Biotin-modified glutathione as a functionalized coating for bioconjugation of CdTe-based quantum dots.

In this study, biotin-conjugated glutathione was synthesized using peptide bonding of the biotin carboxy group and amino group of the γ-glutamic acid to prepare an alternative coating for CdTe quantum dots (QDs). This type of coating combines the functionality of the biotin with the fluorescent properties of the QDs to create a specific, high-affinity fluorescent probe able to react with avidin, streptavidin and/or neutravidin. Biotin-functionalized glutathione-coated CdTe QDs were prepared by a simple one-step method using Na₂ TeO₃ and CdCl₂. Obtained QDs were separated from the excess of the…

Synchronous determination of mercury (II) and copper (II) based on quantum dots-multilayer film.

A sensitive sensor for mercury (II) and copper (II) synchronous detection was established via the changed photoluminescence of CdTe quantum dots (QDs) multilayer films in this work. QDs were deposited on the quartz slides to form QDs-multilayer films by electrostatic interactions with poly(dimethyldiallyl ammonium chloride) (PDDA). Hg(2+) or Cu(2+) could quench the photoluminescence of the QDs-multilayer films, and glutathione (GSH) was used to remove Hg(2+) or Cu(2+) from QDs-multilayer films due to strong affinity of GSH-metal ions, which resulted in the recovered photoluminescence of QDs-mu…

Two-fold emission from the S-shell of PbSe/CdSe core/shell quantum dots.

The optical properties of PbSe/CdSe core/shell quantum dots with core sizes smaller than 4 nm in the 5-300 K range are reported. The photoluminescence spectra show two peaks, which become increasingly separated in energy as the core diameter is reduced below 4 nm. It is shown that these peaks are due to intrinsic exciton transitions in each quantum dot, rather than emission from different quantum dot sub-ensembles. Most likely, the energy separation between the peaks is due to inter-valley coupling between the L-points of PbSe. The temperature dependence of the relative intensities of the peak…

Preparation and biological effect of nucleotide-capped CdSe/ZnS quantum dots on Tetrahymena thermophila.

In this paper, we described the preparation and characterization of different types of modified CdSe/ZnS quantum dots (QDs) and explored the biological effects of QDs with different surface modifications on the whole growth of unicellular protozoan Tetrahymena thermophila BF(5) using a thermal activity monitor air isothermal microcalorimeter. Our results demonstrated that adenosine 5'-monophosphate (AMP) showed stronger interaction with QDs than other types of nucleotide. AMP-QDs could stimulate the growth of T. thermophila while mercaptoacetic acid-capped CdSe/ZnS quantum dots inhibited it. I…

Mechanisms underlying toxicity induced by CdTe quantum dots determined in an invertebrate model organism.

A systematic and thorough quantitative analysis of the in vivo effects of inorganic nanoparticles is extremely important for the design of functional nanomaterials for diagnostic and therapeutic applications, better understanding of their non-specificity toward tissues and cell types, and for assessments of their toxicity. This study was undertaken to examine the impact of CdTe quantum dots (QDs) on an invertebrate freshwater model organism, Hydra vulgaris, for assessment of long term toxicity effects. The continuous exposure of living polyps to sub-lethal doses of QDs caused time and dose dep…

Probing the dynamic effect of cys-CdTe quantum dots toward cancer cells in vitro.

The application of quantum dots (QDs) in various biomedical areas requires detailed studies of their toxicity. We report a new strategy for probing the biocompatibility of these nanocrystals, namely, a dynamic investigation of cellular uptake images, cell growth curves, metabolic activity changes, and apoptosis aspects of cadmium telluride QDs capped with cysteamine (Cys-CdTe QDs) on human hepatocellular carcinoma SMMC-7721 cells. We used a real-time cell electronic sensing (RT-CES) system in combination with fluorescence microscopy, 3-(4,5-dimethyl-thiazol-zyl)-2,5-diphenyltetrazolium bromide…

Interaction and Cooperative Nucleation of InAsSbP Quantum Dots and Pits on InAs(100) Substrate.

An example of InAsSbP quaternary quantum dots (QDs), pits and dots-pits cooperative structures' growth on InAs(100) substrates by liquid phase epitaxy (LPE) is reported. The interaction and surface morphology of the dots-pits combinations are investigated by the high-resolution scanning electron microscope. Bimodal growth mechanism for the both QDs and pits nucleation is observed. Cooperative structures consist of the QDs banded by pits, as well as the "large" pits banded by the quantum wires are detected. The composition of the islands and the pits edges is found to be quaternary, enriched by…

Fluorescence sensing of nitric oxide in aqueous solution by triethanolamine-modified CdSe quantum dots.

The water-soluble luminescent CdSe quantum dots were prepared by ligand exchange with triethanolamine (TEA). Oxygen can reversibly enhance the fluorescence of the synthesized quantum dots (TEA-CdSe-QDs) in aqueous solution. Nitric oxide radical (NO) can react easily with dissolved oxygen in water and was found to have a significant quenching effect on the fluorescence of the TEA-CdSe-QDs. The fluorescence responses were concentration-dependent and can be well described by the typical Stern-Volmer equation. A good linear relationship (R(2 )= 0.9963) was observed over the range 5.92 x 10(-7 )to …

UV-enhanced cytotoxicity of thiol-capped CdTe quantum dots in human pancreatic carcinoma cells.

Quantum dots (QDs) have been gaining popularity due to their potential application in cellular imaging and diagnosis, but their cytotoxicity under light illumination has not been fully investigated. In this study, green and red mercaptopropionic acid capped CdTe quantum dots (MPA-CdTe QDs) were employed to investigate their cytotoxicity in human pancreatic carcinoma cells (PANC-1) under UV illumination. MPA-CdTe QDs exhibited excellent photostability under UV illumination and could be easily ingested by cells. The cytotoxicity of MPA-CdTe QDs was significantly enhanced under UV illumination, w…

Synchrotron-based X-ray fluorescence imaging of human cells labeled with CdSe quantum dots.

Synchrotron-based X-ray fluorescence (S-XRF) is a powerful technique for imaging the distribution of many biologically relevant elements as well as of "artificial" elements deliberately introduced into tissues and cells, for example, through functionalized nanoparticles. In this study, we explored the potential of S-XRF for chemical nanoimaging (100 nm spatial resolution, nanoXRF) of human cells through the use of functionalized CdSe/ZnS quantum dots (QDs). We used a commercially available QD-secondary antibody conjugate to label the cancer marker HER2 (human epidermal growth factor receptor 2…

Enhanced biocompatibility and biostability of CdTe quantum dots by facile surface-initiated dendritic polymerization.

The synthesis of stable, low toxic, multifunctional, and water-soluble quantum dots (QDs) is of crucial importance for nanobiotechnology. An in situ anionic ring-opening polymerization strategy was successfully employed to grow multihydroxyl hyperbranched polyglycerol (HPG) from surfaces of aqueous synthesized QDs directly, affording multifunctional CdTe@HPG nanohybrids. The grafted HPG content can be adjusted from about 25 to 80 wt % by manipulating the feed ratio of glycidol monomer to QDs. The resultant CdTe@HPGs still show strong fluorescence and well water-solubility, and can conjugate fu…

Systematic study of the interaction of cobalt ions with different-sized CdTe quantum dots.

Five sizes of water-dispersed CdTe quantum dots (QDs) stabilized by thioglycolic acid (TGA) with a high photoluminescence (PL) quantum yield were synthesized and a size dependent quenching of the fluorescence by cobalt ions was also observed. No matter for smaller or larger particles, obvious quenching effect was observed, and the fluorescence quenching of CdTe nanoparticles depended on the concentration of cobalt ions solution. However, CdTe QDs with different size showed dramatically different quenching efficiency, sensitivity, linear range and selectivity. With the increase of size, the que…

Zwitterionic biocompatible quantum dots for wide pH stability and weak nonspecific binding to cells.

Applications of water-soluble quantum dots (QDs) in the life sciences are limited by their poor colloidal stability in physiological media and nonspecific interaction with biomatter, particularly cell membranes. We have studied colloidal stability and nonspecific interactions with living cells for zwitterionic d-penicillamine-coated QDs (DPA-QDs) and the traditionally used carboxylated 11-mercaptoundecanoic acid-coated QDs (MUA-QDs) and found clear advantages of DPA-QDs. In single molecule fluorescence experiments, DPA-QDs showed no aggregation over the physiologically relevant pH range of 5-9…

Donor-acceptor systems: energy transfer from CdS quantum dots/rods to Nile Red dye.

We demonstrate strong evidence of shape-dependent efficient resonance energy transfer between CdS quantum dots (QDs) and quantum rods (QRs) (donor) to Nile Red dye (acceptor). We also report a simple solution-based method for the preparation of high quality CdS QDs and CdS QRs at relatively low temperature. The observed quenching of PL intensities are 78.8 % and 63.8 % for CdS QDs and QRs, respectively in the presence of Nile Red dye. The calculated energy-transfer efficiencies are 45 % and 19 % from QDs and QRs to dyes, respectively. The energy transfer varies with changing the shape of the n…

Fluorescence quenching of CdSe quantum dots by tertiary amines and their surface binding effect.

The effect of different amines on the fluorescence of quantum dots has been discussed in the literature but the mechanism involved is not fully understood. We have studied the interaction of core CdSe quantum dots of different sizes and core-shell CdSe quantum dots with tertiary amines, such as aliphatic amines (triethylamine) and bicyclic amines (DABCO and quinuclidine), using fluorescence techniques. Fluorescence quantum yield decreases with increasing concentration of the amine following a linear Stern-Volmer behaviour (Kq=80-330 M(-1)), but relatively small changes were observed in the flu…
