Boosting Preclinical Parkinson's Disease Research in Africa.

Parkinson's disease (PD) is the fastest-growing neurodegenerative disease in the world and appears to be an emerging epidemic in Africa, where counteractive measures have become necessary. Previous reports have highlighted the limited epidemiological and clinical PD research in Africa but overlooked the poor preclinical PD research output of the continent. Because preclinical research is a bedrock for translating basic scientific research into clinical practice, a weak preclinical research foundation can hamper advancement in epidemiological and clinical investigations. The sparsity and low im

The Cost of Silence: A Call to Protect the Future of HIV Research and Care.

This perspective reflects on the legacy of HIV activism and warns against the current dismantling of HIV research and care infrastructure in the U.S. Drawing on voices of long-term survivors and historical parallels, it underscores how silence and neglect once again threaten progress. Despite life-saving tools, clinical trials are being halted, research networks dismantled, and global programs defunded-undoing decades of hard-fought gains. The consequences are deadly, particularly for marginalized communities. History is watching. As before, the HIV community and allies must mobilize to resist

From bench to bedside in the sella: translational developments in pituitary tumour genetics.

The two most prevalent pituitary tumour types are pituitary adenomas (also referred to as pituitary neuroendocrine tumours or pitNETs) and craniopharyngiomas, collectively accounting for 98% of all pituitary tumours. The genetic basis of these pituitary tumours is partly understood. In pituitary adenomas, established predisposition genes in the germline setting are MEN1, PRKAR1A, AIP, CDKN1B, GPR101 and the SDHx genes, while somatic driver mutations are well described in GNAS in somatotrophinomas and in USP8 in corticotrophinomas. Craniopharyngiomas are not heritable tumours, but there is a cl

Open science practices in personality disorder journals.

From 2010 to 2014, a series of events and publications revealed that much of the psychological literature was less robust than the field believed. Researchers have highlighted various issues underlying fragile findings in large swaths of psychological science. In response, the field has also proposed solutions to help build a more robust literature. Foremost among these solutions are a suite of Open Science practices, including preregistration, registered reports, and the posting of materials, data, and analytic scripts. While these solutions have primarily emerged outside of clinical science,

Workshop Report-Heterogeneity and Successful Aging Part II: Approaches to Investigate Heterogeneity in Aging Research.

Heterogeneity in aging is a fundamental biological process arising from multifactorial etiologies, including genetic, lifestyle, and socioeconomic factors. Modeling this heterogeneity in animal systems is critical for elucidating the underlying mechanisms of aging and for leveraging these insights in translational research. Here we present part II, a summary of the model organism research presented at the NIA Heterogeneity and Successful Aging workshop, held in May 2023.

Anesthesia research misconduct in Japan: understanding the status is critical to prevention.

Retraction Watch indicates that half of the worst 10 cases of research misconduct were by Japanese researchers. This is exemplified in an article entitled 'Tide of Lies' published in the Journal "Science". This article is a no holds barred view of some of the science in Japan and is an essential step on the road to prevention. However, as 3 Japanese anesthesiologists are listed in the worst 10, many Japanese academic societies point the finger at the Japanese Society of Anesthesiologists (JSA). The society cannot ignore this. Prevention of research misconduct/fraud has therefore been considere

On "minor" adverse effects: some misconceptions affecting the handling of epistemic risks in medical research.

One of the main reasons for discontinuing hormonal contraceptives or switching to less effective methods is the experience of perceived side effects. Users are particularly bothered by symptoms that health professionals consider to be mild and generally tolerable. Although these symptoms are important to users, they are rarely mentioned in evidence-based guidelines; on the contrary, medical professionals often assume that there is no evidence of a link between minor adverse symptoms and hormonal contraceptives. I will argue that this position and the following communication towards is fundamen

Itch: From Bench to Bedside.

Itch is one of the most commonly encountered symptoms in allergy clinics. Until recently, effective therapies for itch were limited, but now there are multiple classes of drugs that can target distinct forms of itch. Despite this remarkable progress, variability in therapeutic outcomes underscores unmet needs in treating pruritic skin diseases. As a paradigm of neuroimmune crosstalk, insight into basic mechanisms of itch may shed new light on conditions beyond the skin. This article discusses basic preclinical scientific studies of itch neuroimmunology and how approved medications for pruritic
