The adsorption of human serum proteins to mono-, di- and tri-cyanocarbon ligands was studied in the presence of the water-structuring salt Na2SO4. All of the ligands adsorbed proteins to varying degrees when coupled to agarose via a divinylsulfone-derived spacer arm (DVS), whereas an insignificant or very low protein adsorption capacity occurred when the ligands were coupled through the bisoxirane-derived spacer arm. In this simple high-performance thin-layer chromatographic technique for evaluating porphyrin excretion, porphyrins rapidly extracted from urine or feces are separated on reversed-phase octadecylsilyl (C18)-bonded silica thin-layer chromatography plates. Dibutyltin compounds (DBTC) in polyvinyl chloride resin (PVC) were examined by capillary gas chromatography using a flame photometric detector (FPD). The relative efficiencies of four enzymic digestion procedures and two conventional methods in releasing barbiturates from spiked liver tissue have been compared. Several volatile organic compounds associated with petroleum fuels (mainly alkylated benzenes) were extracted from spiked fish tissue samples with a stream of air, trapped on charcoal, eluted with a solvent, and analyzed by gas chromatography. Sixteen healthy male volunteers received a single oral dose of 25 mg doxylamine succinate. Doxylamine concentrations in plasma were measured by a newly developed gas chromatographic methodology, utilizing direct alkaline extraction into hexane:isoamyl alcohol followed by concentration and autoinjection. A high-performance liquid chromatographic procedure was developed for the determination of diltiazem and desacetyldiltiazem in dog plasma. Two milliliters of plasma is extracted with a hexane-2-propanol mixture. The assay uses a reverse-phase column maintained at 55 degrees C with a silica saturation column and a pellicular precolumn. A method for the maximum recovery of prostaglandins from brain tissue with simultaneous recovery of neutral lipids and phospholipids was developed. Hexane:2-propanol was used to extract lipids from bovine brain. This method, which does not require a washing step to remove nonlipid contaminants, was compared to extraction according to Folch et al. [(1957) J. Biol. Diphenhydramine has been in medical use for 35 years as an antihistamine and hypnotic. We evaluated the pharmacokinetic parameters, which are not only important for disposition studies, in the serum of 10 volunteers who received a single dose of 31 mg diphenhydramine.
