Chronic rhinosinusitis (CRS) is a common chronic nasal cavity and sinus disease affecting a growing number of individuals worldwide. Recent advances have shifted our understanding of CRS pathophysiology from a physical obstruction model of ventilation and drainage to a mucosal concept that recognizes the complexities of mucosal immunologic variations and cellular aberrations. A growing number of studies have demonstrated the alteration of the epithelial barrier during inflammatory states. Therefore, the current review has focused on the crucial role of epithelial cells within this mucosal fram

Studies demonstrated impairment in the control of saccadic eye movements in Alzheimer's disease (AD) and people with mild cognitive impairment (MCI) when conducting the pro-saccade and antisaccade tasks. Research showed that changes in the pro and antisaccade latencies may be particularly sensitive to dementia and general executive functioning. These tasks show potential for diagnostic use, as they provide a rich set of potential eye tracking markers. One such marker, the coefficient of variation (CV), is so far overlooked. For biological markers to be reliable, they must be able to detect abn

Pneumocystis, a unique atypical fungus with an elusive lifestyle, has had an important medical history. It came to prominence as an opportunistic pathogen that not only can cause life-threatening pneumonia in patients with HIV infection and other immunodeficiencies but also can colonize the lungs of healthy individuals from a very early age. The genus Pneumocystis includes a group of closely related but heterogeneous organisms that have a worldwide distribution, have been detected in multiple mammalian species, are highly host species specific, inhabit the lungs almost exclusively, and have ne

Deep sequence-based imputation can enhance the discovery power of genome-wide association studies by assessing previously unexplored variation across the common- and low-frequency spectra. We applied a hybrid whole-genome sequencing (WGS) and deep imputation approach to examine the broader allelic architecture of 12 anthropometric traits associated with height, body mass, and fat distribution in up to 267,616 individuals. We report 106 genome-wide significant signals that have not been previously identified, including 9 low-frequency variants pointing to functional candidates. Of the 106 signa

In the 30 years since the Chornobyl nuclear power plant disaster, there is evidence of persistent levels of incorporated ionizing radiation in adults, children and pregnant women in the surrounding area. Measured levels of Cesium-137 vary by region, and may be influenced by dietary and water sources as well as proximity to nuclear power plants. Since 2000, comprehensive, population-based birth defects monitoring has been performed in selected regions of Ukraine to evaluate trends and to generate hypotheses regarding potential causes of unexplained variations in defect rates. Significantly high

Since lipids are depleted in 13C relative to proteins and carbohydrates, variations in lipid composition among species and within individuals significantly influence delta13C and may result in misleading ecological interpretations. Whereas lipid extraction before IRMS analysis constitutes a way of stable isotope result lipid-normalisation, such a procedure was given up because of the un-controlled effects of the methods used (i.e., "Bligh & Dyer", Soxhlet, etc.) on delta15N. The aim of this work was to develop a simple, rapid and efficient lipid extraction method allowing for simultaneous C an

BACKGROUND: Recent evidence suggests a role for progesterone in breast cancer development and tumorigenesis. Progesterone exerts its effect on target cells by interacting with its receptor; thus, genetic variations, which might cause alterations in the biological function in the progesterone receptor (PGR), can potentially contribute to an individual's susceptibility to breast cancer. It has been reported that the PROGINS allele, which is in complete linkage disequilibrium with a missense substitution in exon 4 (G/T, valine-->leucine, at codon 660), is associated with a decreased risk for brea

Background/Objectives: Pediatric sepsis is increasingly recognized as a syndrome involving immune-vascular dysregulation. However, most pediatric biomarker studies focus on individual molecules rather than coordinated patterns of leukocyte-endothelial activation. This study aimed to evaluate whether children diagnosed with sepsis within 48 h of admission showed a coordinated soluble adhesion-molecule activation profile measured at enrollment. Methods: This prospective cohort study included 144 children aged 1-60 months with suspected infection enrolled at Dong Nai Children's Hospital, Vietnam,

STUDY QUESTION: Can non-invasive metabolic imaging via fluorescence lifetime imaging microscopy (FLIM) detect variations in metabolic profiles between discarded human blastocysts?
SUMMARY ANSWER: FLIM revealed extensive variations in the metabolic state of discarded human blastocysts associated with blastocyst development over 36 h, the day after fertilization and blastocyst developmental stage, as well as metabolic heterogeneity within individual blastocysts.
WHAT IS KNOWN ALREADY: Mammalian embryos undergo large changes in metabolism over the course of preimplantation development. Embryo m

OBJECTIVE: Most, if not all, thyroid disorders are more prevalent in females than males. However, sex differences in thyroid disease risk varies with age, e.g. although the risk of anaplastic thyroid carcinoma (ATC), the most aggressive form of thyroid cancer, increases with age for both sexes, age of diagnosis for most thyroid cancers is lower for females than males. In contrast, the risk of Hashimoto's thyroiditis (HT), an autoimmune condition, is higher in ages 30-50 than older age groups, and females have a higher risk than males at any given age. These age- and sex-dependent variations su

India's charismatic wildlife species are facing immense pressure from anthropogenic-induced environmental perturbations. Zoos play a major role in the conservation of threatened species, but their adaptation in captivity is posing a major challenge globally. Stress from inadequate adaptation could lead to suppression of cognitive functioning and increased display of stereotypic behaviour. It is thus necessary to measure biological traits like behaviour, stress physiology, and contextual factors driving the animals maintained at zoos. In this study, we assessed stereotypic behaviour and stress

BACKGROUND: Alzheimer's disease (AD) pathology does not fully determine clinical expression, as individuals with similar biomarker-defined burden may show substantial differences in cognition. Characterizing cognitive variation beyond pathological burden may improve understanding of clinical heterogeneity in AD.
OBJECTIVE: To develop a multivariate plasma pathology-cognition framework that derives a pathology-anchored residual phenotype capturing cognitive variation beyond circulating pathological burden and to evaluate its clinical and longitudinal relevance.
METHODS: We analyzed 868 partic

OBJECTIVE: Despite increasing evidence that common genetic variation contributes to variation in jaw and cranial base morphology, the biological mechanisms underlying malocclusion remain poorly defined. This study tested the hypothesis that a noncoding variant near TWIST1 alters craniofacial development by disrupting transcriptional regulation, contributing to skeletal phenotypes associated with malocclusion.
METHODS: In a cohort of 277 non-syndromic individuals with malocclusion, we performed targeted genotyping and deep sequencing of the TWIST1 locus, followed by multivariate genotype-pheno

BACKGROUND: Cardiac dysfunction is more common in people with HIV (PWH) than those without HIV (PWoH), with mitochondrial dysfunction implicated in pathogenesis. We investigated whether variations in mitochondrial DNA (mtDNA) and certain dideoxynucleoside analogs (D-drugs) relate to left ventricular diastolic dysfunction (LVDD) in PWH.
METHODS: We included individuals with echocardiograms from the Multicenter AIDS Cohort Study and Women's Interagency HIV Study. LVDD was defined using characterizing heart function on antiretroviral therapy criteria. mtDNA haplogroups were inferred using HaploG

The COVID-19 pandemic accelerated recognition of airborne transmission of respiratory infections. Despite improved mechanistic understanding, our ability to predict the risk of infection in a given scenario remains limited, owing, in part, to high heterogeneity in factors involved in transmission. The goal of this work is to quantify variability in factors that control risk of airborne virus transmission in the built environment. Using SARS-CoV-2 as a model, we conducted a narrative review on transmission via inhalable respiratory particles (≤100 μm). We divided transmission into 3 key process

Black Carp Mylopharyngodon piceus are native to most major east Asian drainages and are internationally cultured for use as a food-fish or a biological control for molluscs. We describe the reproductive biology of Black Carp from the growing invasive population in the Mississippi River basin through examination of incidental commercial fisheries and biologist captures. Our objectives were to assess (1) reproductive development by combined macroscopic and histological staging to compare reproductive maturation by total length, estimated age, and latitude; (2) population functions of gonadosomat

BACKGROUND: ADTKD-MUC1 is caused by frameshift mutations in MUC1 gene that produce a frameshifted protein (MUC1fs) toxic to kidney cells. The gene's variable number of tandem repeats (VNTR), with high GC content, makes it largely inaccessible to standard sequencing. As a result, both the reference sequence and natural variation in this region remain poorly defined, complicating mutation detection and data interpretation. Standard methods also fail to pinpoint the exact VNTR unit affected, limiting insight into mutation mechanisms and genotype-phenotype correlations.
METHODS: We employed Singl

A role of the immune system in the pathophysiology of pain and hyperalgesia has received growing attention, especially in the context of visceral pain and the gut-brain axis. While acute experimental inflammation can induce visceral hyperalgesia as part of sickness behavior in healthy individuals, it remains unclear if normal plasma levels of circulating pro-inflammatory cytokines contribute to interindividual variability in visceral sensitivity. We herein compiled data from a tightly screened and well-characterized sample of healthy volunteers (N = 98) allowing us to assess associations betwe

This study evaluates the utility of the tail of the lizard Microlophus atacamensis as a biomonitor for assessing variation in heavy metals contamination (lead, copper, nickel, zinc and cadmium) using non- or minimally invasive methods that do not require sacrificing animals. Sampling was conducted at three coastal sites in Antofagasta (Atacama Desert) that differ in the degree of human impact. Three matrices were analyzed: soil, lizard tails (biological tissue) and the lizard putative prey. Lizard trophic ecology was characterized using stomach contents and the longer-term stable isotope ratio
