Effects of probiotic fermentation on Gracilaria bailinae polysaccharides: Characterization and immunomodulatory potency.

Gracilaria bailinae polysaccharides have limited applications due to their high viscosity, large molecular weight, and poor solubility. Microbial fermentation emerges as an efficient strategy to enhance the bioactivity of plant polysaccharides. This study aimed to prepare fermented water-soluble crude polysaccharide (GBV) from G. bailinae via fermentation with Bacillus velezensis, using water-extracted polysaccharide (GBP) as a control. The physicochemical properties, immunomodulatory activity, and trained immunity effects of GBV were systematically investigated. A significant decrease in mole

Salmonella Biofilm Formation, Chronic Infection, and Immunity Within the Intestine and Hepatobiliary Tract.

Within the species of Salmonella enterica, there is significant diversity represented among the numerous subspecies and serovars. Collectively, these account for microbes with variable host ranges, from common plant and animal colonizers to extremely pathogenic and human-specific serovars. Despite these differences, many Salmonella species find commonality in the ability to form biofilms and the ability to cause acute, latent, or chronic disease. The exact outcome of infection depends on many factors such as the growth state of Salmonella, the environmental conditions encountered at the time o

Interleukin-1 and cutaneous inflammation: a crucial link between innate and acquired immunity.

As our primary interface with the environment, the skin is constantly subjected to injury and invasion by pathogens. The fundamental force driving the evolution of the immune system has been the need to protect the host against overwhelming infection. The ability of T and B cells to recombine antigen receptor genes during development provides an efficient, flexible, and powerful immune system with nearly unlimited specificity for antigen. The capacity to expand subsets of antigen-specific lymphocytes that become activated by environmental antigens (memory response) is termed "acquired" immunit

Terminalia brownii Fresen: Stem Bark Dichloromethane Extract Alleviates Pyrogallol-Induced Suppression of Innate Immune Responses in Swiss Albino Mice.

Terminalia brownii is widely used in folklore medicine and has diverse biological activities. However, its effect on the immune system is yet to be studied. Therefore, our study evaluated the immunomodulatory effect of T. brownii on nonspecific immunity. Innate immunity is the initial defence phase against pathogens or injuries. Dichloromethane plant extracts were tested on female Swiss albino mice and Wister rats. The effect of the extract on innate immunity was assessed via total and differential leukocyte counts, tumor necrosis factor-alpha, and nitric oxide production by mouse macrophages.

Long-term supplementation of Rhodiola kirilowii extracts during pregnancy and lactation does not affect mother health status.

Antibiotics treatment during pregnancy and lactation is problematic. The alternative to the antibiotic treatment is the use of plant-derived supplements, which stimulate immune system to prevent and eliminate bacterial infection. Here, we evaluated the effect of long-term use of Rhodiola kirilowii on the health of mouse mothers. Pregnant mice were fed daily, for whole pregnancy and for 28 days after giving birth, with Rhodiola kirilowii water (RKW) or hydroalcoholic extract (RKW-A) (at 20 mg of extracts/kg). The control group received sterile water. There was no significant change in the total

A Dual-Role Transcription Factor FpCys3 Couples Glucose Sensing to Methionine Biosynthesis and Virulence in Fusarium pseudograminearum.

Fusarium pseudograminearum is a soil-borne phytopathogen that causes Fusarium crown rot (FCR) in wheat and barley, posing a severe threat to global grain yield and quality. A deeper understanding of its pathogenicity is essential for developing effective control strategies. Methionine (Met) has been reported that involved in plant-pathogen interactions at multiple levels, regulating both plant immunity and fungal pathogenicity. Here, we identified a conserved bZIP transcription factor, FpCys3, which is a key mediator of methionine biosynthesis in F. pseudograminearum. The FpCys3 mutant exhibit…

Effector innovation in genome-reduced phytoplasmas and other host-dependent mollicutes.

Obligate host-associated bacteria with reduced genomes, such as phytoplasmas, face strong evolutionary constraints, including metabolic dependence on hosts, limited opportunities for horizontal gene transfer (HGT), and frequent population bottlenecks. Despite these limitations, phytoplasmas, which are parasitic, insect-transmitted plant pathogens, maintain a diverse arsenal of secreted effectors that manipulate both plant and insect hosts to promote infection and transmission. These effectors can suppress immunity and reprogram plant development, inducing alterations such as witch's broom and

BSMT1, SABP3, and MAT2 assemble into a ternary complex vital for methyl salicylate biosynthesis and airborne defense.

When plants encounter biotic and abiotic stresses, they emit various volatile organic compounds to communicate with nearby plants and activate airborne defense (AD). One critical compound in this process is methyl salicylate (MeSA). Previous studies have mostly examined how stress triggers the production of MeSA at the gene level. In our study, we found that MeSA plays a key role in AD during bacterial infections and determined how plants boost MeSA production through a protein complex. Infection by Pseudomonas syringae pv. tomato DC3000 in Arabidopsis thaliana increased salicylic acid levels,

Plant hypersensitive response vs pathogen ingression: Death of few gives life to others.

The hypersensitive response (HR) is a defense action against pathogen ingression. Typically, HR is predictable with the appearance of the dead, brown cells along with visible lesions. Although death during HR can be limited to the cells in direct contact with pathogens, yet cell death can also spread away from the infection site. The variety in morphologies of plant cell death proposes involvement of different pathways for triggering HR. It is considered that, despite the differences, HR in plants performs the resembling functions like that of animal programmed cell death (PCD) for confining p

ADS1 encodes a MATE-transporter that negatively regulates plant disease resistance.

Multidrug and toxic compound extrusion (MATE) proteins comprise the most recently identified family of multidrug transporters. In plants, the numbers of MATE proteins has undergone a remarkable expansion, underscoring the importance of these transporters within this kingdom. Here, we describe the identification and characterization of Activated Disease Susceptibility 1 (ADS1) which encodes a putative MATE transport protein. An activation tagging screen uncovered the ads1-Dominant (ads1-D) mutant, which was subsequently characterized by molecular, genetic and biochemical approaches. The ads1-D

Obesity related lung complications and a possible phytochemical therapeutic solution.

Proteases, especially neutrophil elastase (NE), enzyme provide innate immunity in the lung tissues against elastin protein disintegration by microbial attacks. Normally, an antiprotease, called alpha-1-antitrypsin (A1AT), is responsible to keep NE activity in normal range. In most of the leptin-resistant obese subjects, A1AT deficiency develops which leads to a concurrent higher activity of NE. Obesity associated antiprotease-protease imbalance induces various lung complications, i.e. asthma, emphysema, chronic obstructive pulmonary disorders (COPD), cancer, etc. Past studies manifested that p

Enteric defensins are essential regulators of intestinal microbial ecology.

Antimicrobial peptides are important effectors of innate immunity throughout the plant and animal kingdoms. In the mammalian small intestine, Paneth cell alpha-defensins are antimicrobial peptides that contribute to host defense against enteric pathogens. To determine if alpha-defensins also govern intestinal microbial ecology, we analyzed the intestinal microbiota of mice expressing a human alpha-defensin gene (DEFA5) and in mice lacking an enzyme required for the processing of mouse alpha-defensins. In these complementary models, we detected significant alpha-defensin-dependent changes in mi

Genomic signatures of host-range divergence in the generalist Beauveria bassiana and the specialist Beauveria brongniartii.

Entomopathogenic fungi of the genus Beauveria are widely used biological control agents that infect diverse insect hosts and can also associate with plants as rhizosphere colonizers and endophytes. Within this genus, Beauveria bassiana is a cosmopolitan generalist, whereas Beauveria brongniartii exhibits a narrower host range, primarily targeting soil-dwelling coleopteran larvae with limited evidence of plant colonization. To explore genomic differentiation associated with this ecological divergence, the commercially exploited B. brongniartii strain BIPESCO2 and B. bassiana ATHUM 4946 were seq

Evolutionary genomics of three agricultural pest moths reveals rapid evolution of host adaptation and immune-related genes.

BACKGROUND: Understanding the genotype of pest species provides an important baseline for designing integrated pest management (IPM) strategies. Recently developed long-read sequence technologies make it possible to compare genomic features of nonmodel pest species to disclose the evolutionary path underlying the pest species profiles. Here we sequenced and assembled genomes for 3 agricultural pest gelechiid moths: Phthorimaea absoluta (tomato leafminer), Keiferia lycopersicella (tomato pinworm), and Scrobipalpa atriplicella (goosefoot groundling moth). We also compared genomes of tomato leafm

The Verticillium dahliae Small Cysteine-Rich Protein VdSCP23 Manipulates Host Immunity.

Verticillium wilt caused by Verticillium dahliae is a notorious soil-borne fungal disease and seriously threatens the yield of economic crops worldwide. During host infection, V. dahliae secretes many effectors that manipulate host immunity, among which small cysteine-rich proteins (SCPs) play an important role. However, the exact roles of many SCPs from V. dahliae are unknown and varied. In this study, we show that the small cysteine-rich protein VdSCP23 inhibits cell necrosis in Nicotiana benthamiana leaves, as well as the reactive oxygen species (ROS) burst, electrolyte leakage and the expr

Orchestration of plant development and defense by indirect crosstalk of salicylic acid and brassinosteorid signaling via transcription factor GhTINY2.

Salicylic acid (SA) and brassinosteroids (BRs) are well known to regulate diverse processes of plant development and stress responses, but the mechanisms by which these phytohormones mediate the growth and defense trade-off are largely unclear. In addition, little is known about the roles of DEHYDRATION RESPONSIVE ELEMENT BINDING transcription factors, especially in biotic stress and plant growth. Here, we identified a cotton (Gossypium hirsutum) APETALA2/ETHYLENE RESPONSIVE FACTOR gene GhTINY2 that is strongly induced by Verticillium dahliae.
