[Effect of deep and shallow electroacupuncture stimulation at "Huantiao"(GB30) on expression of phosphorylated-p38 and phosphorylated-p53 proteins and apoptosis in dorsal root ganglia in sciatic nerve injury rats].

OBJECTIVE: To observe the effect of deep electroacupuncture (EA) stimulation at "Huantiao"(GB30) on hindlimb motor function and expression of p38 mitogen-activated protein kinase (p38 MAPK ) and p53 proteins in dorsal root ganglia (DRG) in rats with chronic constrictive injury (CCI) of sciatic nerve. METHODS: Forty-eight SD rats (half male and half female) were randomly divided into control, model, shallow EA (SEA) stimulation and deep EA (DEA) stimulation groups (n=12 in each group). The CCI model was constructed by implanting a silicone tube close to the sciatic nerve of the left hind limb.

Impaired ionizing radiation-induced activation of a nuclear signal essential for phosphorylation of c-Jun by dually phosphorylated c-Jun amino-terminal kinases in ataxia telangiectasia fibroblasts.

The c-Jun amino-terminal kinases (JNKs) participate in intracellular signaling in response to cytokines and cellular stresses. JNKs are activated by phosphorylation on two critical residues, the threonine 183 and tyrosine 185, within the TPY motif. The activated JNKs, in turn, phosphorylate the nuclear protein c-Jun, a major component of the transcription factor AP1. In vitro studies have revealed a defect in ionizing radiation-induced activation of the JNK signaling pathway in lymphoblastoid cells from individuals with ataxia telangiectasia (AT). However, the biochemical basis for this signal…

AKT1 phosphorylates PRMT7 to promote GLUD1 methylation and gastric cancer progression.

Glutamine metabolism has emerged as an essential metabolic driver of tumor progression. Glutamate dehydrogenase 1 (GLUD1), a key enzyme in glutaminolysis, is frequently overexpressed in malignancies. Post-translational modifications (PTMs) are crucial for regulating protein function and tumor progression. However, the PTMs of GLUD1, particularly arginine methylation, remain unexplored. Here we report that protein arginine methyltransferase 7 (PRMT7) mediates monomethylation of GLUD1 at arginine 76 (R76), enhancing its protein stability by antagonizing ubiquitin-dependent degradation. Moreover,

Phosphorylation landscape of dengue virus proteins and their implications in protein-protein interactions.

Phosphorylation is one of the most ubiquitous, reversible post-translational modifications through which cells communicate external signals from the membrane to the nucleus. However, viruses replicate in the host cells by hijacking the phosphorylation signaling to evade immune responses, as shown previously for Ebola and HIV. Here, we characterized the potential phosphorylation sites, the kinases involved, and their location in the structure of the Dengue virus proteins. We also show that the phosphosites in the four Dengue serotypes are evolutionarily conserved across other flaviviruses. Furt…

Modulation of Lung Adenocarcinoma by Phosphorylated FOXN3-Mediated Transcriptional Inactivation of p53.

As a pivotal tumor suppressor, p53 plays a critical role in the progression of lung adenocarcinoma (LUAD). However, the mechanisms through w … n, and tumorigenesis. Importantly, the regulation of p53 transcriptional activity by FOXN3 requires phosphorylation at the S83 and S85 sites. This phosphorylation induces the dissociation of FOXN3 from chromatin, thereby inhibiting p53 transcriptional recruitment and activation. Ablation of FOXN3 S83 and S85 phosphorylation impedes the progression of LUAD. Furthe …

[Advances in measurements and standardization of plasma phosphorylated tau217 as a biomarker for Alzheimer's disease].

With the approval of targeted drugs for modification therapy of Alzheimer's disease, the role of blood markers in the diagnosis and treatment of Alzheimer's disease is becoming more and more important. The economical and easily accessible plasma phosphorylated tau217 (p-tau217) is one of the most promising biomarkers for clinical application. Since it was first found in 2020, it has attracted wide attention in just a few years, rapidly becoming a research hotspot in this field, and is adopted as one of the core diagnostic biomarkers. This article reviewed the clinical measurements of plasma p-

A Novel Phosphorylated Tau Conformer Implicated in the Tauopathy Pathogenesis of Human Neurons.

Alzheimer's disease (AD) is a neurodegenerative disorder with no effective treatments. Hyperphosphorylation of tau protein contributes to neurodegeneration in AD. Previous studies have identified pT231-tau in the cis conformation as an early driver of neurodegeneration in tauopathy models. Here, we identify a novel neurotoxic pT231-tau conformer in human AD neurons, distinct from both cis and trans conformations, which we propose as the gauche pT231-tau conformer. Notably, levels of this conformer were elevated in neurons subjected to aging-associated stress. In order to confirm the stress, we…

Plasma Phosphorylated Tau 217 as a Discriminative Biomarker for Cerebral Amyloid Angiopathy.

BACKGROUND: Blood-based biomarkers may offer a non-invasive approach to diagnose cerebral amyloid angiopathy (CAA), especially in early-stage. We evaluated the ability of plasma phosphorylated tau-217 (p-tau 217) to differentiate CAA from Alzheimer's disease (AD) and deep perforator arteriopathy (DPA). METHODS: Patients with AD (age 73.7 ± 8.1 years), probable CAA (74.8 ± 6.9 years), or DPA (66.1 ± 10.4 years) were enrolled from memory and stroke clinics at a medical center in Taiwan. All participants received amyloid and tau PET scans. Plasma biomarkers were measured via a SIMOA immunoassay

EPHA5 regulates antifungal innate immunity by phosphorylating EPHB2 and Dectin-1.

Invasive fungal infections (IFIs) have emerged as a significant health threat and cause approximately 3.75 million deaths per year globally. Understanding the detailed mechanism of the immune response to eliminate invasive fungal pathogens may help to provide new insights for the development of antifungal methods and drugs. Previously, we reported that the tyrosine kinase receptor EPH receptor B2 (EPHB2) is a coreceptor of β-glucan and phosphorylates Syk to activate the antifungal downstream signaling pathway. However, how EPHB2 is activated after fungal infection is still unknown. In this stu

Cutaneous phosphorylated-synuclein: an early diagnostic biomarker for pure autonomic failure.

BACKGROUND: Pure autonomic failure (PAF) presents with progressive autonomic failure without other neurological features. Atypical presentations may lead to diagnostic uncertainty. We studied whether cutaneous phosphorylated-alpha-synuclein (p-syn) could distinguish between PAF, multiple system atrophy (MSA) and non-synucleinopathy-related autonomic failure, and examined its relationship with quantitative markers of cardiovascular autonomic failure. METHODS: All individuals underwent Composite Autonomic Symptom Score-31 autonomic questionnaires, cardiovascular autonomic testing and bilateral

Identifying an Abnormal Phosphorylated Adaptor by Viral Kinase Using Mass Spectrometry.

Mass spectrometers are widely used to identify protein phosphorylation sites. The process usually involves selective isolation of phosphoproteins and subsequent fragmentation to identify both the peptide sequence and phosphorylation site. Immunoprecipitation could capture and purify the protein of interest, greatly reducing sample complexity before submitting it for mass spectrometry analysis. This chapter describes a method to identify an abnormal phosphorylated site of the adaptor protein by a viral kinase through immunoprecipitation followed by LC-MS/MS.

E3 ligase RAD18 targets phosphorylated IRF3 to terminate IFNB1 transcription.

The transcription factor interferon regulatory factor 3 (IRF3) initiates type I interferon transcription, which is required for host defense. Here, we identify RAD18 as a central E3 ubiquitin ligase that selectively targets phosphorylated IRF3 (p-IRF3) for autophagic degradation. RAD18 specifically promotes the dissociation of p-IRF3 from the IFNB promoter and in turn terminates its transcriptional activity. Mechanistically, RAD18 binds the p-IRF3 dimer located on the IFNB promoter and triggers K63 polyubiquitylation of p-IRF3 at Lys 193. The ubiquitylated p-IRF3 dimer consequently dissociates

Phosphorylation status and prognostic impacts of RSK2, PDPK1, and AKT in malignant lymphoma.

Despite the oncogenic roles of the serine/threonine kinase PDPK1 and its key effectors, RSK2 and AKT, their activation status and prognostic significance remain unexamined in B cell lymphomas (BCLs). This study evaluated the phosphorylation states of PDPK1, the RSK2-N-terminal kinase domain (NTKD), and AKT through immunohistochemical analyses of 468 biopsied samples from patients with nine subtypes of MLs, including diffuse large B cell lymphoma (DLBCL) (n = 277) and follicular lymphoma (FL) (n = 121). PDPK1 was frequently phosphorylated in most subtypes, showing 98% in DLBCL and 76% in FL. RS…

PIN1 promotes the metastasis of cholangiocarcinoma cells by RACK1-mediated phosphorylation of ANXA2.

BACKGROUND: Cholangiocarcinoma (CCA), a primary hepatobiliary malignancy, is characterized by a poor prognosis and a lack of effective treat … ll proliferation and migration. Mechanistically, PIN1 functioned as an oncogene by regulating ANXA2 phosphorylation, thereby promoting CCA adhesion. Notably, the interaction between PIN1 and ANXA2 was facilitated by RACK1. Importantly, pharmacological inhibition of PIN1 using the FDA-approved drug all-trans retinoic acid (ATRA) effectively suppressed the metastat …
