Investigation of genetic and phenotypic heterogeneity in 37 Turkish patients with Kabuki and Kabuki-like phenotype.

Kabuki syndrome (KS) is a rare disorder characterized by distinct face, persistent fingertip pads, and intellectual disability (ID) caused by mutation in KMT2D (56%-76%) or KDM6A (5%-8%). Thirty-seven children aged 1-16 years who followed for median of 6.8 years were included in this study, which aimed to investigate the genetic and clinical characteristics of KS patients. KMT2D and KDM6A were evaluated by sequencing and multiplex-ligation-dependent probe amplification in 32 patients. Twenty-one pathogenic variants in KMT2D, of which 17 were truncated and nine were novel, one frame-shift novel

Annular epidermolytic ichthyosis with palmoplantar keratosis: a unique phenotype associated with interfamilial phenotypic heterogeneity.

BACKGROUND: Annular epidermolytic ichthyosis (AEI) is a rare autosomal dominant ichthyosis that was recently described in 10 separate families in the English literature. There are no reports on the phenotypic heterogeneity of AEI. OBJECTIVES: We investigated, for the first time, a large Chinese AEI pedigree exhibiting interfamilial phenotypic heterogeneity. MATERIALS AND METHODS: We collected clinical data and DNA from the members of the family, and skin lesions were obtained from two patients with different phenotypes. Skin imaging examinations were performed. Whole-exome sequencing (WES) a…

A comprehensive and comparative phenotypic analysis of the collaborative founder strains identifies new and known phenotypes.

The collaborative cross (CC) is a large panel of mouse-inbred lines derived from eight founder strains (NOD/ShiLtJ, NZO/HILtJ, A/J, C57BL/6J … cantly different between strains. This study highlights the value of the founder and CC strains for phenotype-genotype associations of many genetic traits that are highly relevant to human diseases. All data described here are publicly available from the mouse phenome database for analyses and downloads.

Phenotypic study in 40 patients with dysferlin gene mutations: high frequency of atypical phenotypes.

OBJECTIVE: To describe the phenotypic spectrum of dysferlin (DYSF) gene mutations (which cause dysferlinopathies, autosomal recessive muscular dystrophies) in patients with a dysferlin protein deficiency. DESIGN: Clinical, biological, and pathological data from 40 patients were reviewed. The diagnosis of dysferlinopathy was based on the absence or strong reduction of dysferlin in muscle, and confirmed by mutational screening of the DYSF gene. SETTING: Two French neuromuscular diseases centers (in Paris and Marseilles). RESULTS: Two main dysferlinopathy phenotypes are well recognized: Miyosh…

Dysregulated NK cell phenotype in exacerbation-prone severe asthma.

BACKGROUND: Natural killer (NK) cell dysfunction and defective inflammation resolution pathways are associated with chronic airway inflammation in asthma. NK cells are targets for regulation by essential polyunsaturated fatty acid-derived specialized pro-resolving mediators (SPMs), including resolvin D2 (RvD2), to stimulate inflammation resolution. METHODS: Multiparameter flow cytometry was utilized to examine expression patterns of select activating and inhibitory receptors, including the RvD2 receptor DRV2, on circulating NK cells in SARP3 severe and non-severe asthma subjects and healthy c

Targeting Inflammation in Osteoarthritis Pain: A Phenotype-Driven Approach.

BACKGROUND: Osteoarthritis (OA) is a chronic joint disease marked by the progressive loss of articular cartilage and associated inflammatory processes, which can lead to significant pain and disability. However, the heterogeneous nature of OA has limited the effectiveness of many anti-inflammatory interventions, highlighting the need for phenotype-driven therapeutic strategies. METHODS: This narrative review was conducted by searching PubMed, Web of Science, and Google Scholar from inception to June 2025. Predefined keywords included "osteoarthritis", "inflammation", "pain", "synovitis", "eff…

Cracking the Code: Genotype-Phenotype Correlation Models in Sarcoglycanopathies.

OBJECTIVE: Sarcoglycanopathies are among the most severe limb-girdle muscular dystrophies (LGMD), though milder presentations have been described. These diseases are primarily caused by missense variants, but the limited predictability of their effect on protein maturation, complex formation, and transport has hindered reliable genotype-phenotype correlations. This study aimed to establish accurate genotype-phenotype correlations for LGMDR3, LGMDR4, and LGMDR5. METHODS: We analyzed the largest sarcoglycanopathy cohort to date (n = 541). Clinical data, including age at symptom onset and loss o…

Development of a Digital Phenotype for Immune-Related Colitis.

PURPOSE: Immune-related colitis (IR-colitis) is a significant side effect of immune checkpoint inhibitor (ICI) therapy for an expanding group of cancers. Timely identification is required to optimize outcomes, but existing approaches to retrospective case detection are resource intensive. We sought to develop an electronic medical record (EMR)-based digital phenotype to identify patients with IR-colitis. METHODS: Adult patients commenced on ICIs from August 1, 2020, to June 30, 2023, at the Peter MacCallum Cancer Centre were studied. ICIs were prescribed as monotherapy or in combination with …

Phenotype-Guided Renal Denervation: the Key to Therapeutic Response.

Renal denervation (RDN) has re-entered the therapeutic landscape of hypertension following contemporary sham-controlled trials demonstrating reproducible reductions in blood pressure. However, variability in treatment response remains a defining feature, raising important questions regarding its clinical applicability. Emerging evidence indicates that this heterogeneity is primarily attributable to differences in patient phenotype rather than limitations of device technology. This narrative review synthesizes evidence from randomized trials, meta-analyses, and key observational studies to exam…

Psoriatic arthritis-evolution of our understanding of the phenotype.

The psoriatic arthritis phenotype has evolved over the past several decades. The original description of 5 clinical patterns has been expanded into 6 domains including peripheral arthritis (which includes 3 of the patterns described by Moll and Wright namely distal, oligoarticular and polyarticular), axial disease, dactylitis, enthesitis, skin and nails. In this article we review the evolution of the PsA phenotype, from the Moll and Wright subtypes described in 1973 and how they might have changed, evolution of our understanding of axial PsA, consider race and geographic differences in disease…

Periodontitis promotes gingival accumulation of cells with MDSC phenotype.

BackgroundPeriodontitis is one of the most common inflammatory diseases in humans, mostly caused by bacterial infection and with diverse populations of immune cells involved. Myeloid-derived suppressor cells (MDSCs), a heterogeneous group of immature myeloid cells derived from hematopoietic precursor cells, have exhibited immunomodulatory functions by production of different molecules such as inducible NO synthase (iNOS) and it is thought to be involved in periodontitis. However, reports of characterization of cells with MDSC phenotypes in gingival tissues are very scarce. This study aimed to …

Epithelial-Predominant Synovial Sarcoma With a Deceptive Neuroendocrine Phenotype.

Synovial sarcoma (SS) is a tumor of uncertain lineage with a characteristic mixture of spindled and epithelial components that is subtyped as biphasic when both spindle and epithelial components are present and as monophasic when composed solely of spindle cells. Epithelial-predominant SS is extremely rare, and neuroendocrine differentiation within this component would be exceptional. Prominent ossification is another uncommon feature in SS, thought to portend a better prognosis. The hallmark genomic alteration in SS is t(X;18;p11;q11), resulting in the formation of an oncogenic fusion gene, u
