Specificity and neutralizing capacity of antibodies elicited by a synthetic peptide of scorpion toxin.

Polyclonal antibodies raised against a synthetic peptide (sequence 50-59) of Androctonus australis Hector toxin II can neutralize the effects of toxin II in vivo. The antigenic specificities of anti-peptide and anti-toxin antibodies were compared by competitive aqueous phase radioimmunoassay by using 125I-toxin II, chemically modified or homologous toxins, and the synthetic peptide 50-59, either free or bound to bovine serum albumin (BSA). The antipeptide and anti-toxin antibodies had a comparable high affinity for the native toxin, but anti-peptide antibodies exhibited a lower binding capacit

Scorpion venom inhibits selectively Ca2+-activated K+ channels in situ.

Using patch-clamp techniques, a study was made of the component of Leiurus quinquestriatus scorpion venom which caused a blockade of one class of membrane potassium channels, the calcium activated potassium (BK) channels. This blockade was obtained on channels in their native lipidic environment and was specific for this class of channels as other types of potassium channels were not affected by this venom.

Polymorphism and quantitative variations of toxins in the venom of the scorpion Androctonus australis Hector.

Using highly specific radioimmunoassays for toxins I, II and III of the scorpion Androctonus australis Hector, the concentrations of these neurotoxins have been determined in pooled as well as in individual samples. Variations were found that support polymorphism of scorpion toxins at an individual level. Radioimmunoassays were also used to detect toxin I of Buthus occitanus tunetanus and toxin II of Androctonus australis Hector and also antigenically homologous toxins in the venoms of several North African scorpions. These results are interpreted in terms of scorpion serotherapy.

Toxicity to crustacea due to polypeptide-phospholipase interaction in the venom of a chactoid scorpion.

The toxic factors to isopods (crustacea) were isolated from the venom of the chactoid scorpion Scorpio maurus palmatus (Scorpionidae) by the aid of column chromatography, and their purity was assessed by disc electrophoresis, analytical ultracentrifugation, isoelectrofocusing, and amino acid analysis. The toxicity to isopods is attributed to two groups of components: (a) Low-molecular-weight basic polypeptides possessing about 3 and 8% of the crude venom lethality and paralytic potency to isopods, respectively. These components are characterized by very similar and unique amino acid compositio

[Disorders of neurohumoral regulation in rats exposed to the poison of the scorpion Buthus eupeus].

The intraperitoneal injection of Buthus eupeus to rats (2,5 mg/kg) results in increasing of catecholamines and cyclic nucleotides content in blood. The hyperinsulinemia is observed simultaneously with the development of hyperglycaemia. It is believed that the activation of the cyclic system is a key mechanism of the neuro-humoral disturbances of homeostasis in case of scorpion venom influence.

Teratogenicity in the rat of the venom from the scorpion Androctonus amoreuxi (Aud. & Sav.).

A. amoreuxi venom caused a high foetal resorption rate in rats, particularly when injected on days 9-11 of gestation. Vertebral and ossification defects and foetal weight loss were observed in many of the viable foetuses obtained from mothers treated with scorpion venom. Treatment of the rats with phentolamine in addition to the venom significantly reduced the venom-induced hyperglycemia. It also conferred some protection against foetal resorption but had only a slight effect on chondrification or foetal weight loss. This shows that hyperglycemia might be responsible for foetal mortality, but

Effects of a purified scorpion toxin (tityustoxin) on the isolated guinea pig heart.

To study the cardiac effects induced by purified scorpion toxin (tityustoxin, TsTX), without circulatory and respiratory influences, isolated guinea pig hearts were used. Single injections of 10, 20 or 30 micrograms TsTX induced positive inotropic and chronotropic effects in the majority of the experiments. The inotropic effects were dose-dependent. Simultaneous recording of the electrical activity of the heart showed sinus tachycardia, T wave inversion and ST segment deviations. Sinus bradycardia and/or A-V block were recorded in some experiments, simultaneously with the positive inotropic ef

[Kinetics of the reaction between scorpion toxin and the sodium channels of nodes of Ranvier].

The kinetics of the scorpion Buthus eupeus venom toxin binding to sodium channels at holding potential -120 mV and subsequent dissociation of the toxin-channel complex with the membrane potential shifts to -60 divided by +120 mV were studied under voltage clamp conditions in the Ranvier node membrane. The dissociation rate is shown to increase exponentially when shifting the membrane potential towards more positive values, with an e-fold increase for each 32.3 mV. The results are consistent with the suggestion that the dissociation of the toxin-channel complex is due to a difference in electri

Mechanism of the pulmonary edema induced by intravenous injection of scorpion toxin in the rat.

The intravenous injection of purified scorpion toxin (tityustoxin, TsTX) into unanesthetized rats induces a severe systemic hypertension followed by a hemorrhagic edema of the lungs. The edema is focal or diffuse, whereas the hemorrhage is always focal and less prominent than the edema. Anesthesia of the rats prevents the appearance of pulmonary edema. It seems likely that this protective action of the anesthesia is due, at least in part, to an interference with the hypertension induced by TsTX. The pulmonary edema is prevented by bilateral adrenalectomy, guanethidine or phenoxybenzamine. It i
