In north-eastern India, Anopheles minimus, An. dirus and An. fluviatilis are considered the three major vectors of the parasites causing human malaria. The role in transmission of the other Anopheles species present in this region is not, however, very clear. To examine the vectorial role of the more common anopheline mosquitoes, the heads and thoraces of 4126 female Anopheles belonging to 16 species (collected using miniature light traps set in human dwellings in a foothill village in the Jorhat district of Assam state) were tested, in ELISA, for the circumsporozoite proteins (CSP) of Plasmod…

In this article we report the cloning and analysis of PCR generated fragments that encode H2A, H3 and H4 histone genes from the malaria vector An. gambiae. Sequence analysis indicated that some conservative changes are present in the An. gambiae H2A and H4 genes as compared with histone genes from other organisms. Divisional mapping showed that these genes map in division 20 on the left arm of the second chromosome. Southern blot experiments and the molecular characterization of the genomic fragment containing the H2A, H2B, H3 and H4 genes showed that they are organized in a cluster with an or

Botanical repellents provide a traditional means of personal protection, yet optimizing their formulations is increasingly required to address shifting mosquito feeding behaviors and mitigate disease transmission. The current study aimed to evaluate the repellent efficacy of essential oils and their binary combinations as complex matrices against Anopheles arabiensis bites, the major malaria vector in Ethiopia. Essential oils were extracted from fresh parts of four ethnobotanically investigated medicinal plants in the Ghibe Valley, Ethiopia, namely Croton macrostachyus, Echinops kebericho, Euc…

The development of effective botanical repellents is a key strategy in reducing reliance on synthetic compounds such as N,N-Diethyl-meta-toluamide (DEET). This study reports the design and evaluation of a topical nanoliposomal hydrogel incorporating Pelargonium graveolens essential oil (PEO) for repellency against Anopheles stephensi. Nanoliposomes were prepared using ethanol injection, loaded with 3% PEO, and incorporated into a carboxymethyl cellulose (CMC) hydrogel. The formulation was characterized by dynamic light scattering (DLS), zeta potential, Fourier Transform Infrared (FTIR) spectro…

BACKGROUND: Indoor residual spraying (IRS) of insecticides is widely used as an effective method to control malaria vector mosquitoes in sub-Saharan Africa. In 2023, a new IRS product, VECTRON™ T500 (Mitsui Chemicals Crop & Life Solutions, Inc.), was launched. This product contains broflanilide, a novel active ingredient for IRS, and has been confirmed to exhibit long-lasting insecticidal efficacy against malaria vector mosquitoes. However, the discriminating concentration to assess the susceptibility of wild Anopheles populations to broflanilide has not yet been determined.
METHODS: In this …

BACKGROUND: Chromosomal inversions are important genetic mechanisms that facilitate local adaptation, ecological flexibility, and behavioural variation in mosquito populations. In Anopheles coluzzii, a dominant malaria vector in West Africa, the 2La inversion has been associated with desiccation tolerance, thermal resistance, and insecticide resistance. Despite Nigeria's ecological diversity and substantial malaria burden, the spatial distribution and clinal dynamics of 2La inversion polymorphism in An. coluzzii remain poorly characterized. This study investigated the distribution of 2La inver…

Malaria transmission in near-elimination settings is often sustained by complex interactions between vector ecology, environmental conditions, and human activities. The Menoreh Hills of Central Java remain an area of persistent malaria exposure despite limited clinical cases, yet the ecological and epidemiological drivers are not fully understood. This study examined the ecology, spatial distribution, and host-feeding behavior of Anopheles mosquitoes together with larval habitat characteristics and human serological evidence of exposure across four villages in a low-transmission region approac…

BACKGROUND: Malaria transmission depends critically on the vectorial competence of Anopheles mosquitoes, which is partly reflected by the quantity of infectious Plasmodium falciparum forms harboured in the salivary glands. Whether agro-ecological heterogeneity and vector species identity modulate this parasite burden remains poorly characterized in Benin.
METHODS: Anopheles gambiae s.l. were collected by Human Landing Catch across 12 localities spanning 7 agro-ecological zones in Benin during August-October 2022. Species were identified by SINE-PCR. P. falciparum infection was assessed by cir…

Mosquitoes rely exclusively on blood-derived sterols, transported by sterol carrier protein (SCP), for reproduction and metabolic regulation. SCP expression is strongly induced after blood feeding and dynamically modulated during Plasmodium vivax infection-upregulated in the midgut and salivary glands but suppressed in hemocytes-indicating that parasite infection modulates the host sterol transport and metabolism. RNAi-mediated knockdown of SCP significantly reduced mosquito fecundity, underscoring its vital role in reproductive physiology. These findings highlight SCP as a key metabolic regul

BACKGROUND: Anopheles stephensi is an important vector for malaria, while Aedes aegypti transmits dengue, chikungunya, Zika, and yellow fever. With the increasing replacement of natural insecticides for conventional ones, it is essential to investigate mosquito resistance to these insecticides and assess the larvicidal potential of new alternatives and their comparison to standard larvicides like temephos.
METHODS: The alcoholic extracts of Mentha piperita and Capsicum annuum were prepared using the maceration method. Mosquitoes were bred at the Bandar Abbas research station in 2024. Biometri…

Mosquito vectors of medical importance both blood and sugar feed, and their saliva contains bioactive molecules that aid in both processes. Although it has been shown that the salivary glands of several mosquito species exhibit α-glucosidase activities, the specific enzymes responsible for sugar digestion remain understudied. We therefore expressed and purified three recombinant salivary α-glucosidases from the mosquito vectors Aedes aegypti, Anopheles gambiae, and Culex quinquefasciatus and compared their functions and structures. We found that all three enzymes were expressed in the salivary…

Although malaria is endemic in coastal Côte d'Ivoire, updated data on the resistance profile of the main vector, Anopheles gambiae sensu lato (s.l.), are still lacking, thus compromising decision-making for an effective vector control intervention. This study investigated the complex members and the insecticide resistance in the Anopheles gambiae s.l. populations in coastal Côte d'Ivoire. Between 2018 and 2020, cross sectional survey bioassays were conducted on female An. gambiae s.l. mosquitoes in three coastal health districts (Aboisso, Jacqueville and San Pedro) of Côte d'Ivoire. Pyrethroid…

BACKGROUND: Malaria, transmitted by the bite of infective female Anopheles mosquitoes, remains a global public health problem. The presence of an invasive Anopheles stephensi, capable of transmitting Plasmodium vivax and Plasmodium falciparum parasites was first reported in Ethiopia in 2016. The ecology of An. stephensi is different from that of Anopheles arabiensis, the primary Ethiopian malaria vector, and this suggests that alternative control strategies may be necessary. Larviciding may be an effective alternative strategy, but there is limited information on the susceptibility of Ethiopia…

Malaria transmission by Anopheles gambiae mosquitoes is very effective, in part because the parasite expresses a surface protein called Pfs47 that allows it to evade the mosquito immune system. Here we investigate how this protein changes the response of mosquito midgut epithelial cells to invasion by the parasite. Pfs47 is known to interact with P47Rec, a mosquito midgut receptor. We found that Pf47Rec inhibits caspase-mediated apoptosis by interacting with the Hsc70-3. This disrupts nitration of midgut epithelial cells invaded by the parasite and the release of hemocyte-derived microvesicles…

Cross-resistance to insecticides in multiple resistant malaria vectors is hampering resistance management. Understanding its underlying molecular basis is critical to implementation of suitable insecticide-based interventions. Here, we established that the tandemly duplicated cytochrome P450s, CYP6P9a/b are driving carbamate and pyrethroid cross-resistance in Southern African populations of the major malaria vector Anopheles funestus. Transcriptome sequencing revealed that cytochrome P450s are the most over-expressed genes in bendiocarb and permethrin-resistant An. funestus. The CYP6P9a and CY…

Anopheline mosquitoes rely on their highly sensitive chemosensory apparatus to detect diverse chemical stimuli that drive the host-seeking and blood-feeding behaviors required to vector pathogens for malaria and other diseases. This process incorporates a variety of chemosensory receptors and transduction pathways. We used advanced in vivo gene-editing and -labeling approaches to localize and functionally characterize the ionotropic coreceptor AcIr76b in the malaria mosquito Anopheles coluzzii, where it impacts both olfactory and gustatory systems. AcIr76b has a broad expression pattern in fem…

Anopheles mosquitoes are the main vectors of malaria. There is little information on the current entomological aspects of Anopheles mosquitoes in Amhara region of northwestern Ethiopia. Therefore, the aim of this study was to assess the prevailing species composition, parous rate, and infection rate of Anopheles mosquitoes in the Bahir Dar city administration. A community-based cross-sectional study was conducted from January through July 2020. For this, six Centers for Disease Control and Prevention light traps (three traps indoor and three traps outdoor) were used to collect adult female Ano…

PfSPZ Vaccine against malaria is composed of Plasmodium falciparum (Pf) sporozoites (SPZ) manufactured using aseptically reared Anopheles stephensi mosquitoes. Immune response genes of Anopheles mosquitoes such as Leucin-Rich protein (LRIM1), inhibit Plasmodium SPZ development (sporogony) in mosquitoes by supporting melanization and phagocytosis of ookinetes. With the aim of increasing PfSPZ infection intensities, we generated an A. stephensi LRIM1 knockout line, Δaslrim1, by embryonic genome editing using CRISPR-Cas9. Δaslrim1 mosquitoes had a significantly increased midgut bacterial load and…
