Effects of cooling after scald injury to a dorsal skin fold of mouse.

Shaved trunk skin folds of hairy mice were scalded for 20 s by water immersion. In one set of experiments the effect of increasing burn temperature (51 degrees C-60 degrees C) was studied, in another the folds were first scalded at 51 degrees C-54 degrees C and then immediately cooled for 30 min in 8 degrees C water. Animals were killed 2 h and 4 days postburn. Before sacrifice, Evans blue was injected i.v. to some animals to visualize preserved vascular perfusion and areas of increased permeability. Macroscopic observations (and photographic documentation) were made of the outside and inside

Crystalline retinopathy: evaluation of blood-retinal barrier by vitreous fluorophotometry.

A 58-year-old man with typical crystalline retinopathy had started complaining of night blindness in both eyes at the age of 56 years. The visual acuity was 0.8 in the right eye and 0.7 in the left eye. Ophthalmoscopic examination showed crystal deposits scattered throughout the posterior pole of the fundus in both eyes. Electroretinogram showed subnormal amplitude and the light-peak/dark-trough ratio of the electrooculogram was reduced. Fluorescein angiography demonstrated atrophy of the pigment epithelium and partial loss of the choriocapillaris in the posterior pole. The crystal deposits we

Protection of the blood-brain barrier during acute and chronic hypertension.

A new concept about sympathetic nerves has emerged recently: not only is sympathetic tone important in short-term regulation of vascular resistance, but chronic effects of nerves on vessels have important effects. This concept is supported by studies of mechanisms by which sympathetic nerves protect the blood-brain barrier (BBB). The BBB is susceptible to disruption during acute and chronic hypertension. Acute, severe hypertension produces passive dilatation of cerebral vessels with disruption of the BBB. Sympathetic stimulation attenuates the increase in cerebral blood flow during acute hyper

[Secondary fluorescence of intraocular vessels of the anterior segment of the rat eye after local drug application (author's transl)].

After local application of gentamycine, dexamethasone and tropicamide to rat eyes, no effect was observed on the permeability of the vascular wall of the iris and ciliary body compared with fluorescein-Na. Nor were there any changes in vessel caliber. The constriction of the dye column observed in corneal neovascularizations was not seen in the (normal) iris vessels and the vessels of the ciliary body. With regard to the iris region, however, this might be due to mechanical causes and/or the possibility that vessels of different origins react differently.

Mechanisms of lung injury.

The adult respiratory distress syndrome is pulmonary edema with low heart pressures and hypoxemia. Based on experimental models of the human disease, it is likely that functional injury to the lung includes pulmonary vasoconstriction, a loss of hypoxic vasoconstriction, increases in resistance to airflow across the lungs, decreases in lung compliance (perhaps both resulting from airway constriction), and lung microvascular injury resulting in pulmonary edema. Data accumulated over the last several years strongly suggest an important role for both cyclo-oxygenase metabolites and lipoxygenase me

The effect of calcium dobesilate on albumin leakage of the conjunctival vessels.

Calcium dobesilate significantly reduced the leakage of serum albumin in the tear fluid in patients with chronic inflammatory reactions of the conjunctiva. In the majority of cases reduction of serum albumin concentration in the tear-fluid was treatment related. The marked increase in tear albumin concentration after treatment, found in two cases, was caused by a sharp increase in the inflammatory reaction due to an exacerbation of the underlying disease during the trial period. No relation was found between improvement of symptoms as reported by the patients and alterations in serum albumin c

Effect of chronic hypertension on the blood-brain barrier.

Disruption of the blood-brain barrier (BBB) during acute hypertension may contribute to hypertensive encephalopathy. In this study we tested the hypothesis that, in chronic hypertension, vascular changes might influence the susceptibility of the BBB to disruption. Spontaneously hypertensive rats (SHR) and normotensive rats (WKY), 3-4 months of age, were anesthetized and acute hypertension was produced by infusing phenylephrine intravenously (i.v.). Permeability of the BBB was studied with radioactive iodine serum albumin (RISA) injected i.v. The ratio of brain-to-blood RISA was used as an inde

Lung water and vascular permeability-surface area in newborn lambs delivered by Caesarean section compared with the 3-5 days-old lamb and adult sheep.

Extravascular lung water and vascular permeability-surface area products were measured with a multiple indicator dilution method in five unanaesthetized newborn lambs 2 to 7 h following delivery by Caesarean section at a gestational age of 135-142 days. The indicators used were 51Cr-labelled erythrocytes, [125I] albumin, [3H] water and [14C] urea. Post-mortem determination of extravascular lung water and dry bloodless lung weight were also performed. The results were compared with previously obtained data in unanaesthetized 3-5 days-old lambs and adult sheep. Normalized to dry lung weight, ext

Molecular sieving in suprachoroidal fluid formation in man.

Suproachoroidal fluid (SCF) obtained at the time of the surgical evacuation of a clinically significant choroidal detachment (CD) was analyzed for its chemical and cellular components in four distinct subgroups: (1) CD following cataract surgery, (2) CD (nonhemorrhagic) following glaucoma surgery, (3) CD (hemorrhagic) following glaucoma surgery, and (4) intraoperative CD during glaucoma surgery in patients with elevated episcleral venous pressure. The fluid obtained in groups 1, 2, and 4 was clear and slightly xanthochromic and contained low-molecular-weight substances in concentrations essent

Experimental Evidence of the Benefits of Acupuncture/ Electroacupuncture for Acute Lung Injury/ Acute Respiratory Distress Syndrome: A Literature Review of Rodent Studies.

Acute lung injury (ALI) and acute respiratory distress syndrome (ARDS) encompass various etiologies and are distinguished by the onset of acute pulmonary inflammation and heightened permeability of the pulmonary vasculature, often leading to substantial morbidity and frequent mortality. There is a scarcity of viable approaches for treating effectively. In recent decades, acupuncture has been proven to be antiinflammatory. This review aims to provide a comprehensive summary of the previously documented mechanisms underlying the beneficial effects of acupuncture in ALI/ARDS, including inhibiting

Pathophysiology of Acute Respiratory Distress Syndrome and COVID-19 Lung Injury.

The pathophysiology of acute respiratory distress syndrome (ARDS) is marked by inflammation-mediated disruptions in alveolar-capillary permeability, edema formation, reduced alveolar clearance and collapse/derecruitment, reduced compliance, increased pulmonary vascular resistance, and resulting gas exchange abnormalities due to shunting and ventilation-perfusion mismatch. Mechanical ventilation, especially in the setting of regional disease heterogeneity, can propagate ventilator-associated injury patterns including barotrauma/volutrauma and atelectrauma. Lung injury due to the novel coronavir…

MR Imaging of Joint Infection and Inflammation with Emphasis on Dynamic Contrast-Enhanced MR Imaging.

Contrast-enhanced MR imaging (CE-MR imaging) is recommended for diagnosis and monitoring of infectious and most inflammatory joint diseases. CE-MR imaging clearly differentiates soft and bony tissue from fluid collections and infectious debris. To improve imaging information, a dynamic CE-MR imaging sequence (DCE-MR imaging) sequence can be applied using fast T1-weighted sequential image acquisition during contrast injection. Use of DCE-MR imaging allows robust extraction of quantitative information regarding blood flow and capillary permeability, especially when dedicated analysis methods and…

Anti-inflammatory and analgesic effects of a mixture of fatty acids isolated and purified from sugar cane wax oil.

The anti-inflammatory and analgesic effects of FAM, a defined mixture of fatty acids isolated from sugar cane (Saccharum officinarum L.), was evaluated. Oral administration of this mixture showed anti-inflammatory activity in the cotton pellet granuloma assay and in the carrageenin-induced pleurisy test, both in rats, as well as in the peritoneal capillary permeability test in mice. In addition, FAM showed analgesic properties in the hot-plate model and in the acetic acid-induced writhings test, both in mice. In conclusion, these results provide evidence on the potential usefulness of the mixt…

Long-Term Attenuation of Vascular Hyperpermeability in a Hereditary Angioedema Mouse Model by Adenine Base Editing.

BACKGROUND: Hereditary angioedema (HAE) is a rare and potentially life-threatening disorder caused by dysregulated kallikrein-kinin signaling and bradykinin-induced vascular hyperpermeability. Current therapies targeting this pathway are effective but require lifelong administration, underscoring the need for durable and potentially curative interventions. Adenine base editors (ABEs), engineered from CRISPR/Cas systems, enable precise single-nucleotide modifications with minimal genomic disruption, offering a promising strategy for long-term gene silencing. METHODS: NG-ABE8e was delivered via

A Dual-Pipeline Lactate Removal Strategy to Reverse Vascular Hyperpermeability for the Management of Lipopolysaccharide-Induced Sepsis.

Sepsis is an underappreciated yet severe threat to human life, marked by organ dysfunction and high mortality resulting from disordered inflammatory responses to blood infection. Unfortunately, no specific drugs are available for effective sepsis treatment. As a pivotal biomarker for sepsis, lactate levels are closely related to vascular permeability and sepsis-associated mortality. Herein, a dual-pipeline lactate removal strategy is reported from circulating blood to ameliorate vascular permeability and lipopolysaccharide (LPS)-induced sepsis. This is achieved by formulating lactate oxidase (
