Adductomics of Newborn Dried Blood Spots Detects Constituents of Maternal Smoking During Pregnancy and Associated Oxidative Stress Exposure.

Maternal Smoking During Pregnancy (MSDP) remains a major source of fetal toxicant exposure. We applied adductomics to profile reactive adducts at the human serum albumin cysteine-34 (HSA-Cys34) locus, which integrates longer-term exposures. HSA-Cys34 adducts formed by acrylonitrile and ethylene oxide, two tobacco-related toxicants previously linked to smoking in adults, were quantified and compared with cotinine and MSDP status. Their relationships with other reactive adducts were also examined. Neonatal dried blood spots (DBS) from 110 children were analyzed. Cotinine and 55 Cys34 adducts wer

Maternal risk factors associated with complex gastroschisis: Cannabis exposure and recurrent urinary tract infections may be modifiable targets.

AIM OF THE STUDY: Maternal risk factors associated with gastroschisis occurrence have been identified, but predisposing factors to the complex variant are unknown. This study aimed to identify factors associated with complex gastroschisis by comparing the maternal risk factor profiles of simple and complex gastroschisis pregnancies. METHODS: All cases of gastroschisis managed in one tertiary care hospital between 1991 and 2022 were identified from a retrospective clinical database. Cases were classified as 'complex' if severe matting, necrosis, atresia or perforation were present; 'simple' ga

Prenatal serotonin reuptake inhibitor exposure and maternal depression symptoms are associated with altered fetal brain and placental development.

Maternal mental health is associated with fetal neurodevelopment. Identifying effective treatments for maternal psychiatric conditions is a public health priority. SRIs (SSRIs and SNRIs) are commonly prescribed for prenatal mental health conditions; however, their impact on fetal brain development remains understudied. In this observational cohort study, we compared fetal brain and placental structures between SRI-exposed and unexposed pregnancies divided by categories of maternal depressive symptom severity from the Edinburgh Postnatal Depression Scale (EPDS). Pregnant women treated with SRIs

Maternal flutamide exposure induces spermatogenic dysfunction in male offspring: The mechanistic role of RARα-Mediated NLRP3 inflammasome activation.

Maternal exposure to endocrine-disrupting compounds (EDCs) is linked to male reproductive decline, yet the mechanisms underlying developmental programming of long-term reproductive toxicity remain elusive. This study investigated how prenatal exposure to the anti-androgenic fungicide flutamide impairs spermatogenesis in offspring. Pregnant Sprague-Dawley rats were treated with flutamide (25 mg/kg/d) during late gestation (GD12-GD20). Testicular morphology and spermatogenic markers (Stra8, PCNA) were assessed in male offspring at PND30 and PND60. RNA-sequencing (RNA-seq) identified differential…

Prenatal neonicotinoid insecticide exposure, maternal steroid hormone levels, and precocious puberty in girls: A nested case-control study.

Neonicotinoid insecticides (NNIs) are globally prevalent environmental contaminants with suspected endocrine-disrupting properties, yet their effects on maternal steroid hormone homeostasis and the subsequent risks of precocious puberty (PP) in offspring remain poorly understood. In this prospective nested case-control study embedded in a birth cohort in Wuhan, China (2012-2014; follow-up to 2023), we included 202 PP cases (115 incomplete [ICPP], 87 central [CPP]) and 606 matched controls. Third-trimester maternal urine samples were analyzed for 19 NNI biomarkers (8 detected in ≥74.1% of the s

Maternal Low-Dose Lipopolysaccharide Exposure and House Dust Mite Challenge Alter Serum IgG N-Glycosylation in Offspring Mice.

Background: The "hygiene hypothesis" and "farm effect" suggest that early-life microbial exposure protects against allergic diseases. Maternal low-dose lipopolysaccharide (LPS) exposure during pregnancy attenuates allergic airway inflammation in offspring, yet the underlying mechanisms remain incompletely understood. Immunoglobulin G (IgG) N-glycosylation critically regulates antibody effector functions and maternal-fetal immune transfer, but its role in transgenerational allergy protection is unknown. Objective: To investigate whether maternal low-dose LPS exposure modulates serum IgG N-glyca

Effects of subchronic late-gestational nicotine exposure on postpartum maternal behavior in Wistar rats: An exploratory pilot study.

OBJECTIVE: This exploratory pilot study evaluated whether subchronic late-gestational oral nicotine exposure alters postpartum maternal behavior in Wistar rats. METHODS: Twelve pregnant Wistar rats were obtained in three successive intake batches and randomly assigned to experimental or control groups. Control animals received a 2% saccharin solution; experimental animals received nicotine (nominal 200 μg/mL) in 2% saccharin solution ad libitum from gestational day 7 through parturition. The initial concentration of 200 μg/mL produced two maternal deaths and one embryonic reabsorption in the

The tissue partition, maternal-fetal transfer and physiologically based pharmacokinetic modelling for silver nanoparticles exposure in pregnant rats.

To date, only a few studies have investigated the biodistribution and kinetics of silver nanoparticles (AgNPs) throughout pregnancy. In this study, pregnant rats were repeatedly exposed to 20 nm and 30 nm polyvinylpyrrolidone-coated AgNPs via intravenous (IV) or oral gavage for 20 days. Results indicated that AgNPs primarily accumulated in the spleen, kidneys, and liver, with silver concentrations ranging from 0.58 to 75.07 μg/g dry weight (dw) across all treated groups. IV exposure resulted in 1.43- to 80.87-fold greater tissue accumulation compared to oral exposure. The liver and kidneys acc

Maternal exposure to tris (2-butoxyethyl) phosphate induces F0 female reproductive toxicity and offspring developmental toxicity in zebrafish.

The toxicity of tris (2-butoxyethyl) phosphate (TBOEP) has been extensively investigated because of its prevalence in the environment. Nevertheless, the risk factors associated with maternal transmission are poorly understood. In this study, sexually mature female zebrafish were treated with TBOEP (0, 20, 100, and 500 μg/L) for 30 days and were mated with unexposed males. Reproduction impairment in exposed adults (F0), including retarded gonado-somatic index (GSI), delayed oocyte maturation, inhibition of reproductive behavior, and subfertility was found. The observed impacts on the F0 generat

Maternal Phthalate Exposure Alters Prostate Proteome in Rat Offspring: Linking Omics Insights to Prostate Cancer Risk in Humans.

BACKGROUND: Phthalates are compounds used as plasticizers to increase the flexibility of plastics and are considered endocrine disruptors. Some studies suggest that the origin of prostate cancer (PCa) may be associated with disturbances during embryo-fetal development. Previous data showed that perinatal exposure to the same phthalate mixture (PM) used here increased the incidence of adenocarcinomas in the prostates of aged rats. Building on our earlier work, this study identifies proteins altered in the prostate proteome by exposure to a PM during gestation and lactation in rats, focusing on

How nature nurtures: prenatal exposure to green space buffers the effects of maternal stress on neonatal BDNF methylation.

Pregnancy constitutes a critical window of vulnerability during which maternal and environmental exposures may shape fetal development through epigenetic mechanisms. While prenatal maternal anxiety and exposure to green spaces have been independently associated with child neurodevelopment, their potential interactive effects on neonatal epigenetic profiles remain largely unexplored. This study examined the independent and interactive effects of maternal trait anxiety and residential green space exposure during pregnancy on neonatal DNA methylation (DNAm) of the brain-derived neurotrophic facto

Mediating role of the maternal placental growth factor levels on association between ozone exposure and adverse pregnancy outcomes.

BACKGROUND: Maternal placental growth factor (PlGF) is closely associated with adverse pregnancy outcomes (APOs), and ozone (O3) exposure is considered a risk factor for APOs. However, whether PlGF mediates the relationship between O3 and APOs remains unclear. This study investigated the mediating role of PlGF in the association between O3 exposure and APOs. METHODS: This study conducted a retrospective cohort analysis of 11,677 pregnant women in Tianjin, China (2017-2023). The association between O3 exposure and APOs was analyzed using Cox proportional hazards models, while multiple linear r

Identification of micro- and nanoplastic particles in postnatal sprague-dawley rat offspring after maternal inhalation exposure throughout gestation.

Micro-nanoplastic particulates (MNPs) have been identified in both indoor and outdoor environments. From these real-world exposures, MNPs have been identified in human fluids and organ tissues, including the placenta and breastmilk. Laboratory studies have identified MNPs are capable of crossing the placental barrier and depositing in fetal tissues; however, it remained unclear if MNPs persist in offspring tissues after birth. Six pregnant Sprague-Dawley rats were divided equally into two groups: control and exposed to polyamide-12 (PA-12) MNP aerosols (11.46 ± 3.78 mg/m3) over an average of 4

Short-Half-Life Chemicals: Maternal Exposure and Offspring Health Consequences-The Case of Synthetic Phenols, Parabens, and Phthalates.

Phenols, parabens, and phthalates (PPPs) are suspected or known endocrine disruptors. They are used in consumer products that pregnant women and their progeny are exposed to daily through the placenta, which could affect offspring health. This review aims to compile data from cohort studies and in vitro and in vivo models to provide a summary regarding placental transfer, fetoplacental development, and the predisposition to adult diseases resulting from maternal exposure to PPPs during the gestational period. In humans, using the concentration of pollutants in maternal urine, and taking the of…

Prenatal exposure to hexafluoropropylene oxide trimer acid (HFPO-TA) disrupts the maternal gut microbiome and fecal metabolome homeostasis.

Initially considered a "safe" substitute for perfluorooctanoic acid (PFOA), hexafluoropropylene oxide trimer acid (HFPO-TA) has been extensively used in the production of fluoropolymers for several years, leading to its environmental ubiquity and subsequent discovery of its significant bio-accumulative properties and toxicological effects. However, the specific impact of HFPO-TA on females, particularly those who are pregnant, remains unclear. In the present study, pregnant mice were exposed to 0.63 mg/kg/day HFPO-TA from gestational day (GD) 2 to GD 18. We then determined the potential effect

How Maternal Trauma Exposure Contributed to Children's Depressive Symptoms following the Wenchuan Earthquake: A Multiple Mediation Model Study.

Although well-established literature has indicated the burden of mental health among victims after the Wenchuan earthquake, no research has focused on the mental health of mothers and their children who experienced the earthquake and were pregnant during or shortly after it. This study investigates the relationship between maternal trauma exposure (TE) and children's depressive symptoms after the Wenchuan earthquake and explores the risk and protective factors underlying this relationship. A sample of 547 mother-child dyads, in which the mother experienced the Wenchuan earthquake, was used to

Maternal urine phthalate metabolite exposure and miscarriage risk: a nested case-control study of the Zunyi Birth Cohort.

Phthalates (PAEs) are widespread persistent organic pollutants and endocrine disruptors. However, the associations between PAE exposure and the risk of miscarriage in humans are unclear, and an insufficient number of studies have evaluated the possible threshold or dose-dependent effects of first trimester PAE exposure on miscarriage risk. Our research measured the levels of mono-methyl phthalate (MMP), mono-ethyl phthalate, mono-isobutyl phthalate, MiBP mono-butyl phthalate (MBP), mono-octyl phthalate, mono-benzyl phthalate, mono(2-ethylhexyl) phthalate, mono(2-ethyl-5-oxohexyl) phthalate, an
