The role of mammalian skin in harbouring and transmitting arthropod-borne protozoan parasites has been overlooked for decades as these pathogens have been regarded primarily as blood-dwelling organisms. Intriguingly, infections with low or undetected blood parasites are common, particularly in the case of Human African Trypanosomiasis caused by Trypanosoma brucei gambiense. We hypothesise, therefore, the skin represents an anatomic reservoir of infection. Here we definitively show that substantial quantities of trypanosomes exist within the skin following experimental infection, which can be t

A cross-sectional study was conducted in Mbarara district, south-western Uganda in May 2012 to determine the burden of African animal trypanosomosis (AAT) in the semi-intensive dairy production systems where pyrethroid acaricides are frequently used in the control of tick-borne diseases (TBDs). A total of 295 cattle blood samples were taken and analysed using a single pair of primers previously designed to amplify internal transcribed spacer (ITS1) of trypanosome ribosomal deoxyribonucleic acid (rDNA). A structured questionnaire was administered to 55 participating livestock farmers to generat

We conducted a situation analysis of human African trypanosomiasis (HAT) in Zambia from January 2000 to April 2007. The aim of this survey was to identify districts in Zambia that were still recording cases of HAT. Three districts namely, Mpika, Chama, and Chipata were found to be still reporting cases of HAT and thus lay in HAT transmission foci in North Eastern Zambia. During the period under review, 24 cases of HAT were reported from these three districts. We thereafter reviewed literature on the occurrence of HAT in Zambia from the early 1960s to mid 1990s. This revealed that HAT transmiss

BACKGROUND: Glossina palpalis palpalis (Diptera: Glossinidae) is widespread in west Africa, and is the main vector of sleeping sickness in Cameroon as well as in the Bas Congo Province of the Democratic Republic of Congo. However, little is known on the structure of its populations. We investigated G. p. palpalis population genetic structure in five sleeping sickness foci (four in Cameroon, one in Democratic Republic of Congo) using eight microsatellite DNA markers.
RESULTS: A strong isolation by distance explains most of the population structure observed in our sampling sites of Cameroon and

Host and vector distribution of Trypanosoma brucei gambiense was studied in relation to habitat types and seasons. Six (19.35%) of the 31 mammal species recorded in Bipindi were reservoir hosts. Cercopithecus nictitans was confined to the undisturbed forest and the low intensive shifting cultivation zones, while Cephalophus monticola, Cephalophus dorsalis, Cricetomys gambianus, Atherurus africanus and Nandinia binotata occurred in all the habitat types. As for vectors of human African trypanosomiasis (HAT), Glossina palpalis palpalis, was the most abundant (99.13%) among tsetse fly species. It

To obtain updated data on and assess the contribution of trypanosomosis to the disease burden of cattle kept at the edge of the Hluhluwe-iMfolozi Park, a survey was conducted at Mvutshini Dip. Use was made of a purposeful sampling strategy by restricting sampling to animals that the livestock owner considered to be in poor condition. Of a total of 76 blood samples collected, 26 were parasitologically positive and 46 were positive on PCR/RFLP. Almost all infections were due to Trypanosoma congolense savannah subgroup. A total of 63 animals had a PCV < or = 24% and were considered to be anaemic.

An in vitro method has been established to obtain metacyclic form populations of Trypanosoma brucei brucei. Trypanosome populations containing more than 98% of metacyclic forms were obtained from cultures which were: 1) initiated with bloodstream forms in primary cultures in the presence of Microtus montanus embryonic fibroblast-like cells (feeder cell layers); 2) maintained in glucose-free Eagle's minimum essential medium supplemented with 10 mM L-proline, 2 mM L-glutamine and 20% (v/v) fetal bovine serum at 27 degrees C without medium change for five days; 3) subcultured in the absence of th

The source of blood meals from 174 fed Glossina morsitans submorsitans Newstead, captured in malaise traps in Savanna woodland in The Gambia, were identified by the precipitin test. Warthog accounted for 90% of the meals and single bushbuck and ox feeds were identified. Nagana is a major problem in the area, but contact between tsetse and livestock is reduced by restricted grazing. In this situation, warthog, with a ubiquitous distribution, appear to be major maintenance hosts for G. m. submorsitans as well as a potential reservoir of trypanosomiasis.

This study builds upon earlier work mapping the potential benefits from bovine trypanosomosis control and analysing the costs of different approaches. Updated costs were derived for five intervention techniques: trypanocides, targets, insecticide-treated cattle, aerial spraying and the release of sterile males. Two strategies were considered: continuous control and elimination. For mapping the costs, cattle densities, environmental constraints, and the presence of savannah or riverine tsetse species were taken into account. These were combined with maps of potential benefits to produce maps of

African trypanosomes, parasites that cause human sleeping sickness, undergo a density-dependent differentiation in the bloodstream of their mammalian hosts. This process is driven by a released parasite-derived factor that causes parasites to accumulate in G1 and become quiescent. This is accompanied by morphological transformation to 'stumpy' forms that are adapted to survival and further development when taken up in the blood meal of tsetse flies, the vector for trypanosomiasis. Although the soluble signal driving differentiation to stumpy forms is unidentified, a recent genome-wide RNAi scr…

Control of major livestock diseases in the tropics, such as theileriosis or trypanosomosis, is still largely based on the control of their vectors. Understanding the distribution of vectors, such as ticks and tsetse flies, is needed in order to improve the efficiency and economical viability of control operations. Technical improvements such as remote sensing and global information systems have allowed valuable improvements for the prediction of large-scale vector distribution (continental to national), but trying to make these predictions at the scale of a landscape is facing other challenges…

Elimination of sleeping sickness from endemic countries like Uganda is key if the affected communities are to exploit the potential of the available human and livestock resources (production and productivity). Trypanosoma brucei rhodesiense, the parasite that causes acute sleeping sickness in humans, is transmitted by tsetse flies and co-exists in non-human animal reservoirs. Uganda by Act of Parliament in 1992 decided to handle the complex approach to control of sleeping sickness and animal trypanosomiasis by establishing the Uganda Trypanosomiasis Control Council (UTCC) and its secretariat t…

African trypanosomosis is a major threat to livestock production in sub-Saharan Africa. Although the disease mainly concerns cattle, dogs can also be infected by Trypanosoma spp. transmitted by tsetse flies. Between 1997 and 2003, the parasite Trypanosoma congolense was identified in French military dogs sent to Africa. On infected dogs, the diagnosis was made during the mission or just after the return to France, depending on when the symptoms appeared. The high incidence and mortality rate among these dogs led veterinarians of the French Health Service to implement a systematic chemoprophyla…

Human African trypanosomiasis (HAT) is caused by sub-species of the parasitic protozoan Trypanosoma brucei and is transmitted by tsetse flies, both of which are endemic only to sub-Saharan Africa. Several cases have been reported in non-endemic areas, such as North America and Europe, due to travelers, ex-patriots or military personnel returning from abroad or due to immigrants from endemic areas. In this paper, non-endemic cases reported over the past 20 years are reviewed; a total of 68 cases are reported, 19 cases of Trypanosoma brucei gambiense HAT and 49 cases of Trypanosoma brucei rhodes…

The epidemiology of vector-borne diseases is complex due to the variability in the ecology of the different actors involved, i.e. hosts, parasites and vectors. The transmission of African animal trypanosomosis in the West-African savannah region is an excellent example of this complexity: riverine tsetse flies have an heterogeneous distribution along the rivers, depending of suitable habitats, and transmit pathogenic trypanosomes were they use domestic animal as feeding hosts. Contrasting epidemiological situations may thus occur at the local scale, and a broad view of the overall environment …
