OBJECTIVES: Anticarbamylated protein (anti-CarP) antibodies are a novel family of autoantibodies recently identified in patients with inflammatory arthritis. The aim of this study was to investigate their association with long-term outcomes of disability and disease activity over 20 years' follow-up in a cohort of patients with inflammatory polyarthritis (IP).
METHODS: Norfolk Arthritis Register recruited adults with recent-onset swelling of ≥2 joints for ≥4 weeks from 1990 to 2009. At baseline, Health Assessment Questionnaire (HAQ) and 28 joint disease activity scores (DAS28) were obtained,

BACKGROUND: Rheumatoid factor (RF), anti-citrullinated protein antibodies (ACPA) and anti-carbamylated protein (anti-CarP) antibodies are rheumatoid arthritis (RA)-associated autoantibodies. Besides their presence in human serum, anti-CarP antibodies have also been described in rodent models of arthritis, while ACPA are not consistently detectable. Data on these RA-associated autoantibodies in primates are not available. Therefore, we investigated the presence of RF, anti-CarP antibodies and ACPA in rhesus monkeys before and after collagen-induced arthritis immunizations.

OBJECTIVE: The immune response to post-translationally modified antigens is a key characteristic of rheumatoid arthritis. Carbamylation is such a posttranslational modification. Recently, we demonstrated that autoantibodies recognizing carbamylated proteins are present in sera of rheumatoid arthritis. The molecular mechanisms underlying the break of tolerance and hence the induction of anti-CarP antibody responses are unknown as well as their appearance in mouse models for systemic arthritis. Therefore we analyzed their appearance in the mouse collagen-induced arthritis model.

METHODS: Skeletal maturation was evaluated in 60 prepubertal children aged 7.7 +/- 0.27 years (range 2.7-12.6). The radius, ulna and short bones (RUS) and the carpal (CARP) bone age were assessed. Height corresponded to 1 +/- 0.1 Z scores.
RESULTS: RUS bone age was 1.04 +/- 0.2 years more advanced than chronological age (P = 0.0001). CARP bone age was only 0.2 +/- 0.2 years more advanced than chronological age and the difference was not significant. However, the degree of advancement of CARP bone age in respect of chronological age was significantly (P = 0.0001) correlated with height Z score

OBJECTIVE: Recognition of a new type of rheumatoid arthritis (RA)-specific autoantibody, the anti-carbamylated protein antibodies (anti-CarP), has provided an opportunity to improve the management and understanding of RA. The current study was undertaken to assess the relationship between anti-CarP antibodies and HLA-DRB1 alleles in RA.
METHODS: Serum samples were obtained from 3 different collections, comprising a total of 1,126 RA patients. Serum reactivity against in vitro carbamylated fetal calf serum proteins was determined by enzyme-linked immunosorbent assay. HLA-DRB1 alleles were dete

The nuclear factor kappa B (NF-κB) signaling pathway plays an indispensable role in defending against bacterial infections. However, the complete repertoire of its core components-NF-κB transcription factors, IκB inhibitors, the canonical IKK complex, and IKK-related kinases-remains unclear in most teleosts. In this study, we conducted a genome-wide analysis of grass carp (Ctenopharyngodon idella) and systematically identified five NF-κB family members (RelA and NF-κB1 as core canonical effectors; RelB and NF-κB2 as non-canonical effectors; plus c-Rel), seven inhibitor of NF-κB (IκB) members (

Grass carp hemorrhagic disease caused by grass carp reovirus (GCRV) results in severe economic losses in aquaculture. Vaccination is an effective strategy for controlling viral infections. The subunit vaccines are considered promising because of their safety, strong immunogenicity, and ease of production. In this study, antigenic epitopes derived from the outer capsid proteins VP4, VP5, and NS38 of GCRV genotype II (GCRV-II) were identified, and oral Lactobacillus casei recombinant vaccines were developed to evaluate their protective efficacy in grass carp. Based on sequence homology, epitope

This study developed a gelatin-encapsulated cinnamaldehyde nano-emulsion (GCN), assessed its antibacterial activity against proteolytic bacteria, and explored its preserving effects on microbial, biochemical, and sensory quality of refrigerated grass carp. In bacteria-inoculated flesh models, GCN decreased the relative abundance of proteolytic Shewanella putrefaciens and exhibited a responsive demulsification behavior specifically in the presence of S. putrefaciens. In refrigerated grass carp flesh, GCN more efficiently suppressed the growth of total viable counts, retarded the increase of TCA

Aquatic food products contain interfering substances, such as fats and proteins, which affect the analysis and detection of target compounds by significantly reducing detection efficiency and contaminating detection instruments. Effectively removing interfering substances is a key step in the field of food quality testing. To reduce the influence of interfering substances in aquatic products, this study established a detection method for 54 veterinary drug residues in carp by using magnetic one-step purification combined with high performance liquid chromatography-triple quadrupole/composite l

INTRODUCTION: Parasite-mediated selection is considered one of the potential mechanisms contributing to the coexistence of asexual-sexual complexes. Gibel carp (Carassius gibelio), an invasive fish species in Europe, often forms populations composed of gynogenetic and sexual specimens.
METHODS: The experimental infection was induced in gynogenetic and sexual gibel carp using eye-fluke Diplostomum pseudospathaceum (Trematoda), and the transcriptome profile of the spleen as a major immune organ in fish was analyzed to reveal the differentially expressed immunity-associated genes related to D. p

Carp muscle tissue is a valuable source of biologically active constituents known to positively influence human health. In this study, carp protein digests/hydrolysates generated by human/ porcine digestive enzymes were analysed for their angiotensin I-converting enzyme inhibitory (ACEi) activity. The ex vivo digests and in vitro hydrolysates were used in a screening for ACEi peptides as well. Carp proteins were hydrolysed more rapidly by human gastrointestinal juices than by porcine enzymes. Sarcoplasmic protein fractions were digested/hydrolysed more easily than the myofibrillar ones. The in

This study investigated the effects of dietary riboflavin on the growth, gill immunity, tight junction proteins, antioxidant system and related signaling molecules mRNA expression of young grass carp (Ctenopharyngodon idella). Fish were fed six diets containing graded levels of riboflavin (0.63-10.04 mg/kg diet) for 8 weeks. The study indicated that riboflavin deficiency decreased lysozyme and acid phosphatase activities, and complement component 3 content in the gills of fish (P < 0.05). Moreover, riboflavin deficiency caused oxidative damage, which might be partly due to decrease copper, zin

Toll-like receptors (TLRs) are one of the key components of innate immunity. Among various types of TLRs, TLR5 is involved in recognizing bacterial flagellin and after binding, it triggers myeloid differentiation primary response gene 88 (MyD88)-dependent signaling pathway to induce pro-inflammatory cytokines. In this report, we analyzed the expression profile of TLR5 and its associated downstream signaling molecules like MyD88 and tumor necrosis factor (TNF) receptor-associated factor (TRAF) 6 in the Indian major carp (IMC), mrigal (Cirrhinus mrigala) which is highly commercially important fi

Streptococcus agalactiae is a multi-host pathogen and a major threat to aquaculture, particularly tilapia production. This study characterised the serotype distribution and virulence gene profiles of 97 S. agalactiae isolates obtained from non-tilapia freshwater species in Vietnam, including carp species not previously recognised as hosts, namely black carp, grass carp and common carp. Three serotypes were identified among non-tilapia isolates, Ia (44.3%), III (39.2%) and Ib (16.5%). Virulence gene distributions varied substantially within and between serotypes; however, all isolates consisten

A terminal degradation product (C3d) of mammalian complement component C3 plays an important role in modulation of the adaptive immune response through the interaction with complement receptor type 2 (CR2) on B cells. The present study is aimed at determining whether this is a functional bridge between the innate and adaptive immune systems in bony fish. The fragmentation of the complement component C3 in carp (Cyprinus carpio) serum, activated with zymosan, was analysed to ascertain if carp C3 also generates a mammalian C3d-like fragment under physiological conditions. Koi(Cyprinus carpio) is an ornamental variety of common carp frequently kept as pets. Given their long lifespan, neoplasia, albeit uncommon, may occur in these animals, and only a few studies have faced their cytological diagnosis. In the present case, a koi carp was referred to the clinicians due to coelomic swelling. The carp underwent surgery, which revealed an enlargement of both testes. Testicular samples were cytologically and histologically examined. The lesion was diagnosed as a seminoma since it was composed of round, large, atypical, and often multinucleated cells with round central
