Tetrahedral-Framework Nucleic Acids Carry Small Interfering RNA to Downregulate Toll-Like Receptor 2 Gene Expression for the Treatment of Sepsis.

Sepsis is caused by the invasion of pathogenic microorganisms, which can lead to excessive expression of toll-like receptors (TLRs) in cells and uncontrollable amplification of the inflammatory response. TLR2, as an essential part of the TLR family, has a significant feature in the identification of innate immune responses. Therefore, blocking the expression and activation of TLR2 can inhibit the synthesis and release of inflammatory factors and avoid the occurrence of excessive inflammatory reactions. Small interfering RNA (siRNA) can selectively target the silencing or downregulation of path

Diversity of the 47-kD HtrA nucleic acid and translated amino acid sequences from 17 recent human isolates of Orientia.

Orientia tsutsugamushi, the etiologic agent of potentially fatal scrub typhus, is characterized by a high antigenic diversity, which complicates the development of a broadly protective vaccine. Efficacy studies in murine and nonhuman primate models demonstrated the DNA vaccine candidate pKarp47, based upon the O. tsutsugamushi Karp 47-kD HtrA protein gene, to be a successful immunoprophylactic against scrub typhus. To characterize 47-kD HtrA protein diversity among human isolates of Orientia, we sequenced the full open reading frame (ORF) of the 47-kD HtrA gene and analyzed the translated amin

Cytology smears from endobronchial ultrasound-guided transbronchial needle aspiration as a source of nucleic acids with adequate yield for next-generation sequencing.

BACKGROUND: Formalin-fixed paraffin-embedded (FFPE) specimens obtained via endobronchial ultrasound-guided transbronchial needle aspiration (EBUS-TBNA) are commonly used for next-generation sequencing (NGS), which has become integral to guiding treatment strategies in advanced non-small cell lung cancer (NSCLC). However, the DNA and RNA extracted from these samples often exhibit suboptimal quality, leading to test failure. Although cytology smears are routinely prepared during the processing of EBUS-TBNA specimens, their utility for DNA and RNA extraction remains insufficiently explored. There

Tetrahedral framework nucleic acids as a promising vehicle for small molecule drugs of traditional Chinese medicine: application to disease prevention and treatment.

The modernization of Traditional Chinese Medicine (TCM) faces considerable challenges, primarily due to the poor water solubility, low chemical stability, and limited oral bioavailability of its active components. These limitations hinder the translation of Natural Compounds from Chinese Herbal Medicine (NCCHMs) from basic research to clinical applications. Recently, the advent of DNA nanotechnology has provided innovative solutions to address these challenges. Among these, tetrahedral framework nucleic acids (tFNAs), a novel class of programmable nanomaterials, show significant promise as ide…

Transmission electron microscopy reveals an optimal HIV-1 nucleocapsid aggregation with single-stranded nucleic acids and the mature HIV-1 nucleocapsid protein.

HIV-1 nucleocapsid protein (NCp7) condenses the viral RNA within the mature capsid. In a capsid-free system, NCp7 promotes an efficient mechanism of aggregation with both RNA and DNA. Here, we show an analysis of these macromolecular complexes by dark-field imaging using transmission electron microscopy. Thousands of mature NCp7 proteins co-aggregate with hundreds of single-stranded circular DNA molecules (ssDNA) within minutes, as observed with poly(rA). These co-aggregates are highly stable but dynamic structures, as they dissociate under harsh conditions, and after addition of potent ssDNA

Comparison of synovial tissue and synovial fluid as the source of nucleic acids for detection of Chlamydia trachomatis by polymerase chain reaction.

OBJECTIVE: Difficulties in detecting Chlamydia trachomatis in human joints by polymerase chain reaction (PCR) may be related to whether syno … y PCR gives positive results more often when synovial tissue rather than SF is the source of target nucleic acids. Although synovial tissue is the source of choice for the most reliable determination of chlamydia in the joint, both synovial tissue and SF should be assayed if possible.

A Novel Voltammetric Point-of-Care Device for Rapid and Accurate Nucleic Acid and Viral Pathogen Detection.

With zoonotic outbreaks on the rise, rapid and accurate infectious disease diagnostics are critical for both human and animal health. Traditional lateral flow assays lack sensitivity and specificity, while isothermal amplification methods like LAMP, though rapid, can be hindered by optical detection issues. We present an electrochemical nucleic acid amplification test (NAAT) that combines isothermal amplification with real-time voltammetric detection. This novel method relies on monitoring of amplification-associated proton release through redox probing of a pH-sensing compound. The method sho

Rapid photo-crosslinking in living cells reveals protein-nucleic acid dynamics on a timescale of minutes.

The activation of chemical reactions in living cells using ultraviolet (UV) light enables the interrogation of biomolecules in their native environment with photoreactive probes or crosslinking reagents. Although numerous photo-crosslinking approaches have been successfully employed, they often suffer from common limitations, including low reaction yields, the need for long exposure times, and irradiation-induced cellular damage from heat, desiccation, or side reactions. We recently showed that 365 nm light-emitting diodes enable rapid, biorthogonal protein-DNA crosslinking in living cells, in

Stimulus-Responsive Polymeric Carriers for Gene Delivery: Balancing Endosomal Escape with Nucleic Acid Release.

BACKGROUND: Polymeric carriers for nucleic acid delivery must overcome two sequential intracellular barriers: escape from the endosome and release of the cargo into the cytosol. These steps are typically treated as independent design problems and optimized separately. OBJECTIVE: This review argues that escape and release are coupled through the same electrostatic and pH-dependent interactions, creating a fundamental tradeoff . The cationic charge that enables a polymer to destabilize endosomal membranes also tightens its grip on the nucleic acid payload, so conditions favoring escape simultan

Universal contamination-free nucleic acid biosensing enabled by guide-independent Argonaute amplicon elimination.

Amplicon contamination remains a major challenge in nucleic acid biosensing, frequently compromising diagnostic accuracy in both clinical testing and field-deployable sensing systems. Although strategies such as uracil-DNA glycosylase (UDG) treatment and CRISPR-based approaches have been developed to mitigate this problem, their broader implementation is often limited by incomplete decontamination, increased assay cost, and the complexity associated with exogenous guide design. Here we introduce CREDIT (chopping-based Argonaute-mediated testing), a molecular diagnostic biosensor platform that
