Change of Flavonoid Content in Wheatgrass in a Historic Collection of Wheat Cultivars.

Wheatgrass is recognized for its nutritional and medicinal properties, partly attributed to its flavonoid content. The objective of this study was to assess the flavonoid content and antioxidant properties of wheatgrass obtained from a wide range of 145 wheat cultivars, which included Chinese landraces (CL), modern Chinese cultivars (MCC), and introduced modern cultivars (IMC). The flavonoids were extracted using a solution of 80% methanol, and their content was evaluated using ultra-high-performance liquid chromatography-mass spectrometry (UHPLC-MS). The results revealed the assessed cultivar

Rising Out of the Ashes: Additive Genetic Variation for Crown and Collar Resistance to Hymenoscyphus fraxineus in Fraxinus excelsior.

Since the early 1990s, ash dieback due to the invasive ascomycete Hymenoscyphus fraxineus is threatening Fraxinus excelsior in most of its natural range. Previous studies reported significant levels of genetic variability in susceptibility in F. excelsior either in field or inoculation experiments. The present study was based on a field experiment planted in 1995, 15 years before onset of the disease. Crown and collar status were monitored on 777 trees from 23 open-pollinated progenies originating from three French provenances. Health status was modeled using a Bayesian approach where spatiote

Application of Molecular Markers for Trait-Specific Breeding of Aroma Accumulation in Rice.

The translation of genetic variation into breeding gain relies on an integrated and sequential pipeline comprising high-quality DNA extraction, precise locus amplification and sequencing, accurate variant discovery, conversion of polymorphisms into diagnostic markers, high-throughput genotyping, and systematic integration into crossing and selection programs. This chapter presents a comprehensive framework for the development and application of molecular markers for aroma improvement in rice, with broader relevance to yield and stress-resilience traits in Oryza sativa. Rice aroma, a premium gr

Genetic dissection of cold tolerance across developmental stages in sorghum reveals stage-specific haplotypes and potential trade-offs.

Cold tolerance in sorghum is largely stage- and environment-specific yet partially shared genetic control allows improvement across developmental stages with mostly modest, trait-specific trade-offs. Cold tolerance is a major constraint to sorghum production in temperate environments, and its improvement is therefore a key breeding objective. However, cold tolerance varies across developmental stages and environments, and selection based on stage-specific phenotypes may overlook shared genetic components. We analysed a diversity panel of 394 sorghum genotypes evaluated across 10 environments r

The impact of protein source and grain inclusion on digestibility, fecal metabolites, and fecal microbiome in adult canines.

This study was conducted to determine the effect of animal protein inclusion rate and grain-free or grain-inclusive diets on macronutrient digestibility, fecal characteristics, metabolites, and microbiota in mixed-breed hounds and Beagles. Four experimental extruded kibble diets were made with varying amounts of animal protein and carbohydrates: 1) high animal protein, grain-inclusive (HA-GI), 2) low animal protein, grain-free (LA-GF), 3) low animal protein, grain-inclusive (LA-GI), and 4) high animal protein, grain-free (HA-GF). Thirty-two Beagles and 33 mixed-breed hounds were assigned to 1

Selecting goats for juniper consumption did not improve their liver Phase I detoxification.

A sodium pentobarbital-induced sleep time study was conducted on 15 adult intact male Boer × Spanish goats selected for high (J+, n = 7) or low (J-, n = 8) juniper consumption (estimated breeding values of 13.1 ± 1.0 and -14.3 ± 0.8, respectively; mean ± standard deviation). Pentobarbital sleep time is an in vivo assay of Phase I hepatic metabolism that can be induced by exposure to barbiturates and monoterpenes. Monoterpenes and pentobarbital are initially oxidized by this pathway; thus, we hypothesized that J+ goats would have shorter sleep times than J- goats. Time to the righting reflex af

Spinal cord protothecosis causing paraparesis in a dog.

Protothecosis, an infectious disease caused by the green algae Prototheca zopfii and P. wickerhamii, occurs sporadically in domestic animals and humans. Diagnosis of CNS protothecosis is based on neurologic signs that indicate multifocal nervous system lesions and that follow a period of chronic diarrhea and weight loss, cytologic observation of algae in fecal culture or histopathology, and detection of the agent by PCR assay of infected tissues. Here, we report a case of a paraparetic dog with CNS protothecosis that was diagnosed definitively antemortem using CSF cytology, PCR, and DNA sequen

Albino Plant Formation in Androgenic Cultures: An Old Problem and New Facts.

High frequency of albino plant formation in isolated microspore or anther cultures is a great problem limiting the possibility of their exploitation on a wider scale. It is highly inconvenient as androgenesis-based doubled haploid (DH) technology provides the simplest and shortest way to total homozygosity, highly valued by plant geneticists, biotechnologists and especially, plant breeders, and this phenomenon constitutes a serious limitation of these otherwise powerful tools. The genotype-dependent tendency toward albino plant formation is typical for many monocotyledonous plants, including c

A genome scan for quantitative trait loci affecting cyanogenic potential of cassava root in an outbred population.

BACKGROUND: Cassava (Manihot esculenta Crantz) can produce cyanide, a toxic compound, without self-injury. That ability was called the cyanogenic potential (CN). This project aimed to identify quantitative trait loci (QTL) associated with the CN in an outbred population derived from 'Hanatee' × 'Huay Bong 60', two contrasting cultivars. CN was evaluated in 2008 and in 2009 at Rayong province, and in 2009 at Lop Buri province, Thailand. CN was measured using a picrate paper kit. QTL analysis affecting CN was performed with 303 SSR markers. RESULTS: The phenotypic values showed continuous varia

The Co-Expression of GmCML27 and GmU2AFb Enhances Tolerance to Alkaline Stress in Lupinus angustifolius.

Soil salinization severely constrains plant growth and agricultural productivity. Identifying key genes conferring alkali tolerance and elucidating their regulatory mechanisms is of great significance for breeding salt-alkali-tolerant leguminous crops. In this study, we investigated the soybean genes GmCML27 and its potential interacting partner GmU2AFb, and explored their functions in response to alkali stress in narrow-leaf lupine (Lupinus angustifolius) through bioinformatics analyses. Protein interaction prediction and experimental validation further supported a potential association betwe

Advances in Plant Antiviral RNAi: From Host DCLs/RDRs to Diversified Viral Counteracting Strategies.

Plant RNA interference (RNAi) is a fundamental antiviral defense that relies on coordinated activities of DICER-like endonucleases (DCLs), Argonaute proteins (AGOs) and RNA-dependent RNA polymerases (RDRs). Over the past decades, studies using model and crop species have uncovered complex and often redundant roles for DCLs and RDRs in generating and amplifying virus-derived small interfering RNAs (vsiRNAs), in addition to connections with transcriptional gene silencing (TGS) and epigenetic defenses against DNA viruses. Concurrently, plant viruses have evolved diverse counterstrategies-proteina

A Positive Regulator CsPR10-9 Confers Resistance to Anthracnose (Colletotrichum gloeosporioides) Is Negatively Regulated by CsMYB72 in Tea Plants.

Anthracnose is a devastating fungal disease in tea plants caused by the pathogen Colletotrichum species. It severely affects tea yield and quality. Pathogenesis-related (PR) genes play indispensable roles in plant resistance to fungal pathogens. Our transcriptome data showed that PR10 family genes are involved in the immune response to anthracnose infection in tea plants; however, the underlying regulatory mechanisms remain unclear. Here, we identified a PR10 family gene, CsPR10-9, which was highly induced by Colletotrichum gloeosporioides infection and salicylic acid (SA)/methyl jasmonate (Me

Plant RNA interference from antiviral silencing to multiplex trait engineering for climate-resilient crops.

RNA interference (RNAi) in plants has evolved from an unexplained antiviral and transgene interference phenomenon into a general regulatory platform for sequence-guided gene suppression, chromatin control, systemic signaling, and phenotypic plasticity. This Review synthesizes six decades of plant RNAi, tracing its progression through conceptual bottlenecks and technological solutions. Early work established that RNA-derived homology could suppress viral infection and transgene expression. Mechanistic studies then revealed a diversified plant silencing system involving Dicer-like proteins, Argo

Identification of Promising RILs for High Grain Zinc Through Genotype × Environment Analysis and Stable Grain Zinc QTL Using SSRs and SNPs in Rice (Oryza sativa L.).

Polished rice is one of the commonly consumed staple foods across the world. However, it contains limited nutrients especially iron (Fe) and zinc (Zn). To identify promising recombinant inbred lines (RILs) for grain Zn and single plant yield, 190 RILs developed from PR116 and Ranbir Basmati were evaluated in two environments (E1 and E2). A subset of 44 contrasting RILs for grain Zn was screened in another two environments (E3 and E4). Phenotypic data was collected for 10 traits, viz., days to 50% flowering, plant height, panicle length, number of tillers, single plant yield (SPY), test weight,

Pangenome graph analysis reveals evolution of resistance breaking in spinach downy mildew.

Filamentous plant pathogens secrete effectors to successfully establish host infections. In resistant crop varieties, plant immunity can be triggered by immune receptors that recognize these effectors. Resistant crop varieties are grown in large-scale monocultures imposing strong selection pressure on pathogens, driving rapid evolution of effector repertoires resulting in the frequent breakdowns of resistance within just a few growing seasons. The oomycete Peronospora effusa, responsible for downy mildew on spinach, is an example of a rapidly adapting pathogen, but it is yet unknown how P. eff

Application of Metabolomics in Defence Responses of Brassica Crops.

Brassica crops, encompassing globally important vegetables and oilseeds, face severe threats from diverse biotic and abiotic stresses. Plant secondary metabolites constitute the chemical foundation of defence, and metabolomics has emerged as an effective systems biology tool for comprehensively dissecting stress-induced metabolic changes. Recent progress in applying metabolomics to elucidate defence mechanisms in Brassica crops is systematically synthesised here. Major stresses confronting Brassica crop production and the metabolic basis of plant defence are first outlined. Current analytical

Advances in the Evolutionary Mechanisms and Genomic Studies of Sexual Differentiation in Lauraceae Plants.

The Lauraceae family, a keystone group in subtropical evergreen broad-leaved forest ecosystems, exhibits exceptional diversity in sexual systems (including hermaphroditic flowers, functionally unisexual flowers, and pseudo-dioecy), serving as a natural model for studying plant sexual differentiation mechanisms. This review synthesizes advances in the evolutionary mechanisms and genomic studies of sexual differentiation in Lauraceae, focusing on three key areas: (1) the evolution of taxonomic classification and floral morphology, (2) molecular trajectories of sexual differentiation, and (3) cha

The Biocontrol and Plant Growth-Promoting Properties of Streptomyces alfalfae XN-04 Revealed by Functional and Genomic Analysis.

Fusarium wilt of cotton, caused by the pathogenic fungal Fusarium oxysporum f. sp. vasinfectum (Fov), is a devastating disease of cotton, dramatically affecting cotton production and quality. With the increase of pathogen resistance, controlling Fusarium wilt disease has become a significant challenge. Biocontrol agents (BCAs) can be used as an additional solution to traditional crop breeding and chemical control. In this study, an actinomycete with high inhibitory activity against Fov was isolated from rhizosphere soil and identified as Streptomyces alfalfae based on phylogenetic analyses. Ne

Comprehensive mapping of abiotic stress inputs into the soybean circadian clock.

The plant circadian clock evolved to increase fitness by synchronizing physiological processes with environmental oscillations. Crop fitness was artificially selected through domestication and breeding, and the circadian clock was identified by both natural and artificial selections as a key to improved fitness. Despite progress in Arabidopsis, our understanding of the crop circadian clock is still limited, impeding its rational improvement for enhanced fitness. To unveil the interactions between the crop circadian clock and various environmental cues, we comprehensively mapped abiotic stress

Physiological and quantitative proteomic analyses unraveling potassium deficiency stress response in alligator weed (Alternanthera philoxeroides L.) root.

Physiological and iTRAQ based proteomic analysis provided new insights into potassium deficiency stress response in alligator weed root. Alligator weed (Alternanthera philoxeroides) has a strong ability to adapt to potassium deficiency (LK) stress. Proteomic changes in response to this stress are largely unknown in alligator weed. In this study, we investigated physiological and molecular mechanisms under LK using isobaric tags for relative and absolute quantitation to characterize proteome-level changes in this plant.
