Electroretinography biomarkers indicate disrupted visual processing in Fragile X syndrome.

BACKGROUND: Objective physiological biomarkers that index underlying neural circuit dysfunction, such as electroretinography (ERG), are needed in Fragile X syndrome (FXS) research. Fragile X syndrome (FXS) is a neurodevelopmental disorder caused by silencing of the FMR1 gene and loss of fragile X messenger ribonucleoprotein (FMRP), leading to synaptic dysfunction and prominent sensory processing abnormalities. This study evaluated whether ERG waveform differences are detectable in FXS using a handheld RETeval® protocol while accounting for key technical and physiological determinants of signal

Evaluating Human Rod Photoreceptor Function Using Pixelwise Intensity-Based Optoretinography.

PURPOSE: More than 50 inherited retinal diseases are known, with rod photoreceptor functional loss serving as an early indicator in nearly half of them. The "optoretinogram" is a relatively new assay that detects optical changes in cells in response to stimuli. This tool has excellent potential for providing insights into the earliest functional changes of rod photoreceptors, with the potential to assist in the early detection, monitoring, and treatment of retinal diseases. METHODS: In this work, we obtained intensity-based optoretinograms (iORGs) from rod photoreceptors using an adaptive opt

An image-computable spatio-chromatic receptive field model of the midget retinal ganglion cell mosaic across the retina.

Image-computable models of primate retinal ganglion cell (RGC) mosaics that are synthesized and constrained jointly by optical, anatomical and physiological properties, and which operate on images defined by their spatial-spectral radiance, do not currently exist. Here, we deploy a novel computational framework which synthesizes mosaics of linear spatio-chromatic receptive fields (RFs) of ON midget RGCs (mRGCs) by integrating published anatomical, physiological, and optical quality measurements, all varying with eccentricity. We use the synthesized mRGC mosaics to simulate both in vivo and in

Subspace reverse-correlation estimation of receptive fields during free viewing.

Accurately mapping receptive fields under naturalistic viewing requires correcting for distortions introduced by eye movements. Current approaches often rely on parameterized "shifter networks" optimized with Poisson generalized linear models, but these methods are computationally intensive and limited to small regions of interest. We present a new framework that exploits the translation properties of Hartley basis functions to model eye position effects directly as phase shifts, eliminating the need for explicit retinal image reconstruction. This formulation admits closed-form gradients, enab

Lexical brain responses in 10-year-old children are impaired in dyslexia: An FPVS-EEG study.

The developmental origin of the left occipitotemporal cortex specialization for automatic lexical access from vision remains unclear. Here we investigated cortical specialization for print processing in children with or without dyslexia, focusing on two distinct tuning levels: coarse-grained tuning for letter/symbol discrimination, and fine-grained tuning for word/pseudoword discrimination. 10-year-old typical readers (n = 24) and children with dyslexia (n = 14) were tested with electroencephalography (EEG) and fast periodic visual stimulation (FPVS), viewing streams of stimuli at a relatively

Conspecific proximity alters crabs' attention to novel visual stimuli.

The ability to prioritize relevant stimuli over distracting ones is essential for adaptive behavior, particularly in species with limited sensory systems. In the mudflat crab Neohelice granulata, a moving object on the ground (dummy) can elicit either avoidance or pursuit, indicating that the same stimulus may be interpreted as predator or prey. Previous studies have shown that the probability of these opposite responses depends on both stimulus features (e.g. size, speed) and intrinsic animal attributes (e.g. sex, hunger level). Here, we investigated how social context - a factor independent

Rhythmic stimulation elicits multiple, concurrent neural responses.

Rhythmic light stimulation offers solutions to innumerable cognitive and neurological disorders. However, like any neuromodulatory technique, responses to rhythmic light stimulation are highly variable, producing challenges in replicating lab-based studies and translating findings to the clinic. Across 3 magnetoencephalography (MEG) and electroencephalography (EEG) experiments, we show that this variability can, in part, be attributed to rhythmic light stimulation eliciting multiple, coexisting neural responses which have separable influences on behavior. Specifically, we find that rhythmic li

Location-Specific and Location-Invariant Neural Representations of Visual Summary Statistics.

Human vision efficiently navigates information-dense environments by extracting summary statistics-the average properties of item groups-to circumvent capacity limits. However, the neural transition from location-specific sensory registration to location-invariant abstract representation remains poorly understood. We recorded high-density EEG while participants of either sex performed an ensemble size discrimination task, using time-resolved multivariate pattern analysis and cross-visual-field generalization to dissociate these two levels of representation. Our results reveal a clear temporal

Compact Monocular Video-Ophthalmoscope to Measure Retinal Reflectance Changes to Flicker Light Stimuli.

This paper describes a compact video-ophthalmoscope (VO) designed for capturing retinal video sequences of the optic nerve head (ONH) under flicker light stimulation. The device uses an OLED display and a fiber optic-coupled LED light source, enabling high-frame-rate video at low illumination intensity (12 μW/cm2). Retinal responses were recorded in 10 healthy subjects during flicker light exposure with a pupil irradiance of 2 μW/cm2. Following 20 s of stimulation, all subjects displayed changes in retinal reflectance and pulsation attenuation, linked to blood flow and volume variations. These

Developmental differences in perceiving arousal and valence from dynamically unfolding emotional expressions.

The development of emotion perception has predominantly been studied using static, unimodal stimuli featuring the faces of young adults. Most findings indicate a processing advantage for positive emotions in children (positivity bias) and a negativity bias in adults, although these results are usually task-dependent. We created a new stimulus database comprising digital avatars from four age groups, dynamically expressing happiness, happy-surprise, anger, and sadness in visual (face only) and visual-verbal (face and voice) conditions. To determine whether previously found biases would re-emerg

Effects of Partial Occlusion on Response Dynamics and Interregional Processing within Primate Superior Temporal Sulcus.

Recognizing partially occluded objects is a critical visual function that primates perform with ease, yet the underlying neural mechanisms remain incompletely understood. Previous studies in the macaque inferotemporal cortex have reported mixed results on whether occlusion delays and reduces responses to partially occluded objects. To address this, we recorded single-unit activity from body-responsive regions of the middle and anterior superior temporal sulcus (STS) in male macaques while presenting body stimuli with varying levels of occlusion using a dot pattern. Occlusion reduced response s

Facial distortions as a critical test for models of the organization of visual function.

Ritchie et al. propose a novel framework for understanding the organization of visual function in the occipitotemporal cortex (OTC). We argue that neuropsychological evidence from individuals with prosopometamorphopsia (PMO), a condition in which faces appear distorted, offers a unique opportunity to test models of OTC. Drawing on findings from a large-scale experiment that used naturalistic stimuli with five individuals with PMO, we show that distortions in two cases appear to be face-specific. These results highlight the value of integrating neuropsychological, computational, and behavioral

Infant's motion adaptation with animal motion.

We investigated motion adaptation with animal motion using optokinetic nystagmus responses in infants. Three movies depicting an animal running were presented, followed by the test random dot kinematogram. We predicted that viewing animal motions would shift OKN responses to the RDK in the opposite direction of animal motion. Results showed that motion adaptation shifted the OKN responses to the RDK in the opposite direction of animal motion in all infants. This suggests that 3-8 monthinfants were able to adapt to animal motion direction. This is the first study to reveal infants' motion adapta

Object continuity through invisible retinal motion at saccadic speed.

Saccadic eye movements rapidly shift the visual scene across the retina, raising the question of how object correspondence is established between gaze fixations. Using submillisecond video projection, we isolated the role of high-speed retinal motion from saccade-related extraretinal signals. We introduced high-speed motion in a quartet motion display, a classic tool for studying long-distance object correspondence. While motion visibility dropped to chance level at saccadic speeds, even invisible motion robustly biased object correspondence. These results demonstrate that retinal motion alone

Remediation of the picture-text problem for learners exhibiting reading deficits.

Early reading materials are replete with pictures. Pictures purportedly improve reading comprehension and motivation; however, the simultaneous presentation of pictures and text can also impede textual control for some readers. Attempts to remediate restricted stimulus control in picture-text compounds suggest that omitting the picture element is most effective, although these arrangements may also be less socially valid. The current study is an evaluation of a novel compound stimulus prompt (CSP) arrangement that required that the learner differentially respond to the underselected (i.e., tex

Nitric oxide modulates contrast suppression in a subset of mouse retinal ganglion cells.

Neuromodulators have major influences on the regulation of neural circuit activity across the nervous system. Nitric oxide (NO) has been shown to be a prominent neuromodulator in many circuits and has been extensively studied in the retina. Here, it has been associated with the regulation of light adaptation, gain control, and gap junctional coupling, but its effect on the retinal output, specifically on the different types of retinal ganglion cells (RGCs), is still poorly understood. In this study, we used two-photon Ca2+ imaging and multi-electrode array (MEA) recordings to measure light-evo

Long-range precise synchronous firing and receptive field encoding in the frog retina.

Long-range synchrony in the frog retina has been reported to be accompanied by oscillations. In this study, we found novel long-range synchronous firing without oscillatory activities. Spatially correlated visual stimuli were presented to the frog retina, after which spike responses from dimming detectors were analyzed to investigate the correlated spike activities and receptive fields (RFs). The results revealed precise synchronous firing from both adjacent cell pairs with overlapping RFs and cell pairs with distant RFs of up to 1,500 µm apart, even in the absence of oscillatory activities. Encoding of cerebellar dentate neuron activity during visual attention in rhesus macaques.

The role of cerebellum in controlling eye movements is well established, but its contribution to more complex forms of visual behavior has remained elusive. To study cerebellar activity during visual attention we recorded extracellular activity of dentate nucleus (DN) neurons in two non-human primates (NHPs). NHPs were trained to read the direction indicated by a peripheral visual stimulus while maintaining fixation at the center, and report the direction of the cue by performing a saccadic eye movement into the same direction following a delay. We found that single-unit DN neurons modulated s

Neural Correlates of Category Learning in Monkey Inferior Temporal Cortex.

Area TE is required for normal learning of visual categories based on perceptual similarity. To evaluate whether category learning changes neural activity in area TE, we trained two monkeys (both male) implanted with multielectrode arrays to categorize natural images of cats and dogs. Neural activity during a passive viewing task was compared pre- and post-training. After the category training, the accuracy of abstract category decoding improved. Single units became more category selective, the proportion of single units with category selectivity increased, and units sustained their category-s

Induced awareness of synesthetic sensations in synesthetically predisposed "Borderline Non-synesthetes".

A long-standing issue concerning synesthesia is whether the trait is continuous or discontinuous with ordinary perception. Here, we found that a substantial proportion of non-synesthetes (>10 % out of >200 unselected participants) spontaneously became aware of their synesthesia by participating in an online survey that forced them to select colors for stimuli that evoke color sensations in synesthetes. Notably, the test-retest consistencies of color sensation in these non-synesthetes were comparable to those in self-claimed synesthetes, revealing their strong though latent synesthetic disposit
