Safety and immunogenicity of myxoma virus as a new viral vector for small ruminants.

Myxoma virus (MYXV), a leporide-specific poxvirus, represents an attractive candidate for the generation of safe and non-replicative vaccine vectors for other species. With the aim of developing new recombinant vaccines for ruminants, we evaluated the safety and the immunogenicity of recombinant MYXV in sheep. In vitro studies indicated that ovine primary fibroblasts were not permissive for MYXV and that infection of ovine peripheral blood mononuclear cells occurred at a low rate. Although non-specific activation significantly improved the susceptibility of lymphocytes, MYXV infection remained…

Natural peste des petits ruminants virus infection: novel pathologic findings resembling other morbillivirus infections.

The present study describes pathologic and virologic findings in 15 sheep and 6 goats that died of natural peste des petits ruminants virus infection in Turkey. Pathologic findings included erosive-ulcerative stomatitis, fibrino-necrotic tracheitis, bronchointerstitial pneumonia, multifocal coagulation necroses in the liver, and severe lymphocytolysis in lymphoid tissues. Syncytial cells were conspicuous, especially in the oral mucosa, pulmonary alveoli, liver, and lymphoid tissues. In addition to the typical tissue distribution, eosinophilic intracytoplasmic and/or intranuclear inclusions wer…

An attenuated Ehrlichia ruminantium (Welgevonden stock) vaccine protects small ruminants against virulent heartwater challenge.

Heartwater is a tick-borne disease of ruminants caused by the intracellular rickettsia Ehrlichia ruminantium. The only commercially available immunization procedure involves infecting animals with cryopreserved sheep blood containing virulent E. ruminantium organisms, followed by treatment with tetracyclines when fever develops. The virulent Welgevonden stock of E. ruminantium was attenuated by continuous propagation of the organisms in a canine macrophage-monocyte cell line (DH82), followed by re-adaptation to grow in a bovine endothelial cell line (BA 886). The material used for the present …

Mycoplasma species and related organisms isolated from ruminants in Britain between 1990 and 2000.

Between 1990 and 2000, more than 1600 mycoplasmas and the related acholeplasmas were identified from ruminant animals by the Mycoplasma Group at the Veterinary Laboratories Agency--Weybridge. Mycoplasma bovis was the most commonly identified pathogen, mostly from pneumonic calves but occasionally from cattle with mastitis and arthritis. Mycoplasma canis was first isolated in Britain in 1995 from pneumonic calves and the number of isolates increased to 18 per cent of the total mycoplasmas isolated from cattle in 1999. The ELISA for antibodies to M. bovis detected 1971 positive samples (22 per c

Does high plant feed magnesium and potassium protect healthy ruminants from atherosclerosis? A review.

The literature data were surveyed to clarify the role of magnesium and potassium in the development of atherosclerosis (AS) in cows and the findings were compared with human data. Special attention was paid to the eastern Finland where AS is very common in humans and absent in the cattle. A hypothesis is proposed that high magnesium and potassium intakes at plant feeding protect from severe AS processes at least in cows in the absence of chronic infections. In about 1500 necropsies in calves and cows on plant feeding, neither antemortem clinical AS symptoms nor postmortal macroscopical AS were

Early immunodiagnosis of fasciolosis in ruminants using recombinant Fasciola hepatica cathepsin L-like protease.

A diagnostic ELISA with recombinant Fasciola hepatica cathepsin L-like protease as antigen was developed to detect antibodies against F. hepatica in sheep and cattle. The recombinant cathepsin L-like protease was generated by functional expression of the cDNA from adult stage F. hepatica flukes in Saccharomyces cerevisiae. Specificity and sensitivity of the cathepsin L enzyme-linked immunosorbent assay (ELISA) was assessed using sera from sheep and calves experimentally or naturally mono-infected with F. hepatica and six-seven other parasites. The sensitivity of the cathepsin L ELISA for sheep

Protection of ruminants by Pasteurella haemolytica A1 capsular polysaccharide vaccines containing muramyl dipeptide analogs.

The capsular polysaccharide (CP) of Pasteurella haemolytica serotype A1 is a poor immunogen for the prevention of pneumonic pasteurellosis of ruminants. To improve CP immunogenicity, vaccines were prepared with 1.0 mg CP dose-1 with and without the synthetic adjuvant, muramyl dipeptide (MDP; range 0.2-1.0 mg) or a lipophilic derivative, MDP-sn-glyceryl-dipalmitoyl (MDP-GDP; range 0.1-1.0 mg). The optimum effective concentration of adjuvant was first determined in lambs and calves and then the efficacy of CP +0.5 mg MDP and CP +1.0 mg MDP-GDP was compared with that of two commercial vaccines in…

[Actinomyces pyogenes: conventional and Api system bacteriologic study of 103 strains isolated from ruminants].

One hundred and three strains of Actinomyces pyogenes isolated from ruminants were examined for morphological, cultural and biochemical properties by standard tests and by the Api 50 CH and Api Coryne methods. No biotype could be demonstrated, but a few atypical non- or practically non-proteolytic strains were detected which should be differentiated from Arcanobacterium haemolyticum. Criteria for laboratory identification of A pyogenes were established: Gram stain; culture on blood agar; deep agar and Loeffler's medium; catalase, nitrate reduction, acid formation from xylose; Hugh and Leifson …

Copper deficiency and posterior paralysis (Shalal) in small ruminants in the Sultanate of Oman.

A posterior ataxia or paralysis in goats and sheep is a syndrome known as Shalal within the Sultanate of Oman. An investigation was carried out into the etiology of the syndrome. Samples of blood and tissues were obtained from normal and Shalal-affected goats and sheep. Samples of feed were collected from the affected areas of the Sultanate. Chemical analyses of samples showed that affected animals suffered from severe copper deficiency in spite of sufficient copper contents in feed. However, concentrations of sulphur and iron in the feed were high. It was concluded that the Shalal syndrome in

More rumination, better cows: Exploring genetic parameters of daily rumination time in Holstein dairy cows around first calving.

Several studies have indicated that rumination could be used as a metric to monitor dairy cows' performance, condition, and health status. Along with applications in cow management, there may be scope to use rumination data in breeding programs. However, the genetics of rumination around first calving and the relationships with other key traits have not been studied. Therefore, the main objectives of this study were to estimate genetic parameters for daily rumination time (DRT) in Holstein Friesian cows around first calving and genetic correlations with 2 production traits, 3 reproduction trai

Influence of Dietary Forage Neutral Detergent Fiber on Ruminal Fermentation, Chewing Activity, Nutrient Digestion, and Ruminal Microbiota of Hu Sheep.

As the key components of dietary carbohydrates, ensuring a balance between forage-neutral detergent fiber (FNDF) and rumen-degradable starch (RDS) is essential for ruminant health. Eight male Hu sheep equipped with rumen cannulas were randomly divided into four groups based on dietary FNDF content: low FNDF (L-FNDF, 6.08%), middle low FNDF (ML-FNDF, 9.47%), middle high FNDF (MH-FNDF, 12.48%), and high FNDF (H-FNDF, 15.68%), while the RDS levels (15.65% of DM on average) were similar among the four groups. A replicated 4 × 4 Latin square design was employed in this study. The results indicated

Comparing rumination activity using the Nedap® rumination sensor against analysis of video recordings in pre-weaned dairy calves.

This Research Communication describes a pilot study to validate a rumination sensor for pre-weaned dairy calves. There is increasing interest in precision livestock farming (PLF) tools to capture behaviours and health parameters in farm animals. However, much of the research has focused on devices suitable for adult animals, and few devices have been validated for young animals. The aim of our pilot study was to validate the rumination estimates from Nedap rumination sensors® (NRS) when worn by dairy calves less than 2 months of age. Eight Norwegian Red dairy calves were raised in a cow-direct

Ruminal fermentation pattern of acidosis-induced cows fed either monensin or polyclonal antibodies preparation against several ruminal bacteria.

This study was designed to evaluate a spray-dried multivalent polyclonal antibody preparation (PAP) against lactate-producing bacteria as an alternative to monensin (MON) to control ruminal acidification. Holstein cows (677 ± 98 kg) fitted with ruminal cannulas were allocated in an incomplete Latin square design with two 20 days period. Cows were randomly assigned to control (CTL), PAP, or MON treatments. For each period, cows were fed a forage diet in the first 5 days (d-5 to d-1), composed of sugarcane, urea and a mineral supplement, followed by a 74% concentrate diet for 15 days (d 0 to d 1

Questionnaires based on natural language processing elicit immersive ruminative thinking in ruminators: Evidence from behavioral responses and EEG data.

Rumination is closely related to mental disorders and can thus be used as a marker of their presence or a predictor of their development. The presence of masking and fabrication in psychological selection can lead to inaccurate detection of psychological disorders. Human language is considered crucial in eliciting specific conscious activities, and the use of natural language processing (NLP) in the development of questionnaires for psychological tests has the potential to elicit immersive ruminative thinking, leading to changes in neural activity. Electroencephalography (EEG) is commonly used

A meta-analysis of the association between event-related rumination and posttraumatic growth: The Event-Related Rumination Inventory and the Posttraumatic Growth Inventory.

Trauma can disrupt an individual's core beliefs about themselves, others, and the world. Posttraumatic growth (PTG) is thought to be the outcome of a reconstruction process involving ruminative processing. This meta-analysis examined the strength of the associations between event-related intrusive and deliberate rumination and PTG. The moderating effects of variables including age, time since trauma exposure, and trauma type were examined. Eight databases were searched for English-language, peer reviewed studies examining the associations between PTG and types of event-related rumination in ad

Repeated ruminal acidosis challenges in lactating dairy cows at high and low risk for developing acidosis: feeding, ruminating, and lying behavior.

An experiment was conducted to determine whether the susceptibility to ruminal acidosis, as defined through differences in days in milk (DIM), milk production level, and ration composition, influences cow feeding, ruminating, and lying behavior and whether these behaviors change during an acute bout of ruminal acidosis. Eight ruminally cannulated cows were assigned to 1 of 2 acidosis risk levels: low risk (LR, mid-lactation cows fed a 60:40 forage:concentrate ratio diet) or high risk (HR, early lactation cows fed a 45:55 forage:concentrate diet). As a result, diets were intentionally confounde

Integrated effects of diet and probiotics on rumen microbiota and host physiology in ruminants.

Rumen microbiota is essential for nutrient digestion, immune function, and metabolic health in ruminants. With growing interest in sustainable animal production, recent studies have focused on the combined use of diet and probiotics in modulating rumen microbial community and its association with host performance. This review summarizes the effects of dietary strategies on microbial composition and fermentation efficiency. This review also discusses how probiotics such as Saccharomyces cerevisiae, Lactobacillus, Lacticaseibacillus, Lactiplantibacillus, and Bacillus spp. stabilize the rumen env…

Immunohistochemical characterization of nerve endings in the vaginal fornix and cervix of small ruminants.

This study assessed nerve endings in the vaginal fornix and the cervix of five Dorper ewes and five Saanen does, quantifying their distribution within epithelial and connective tissues, providing a preliminary assessment of the potential painful conditions they represent. Nerve endings were detected in all animals, with variations in their quantity among the organs and tissues analyzed. Overall, the presence and distribution of nerve endings suggest a sensory role, implicating a potential sensitivity to nociceptive stimuli in these organs.

Integrating Epidemiology, Immunology, and Host Genetics for Controlling Peste des Petits Ruminants in Goats.

Peste des petits ruminants (PPR) is a highly contagious transboundary viral disease of sheep and goats that causes major economic losses and livelihood disruption across Africa, the Middle East, and Asia. Goats are often more severely affected than sheep, particularly in endemic smallholder systems, where high turnover, nutritional stress, and limited veterinary infrastructure sustain viral transmission. Despite effective live attenuated vaccines and the ongoing FAO-WOAH Global Eradication Programme, PPR remains widely distributed, indicating that vaccination alone is insufficient without unde…
