Platelet serotonin uptake in panic disorder patients: a replication study.

Platelet serotonin uptake was measured in 29 patients with DSM-III panic disorder or agoraphobia with panic attacks and compared to values obtained in 23 controls. Both the affinity constant (Km) and the maximal rate of uptake (Vmax) were determined in a buffered medium using 14C-serotonin. Patients and controls did not differ significantly with respect to age or Km values. A statistically significant difference was observed for Vmax (mean +/- SD = 65 +/- 22 pmol/10(8) platelet/min in patients vs. 44 +/- 13 pmol/10(8) platelets/min in controls). This finding suggests an overactivity of periphe…

Alpha-adrenergic control of serotonin release from rat pineal glands.

This study was designed to determine if norepinephrine (NE) stimulated release of pineal serotonin (5-HT) is receptor mediated and to identify the receptor(s) most influential in the release process. Efflux of 3H-5-HT from rat pineals in vitro was significantly increased within 5 min after NE was added to perifusion buffer in concentrations ranging from 0.1 to 3 microM. 3H-5-HT was not simply displaced from pinealocytes by NE, since buffer containing 3 microM 5-HT had no effect on 3H-5-HT efflux. Furthermore, NE enhanced 3H-5-HT secretion by a stereospecific process, since d-NE was significant…

Neural serotonin stimulates chloride transport in the rabbit corneal epithelium.

Evidence is presented that serotonin acts as a neurotransmitter in the cornea of the adult rabbit. Serotonin was localized to granules in a sparse population of subepithelial corneal nerves by an electron microscopic histochemical procedure. Significant endogenous levels of serotonin and its principal metabolite, 5-hydroxyindoleacetic acid, were detected in the central cornea by a fluorometric assay. Exogenous serotonin stimulated ion transport by corneal epithelium. This effect was potentiated by monoamine oxidase inhibition and was unaffected by an alpha-adrenergic receptor antagonist. Serot…

A selective serotonin reuptake inhibitor (SSRI) - induced case of catathrenia.

Catathrenia is currently classified as a sleep-related breathing disorder characterized by a deep inspiration followed by a prolonged, groaning expiration. While antidepressant- and antipsychotic-induced akathisia and restless legs syndrome (RLS) are well-documented, the potential for these medications to trigger catathrenia remains unrecognized. Selective serotonin reuptake inhibitors (SSRIs) are known to exacerbate or induce various sleep disturbances, including periodic leg movement disorder (PLMD), RLS, bruxism, and REM sleep without atonia. However, an association between SSRIs and catath…

Synchronous activation of striatal cholinergic interneurons induces local serotonin release.

Striatal cholinergic interneurons (CINs) can drive local dopamine release via nicotinic acetylcholine receptors (nAChRs) expressed on dopaminergic axons, but their role in modulating serotonin (5-HT) signaling is poorly understood. Here, we show that synchronous activation of CINs directly triggers local 5-HT release in the dorsal striatum via nAChRs expressed on serotonergic axons. This CIN-5-HT coupling is not detectable in the ventral striatum, despite its substantially denser serotonergic innervation. The nAChR-dependent release not only increases 5-HT levels in the dorsal striatum, but al…

Neonatal Outcomes Following Selective Serotonin Reuptake Inhibitor Use During Pregnancy.

IMPORTANCE: Evaluating the safety of selective serotonin reuptake inhibitors (SSRIs) during pregnancy is challenging due to ethical barriers to randomized trials and potential confounding in observational studies. OBJECTIVE: To compare neonatal outcomes for pregnancies with SSRI continuation vs those with SSRI discontinuation using a target trial framework. DESIGN, SETTING, AND PARTICIPANTS: This cohort study comprised target trials using electronic health records of linked mothers and neonates from a single academic center. The periconception trial examined congenital anomalies with first-t…

Clozapine-Associated Serotonin Syndrome: A Case Report and Literature Review.

Serotonin syndrome (SS) is a rare but potentially life-threatening condition resulting from increased serotoninergic activity in the central nervous system, either from exposure to a serotonin agonist or from withdrawal of a serotonin antagonist. Clozapine withdrawal has been implicated in cases of serotonin syndrome; however, clozapine exerts both agonist and antagonist properties at the serotonin receptor. We present the case of a 37-year-old male with unspecified psychosis and a structural neurological deficit who developed SS according to the Hunter Criteria while on a regimen of clozapine…

Fluoxetine Disrupts Ovarian Serotonin Signaling and Oocyte Competence in Mice.

Background: Selective serotonin reuptake inhibitors (SSRIs) are widely prescribed, yet their direct impact on ovarian function remains poorly understood. While serotonin signaling is known to occur within the ovarian follicle, the specific molecular consequences of its disruption by SSRIs are unclear. This study aimed to elucidate the direct, intra-ovarian mechanisms by which fluoxetine, a common SSRI, affects follicular development and oocyte competence. Methods: We administered fluoxetine (20 mg/kg) or vehicle daily for seven days to both prepubertal and adult female mice to model short-term…

Association of Umbilical Cord Blood Serotonin Levels with Neurodevelopmental Outcomes.

INTRODUCTION: Serotonin (5-hydroxytryptamine [5-HT]) plays an important role in early development, and fetal 5-HT has been reported to arise from placental and maternal sources. Previous human studies have established an association between maternal 5-HT levels and neurodevelopmental outcomes in populations with autism. In this study, we analyze umbilical cord blood and placental 5-HT levels at birth to further investigate the relationship of gestational 5-HT levels with birth outcomes and offspring cognitive development. METHODS: Participants were enrolled in the Safe Passage Study conducted…

Selective serotonin reuptake inhibitors enhance cancer immunosurveillance by pleiotropic mechanisms.

In a recent paper published in Cell, Li et al. suggest that selective serotonin reuptake inhibitors (SSRIs) antidepressants act on serotonin transporters on CD8+ T cells to enhance antitumor immunity. Beyond this mechanism, SSRIs can act on malignant cells, as well as on other immune cells, to improve cancer immunosurveillance.

Serotonin receptor 5-HT7 modulates inflammatory-associated functions of macrophages.

The hormone and neurotransmitter serotonin regulates numerous physiological functions within the central nervous system and in the periphery upon binding to specific receptors. In the periphery, the serotonin receptor 7 (5-HT7R) is expressed on different immune cells including monocytes and macrophages. To investigate the impact of 5-HT7R-mediated signaling on macrophage properties, we used human THP-1 cells and differentiated them into pro-inflammatory M1- and anti-inflammatory M2-like macrophages. Pharmacological 5-HT7R activation with the specific agonist LP-211 especially modulates morphol…

Context-dependent serotonin signaling links dietary quality to foraging decisions.

Animals sense their metabolic needs to guide adaptive behaviors partly through serotonin, a neurotransmitter associated with feeding in many species. Here we investigate the ability of the serotonin system to evaluate and interpret diverse diets by studying long-term foraging behaviors of the nematode C. elegans on bacteria. Behavioral screens on a genome-wide collection of E coli strains identified 22 metabolic mutants that induce behavioral aversion and stress responses in C. elegans. We show that different classes of serotonergic neurons promote aversion to non-preferred E. coli diets and r…

Baseline peripheral serotonin levels in two sympatric Podarcis lizard species.

Serotonin (5-hydroxytryptamine, 5HT) plays a crucial role in modulating physiological and behavioral traits in vertebrates, nonetheless little is known about its peripheral function in reptiles. Differences in central serotonin levels and behavior under serotonergic control have been observed in two sympatric lacertid lizard - Podarcis siculus and P. melisellensis. We compared baseline peripheral serotonin and tryptophan (Trp) levels, along with intestinal morphology, in these two species known to differ in behavior and invasiveness. Serum 5HT and Trp were measured using High-performance liqui…

Effects of selective serotonin reuptake inhibitors on endocrine system (Review).

Selective serotonin reuptake inhibitors (SSRIs) are typically prescribed for treating major depressive disorder (MDD) due to their high efficacy. These drugs function by inhibiting the reuptake of serotonin [also termed 5-hydroxytryptamine (5-HT)], which raises the levels of 5-HT in the synaptic cleft, leading to prolonged activation of postsynaptic 5-HT receptors. Despite the therapeutic benefits of SSRIs, this mechanism of action also disturbs the neuroendocrine response. Hypothalamic-pituitary-adrenal (HPA) axis activity is strongly linked to both MDD and the response to antidepressants, ow…

Serotonin Transporter Deficiency Induces Metabolic Alterations in the Ileal Mucosa.

Serotonin transporter (SERT) deficiency has been implicated in metabolic syndrome, intestinal inflammation, and microbial dysbiosis. Interestingly, changes in microbiome metabolic capacity and several alterations in host gene expression, including lipid metabolism, were previously observed in SERT-/- mice ileal mucosa. However, the precise host or microbial metabolites altered by SERT deficiency that may contribute to the pleiotropic phenotype of SERT KO mice are not yet understood. This study investigated the hypothesis that SERT deficiency impacts lipid and microbial metabolite abundances in…
