques. Multiple enzyme activities were demonstrated by substrate gel electrophoresis, associated with proteins of molecular weights of 120 and 32 kDa. The enzyme activity was inhibited both in substrate gels and in a radiogelatin microplate assay by phenylmeth…

Moulting and exsheathment of the infective larvae of Onchocerca lienalis (Filarioidea) in vitro.

Fourth-stage larvae (L4) of the cattle filarial parasite Onchocerca lienalis were produced from third stage forms (L3) and maintained in vitro using a variety of culture media and feeder cell layers. Up to 50% of the L3 larvae moulted to the fourth stage in the presence of two jird (Meriones unguiculatus) cell lines, a monkey kidney cell line and also with Vero cells. Moulting took place 2-5 days after of the initiation of the cultures: These L4 parasites could be maintained in a healthy condition for up to 88 days, although no significant increases in size of these larvae were observed during…
