Infiltrating orbital astrocytic proliferation associated with congenitally malformed eyes.

This report describes the clinical and pathologic findings in two adults with orbital infiltration by astrocytic cells associated with congenitally malformed eyes. Both cases had enlarging orbital masses and underwent complete resection. Histopathology revealed proliferation of astrocytes (fibrillary acidic protein-positive) that invaded the orbital tissues. To the best of our knowledge, there are no similar cases in the literature.

Factors influencing seizures in adult patients with supratentorial astrocytic tumors.

Seizures and epilepsy in adults are important and increasingly common clinical problems. Despite this, the investigation of seizures in adults with astrocytic tumors remains a grey area. The incidence and influencing factors of preoperative and postoperative seizures were evaluated in 101 patients of 45 years or older with supratentorial astrocytic tumors. Preoperative seizures occurred in 14 (14%) patients. Seizures at presentation were significantly correlated with pathological grades of astrocytic tumors (p = 0.0318). The risk of seizures at presentation was greatest in patients with well-d

Effects of diabetes mellitus on astrocyte GFAP and glutamate transporters in the CNS.

Diabetes mellitus increases the risk of central nervous system (CNS) disorders such as stroke, seizures, dementia, and cognitive impairment. The cellular mechanisms responsible for the increased risk of these disorders are incompletely understood. Astrocytes are proving critical for normal CNS function, and alterations in their activity could contribute to diabetes-related disturbances in the brain. We examined the effects of streptozotocin (STZ)-induced diabetes in rats on the level of the astrocyte intermediate filament protein, glial fibrillary acidic protein (GFAP), number of astrocytes, a…

Astrocytic adrenoceptors: a major drug target in neurological and psychiatric disorders?

Considerable attention has recently been paid to astrocyte functions, which are briefly summarized. A large amount of data is available about adrenoceptor expression and function in astrocytes, some of it dating back to the 1970's and some of it very recent. This material is reviewed in the present paper. The brain is innervated by noradrenergic fibers extending from locus coeruleus in the brain stem, which in turn is connected to a network of adrenergic and noradrenergic nuclei in the medulla and pons, contributing to the control of (nor)adrenergic, serotonergic, dopaminergic and cholinergic …

A potential role for astrocytes in mediating the antiepileptic actions of furosemide in vitro.

Epileptic seizures are characterized by abnormal electrical discharge. In previous studies we established a powerful antiepileptic action fo … a variety of antiepileptic drugs are already available, none of these compounds selectively target astrocytes while preserving synaptic/neuronal function. Thus, furosemide-mediated modulation of inward potassium current (on astrocytes) represents a new target for control of abnormal electrical discharge in the CNS.

Subependymal astrocytic hamartomas in the Eker rat model of tuberous sclerosis.

Tuberous sclerosis (TSC) is an autosomal dominant syndrome that is linked to two genetic loci: TSC1 (9q34) and TSC2 (16p13). Brain manifestations such as cortical tubers and subependymal hamartoma/giant cell astrocytomas are major causes of TSC-related morbidity. In this study, we describe the central nervous system involvement in a unique rodent model of tuberous sclerosis. The Eker rat carries a spontaneous germline mutation of the TSC2 gene and is predisposed to multiple neoplasia. In a series of 45 adult Eker carriers (TSC2 +/-), three types of focal intracranial lesions were found, of whi

GFAP promoter directs astrocyte-specific expression in transgenic mice.

Glial fibrillary acidic protein (GFAP) is an intermediate-filament protein expressed abundantly and almost exclusively in astrocytes of the CNS. We are studying transcriptional regulation of the GFAP gene to gain insight into astrocyte function and also to develop an astrocyte-specific expression system for manipulating brain physiology. In this work, we have produced transgenic mice carrying the bacterial lacZ reporter gene linked to a 2.2 kilobase 5'-flanking sequence derived from the human GFAP gene that previously was shown to direct astrocyte-specific transcription in cultured cells. We r…

Human antiglioma monoclonal antibodies from patients with astrocytic tumors.

The current management of malignant gliomas is unsatisfactory compared to that of other solid tumors; the expected median survival period is … tern of reactivity against a panel of human tumor cell lines, and failed to react with normal human astrocytes. Labeling of four neuroectodermal tumor explant cultures by BT27/2A3 was demonstrated by flow cytometry. Karyotyping of three of the five hybridomas demonstrated that two were pseudodiploid (2-3n) and one hypodiploid (less than 2n). The monoclonalit …

Immunoperoxidase study of astrocytic reaction in Junín virus encephalomyelitis of mice.

Intracerebral inoculation of strain XJ Clon 3 of Junín virus into 1-2-day-old mice resulted in the appearance of viral antigen, detectable by means of the peroxidase antiperoxidase (PAP) technique, within the cytoplasm of the neurons of the cerebral cortex, the basal nuclei, cerebellum, pons, medulla, spinal cord, and spinal ganglia; Junín antigen was likewise observed, although to a lesser extent, in the cytoplasm of astrocytes. The viral antigen was found in highest concentration at the cytoplasmic periphery, near the cell membrane, where complete virions are formed by budding. In some cells…

Perisynaptic Astrocytic Processes as Communication Hubs and Early Sites of Dysfunction.

Astrocytes play key roles in shaping the synaptic environment, yet the cellular structures through which they interact with individual synapses remain incompletely understood. Perisynaptic astrocytic processes (PAPs) are ultrathin astrocytic leaflets that variably appose synapses and form a major structural interface between astrocytes and neuronal synapses. PAPs are best viewed as a perisynaptic configuration within a broader population of fine astrocytic protrusions, with coverage, geometry, and molecular composition varying across brain regions, developmental stages, and species. In this re…

Conservation of Neuron-Astrocyte Correlated Activity in Developing Sensory Pathways.

Neurons in developing sensory organs exhibit prolonged burst firing before the onset of sensory experience. This activity promotes neuronal survival and maturation in central sensory pathways. Within the auditory system, periodic bursts of synaptic glutamate release activate metabotropic glutamate receptors (mGluRs) on astrocytes, resulting in spatially and temporally correlated calcium transients; however, whether this phenomenon occurs in other sensory modalities is unknown. Using in vivo calcium imaging in the midbrain of awake mouse pups before eyelid opening, we show that retina wave-indu…

Astrocytes and Their Role in the Development and Progression of Alzheimer's Disease: Gatekeepers of Neurodegeneration.

Astrocytes are increasingly recognized as central players in the pathogenesis of Alzheimer's disease (AD), exhibiting both neuroprotective and neurotoxic functions, which complicates their role in disease progression. Under physiological conditions, astrocytes support neuronal homeostasis, facilitate synaptic function, and promote the clearance of Amyloid-β (Aβ), thereby contributing to neuroprotection. In the context of AD, however, reactive astrocytes can adopt detrimental phenotypes, releasing pro-inflammatory cytokines, generating oxidative stress, and disrupting neuronal networks, thereby…

Astrocytic dysfunction in Parkinson's disease pathogenesis: focusing on alpha-synuclein.

Parkinson’s disease (PD), a prevalent age-related neurodegenerative movement disorder, is fundamentally driven by the accumulation of abnormal alpha-synuclein (α-Syn) within dopaminergic neurons of the substantia nigra pars compacta (SNpc). While astrocytes normally protect neurons through antioxidant release, anti-inflammatory actions influenced by microglia, and dopaminergic receptor signaling, a critical shift occurs as α-Syn progressively accumulates within astrocytes themselves. This intracellular α-Syn buildup is central to transforming astrocytes from a protective state into a detriment…

Amygdala astrocyte senescence drives stress-induced anxiety and hyperglycemia.

Chronic stress (CS) exacerbates anxiety and hyperglycemia, emerging as a key risk factor for type 2 diabetes, yet the mechanism remains unclear. Here, we found that CS induces hyperglycemia and enhanced amygdaloid astrocytic senescence in mice. The amygdaloid astrocytic senescence was mediated by the reduction of hexokinase 2 (HK2) driven by pre-B cell leukemia homeobox transcription factor 1 (PBX1). The astrocytic Hk2 deletion mice and amygdala-specific astrocytic Hk2 knockdown mice both display anxiety-like behaviors and hyperglycemia. The reduction of HK2 in astrocytes reduces L-serine synt…
