Domain Knowledge Constrained Symbolic Regression for Optimising Dermal Drug Formulations.

PURPOSE: Interpretable dynamic models that remain physically admissible under extrapolation are essential for dermal formulation optimisation. Mechanistic models are often labour-intensive to derive, whereas unconstrained data-driven models may violate physical constraints and degrade beyond observed conditions. METHODS: We developed a domain knowledge constrained symbolic regression (DKC-SR) framework to discover compact, interpretable dynamic models for cumulative release directly from time-series experiments. Domain knowledge was embedded through a restricted operator set and feasibility c

A review on microneedle patch as a delivery system for proteins/peptides and their applications in transdermal inflammation suppression.

Transdermal delivery is one of the most recent modes of administration studied due to several shortfalls observed for intra-venous, and oral drug administrations. Among, microneedle-based transdermal delivery is the popular choice due to non-invasive procedure and minimal toxicological effects. Microneedle devices consist of micron scaled needle patch entrapped with the target specific drug molecules. Due to body's immune response and occasional pathogen attack, various inflammatory diseases are developed such as psoriasis, dermatitis, rashes, rheumatoid arthritis, gouty arthritis, and fibrosi

Study on the synthesis, physicochemical properties and transdermal absorption of scutellarein prodrugs.

In this research, we aimed to improve the lipid solubility and transdermal absorbability of scutellarein through a prodrug strategy, and then improve its efficacy for treating psoriasis. Firstly, we designed and synthesized 19 prodrugs of scutellarein, including AC (alkylcarbonyl), AAC (alkylaminocarbonyl) and AOC (alkyloxycarbonyl) series. Then, we selected three prodrugs (AC-4, AAC-2 and AOC-SEC-4) with proper log P and favorable solubility for subsequent transdermal absorption study, and the results demonstrated the three prodrugs significantly improved the transdermal flux and the drug ret

A review on recent advances and challenges of microneedle technology for enhanced topical treatment of skin disorders.

Microneedles are tiny, minimally invasive needles that are made to create microchannels over a subcutaneous layer of the skin. These micro-injuries encourage the secretion of growth factors and prompt collagen synthesis. The reasons for using microneedling therapy have increased significantly, leading to its widespread adoption in dermatology. This review article discussed recent advances and challenges of microneedle technology for enhanced topical treatment of skin disorders. It discussed the effective and safe use of microneedles for treating different types of skin conditions like acne, sc

Finite Dose In Vitro Permeation Testing: Significance of Occluding the Donor compartment, a Case study.

The evaporation of the solvent induces significant changes in formulation, directly impacting its performance. The performance of topical products is determined by the interplay between their inherent quality attributes and the transformations that occur due to solvent evaporation when applied to the skin in clinically relevant doses. To accurately assess, in vivo performance, it is advisable to apply smaller doses to the skin and keep the donor compartment open to enable evaporation of solvents while carrying out in vitro permeation tests. This manuscript highlights the critical role of solve

Cutaneous pharmacokinetics of a volatile drug post-application to the skin.

This research explored whether in vitro release and skin permeation tests, combined with in vitro and in vivo stratum corneum (SC) sampling, can quantify the "input rate" of a volatile drug into and through the skin. Two topical methyl salicylate (MeSA) products were studied. Drug release from the formulations across a silicone membrane was similar, with ~ 65% of the drug load being released in 6 h. In vitro porcine skin permeation tests showed that ~ 50% of the dose crossed the skin in 24 h from both products. In SC sampling experiments, the mass of MeSA in the SC at one 'uptake' and three 'c

Polymeric microneedle advancements in macromolecule drug delivery: current trends, challenges, and future perspectives.

Microneedles (MNs) offer a transformative solution for delivering macromolecules, including proteins, RNA, and peptides. These are critical in treating complex diseases but face significant challenges such as immunogenicity, poor stability, high molecular weight, and delivery efficiency. Unlike conventional methods, MNs efficiently bypass biological barriers like the stratum corneum, enabling precise and minimally invasive transdermal drug delivery. This review explores various MN types such as solid, coated, hollow, hydrogel-forming, and dissolving and their therapeutic applications in cancer

Research progress of biomaterial-based hydrogels in the treatment of psoriasis.

As a key barrier in psoriatic skin, the thickened stratum corneum limits the efficacy of topical therapies, prompting the development of biomaterial-based drug delivery systems. Although systems such as microneedles, nanofibers, and nanoparticles enhance transdermal delivery, single-component approaches often face limitations like restricted drug-loading capacity, uncontrolled release, and poor mechanical properties. Hydrogels, despite their extracellular matrix-mimicking structure, high biocompatibility, and tunable properties, are limited by weak hydrophobic drug encapsulation and insufficie

Minoxidil Nanosuspension-Loaded Dissolved Microneedles for Hair Regrowth.

Minoxidil (MIN) is used topically to treat alopecia. However, its low absorption limits its use, warranting a new strategy to enhance its delivery into skin layers. The objective of this study was to evaluate the dermal delivery of MIN by utilizing dissolved microneedles (MNs) loaded with MIN nanosuspension (MIN-NS) for hair regrowth. MIN-NS was prepared by the solvent-antisolvent precipitation technique. The particle size of MIN-NS was 226.7 ± 9.3 nm with a polydispersity index of 0.29 ± 0.17 and a zeta potential of -29.97 ± 1.23 mV. An optimized formulation of MIN-NS was selected, freeze-dri

Rational development of topical climbazole formulations.

Dandruff, or pityriasis capitis simplex, is a common scalp condition associated with excessive flaking and scaling of the epidermal tissue. Other features include irregular corneocyte turnover, irritation, itching and an impaired skin barrier function. Previously we reported the characterization of climbazole (CBZ), an antifungal agent used in the management of dandruff. Skin permeation of CBZ from neat solvents was also investigated. In the present work we evaluated CBZ permeation in human skin in vitro from more complex formulations that better represent products used by consumers. The vario

Microneedle transdermal drug delivery as a candidate for the treatment of gouty arthritis: Material structure, design strategies and prospects.

Gouty arthritis (GA) is caused by monosodium urate (MSU) crystals deposition. GA is difficult to cure because of its complex disease mechanism and the tendency to reoccur. GA patients require long-term uric acid-lowering and anti-inflammatory treatments. In the past ten years, as a painless, convenient and well-tolerated new drug transdermal delivery method, microneedles (MNs) administration has been continuously developed, which can realize various drug release modes to deal with various complex diseases. Compared with the traditional administration methods (oral and injection), MNs are more

BACKGROUND: When administered transdermally, desonide is ineffective due to its poor solubility. As a new transdermal delivery system, nanoemulsion gel has demonstrated significant advantages for drug delivery over conventional formulations. We have established desonide nanoemulsion gel (DES NE gel) for better transdermal absorption, but its efficacy and safety still need to be evaluated. This study aims to provide additional evidence demonstrating the improved pharmacodynamics and safety of transdermal delivery of Desonide via nanoemulsion gel. METHODS: Pharmacodynamics and safety of Desonid

Resident cutaneous memory T cells: a clinical review of their role in chronic inflammatory dermatoses and potential as therapeutic targets.

Resident memory T cells (T-RMs) remain in epithelial barrier tissues after antigen exposure and the initial effector phase. These T-RMs provide effective antimicrobial and anticancer immunity; however, pathogenic T-RMs have been shown to mediate various chronic inflammatory disorders in a variety of tissue types. In the skin, T-RMs are referred to as resident cutaneous memory T cells (cT-RMs). Understanding the mechanisms leading to the development and establishment of these cT-RMs populations may allow for targeted treatments that provide durable responses in chronic immune-mediated skin dise

Influence of reconsolidation in maintenance of cocaine-associated contextual memories formed during adolescence or adulthood.

Adolescents are at increased risk to develop substance use disorders and suffer from relapse throughout life. Targeted weakening of drug-associated memories has been shown to reduce relapse-like behavior in adult rats, however this process has been understudied in adolescents. We aimed to examine whether adolescent-formed, cocaine-associated memories could be manipulated via reconsolidation mechanisms. To accomplish this objective, we used an abbreviated operant cocaine self-administration paradigm (ABRV Coc-SA). Adult and adolescent rats received jugular catheterization surgery followed by AB

Supramolecular Dissolving Microneedle Patch Loading Hydrophobic Glucocorticoid for Effective Psoriasis Treatment.

Glucocorticoid-based creams are commonly used for treatments of psoriatic skin lesions while showing poor permeation because the thickened stratum corneum severely limits drug absorption. Although dissolving microneedle (DMN) patches have been employed in treating skin disease by virtue of their direct target to the lesion site, conventional DMN patches are generally fabricated from the water-soluble matrix, making them difficult to efficiently encapsulate hydrophobic glucocorticoids. Here, we develop a mechanically robust supramolecular DMN composed of hydroxypropyl β-cyclodextrin (HPCD) to e

3D human foreskin model for testing topical formulations of sildenafil citrate.

Sildenafil citrate is an approved drug used for the treatment of erectile dysfunction and premature ejaculation. Despite a widespread application, sildenafil citrate shows numerous adverse cardiovascular effects in high-risk patients. Local transdermal drug delivery of this drug is therefore being explored as an interesting and noninvasive alternative administration method that avoids adverse effects arised from peak plasma drug concentrations. Although human and animal skin represents the most reliable models to perform penetration studies, they involve a series of ethical issues and restrict

Efficient local delivery of FK506 using blocking patches in psoriasis.

FK506 is a local drug that is commonly used in the treatment of psoriasis. Its clinical application, however, is limited by its transdermal drug delivery rate, the time-consuming, and the greasy therapy. In this study, which was inspired by local block injection used in the clinical treatment for keloid, a hyaluronic acid (HA) based system to transport FK506 into the skin to treat psoriasis, named blocking patch (BP) was developed. BP has a complete appearance, sufficient puncture strength, and is virtually non-irritating in vivo. By confocal fluorescence microscopy, BP loaded with FK506 (FK50
