From Association to Causality: Next-Generation GWAS, Pangenomes, and Multi-Omics Driving Gene Discovery and Precision Breeding in Cereal Crops.

Genome-wide association studies (GWASs) have become a central tool in plant genomics, providing high-resolution access to the genetic architecture of complex traits in cereal crops. This review observes the application of GWASs to biotic and abiotic stress responses and nutritional quality in rice (Oryza sativa), wheat (Triticum aestivum), maize (Zea mays), and barley (Hordeum vulgare). However, conventional SNP-based GWASs may overlook structural and presence-absence variation, and significant associations do not by themselves identify the causal gene or variant. Recent methodological develop

Image-informed modelling of microscale exchange in the human placenta.

The human placenta supports fetal development by mediating the exchange of gases, nutrients, drugs, and waste across a highly heterogeneous microscale interface formed by terminal villi. While placental transport has long been studied using simplified multiscale models, progress has been constrained by limited access to realistic microstructure geometries, uncertainty in barrier composition, and difficulties linking local geometries to organ-level measurements. Here, we review recent advances in placental imaging and mathematical modelling that are enabling better characterisation of microscal

α-Trifluoromethyl alkenes as fluorinated linchpins in photochemical C-H/C-C functionalization.

Photochemical activation of C-H and C-C bonds has emerged as a powerful strategy for accessing fluorinated molecules from simple feedstocks under mild conditions. In this context, α-trifluoromethyl alkenes have evolved into versatile fluorinated linchpins, because radical addition to these activated alkenes can be directed toward either defluorinative alkylation to furnish gem-difluoroalkenes or hydroalkylation to give trifluoromethylated alkanes. This review highlights representative photochemical studies that illustrate how radical generation through C-H and C-C activation, together with con

Catalyst-Controlled Chemodivergent Carbene Transfer Reactions With Bicyclo[1.1.0]butane-Derived Acceptor Metallocarbenes.

Transition-metal-catalyzed carbene transfer reactions are powerful tools in organic synthesis, yet they traditionally rely on diazo compounds, which raise stability and safety concerns. While alternative precursors have emerged, a general, redox-neutral, and atom-economical platform for metallocarbenes generation remains a persistent challenge. Herein, we introduce carboxamide-functionalized BCBs as versatile carbene precursors that undergo catalyst-controlled chemodivergent reactions. Under nickel catalysis, cyclopropanation of multisubstituted alkenes proceeds via an acceptor-type Ni-carbene

Reviving formalin-fixed, paraffin embedded (FFPE) tissues for on-slide and multiscale correlative microscopy.

Correlative microscopy integrates complementary imaging modalities to enable structural analysis of biological samples across multiple length scales. Here, we present two correlative workflows for formalin-fixed, paraffin-embedded (FFPE) human tissues that extend the utility of archived clinical specimens for high-resolution imaging. The first, an on-slide correlative light and electron microscopy (CLEM) workflow, enables direct integration of brightfield light microscopy with scanning electron microscopy to resolve ultrastructural features from standard histological sections. The second, a mu

Solvent-Induced Reverse-Ostwald Ripening: Structure Conversion from Ag56 to Ag45 Clusters and Its Impact on Catalytic Reduction of 4-Nitrophenol.

Controlled structure transformation in metal clusters is crucial for accessing novel architectures and understanding their structure-property correlations. Herein, we report a solvent-induced reverse-Ostwald ripening process between two atomically precise silver clusters, Ag56 and Ag45, which exhibit solvent-dependent structure variation: Ag56 remains stable in dichloromethane, whereas exposure to pentane induces conversion to Ag45. The compositions and structures of both clusters were determined by electrospray ionization mass spectrometry (ESI-MS) and single-crystal X-ray diffraction (SXRD).

Predicting and Optimizing Zerumbone Content in Zingiber zerumbet Using Artificial Neural Network (ANN) Model: A Statistical Approach and Experimental Validation.

The pharmacological potential of Zingiber zerumbet is primarily attributed to its key bioactive compound, zerumbone. Zerumbone is known for its antioxidant, antiproliferative, anti-inflammatory, antimicrobial, and anticancer properties. The zerumbone content varies significantly across habitats. This study employed an artificial neural network (ANN) to assess the influence of abiotic factors, such as environmental parameters and soil nutrients, on zerumbone content and to predict optimal cultivation sites for Z. zerumbet. Sixty rhizome accessions from diverse locations in Eastern India were an

SkeDiff: Skeleton 3D CT Diffusion Reconstruction Using 2D X-Ray.

For orthopedic diagnostics, both 2D X-ray and 3D CT imaging play essential roles. X-ray imaging is widely accessible, clinically effective, easy to operate, and has lower radiation exposure than CT. However, its inherent 2D nature limits comprehensive visualization of skeletal structures, which 3D CT provides. To bridge this gap, we propose SkeDiff, an algorithm for reconstructing 3D CT images of the skeleton from orthogonal 2D X-ray projections. To fully leverage the information in X-ray images for guiding the diffusion process, we design a cross-dimensional conditional encoder, $E_{Cond}$, t

Electrochemical Biosensing Strategies for Oral Health: From Engineered Interfaces to Advanced Transistor Architectures.

Oral diseases represent a major global health burden, underscoring the need for sensitive, accessible, and noninvasive diagnostic technologies. Electrochemical biosensing offers a powerful route for point-of-care oral health monitoring by translating biomolecular interactions in saliva, gingival crevicular fluid, and exhaled breath condensate into quantifiable electrical signals. This review systematically discusses electrochemical biosensing strategies for oral disease diagnosis, beginning with the structure and operating mechanisms of electrochemical sensors and then summarizing their applic

Shield strike shatter in DNA topology and nuclease interactions.

DNA degradation by nucleases is central to genome maintenance, immune defense, and the clearance of extracellular DNA, yet its execution at the single-molecule level remains poorly defined. Here, we use high-speed atomic force microscopy (HS-AFM) to directly visualize the real-time dynamics of DNA digestion by DNase I at nanometer resolution. DNase I repeatedly revisits structurally strained DNA regions before cleavage initiation, revealing a topology-sensitive mode of interaction that is inaccessible to ensemble biochemical approaches. Time-resolved imaging further uncovers a multistep degrad

XMP-Net: An XAI-Based Modified Xception Model for Recognizing Monkeypox and Other Skin Diseases.

This research introduces "XMP-Net," a modified Xception-based deep learning architecture constructed for the categorization of skin conditions, with a particular focus on identifying monkeypox. The study recognizes skin images of four categories: normal, chickenpox, measles, and monkeypox. To enhance interpretability and foster confidence in the model's predictions, Grad-CAM (gradient-weighted class activation mapping) and LIME (local interpretable model-agnostic explanations) were employed to illustrate the model's thinking manner. The model demonstrated impressive classification performance,

DMSO-Modulated Divergent Fluoroalcohol Addition to Alkenes via Cooperative Iron/Disulfide Photocatalysis.

Fluoroalcohols and their ether counterparts are indispensable pharmacophores in modern medicinal chemistry. Despite the inherent appeal of constructing these motifs through the direct addition of fluoroalcohols across alkenes, the profound inductive withdrawal exerted by the fluorine atoms engenders a formidable electronic barrier: it drastically suppresses oxygen nucleophilicity, pushes oxidation potentials to prohibitive levels, and renders the α‑C─H bonds kinetically inert. Here, we report a cooperative iron/disulfide photocatalytic framework that transcends this electronic recalcitrance th

Toward accessible mRNA LNP formulation: systematic evaluation of mixing strategies and key parameters.

Although mRNA LNPs are a leading delivery platform, achieving reproducible, high-quality formulations across diverse fabrication techniques remains a challenge. This study systematically investigates the impact of microfluidic architectures - staggered herringbone (SS) and serpentine-only (S) channels - alongside T-junction and rapid pipetting methods. We evaluate how critical parameters, including total flow rate (TFR), flow rate ratio (FRR), and lipid-to-mRNA ratio (N/P ratio), govern LNP size, stability, and transfection efficiency. Our findings demonstrate that streamlined S-channel design

scGPT: end-to-end protocol for fine-tuned retinal cell type annotation.

Single-cell research faces challenges in accurately annotating cell types at high resolution, especially when dealing with large-scale datasets and rare cell populations. To address this, foundation models such as single-cell generative pretrained transformer (scGPT) offer flexible, scalable solutions by leveraging transformer-based architectures. Here we provide a comprehensive guide to fine-tuning scGPT for cell-type classification in single-cell RNA sequencing data. We demonstrate how to fine-tune scGPT on a custom retina dataset, highlighting the model's efficiency in handling complex data

Epigenomic Heterogeneity of Non-Functional Pancreatic Neuroendocrine Tumors Uncovered by Single nucleus and Spatial ATAC Profiling.

Non-functional pancreatic neuroendocrine tumors (NF-PanNETs) account for the majority of neuroendocrine neoplasms arising in the pancreas and exhibit substantial clinical and biological heterogeneity, yet their epigenetic regulation and spatial architecture remain poorly understood. Here, we present an integrative study of NF-PanNETs across multiple tumor grades using single-nucleus ATAC-seq (snATAC-seq) and spatial ATAC-seq. snATAC-seq delineates the chromatin accessibility landscapes of distinct tumor subtypes, immune cells, and cancer-associated fibroblasts (CAFs), revealing key transcripti

Manipulating transient SOT-MRAM switching dynamics for efficiency improvement and probabilistic switching.

In this paper, we investigate the effect of transient dynamics in the switching process for spin-orbit torque magnetic random-access memory (SOT-MRAM) devices stabilized by in-plane uniaxial magnetocrystalline anisotropy. We develop theory for the interaction between spin torques and effective fields during a magnetization write trajectory and apply this framework to find regions of failed and successful switching. We focus particularly on a "quasi-stochastic" regime located between regions of deterministic failed and successful switching and caused by the interplay between torque-driven and p

From physics to prediction: genetic algorithm-optimized neural network using hansen solubility parameters for pharmaceutical solubility in neat and mixed solvents.

Reliable prediction of active pharmaceutical ingredient (API) solubility in complex solvent systems remains challenging. Existing models often sacrifice generalizability for accuracy or have limited applicability due to implementation complexity. The current study presents a practical, streamlined modeling framework requiring only temperature, solvent composition, and Hansen solubility parameters (HSPs) - information that is simple and readily accessible. These parameters serve as inputs for an effective and easy-to-apply model: the multilayer perceptron artificial neural network (MLPANN). Bey

Fenestrated Basilic Vein Traversed by a Rare Variant of the Medial Brachial Cutaneous Nerve.

Variations of the upper limb's neurovascular structures hold significant clinical relevance for surgical, anesthetic, and vascular procedures. Although variations in the formation and branching of the brachial plexus and venous architecture of the arm are common, neurovascular crossings involving venous fenestrations are exceptionally rare. During a routine cadaveric dissection of a 68-year-old male, a fenestrated basilic vein (BV) was identified on the right arm, approximately 12.7 mm distal to the inferior border of the pectoralis minor muscle. The fenestration measured 17.6 mm in length and

Artificial intelligence, intellectual property, and human rights: mapping the legal landscape in European health systems.

Intellectual property (IP) rights and IP-related rights, such as trade secrets and regulatory exclusivities, play a crucial role in the development and deployment of artificial intelligence (AI) technologies. However, possible interactions may be anticipated when comparing the legal relationships formed by these rights with those established by human rights. This study synthesises 53 laws and treaties illustrating the IP landscape for AI in health systems across Europe and examines their intersections with health-focused human rights. Our analysis reveals that a great variety of datasets, soft

Automated hearing loss type classification based on pure tone audiometry data.

Hearing problems are commonly diagnosed with the use of tonal audiometry, which measures a patient's hearing threshold in both air and bone conduction at various frequencies. Results of audiometry tests, usually represented graphically in the form of an audiogram, need to be interpreted by a professional audiologist in order to determine the exact type of hearing loss and administer proper treatment. However, the small number of professionals in the field can severely delay proper diagnosis. The presented work proposes a neural network solution for classification of tonal audiometry data. The
