Neonatal hypoxia-ischemia alters the events governing the hippocampal critical period of postnatal synaptic plasticity leading to deficits in working memory in mice.

Postnatal critical periods of synaptic plasticity (CPsp) are characterized by profound neural network refinement, which is shaped by synaptic activity and sculpted by maturation of the GABAergic network. Even after therapeutic hypothermia (TH), neonatal hypoxia-ischemia (HI) impairs two triggers for the initiation of the CPsp in the hippocampus: i) PSA-NCAM developmental decline and ii) parvalbumin (PV) + interneuron (IN) maturation. Thus, we investigated whether neonatal HI despite TH disturbs other events governing the onset, consolidation and closure of the postnatal CPsp in the hippocampus

Insulin signaling represents a gating mechanism between different memory phases in Drosophila larvae.

The ability to learn new skills and to store them as memory entities is one of the most impressive features of higher evolved organisms. However, not all memories are created equal; some are short-lived forms, and some are longer lasting. Formation of the latter is energetically costly and by the reason of restricted availability of food or fluctuations in energy expanses, efficient metabolic homeostasis modulating different needs like survival, growth, reproduction, or investment in longer lasting memories is crucial. Whilst equipped with cellular and molecular pre-requisites for formation of

Semantic distances of WAIS Similarities word pairs in non-demented adults: An item-level index of semantic memory granularity.

Innovative approaches to test scoring can help neuropsychologists detect subtle semantic memory alterations. We focussed on the Wechsler Adult Intelligence Scale Similarities (SIM) test and calculated an item-level score based on the 'Leacock & Chodorow '(LCH) semantic distance expressed by each SIM item. We hypothesised that LCH would predict 1) performance on standard semantic memory tests; 2) an Alzheimer aetiology; 3) transentorhinal grey-matter integrity. Six hundred sixty-nine non-demented participants completed a neuropsychological battery inclusive of SIM and consolidated tests of sema

Sodium butyrate exerts a neuroprotective effect in rats with acute carbon monoxide poisoning by activating autophagy through the mTOR signaling pathway.

Acute carbon monoxide (CO) poisoning is a prevalent type of poisoning that causes significant harm globally. Delayed encephalopathy after acute carbon monoxide poisoning (DEACMP) is a severe complication that occurs after acute CO poisoning; however, the exact underlying pathological cause of DEACMP remains unclear. Accumulating evidence indicates that abnormal inflammation and immune-mediated brain damage, cellular apoptosis and autophagy, and direct neuronal toxicity are involved in the development of delayed neurologic sequelae. Sodium butyrate, a histone deacetylase inhibitor, has gained i

Glucocorticoids and cortical decoding in the phobic brain.

Glucocorticoids reduce phobic fear in anxiety disorders and enhance psychotherapy, possibly by reducing the retrieval of fear memories and enhancing the consolidation of new corrective memories. Glucocorticoid signaling in the basolateral amygdala can influence connected fear and memory-related cortical regions, but this is not fully understood. Previous studies investigated specific pathways moderated by glucocorticoids, for example, visual-temporal pathways; however, these analyses were limited to a-priori selected regions. Here, we performed whole-brain pattern analysis to localize phobic s

Repeated stimulation of dopamine D1-like receptor and hyperactivation of mTOR signaling lead to generalized seizures, altered dentate gyrus plasticity, and memory deficits.

The acute activation of the dopamine D1-like receptors (D1R) is involved in a plethora of functions ranging from increased locomotor activity to the facilitation of consolidation, storage, and retrieval of memories. Although much less characterized, epileptiform activities, usually triggered by disruption of the glutamate and GABA balance, have also been reported to involve the dopaminergic transmission. Using a combination of biochemical, immunohistochemical, electrophysiological, and behavioral approaches we have investigated the consequences of repeated stimulation of D1R using the selectiv

The influence of anesthesia and surgery on fear extinction.

Accumulating evidence has demonstrated significant clinical post-traumatic stress disorder (PTSD) symptoms after anesthesia or surgery. Fear extinction dysfunction is a notable feature of PTSD. Although anesthetics and surgery profoundly affect memory processes, their designated effects on fear extinction have not been dissertated. Previous studies have suggested that innate immune system activation disrupts fear extinction, and surgery has been shown to increase the inflammatory response. Thus, in the current study, we examined the effects of propofol, sevoflurane, dexmedetomidine and surgery

Sound asleep, yet still listening: auditory processing during human sleep.

Sleepers can wake to their name, but ignore louder irrelevant sounds, a paradox that suggests more than simple sensory gating. Growing evidence reveals flexible auditory coupling during human sleep, ranging from complete disconnection to high-level cognitive processing and even to physical responses. Here, we synthesize the multiple cognitive levels at which auditory stimuli are processed (or not) during human sleep and outline factors modulating this processing. Moments of sensory isolation may promote restoration and memory consolidation, while moments of environmental coupling enable threat…

Local Protein Synthesis at Synapses: A Driver for Synapse Diversification.

Local protein synthesis within neuronal processes seems to be crucial for the rapid and dynamic remodeling of the proteome at synaptic compartments. Indeed, this capability enables neurons to swiftly adapt their synaptic functions in response to activity. In this review, we first explore the diverse mechanisms that allow the targeted transport of mRNAs into both dendrites and axons. Then, we report evidence that local mRNAs are actively recruited for protein synthesis during plasticity. Finally, we highlight how this molecular complexity contributes to the establishment and stabilization of me

Role of cerebellum in sleep-dependent memory processes.

The activities and role of the cerebellum in sleep have, until recently, been largely ignored by both the sleep and cerebellum fields. Human sleep studies often neglect the cerebellum because it is at a position in the skull that is inaccessible to EEG electrodes. Animal neurophysiology sleep studies have focussed mainly on the neocortex, thalamus and the hippocampus. However, recent neurophysiological studies have shown that not only does the cerebellum participate in the sleep cycle, but it may also be implicated in off-line memory consolidation. Here we review the literature on cerebellar a…

Rhinal cortex, but not medial thalamic, lesions cause retrograde amnesia for objects in rats.

Male Long-Evans rats were trained on five separate object discrimination problems at different times prior to surgery. Following surgery, retrograde amnesia was assessed by measuring retention of the preoperatively learned discrimination problems in lesioned rats and controls. Rats with rhinal cortex lesions displayed temporally graded retrograde amnesia; retention of object discriminations acquired in the recent past (i.e. 2 or 9 days prior to surgery) was significantly impaired, whereas retention of object discriminations acquired more remotely (i.e. 16, 37, or 58 days prior to surgery) was

A new case of retrograde amnesia with abnormal forgetting rate.

The classic amnesic syndrome is characterised by a severe anterograde amnesia and a less important retrograde amnesia with sparing of the semantic component. We report the case of a patient who showed a global amnesic syndrome following a mild head injury. Initially, amnesia was both anterograde and retrograde, and also involved semantic knowledge. Two years later the picture had changed remarkably. The retrograde deficit for autobiographical events was still total, while semantic memory had recovered to a large extent. Learning had also greatly improved, but only if assessed after a short del

Focal retrograde amnesia and the attribution of causality: An exceptionally critical view.

A detailed critique of the literature on focal retrograde amnesia is provided. Some of the cases commonly cited in this literature had, in fact, severely impaired anterograde memory, most often involving visuospatial material. Other cases showed poor anterograde memory in more moderate or subtle form, begging the question of whether "like" had really been compared with "like" across the retrograde and anterograde domains: there may be alternative explanations for the observed patterns of performance. One suggestion is that these patients suffer an impairment of long-term consolidation, an attr

Single-cell analysis of bronchoalveolar cells in inflammatory and fibrotic post-COVID lung disease.

BACKGROUND: Persistent radiological lung abnormalities are evident in many survivors of acute coronavirus disease 2019 (COVID-19). Consolidation and ground glass opacities are interpreted to indicate subacute inflammation whereas reticulation is thought to reflect fibrosis. We sought to identify differences at molecular and cellular level, in the local immunopathology of post-COVID inflammation and fibrosis. METHODS: We compared single-cell transcriptomic profiles and T cell receptor (TCR) repertoires of bronchoalveolar cells obtained from convalescent individuals with each radiological patte

Reduced plasma BDNF concentration and cognitive decline in veterans with PTSD.

Post-traumatic stress disorder (PTSD) is a trauma and stress related disorder frequently associated with cognitive decline. War veterans with PTSD have a higher risk of developing dementia than healthy subjects. Brain derived neurotrophic factor (BDNF) is an important protein that modulates plasticity, memory consolidation and cognitive processes. Lower circulating BDNF levels were related to memory impairment and cognitive deterioration. The aim of this study was to evaluate cognitive deterioration and plasma BDNF concentration in 120 veterans with combat related PTSD, 120 healthy controls, 4…

Expression of renewal is dependent on the extinction-test interval rather than the acquisition-extinction interval.

A recent finding suggested that when extinction occurs shortly after acquisition, renewal of an extinguished fear response (fear-potentiated startle) to a light conditioned stimulus (CS) is diminished (Myers, Ressler, & Davis, 2006). The present study attempted to extend this finding using a white-noise CS and freezing as the behavioral measure of fear. In Experiments 1A and 1B, we observed renewal whether extinction occurred 10 min or 24 hr after acquisition. In contrast, renewal was not observed if test occurred 10 min after extinction (Experiment 2). Experiment 3 demonstrated that expressio
