Rats given linseed oil in microemulsion forms enriches the brain synaptic membrane with docosahexaenoic acid and enhances the neurotransmitter levels in the brain.

Long chain n-3 fatty acids such as docosahexaenoic acid (DHA) are essential for the normal functioning of the brain. The vegetarian sections of the population get only alpha-linolenic acid (ALA) through their diet as a source of n-3 fatty acids. Hence, in this group of the population, the ALAs need to be converted to DHA through the action of the desaturase and the elongase enzymes. However, the conversion of the ALA to the DHA is very minimal (<2%) in mammals. Our recent studies have shown that the conversion of the ALA to the DHA can be enhanced significantly when given in the microemulsion

Association of erythrocyte membrane fatty acids with changes in glycemia and risk of type 2 diabetes.

BACKGROUND: The significance of erythrocyte membrane fatty acids (EMFAs) and their ratios to predict hyperglycemia and incident type 2 diabetes is unclear. OBJECTIVE: We investigated EMFAs as predictors of the worsening of hyperglycemia and incident type 2 diabetes in a 5-y follow-up of a population-based study. DESIGN: We measured EMFAs in 1346 Finnish men aged 45-73 y at baseline [mean ± SD age: 55 ± 6 y; body mass index (in kg/m(2)): 26.5 ± 3.5]. Our prospective follow-up study included only men who were nondiabetic at baseline and who had data available at the 5-y follow-up visit (n = 73

Docosahexaenoic acid (DHA) and the developing central nervous system (CNS) - Implications for dietary recommendations.

The accretion of docosahexaenoic acid (DHA) in membranes of the central nervous system is required for the optimum development of retina and brain functions. DHA status is determined by the dietary intake of n-3 polyunsaturated fatty acids (PUFA), both the metabolic precursor α-linolenic acid (α-LNA) and DHA. Clinical studies have shown that feeding term or premature infants with formula low in total n-3 PUFA may alter the maturation of visual acuity. Moreover, feeding infants over the first 6 mon of life with formula containing adequate α-LNA, but no DHA, did not sustain the same cerebral acc

ALA/LA ameliorates glucose toxicity on HK-2 cells by attenuating oxidative stress and apoptosis through the ROS/p38/TGF-β1 pathway.

BACKGROUND: Growing evidence indicates that oxidative stress (OS) plays a pivotal role in Diabetic nephropathy (DN). In a previous study we demonstrated that ALA/LA protected HK-2 cells against high glucose-induced cytotoxicity. So we aimed to establish the glucose injury model of HK-2 cells and investigate the beneficial effects of ALA/LA on high glucose-induced excessive production of TGF-β1 and the possible mechanisms mediating the effects. METHODS: The expression of OS markers in high glucose-induced HK-2 cells treated with ALA/LA., including the antioxidant enzymes and reactive oxygen sp

Properties of cholesterol-reduced butter made with beta-cyclodextrin and added evening primrose oil and phytosterols.

The present study was carried out to examine changes in the chemical and sensory properties of butter in which the cholesterol was reduced and to which evening primrose oil (EPO) and phytosterols were added. Crosslinked beta-cyclodextrin (beta-CD) made from adipic acid was used, and approximately 90% of the cholesterol was removed. The color measurement values "L" and "a" were significantly different between the control (butter with no beta-CD treatment and no added EPO and phytosterols) and treatment A (butter treated with 10% crosslinked beta-CD); however, the color values for "L" and "a" we

Differential effects of n-3 polyunsaturated fatty acids on metabolic control and vascular reactivity in the type 2 diabetic ob/ob mouse.

Diets rich in monounsaturated fatty acids (MUFA) are recommended for individuals with type 2 diabetes mellitus (T2DM). The American Heart Association recommends increasing intakes of n-3 polyunsaturated fatty acids (PUFA) to reduce the risk of vascular disease in high-risk individuals; however, the long-term effects of these bioactive fatty acids on glucose metabolism in insulin resistance are controversial. The present studies were conducted to evaluate the effects of diets rich in both MUFA and alpha linolenic acid (C18:3n-3, ALA), eicosapentaenoic acid (C20:5n-3, EPA), or docosahexaenoic ac

Lipidome Characterization Reveals Alterations of Fatty Acid Metabolism in Helicobacter pylori Infection.

BACKGROUND: Helicobacter pylori (H. pylori) is a pathobiont that infects around two-thirds of the global population and has demonstrated a rise in antibiotic resistance, warranting a search for alternative treatments. As fatty acid biosynthesis is central to membrane structure and function, and H. pylori is correlated with the erosion of the mucosal barrier, lipidome analysis can elucidate the role of fatty acid metabolism in H. pylori infection and yield potential targets for intervention. MATERIALS AND METHODS: Fecal samples from 68 H. pylori patients and 35 healthy control subjects were an

Differences in honeybee queen pheromones revealed by LC-MS/MS: Reassessing the honest signal hypothesis.

The honest signal hypothesis posits that social insect queens emit pheromonal signals that convey information about fecundity, which workers use to make decisions around investing in direct or indirect fitness. We used liquid chromatography-tandem mass spectrometry to measure honeybee (Apis mellifera) queen retinue pheromone (QRP) in relation to age, laying status, and acceptance using a protocol that enables concurrent metabolomic and lipidomic analyses. Older queens produced higher levels of the QRP components 9-R-hydroxydec-2(E)-enoic acid (9(R)-HDA), linolenic acid (LEA), and 4-hydroxy-3-m

Cord blood eicosanoid signatures and newborn gestational age.

Beyond prostaglandins, the function of eicosanoids and other oxylipins in pregnancy and labor is poorly understood. In contrast to earlier work focusing on preterm infants, we investigated how oxylipin levels in newborns (measured in cord blood) vary during the last weeks of pregnancy in 190 mother-newborns (≥37 weeks of gestation) of the ENVIRONAGE birth cohort, Belgium. We found increased levels of PGE2 (p=0.003), PGF2α (p=0.042), 8,9-DHET (p=0.037), 11-HETE (p=0.034), and 15-HETrE (p=0.008) associated with full term pregnancy compared to early term labor. Furthermore, late vs early term was

Metabolomic analysis of follicular fluid from women with Hashimoto thyroiditis.

Hashimoto thyroiditis is an autoimmune disease characterized by hypothyroidism and a high level of anti-thyroid autoantibodies. It has shown to negatively impact female fertility; however, the mechanisms are unclear. Ovarian follicular fluid appears to be the key to understanding how Hashimoto thyroiditis affecst fertility. Thus, we aimed to evaluated the metabolic profile of follicular fluid and antithyroid autoantibody levels in the context of Hashimoto thyroiditis. We collected follicular fluid from 61 patients, namely 38 women with thyroid autoantibody positivity and 23 women as negative c

Ethanol dependent interaction between prostaglandins and lipoxygenase products in human peripheral lymphocytes.

The effect of lipoxygenase products, 12-L-HETE and 15-L-HPETE, on cyclic AMP levels in human peripheral lymphocytes was examined in the absence and in the presence of a prostaglandin of the E-type (0.6-3.0 microM) or isoprenaline (33 microM). The studies were performed either in the absence or in the presence of 6 per cent ethanol. For comparison the effect of arachidonic acid and linolenic acid were studied. In the absence of ethanol 12-L-HETE and 15-L-HPETE had no significant effect on cyclic AMP accumulation. However, in the presence of ethanol 12-L-HETE (above 1 microM) inhibited prostagla

Mechanistic insights into inulin-fatty acid conjugates as β-carotene carriers for enhancing stability.

BACKGROUND: To mitigate the low stability of β-carotene (βC), this study developed amphiphilic inulin (INU)-fatty acid (FA) conjugates by hydrophobically modifying INU with stearic acid, oleic acid (OA), linoleic acid, and linolenic acid, which were subsequently employed for βC encapsulation. RESULTS: Fourier transform infrared spectroscopy and 1H-nuclear magnetic resonance verified successful synthesis, with critical micelle concentrations ranging from 0.014 to 0.035 mg mL-1. Among all the βC-INU-FA micelles, βC-INU-OA showed the strongest stability and homogeneity, with its encapsulation ef

Polystyrene Microplastics and Bisphenol A Exposure Worsen Intestinal Injury in Diabetic Mice by Disrupting Gut Microbiota and Metabolites.

Environmental pollutants can induce multiorgan damage, with the digestive tract particularly susceptible. Diabetic enteropathy is a significant complication of type 2 diabetes mellitus (T2D). However, the relationship between environmental pollutant exposure and T2D-associated intestinal injury has not been previously explored. In this study, T2D mice were subjected to polystyrene microplastics (PS-MPs, 100 μg/day, 3 weeks) and bisphenol A (BPA, 100 μg/kg/day, 2 weeks). Metabolomics and 16S rRNA sequencing were used to detect changes in colonic metabolites and gut microbial composition. Caco-2

Screening of metabolites in the treatment of liver cancer xenografts HepG2/ADR by psoralen-loaded lipid nanoparticles.

OBJECTIVE: Our study aimed to find potential biomarkers for drug resistance in liver cancer cells using metabolomics and further to evaluate the potential of psoralen-loaded polymer lipid nanoparticles (PSO-PLNs) to reverse the resistance of cells to doxorubicin. METHODS: We used LC-MS-based non-targeted metabolomics, also known as global metabolite profiling, to screen in serum and urine of mice engrafted with a liver cancer cell line sensitive (HepG2/S) or resistant to doxorubicin (HepG2/ADR) for differentially regulated metabolites. We subsequently quantified the abundance of these metabol

Increasing omega-3 desaturase expression in tomato results in altered aroma profile and enhanced resistance to cold stress.

One of the drawbacks in improving the aroma properties of tomato (Solanum lycopersicum) fruit is the complexity of this organoleptic trait, with a great variety of volatiles contributing to determine specific quality features. It is well established that the oxylipins hexanal and (Z)-hex-3-enal, synthesized through the lipoxygenase pathway, are among the most important aroma compounds and impart in a correct proportion some of the unique fresh notes in tomato. Here, we confirm that all enzymes responsible for the synthesis of these C6 compounds are present and active in tomato fruit. Moreover,

UHPLC-MS/MS-based analysis of lipidomic profiling reveals age-group-associated and species-specific lipid characteristics in Anabarilius grahami.

Anabarilius grahami is a vulnerable endemic cyprinid fish in Yunnan Province, yet its muscle-lipid metabolic features remain poorly understood. In this study, UHPLC-MS/MS-based lipidomics was applied to compare the lipidomes of Anabarilius grahami at three age groups with those of Carassius auratus from a single age group. A total of 1703 lipid metabolites were detected, with glycerophospholipids accounting for the largest proportion (59.84%). Seven differentially abundant lipids (DALs) were identified as potential biomarkers for distinguishing C. auratus and A. grahami. In addition, three DAL
