Signal transduction pathways: new targets for treating rheumatoid arthritis.

Biotherapies and other new treatments introduced over the last few years have considerably enriched the therapeutic armamentarium for rheumatoid arthritis. Nevertheless, primary refractoriness or secondary escape phenomenon may occur, indicating a need for identifying new treatment targets. Promising candidates can be found among compounds involved in signal transduction pathways, most notably protein kinases (mitogen-activated protein kinase, MAPK and phosphatidylinositol-3 protein kinase, PI3) and transcription factors (nuclear factor kappa B, NF-kappaB; activating protein 1, AP-1; CCAAT/enh

Multi-Omics Analysis Uncovers the Salicylic Acid and Ethylene-Responsive Transcription Factor-Mediated Defence Mechanism of Panax notoginseng to Panax notoginseng Virus Y.

Panax notoginseng is an edible traditional medicinal plant, but it is highly susceptible to Panax notoginseng virus Y (PnVY). This study revealed that salicylic acid (SA) treatment decreases this susceptibility. The underlying molecular mechanism was elucidated by integrating physiological, multi-omics and genetic approaches. SA application alleviated PnVY-induced photosynthetic inhibition and activated antioxidant enzymes, thereby decreasing reactive oxygen species levels and maintaining redox homeostasis. Upon PnVY inoculation, SA triggered a range of defence responses. The exogenous applica

Knockout of OsSAP5 Improves Resistance to Pre-harvest Sprouting by Modulating OsSLR1 Protein Abundance in Rice.

Pre-harvest sprouting (PHS), defined as the premature germination of grains on the panicle before harvest, significantly compromises grain yield and seed quality. Optimizing seed dormancy is therefore crucial for mitigating PHS. Here, we report that loss of function in Stress-associated Protein 5 (OsSAP5) enhances resistance to PHS without grain yield reduction. Mechanistically, OsSAP5 participates in the GA-mediated modulation of the DELLA protein OsSLR1 abundance to orchestrate downstream responses; it accelerates GA signaling transduction to induce amylase gene expression and elevate solubl

CFTR gene delivery to human airway epithelia using parainfluenza virus 5 amplifying virus-like particles.

Parainfluenza virus 5 (PIV5)-derived amplifying virus-like particles (PIV5-AVLPs) are an efficient gene delivery platform with broad cell tropism. To investigate the feasibility of using PIV5-AVLPs for cystic fibrosis (CF) gene therapy, we generated AVLPs expressing enhanced green fluorescent protein (AVLP-eGFP) or a codon-optimized human cystic fibrosis transmembrane conductance regulator (CFTR) coding sequence and an mCherry reporter (AVLP-CFTR). We examined the transduction efficiency and persistence of transgene expression of AVLP-eGFP in primary cultures of non-CF and CF human airway epit

Optimization of systemic AAV9 gene therapy in Niemann-Pick disease, type C1 mice.

Niemann-Pick disease, type C1 (NPC1), is a rare, fatal neurodegenerative disorder caused by pathological variants in NPC1, which encodes a lysosomal cholesterol transporter. FDA-approved treatments are limited and do not target the underlying genetic defect. Both systemic and central nervous system delivery of AAV9-hNPC1 have shown significant disease amelioration in NPC1 murine models. To assess the impact of dose in null Npc1 m1N/m1N mice, we systemically administered three different doses of AAV9-hNPC1 at 4 wk old. Then, to assess the impact of age, we administered the medium dose before ph

Nephronophthisis: Current clinical spectrum and molecular pathogenesis.

Nephronophthisis (NPH) is a ciliopathy primarily affecting renal tubules and interstitial tissue, ultimately progressing to end-stage kidney disease (ESKD). The clinical spectrum of NPH is broad and can be classified according to the age at onset into infantile, juvenile, adolescent, and late-onset forms. Histopathologically, NPH is characterized by corticomedullary cysts, tubular atrophy, interstitial fibrosis, and cystic dilatation of distal tubules. The kidneys may appear normal in early stages but gradually shrink with disease progression. More than 20 causative genes have been identified,

St. John's Wort for Depression: From Neurotransmitters to Membrane Plasticity.

Depression is a multifactorial disorder shaped by genetic, psychosocial, and biological influences, with hypotheses ranging from monoamine deficiency and neuroplasticity deficits to inflammation and stress-induced dysregulation. St. John's wort (Hypericum perforatum L.) has long been used as an herbal antidepressant and is supported by clinical evidence for efficacy and safety in mild-to-moderate depression. While its multimodal mechanisms have been linked to neurotransmitter reuptake inhibition, neuroendocrine regulation, and modulation of neuroplasticity, recent findings suggest an additiona

Rapid cochlear gene therapy in adult deaf mice: Vglut3 rescue via AAV8 achieves day-1 hearing restoration.

Genetic hearing loss, caused by mutations in critical auditory genes, has seen promising advances through gene therapy, yet the temporal dynamics of early-stage auditory functional recovery and therapeutic transgene expression patterns following intervention remain uncharacterized in preclinical deafness models. This study systematically investigates the post-treatment progression of cochlear functional restoration and spatially resolved transgene expression kinetics in adult Vglut3 knockout (Vglut3 KO ) mice following adeno-associated virus (AAV)-mediated inner ear gene therapy. Vegfa signaling ameliorates cardiac fibrosis and restores endothelial regeneration in a point-mutated zebrafish model of pseudoxanthoma elasticum.

The ABCC6 gene is associated with pseudoxanthoma elasticum (PXE), a genetic disorder characterized by ectopic calcification in soft tissues, notably affecting the cardiovascular system. However, the underlying cardiovascular pathological mechanisms remain elusive and effective treatments are currently unavailable. Here, we employed a point-mutated zebrafish model harboring the abcc6aR1463C/R1463C mutation (orthologous to the human ABCC6R1459C variant) to investigate the cardiac homeostasis and regeneration. We find that abcc6a deficiency leads to enhanced TGF-β/Smad3 signaling and vegfaa overe

[Research progress in Runt-related transcription factor 2 regulation of bone remodeling and tooth eruption].

Cleidocranial dysplasia, a rare genetic disorder primarily caused by Runt-related transcription factor 2 (RUNX2) heterozygous mutation, serves as a representative model for investigating regulatory mechanisms of RUNX2 in bone remodeling and tooth eruption. As a master transcription factor governing mineralized tissue development, RUNX2 orchestrates bone remodeling and tooth eruption through diverse regulatory networks. It drives alveolar bone formation via transcriptional activation, integration of multiple signaling cascades, and epigenetic modifications, thereby generating the biomechanical

The Leukemic Isocitrate Dehydrogenase (IDH) 1/2 Mutations Impair Myeloid and Erythroid Cell Differentiation of Primary Human Hematopoietic Stem and Progenitor Cells (HSPCs).

How hematopoietic stem and progenitor cell (HSPC) fate decisions are affected by genetic alterations acquired during AML leukemogenesis is poorly understood and mainly explored in animal models. Here, we study isocitrate dehydrogenase (IDH) gene mutations in the human model of HSPC and discuss the available literature on this topic. IDH1/2 mutations occur in ~20% of AML cases, are recognized among the mutations earliest acquired during leukemogenesis, and are targets of specific inhibitors (ivosidenib and enasidenib, respectively). In order to investigate the direct effects of these mutations

Designing and optimizing AAV-mediated gene therapy for neurodegenerative diseases: from bench to bedside.

Recombinant adeno-associated viruses (rAAVs) have emerged as an attractive tool for gene delivery, and demonstrated tremendous promise in gene therapy and gene editing-therapeutic modalities with potential "one-and-done" treatment benefits compared to conventional drugs. Given their tropisms for the central nervous system (CNS) across various species including humans, rAAVs have been extensively investigated in both pre-clinical and clinical studies targeting neurodegenerative disease. However, major challenges remain in the application of rAAVs for CNS gene therapy, such as suboptimal vector

People with higher systemizing traits have wider right hands.

INTRODUCTION: Various genetic mutations have been implicated in autism spectrum disorder (ASD). Some candidate genes for ASD are known to be related to signal transduction and may be involved in hand development as well as neurodevelopment. Therefore, although subtle, anatomical variations in hand configurations may be observed in individuals with ASD. However, except for research on the finger ratio, which has been suggested to be related to prenatal sex hormone exposure, only few studies have been conducted. Given the spectrum characteristics of ASD, we explored whether hand configurations a

HAX1-Overexpression Augments Cardioprotective Efficacy of Stem Cell-Based Therapy Through Mediating Hippo-Yap Signaling.

Although stem/progenitor cell therapy shows potential for myocardial infarction repair, enhancing the therapeutic efficacy could be achieved through additional genetic modifications. HCLS1-associated protein X-1 (HAX1) has been identified as a versatile modulator responsible for cardio-protective signaling, while its role in regulating stem cell survival and functionality remains unknown. In this study, we investigated whether HAX1 can augment the protective potential of Sca1+ cardiac stromal cells (CSCs) for myocardial injury. The overexpression of HAX1 significantly increased cell proliferat

Molecular Engineering of Virus Tropism.

Engineered viral vectors designed to deliver genetic material to specific targets offer significant potential for disease treatment, safer vaccine development, and the creation of novel biochemical research tools. Viral tropism, the specificity of a virus for infecting a particular host, is often modified in recombinant viruses to achieve precise delivery, minimize off-target effects, enhance transduction efficiency, and improve safety. Key factors influencing tropism include surface protein interactions between the virus and host-cell, the availability of host-cell machinery for viral replica

β-arrestin2 mediates the hippocampal dopaminergic system in autistic mouse through the ERK signaling pathway.

Autism is a complex neurodevelopmental disease that may be caused by genetic and environmental factors, that are incompletely understood. Overactivation of dopaminergic receptors can lead to autistic-like behavior. β-arrestin2 (Arrb2) is a scaffolding protein of the arrestin family, which function as cytosolic multifunctional adapter proteins that activate cell signal transduction and mediate the signal termination and endocytosis of G-protein-coupled receptors (GPCRs) complexes. In this study, we established an Arrb2 knockout (Arrb2-/-) mouse to explore the biological function of Arrb2 in aut

Gene Replacement Therapies for Genodermatoses: A Status Quo.

Epidermolysis bullosa (EB) is a genodermatosis, characterized by the formation of extended blisters and lesions on the skin and mucous membranes upon minimal mechanical trauma. The disease is caused by mutations in genes encoding proteins that are essential for skin stability. Functional impairment, reduction, or absence of one of these proteins results in skin fragility due to reduced connectivity between dermis and epidermis. Currently, gene therapy represents the only treatment option with the potential to cure this severe blistering skin disease. Two promising forms of gene therapy are pot

The progress of AAV-mediated gene therapy in neuromuscular disorders.

INTRODUCTION: The well-defined genetic causes and monogenetic nature of many neuromuscular disorders, including Duchenne muscular dystrophy (DMD) and spinal muscular atrophy (SMA), present gene therapy as a prominent therapeutic approach. The novel variants of adeno-associated virus (AAV) can achieve satisfactory transduction efficiency of exogenous genes through the central nervous system and body-wide in skeletal muscle. AREAS COVERED: In this review, we summarize the strategies of AAV gene therapy that are currently under preclinical and clinical evaluation for the treatment of degenerativ

PI3K/mTOR mediate mitogen-dependent HDAC1 phosphorylation in breast cancer: a novel regulation of estrogen receptor expression.

Histone deacetylase 1 (HDAC1) is an important epigenetic controller involved in transcriptional regulation through modification of chromatin structure. Genetic and epigenetic changes and deregulation of signal transduction pathways have been implicated in the development of breast cancer. Downregulation of estrogen receptor α (ERα) expression is one of the mechanisms behind the acquisition of endocrine resistance. Sustained and increased hormone and growth factor receptor signaling in breast cancer cells contribute to resistance to endocrine therapy. Both HDACs and the PI3K/mTOR signaling path

Regulation of YAP by mTOR and autophagy reveals a therapeutic target of tuberous sclerosis complex.

Genetic studies have shown that the tuberous sclerosis complex (TSC) 1-TSC2-mammalian target of Rapamycin (mTOR) and the Hippo-Yes-associated protein 1 (YAP) pathways are master regulators of organ size, which are often involved in tumorigenesis. The crosstalk between these signal transduction pathways in coordinating environmental cues, such as nutritional status and mechanical constraints, is crucial for tissue growth. Whether and how mTOR regulates YAP remains elusive. Here we describe a novel mouse model of TSC which develops renal mesenchymal lesions recapitulating human perivascular epit
