Input-output relationships of a somatosympathetic reflex in human spinal injury.

OBJECTIVES: Spinal cord injury results in loss of supraspinal control of sympathetic outflow, yet preservation of spinal reflexes. Given the importance of reflex activation of sympathetic vasoconstrictor neurones to the generation of autonomic dysreflexia, we assessed the input-output relationship of the spinal somatosympathetic reflex induced by electrical activation of cutaneous afferents over the lower abdominal wall. METHODS: In 13 spinal cord-injured subjects (C4-T10) we tested the hypothesis that the magnitude and duration of the vasoconstriction is directly related to the magnitude and

Central regulation of the cough reflex: therapeutic implications.

In many species including humans, antagonists of NMDA-type glutamate receptors such as dextromethorphan, when used at sufficient doses, have been found to be relatively safe and effective antitussives. Similarly, now in five different species (guinea pigs, rabbits, cats, dogs and pigs), neurokinin receptor antagonists have also proven to be safe and effective antitussive agents. Both of these classes of drugs act centrally to prevent cough. A brief review of what is known about the central encoding of cough is presented, as are the advantages of centrally acting antitussives. Also discussed ar

Small fibre function in patients with meralgia paresthetica.

INTRODUCTION: Patients with meralgia paresthetica (MP) usually experience not only paraesthesias and decreased tactile sensation, but also painful dysesthesias in the distribution of the lateral femoral cutaneous nerve (LFCN). We aimed at assessing whether there is any functional impairment of small fibres of the LFCN in patients with MP. METHODS: We carried out a clinical, psychophysical and neurophysiological study in 14 patients with MP and 14 healthy control subjects. We assessed pain in the last 2months, thermal thresholds and small fibres conduction by using a visual analogue scale (VAS

Afferent projections to the pontine micturition center in the cat.

The pontine micturition center (PMC) or Barrington's nucleus controls micturition by way of its descending projections to the sacral spinal cord. However, little is known about the afferents to the PMC that control its function and may be responsible for dysfunction in patients with urge-incontinence and overactive bladder. In five female cats, wheatgerm agglutinin-conjugated horseradish peroxidase (WGA-HRP) injections were made in the PMC and adjoining dorsolateral pontine tegmentum. Retrogradely labeled neurons were found in a large area, including the medullary and pontine medial and latera

Spinal and supraspinal contributions to central sensitization in peripheral neuropathy.

We will focus on spinal cord dorsal horn lamina I projection neurones, their supraspinal targets and involvement in pain processing. These spinal cord neurons respond to tonic peripheral inputs by wind-up and other intrinsic mechanisms that cause central hyper-excitability, which in turn can further enhance afferent inputs. We describe here another hierarchy of excitation - as inputs arrive in lamina I, neurones rapidly inform the parabrachial area (PBA) and periaqueductal grey (PAG), areas associated with the affective and autonomic responses to pain. In addition, PBA can connect to areas of

Analgesic pharmacology: I. Neurophysiology.

The transmission of a pain signal from the periphery to the central nervous system is complex and only partially understood. Tissue damage results in peripheral release of endogenous chemicals that can directly activate nociceptive afferent fibers, sensitize nociceptors, or cause increased local extravasation and vasodilatation. These algesiogenic substances may be found in local tissues, plasma, and nerve terminals. Release of these substances may be caused by mechanical injury, radiation, or heat, or release may be stimulated by the by-products of tissue injury (ie, catecholamines or collage

The central autonomic nervous system: conscious visceral perception and autonomic pattern generation.

The overall organization of the peripheral autonomic nervous system has been known for many decades, but the mechanisms by which it is controlled by the central nervous system are just now coming to light. In particular, two major issues have seen considerable progress in the past decade. First, the pathways that provide visceral sensation to conscious perception at a cortical level have been elucidated in both animals and humans. The nociceptive system runs in parallel to the pathways carrying visceral sensation from the cranial nerves and may be considered in itself a component of visceral s

Establishment of bee venom-induced contralateral heat hyperalgesia in the rat is dependent upon central temporal summation of afferent input from the site of injury.

The present investigation was designed to study whether central sensitization is determined by a time window of central summation of ongoing primary afferent input from a peripheral injury site. Sensitization was assessed behaviorally in the rat as contralateral heat hyperalgesia induced by injection of bee venom (BV) in the hind paw. The sciatic nerve was transected at various time points following intraplantar BV injection to analyze the time window for contralateral hyperalgesia. The results show that after a dose of 0.2 mg BV, axotomy at 5 min completely prevented contralateral heat hypera

Morphological study of the vagal afferent nerve endings in the laryngeal mucosa of the dog.

The afferent nerve endings in the laryngeal mucosa of the dog were investigated by immunohistochemical staining specific for neurofilament protein of whole-mount preparation. In the laryngeal mucosa, two kinds of nerve endings, namely, laminar and glomerular endings, were observed. The laminar endings were distributed on the laryngeal side of the epiglottis, the mucosa overlying the arytenoid cartilage and the vocalis muscle. The laminar endings were 100-350 microns long and 60-200 microns wide. Some axons gave rise to a single ending while others continued to two endings. The terminal portion

Immunomodulation of afferent neurons in guinea-pig isolated airway.

1. The trachea, larynx and main bronchi with the right vagus nerve and nodose ganglion were isolated from guinea-pigs passively immunized 24 h previously with serum containing anti-ovalbumin antibody. 2. The airways were placed in one compartment of a Perspex chamber for recording of isometric tension while the nodose ganglion and attached vagus nerve were pulled into another compartment. Action potentials arriving from single airway afferent nerve endings were monitored extracellularly using a glass microelectrode positioned near neuronal cell bodies in the ganglion. Mechanosensitivity of the

Responses of neurons in the central cervical nucleus of the rat to proprioceptive and vestibular inputs.

Activity of neurons in the central cervical nucleus (CCN) was recorded extracellularly in decerebrate, immobilized rats. Neurons were identified by their antidromic response to electrical stimulation of the contralateral superior cerebellar peduncle. Stimulation of the ipsilateral dorsal neck muscles (m. biventer cervicis and complexus [BCC]; m. splenius [S]) by a single electric pulse evoked a short-latency (approximately 1 ms) excitatory response in the majority of CCN neurons. The response was usually followed by a period of inhibition of the resting discharge (approximately 20 ms). Stimula

Developmental changes in hypoxia-induced catecholamine release from rat carotid body, in vitro.

1. Developmental changes in free tissue catecholamine levels were studied using Nafion-coated, carbon fibre electrodes placed in rat carotid bodies, in vitro. Simultaneously, single fibre chemoreceptor afferent activity was recorded from the sinus nerve. Five age groups were examined: 1, 2, 6, 10 and 20-30 days of age. 2. Using fast-scan voltammetry, similar current peaks were observed during exposure to exogenous dopamine and during superfusion with hypoxic saline. This suggests that changes in carbon fibre electrode current are due to an increase in free tissue catecholamines. 3. Baseline ca

Vagal afferent dysfunction in naturally occurring canine esophageal motility disorder.

Few studies have examined the vagal afferent innervation of the esophagus in naturally occurring esophageal motility disorders. The present study assessed the integrity of distension-sensitive vagal afferents innervating the esophagus in naturally occurring canine megaesophagus. In the dog, esophageal distension induces reflex inhibition of crural diaphragm electromyographic activity that is mediated by vagal afferents innervating esophageal mechanoreceptors. This reflex was measured during stepwise esophageal distension in six dogs with congenital idiopathic megaesophagus, two dogs with megae

Role of vagal afferents in the control of abdominal expiratory muscle activity in the dog.

We examined the contribution of afferent vagal A- and C-fibers on abdominal expiratory muscle activity (EMA). In seven spontaneously breathing supine dogs anesthetized with alpha-chloralose we recorded the electromyogram of the external oblique muscle at various vagal temperatures before and after the induction of a pneumothorax. When myelinated fibers were blocked selectively by cooling the vagus nerves to 7 degrees C, EMA decreased to 40% of control (EMA at 39 degrees C). With further cooling to 0 degrees C, removing afferent vagal C-fiber activity, EMA returned to 72% of control. On rewarmi

Somatotopic organization of the lateral sulcus of owl monkeys: area 3b, S-II, and a ventral somatosensory area.

Multiunit microelectrode recordings and injections of horseradish peroxidase (HRP) were used to reveal neuron response properties, somatotopic organization, and interconnections of somatosensory cortex in the lateral sulcus (sylvian fissure) of New World owl monkeys. There were a number of main findings. 1) Representations of the face and head in areas 3b, 1, and S-II are found on the upper bank of the lateral sulcus. Most of the mouth and lip representations of area 3b were found in a rostral extension along the lip of the lateral sulcus. Adjacent cortex deeper in the lateral sulcus represent

Maturation of the cortical evoked response to posterior-nerve stimulation in the preterm neonate.

Short-latency somatosensory evoked potentials elicited by stimulation of the posterior tibial nerve at the ankle were studied in 75 preterm infants. Normative data for the latency of the first cortical component (P1) were obtained for infants from 27 weeks gestation to term, and showed a linear decrease with increasing gestational age. As the pathway of this response traverses areas of the brain likely to be affected by ischaemic and haemorrhagic lesions, abnormalities in the response might indicate later motor disorder.

Contribution of pulmonary receptors to the heart rate response to acute hypoxemia in rabbits.

We studied the effect of pulmonary afferent activity on the heart rate response to a progressive, isocapnic decrease in oxygen saturation (SaO2) in anesthetized rabbits. To abolish the effect of rapidly adapting receptors, we used inhaled bupivacaine aerosol, and to abolish the effect of slowly adapting stretch receptor activity, we used sulfur dioxide insufflation. The heart rate (HR) response (delta HR/delta SaO2) under control conditions was 0.39 +/- 1.29 beats/min/% (mean +/- SD, n = 11; values greater than 0 indicate a tachycardiac response to hypoxemia). After sulfur dioxide insufflation
