Nasal-cannula-integrated Fabry-Pérot fiber sensor for respiration-driven Morse code communication.

Spontaneous breathing offers a viable non-limb-based interaction channel for patients with severe motor impairments. However, traditional electronic monitors are often limited by electromagnetic susceptibility and bulky designs. We report a flexible, wearable human-computer interface that incorporates a miniature Fabry-Pérot interferometer (FPI) fiber-optic sensor directly embedded within a standard nasal cannula. By measuring temperature-induced changes in cavity length, the sensor tracks exhaled airflow with high sensitivity. The sensor achieves an experimental temperature sensitivity of 0.2

Amyloid precursor protein interacts with the mitochondrial phosphatase PGAM5 and regulates mitochondrial respiration.

Amyloid Precursor Protein (APP) has been reported to partially localize to mitochondria, and mitochondrial dysfunction is a key feature of A … lated factor 2 (Nrf2) through Keap1 at the outer mitochondrial membrane and regulates mitochondrial respiration and stress responses. We found that the Nrf2-regulated genes Hmox1 (Heme oxygenase-1) and Nqo1 (NADH:quinone oxidoreductase 1), which are involved in mitochondrial respiration, are downregulated in APP KO astrocytes. Accordingly, mitochondria isolat …

Targeting MEK1/2 inhibits mitochondrial respiration and redox homeostasis in resistant rectal cancer.

Chemo-resistance is a major challenge in rectal cancer treatment. This study investigates the therapeutic potential of MEK inhibitors, cobim … markers, including protein carbonyl and malondialdehyde (MDA), while also decreasing mitochondrial respiration, as evidenced by reduced oxygen consumption rates (OCR). Apoptotic induction was significantly reduced in mitochondrial respiration-deficient p⁰ cells, supporting the role of mitochondrial respiration in MEK inhibitor activity. Genetic MEK1/2 knockd …

A reversible rat model of hyposmia affects respiration-linked brain oscillations and behavior.

Rhythmic breathing coordinates neural activity across olfactory and limbic circuits by coupling nasal airflow to oscillations in the olfactory bulb (OB) and downstream networks. To test how reduced peripheral olfactory input reshapes this respiration-linked coordination, we developed a reversible hyposmia model in rats using intranasal gadolinium chloride. This manipulation impaired hidden cookie performance for around 1 week and was accompanied by nasal epithelial damage. Hyposmia robustly decreased OB respiration-linked (1-10 Hz) and gamma (30-90 Hz) power, with smaller reductions in prefron…

Galactokinase 1 Positively Regulates Mitochondrial Respiration by Phosphorylating TIMM13 as a Protein Kinase.

Galactokinase 1 (GALK1) is a metabolic kinase that phosphorylates galactose. Mutations in GALK1 cause type II galactosemia, which manifests … ations showed compromised activity in TIMM13 phosphorylation, subsequently repressing mitochondrial respiration. Collectively, the GALK1-TIMM13 axis appears to provide a potential link between galactose metabolism and mitochondrial respiration.

A scalable plate reader-based protocol to measure mitochondrial respiration in embryonic hearts.

Mitochondrial function is central to embryonic development, yet its assessment in the embryonic heart is hampered by limited tissue availability. Here, we present a protocol to measure mitochondrial oxygen consumption rates directly from embryonic mouse hearts using a fluorescence-based plate reader assay. We describe steps for heart isolation, plate setup, and acquisition of signal profiles. We then detail procedures for normalization and rate conversion. This protocol enables real-time readings with a low input requirement of one heart.

Extracellular electron transfer in cable bacteria enables growth rates comparable to aerobic respiration.

Cable bacteria are filamentous sulphide-oxidising bacteria that transport electrons through conductive periplasmic fibres over centimetre-scale distances in redox-stratified sediments to respire oxygen. Here, we show that the freshwater cable bacterium Electronema aureum GS employs a versatile extracellular electron transfer (EET) system to respire insoluble electron acceptors under anoxic conditions, achieving growth rates comparable to aerobic respiration. Electrochemical and molecular analyses reveal that E. aureum GS performs both direct and riboflavin-mediated EET to electrodes, including…

Cox10-mediated mitochondrial respiration in brown adipocytes regulates adaptive thermogenesis and systemic metabolism.

Mitochondrial respiration is essential for Ucp1-mediated thermogenesis in brown adipocytes, where heat production depends on oxygen-driven mitochondrial activity. To define the role of complex IV, we generated brown-adipocyte-specific Cox10-knockout mice (Cox10BKO), as Cox10 is required for cytochrome c oxidase assembly. Cox10-deficient brown adipocytes exhibited markedly reduced complex IV activity and impaired Ucp1-dependent thermogenesis. Although ATF4 signaling was strongly induced, the alternative ATF4-dependent thermogenic pathway failed due to suppression of global protein synthesis, co…

Tyrosine 48 Phosphorylation of Cytochrome c Alters Mitochondrial Respiration, ROS Production, and Apoptosis.

Cytochrome c (Cytc) tyrosine 48 (Y48) has been previously shown to be phosphorylated in bovine liver, and phosphomimetic substitution (Y48E) inhibits key functions of Cytc in vitro, including respiration and apoptosis. In this study, we investigated the effect of Y48 modification in a double-knockout cell culture model that stably expressed either unphosphorylated wild-type (WT) Cytc, control Y48F Cytc, or phosphomimetic Y48E Cytc. Our findings revealed that Y48E Cytc caused partial inhibition of mitochondrial respiration in intact cells, which corresponded with lower mitochondrial membrane po…

Multilevel impairment of mitochondrial respiration with sex-specific signatures in inclusion body myositis.

BACKGROUND: Oxidative phosphorylation (OXPHOS) is a central function and a key indicator of mitochondrial fitness, yet studies in human tissue remain limited. Inclusion body myositis (IBM) is a progressive myopathy that lies at the intersection of aging, inflammation and mitochondrial dysfunction. We aimed to perform a comprehensive profiling of mitochondrial respiration in muscle tissue from patients with IBM. METHODS: A wide battery of complementary tests from RNA level to high-resolution respirometry on permeabilized muscle fibers was performed. The relationship between respiration, mitoch…

Targeting enhanced mitochondrial respiration chain activity as a potential therapeutic approach for endometriosis.

Endometriosis is a chronic condition defined by the presence of endometrial-like tissue outside the uterus. Since endometriotic cells share similarities with cancer cells, including uncontrolled cell growth and invasion, we investigated whether cancer cell-specific rewiring of mitochondrial signaling is also present in endometriotic cells. We utilized the endometriotic cell line 12Z and investigated its mitochondrial function in comparison with the uterine cancer cell line SK-UT-1 and the mammary epithelial cell line hTERT-HME1. We could show that the endometriotic 12Z cells share structural s

BLOC1S1 depletion via splice-switching oligonucleotides improves mitochondrial respiration and rescues ALS phenotypes.

Amyotrophic lateral sclerosis (ALS) is a rapidly progressing and debilitating neurodegenerative disease, yet the mechanisms underlying disease onset and progression remain poorly understood, particularly in sporadic ALS. Emerging evidence suggests that mitochondrial dysfunction and metabolic dysregulation are central to ALS pathophysiology. A key feature of ALS motor neurons (MNs) is hyper-acetylation of mitochondrial proteins, which disrupt mitochondrial respiration and energy homeostasis. In this study, we identify BLOC1S1 (also known as GCN5L1) as a novel regulator of mitochondrial acetylat…

Exploring the Molecular Interplay Between Oxygen Transport, Cellular Oxygen Sensing, and Mitochondrial Respiration.

Significance: The mitochondria play a key role in maintaining oxygen homeostasis under normal oxygen tension (normoxia) and during oxygen deprivation (hypoxia). This is a critical balancing act between the oxygen content of the blood, the tissue oxygen sensing mechanisms, and the mitochondria, which ultimately consume most oxygen for energy production. Recent Advances: We describe the well-defined role of the mitochondria in oxygen metabolism with a special focus on the impact on blood physiology and pathophysiology. Critical Issues: Fundamental questions remain regarding the impact of mitocho

Microplastic-Derived Dissolved Organic Matter Regulates Soil Carbon Respiration via Microbial Ecophysiological Controls.

Microbial regulation of soil carbon sequestration is vulnerable to anthropogenic stressors, notably microplastic pollution. Microplastics can release carbon-based compounds that serve as potential substrates for soil heterotrophic microbes. The impact of this novel microplastic-derived dissolved organic matter (MP-DOM) on soil carbon cycling and the underlying mechanisms remain largely unexplored. Here, short-term soil microcosm experiments were conducted with MP-DOM sourced from pristine and aged microplastics to compare their effects on soil carbon respiration against natural dissolved organ…

UDP-glucose regulates dendritic cell mitochondrial respiration via a nitric oxide-dependent mechanism.

Bone marrow-derived dendritic cell (BMDC) activation is associated with rewiring of cellular metabolism and concurrent large-scale changes in gene expression promoting a proinflammatory program characterized by expression of inducible nitric oxide synthase and the production of nitric oxide (NO). NO inhibits vital cellular activities including mitochondrial respiration. Mitochondrial respiration inhibition via NO occurs at discrete levels of activating stimulus, termed the mitochondrial respiration threshold, and regulation of this threshold is not fully understood. In this work, we characteri…
