Integrating in vitro chemical transplacental passage into a generic PBK model: A QIVIVE approach.

With the increasing application of cell culture models as primary tools for predicting chemical safety, the quantitative extrapolation of the effective dose from in vitro to in vivo (QIVIVE) is of increasing importance. For developmental toxicity this requires scaling the in vitro observed dose-response characteristics to in vivo fetal exposure, while integrating maternal in vivo kinetics during pregnancy, in particular transplacental transfer. Here the transfer of substances across the placental barrier, has been studied using the in vitro BeWo cell assay and six embryotoxic compounds of diff

Biokinetics and effects of barium sulfate nanoparticles.

BACKGROUND: Nanoparticulate barium sulfate has potential novel applications and wide use in the polymer and paint industries. A short-term inhalation study on barium sulfate nanoparticles (BaSO₄ NPs) was previously published [Part Fibre Toxicol 11:16, 2014]. We performed comprehensive biokinetic studies of ¹³¹BaSO₄ NPs administered via different routes and of acute and subchronic pulmonary responses to instilled or inhaled BaSO₄ in rats. METHODS: We compared the tissue distribution of ¹³¹Ba over 28 days after intratracheal (IT) instillation, and over 7 days after gavage and intravenous (IV) i

Inorganic mercury poisoning associated with skin-lightening cosmetic products.

INTRODUCTION: Mercury and mercury salts, including mercurous chloride and mercurous oxide, are prohibited for use in cosmetic products as sk … rption as the subject headings. These searches identified 118 citations of which 31 were relevant. TOXICOKINETICS: The rate of dermal absorption increases with the concentration of mercury and prior hydration of the skin. The degree of dermal absorption varies with the skin integrity and lipid solubility of the vehicle in the cosmetic products. Ingestion may oc …

Critical period for a teratogenic VLA-4 antagonist: Developmental effects and comparison of embryo drug concentrations of teratogenic and non-teratogenic VLA-4 antagonists.

BACKGROUND: Integrins such as VLA-4 (Very late antigen 4, integrin alpha4beta1) play key roles in cell-cell interactions that are critical for development. Homozygous null knockouts of the VLA-4 alpha4-subunit or VCAM-1 (VLA-4 cell surface ligand) in mice result in failure of the allantois and chorion to fuse leading to interrupted placentation and cardiac development and embryo lethality. Embryo-fetal studies of three VLA-4 antagonists, IVL745, IVL984, and HMR1031 [Crofts et al., Birth Defects Res B 71:55-68 (this issue), 2004] with exposure on gestation days (GD) 6-17 (rat), 6-18 (rabbit) or

L-citrulline: A preclinical safety biomarker for the small intestine in rats and dogs in repeat dose toxicity studies.

INTRODUCTION: Gastrointestinal (GI) toxicity is still an issue within drug development, especially for novel oncology drugs. The identification of GI mucosal damage at an early stage with high sensitivity and specificity across preclinical species and humans remains difficult. To date, in preclinical studies, no qualified mechanistic, diagnostic or prognostic biomarkers exist for GI mucosal toxicity. L-citrulline is one of the most promising biomarker candidates used in clinical settings to quantify enterocyte integrity in various small intestinal diseases. L-citrulline is an intermediate meta

Their control is exceedingly difficult due to their mu … promutagenic lesions in different organs are the composite result of several factors, including (a) toxicokinetics (first-pass effect); (b) local and distant metabolism; (c) efficiency and fidelity of DNA repair; and (d) cell proliferation rate. Use of the molecular adsorbent recirculating system (MARS™) for the management of acute poisoning with or without liver failure.

INTRODUCTION: There is an increasing interest in recent developments in bioartificial and non-bioartificial devices, so called extracorporeal liver assist devices, which are now used widely not only to increase drug elimination, but also to enhance the removal of endogenous substances in acute liver failure. Most of the non-bioartificial techniques are based on the principle of albumin dialysis. The objective is to remove albumin-bound substances that could play a role in the pathophysiology of acute liver failure by dialysing blood against an albumin-containing solution across a high flux per

The reproductive and developmental toxicity of the antifungal drug Nyotran (liposomal nystatin) in rats and rabbits.

Nyotran is a liposomally encapsulated i.v. formulation of the antifungal polyene nystatin. This drug was evaluated in a series of reproductive toxicity studies, according to the guidelines outlined by the International Conference on Harmonization (ICH). A fertility and early embryonic development study (SEG I) and a prenatal and postnatal development (SEG III) study were conducted in rats, and embryo-fetal development (SEG II) studies were conducted in rats and rabbits. Nyotran was administered iv in all studies. In SEG I and SEG III, rats were administered daily doses of 0.5, 1.5, or 3.0 mg/k

Impacts of Uremic Toxins on the Population Pharmacokinetics of Total Mycophenolic Acid and its Glucuronide Metabolite in Adult Kidney Transplant Recipients.

BACKGROUND AND OBJECTIVE: Mycophenolic acid (MPA) exhibits considerable inter-individual variability in drug exposure, which can result in acute graft rejection as well as hematological or infectious adverse effects. Evidence from our group and others indicates that certain uremic toxins may contribute to this variability through pharmacokinetic (PK) interactions. This study aimed to develop a novel population PK (popPK) model to investigate how conjugated metabolites of p-cresol and indole (i.e., toxicokinetically important uremic toxins) affect total MPA PK, and to conduct model-based simula

Approaches to the treatment of nerve agent poisoning with oximes - from experimental studies to the intensive care unit.

INTRODUCTION: Organophosphorus poisoning is still a continuing threat to military forces and the civilian health sector. The therapeutic value of clinically used oxime antidotes is frequently a matter of controversy. In this narrative review, we will focus on nerve agent poisoning in particular and show how experimental results can be integrated into the therapy of patients. METHODS: This narrative review is based primarily on the extensive studies conducted by the authors over many years, supplemented by appropriate literature to provide the reader with currently available knowledge of the e

NTP Toxicology and Carcinogenesis Studies of Methyleugenol (CAS NO. 93-15-2) in F344/N Rats and B6C3F1 Mice (Gavage Studies).

Methyleugenol is used as a flavoring agent in jellies, baked goods, nonalcoholic beverages, chewing gum, candy, pudding, relish, and ice cream. It is also used as a fragrance in perfumes, creams, lotions, detergents, and soaps. Methyleugenol has also been used as an insect attractant in eradication programs and as an anesthetic in rodents. Methyleugenol was nominated for testing because of its widespread use and because of its structural resemblance to safrole, a known carcinogen, and isosafrole and estragole. Male and female F344/N rats and B6C3F1 mice received methyleugenol (approximately 99

Nonclinical Safety Assessment of Digadoglucitol, a Novel Magnetic Resonance Imaging Contrast Agent for the Central Nervous System.

OBJECTIVE: Digadoglucitol is a new macrocyclic gadolinium-based contrast agent for magnetic resonance imaging, intended to be used in central nervous system (CNS) and whole body clinical indications. The product features a unique dimeric structure with Gd(III) complexed by 2 macrocyclic chelators, setting it apart from other macrocyclic gadolinium-based contrast agents on the market. The safety profile of digadoglucitol in the context of single diagnostic use in humans was examined using a range of nonclinical studies. MATERIALS AND METHODS: All safety pharmacology and toxicology studies, exc
