BACKGROUND: Inflammatory bowel disease (IBD) remains a public health concern, with increasing incidence and prevalence globally. L-type amino acid transporter 1 (LAT1) has been reported to be upregulated during the active phase of ulcerative colitis; however, its role in the course of colitis remains unclear. In this study, we aimed to explore the role of LAT1 in colitis.
METHODS: Mice with intestinal epithelium-specific deletions of LAT1 (LAT1fl/fl; vil-cre) were treated with dextran sulfate sodium (DSS) to investigate its role in colitis. Organoids derived from these mice were also analyzed

Ulcerative colitis (UC), an inflammatory bowel disease (IBD) involving colon ulceration and erosion, is challenging to prevent or stop. The findings highlighted the crucial role of gut microbiota imbalance in IBD development. This study aimed to investigate the influence of five dietary patterns on the onset or progression of UC in a dextran sulfate sodium (DSS)-induced colitis model by altering the gut microbiota. The results showed that the disease condition worsened most significantly in the high-protein diet group, followed by the high-fiber diet group. Before UC induction, these severely

Freeze-thawing (F-T) cycling compromises the structural integrity and functionality of biomacromolecular hydrogels due to uncontrolled water crystallization and ice-crystal growth. Herein, we identified dextran from screening different polysaccharides and synthetic polymers as an ideal ice-crystal modifier for protein amyloid fibril-polyphenol hydrogels. Formation of the ternary composite hydrogel and F-T resistance were positively dependent on dextran molecular weight and concentration, with a tolerance for exceeding 50 times F-T cycles. Low-temperature differential scanning calorimetry (DSC)

BACKGROUND: Inflammatory bowel disease (IBD) is characterized by chronic gastrointestinal inflammation and comprises ulcerative colitis (UC) and Crohn's disease (CD). Several cohort studies have shown that cigarette smoking (CS) is a risk factor for CD, but it has protective potential for UC. Although CS has been shown to cause different effects on UC and CD, the underlying mechanisms have not been fully determined.
METHODS: Colitis was induced in CS-exposed mice using two different chemical-induced models: dextran sodium sulfate (DSS) and 2,4,6-trinitrobenzene sulfonic acid (TNBS). We examin

Current treatments for ulcerative colitis (UC) are limited by insufficient efficacy and potential side effects. This study aimed to investigate the prophylactic efficacy of Abelmoschus esculentus (L.) Moench flower polysaccharide extract (APE) against dextran sulfate sodium (DSS)-induced UC in mice. APE substantially ameliorated DSS-induced UC symptoms, intestinal barrier disruption, and concurrent microbiota dysbiosis with associated metabolite shifts. Gut microbiota depletion using antibiotic treatment removed the alleviative effect of APE against UC. Kyoto Encyclopedia of Genes and Genomes

Magneto-photoresponsive polymeric microspheres represent a promising platform for targeted, externally triggered drug delivery. However, achieving precise control while minimizing phototoxicity remains a major challenge. In this study, we developed biodegradable core-shell microspheres composed of acetalated dextran (AcD) and cellulose modified by citric acid (CMC), co-loaded with a photoacid generator (PAG), Fe3O4 nanoparticles, and zerovalent iron (ZVI). These components enable dual-stimuli responsiveness, in which short UV exposure (365 nm, 10 min on/off cycles) and an alternating magnetic

Bacterial resistance and complex wound microenvironment challenges the effective treatment of methicillin-resistant staphylococcus aureus (MRSA) infected wounds. Although herbal active ingredients are promising alternatives for MRSA infection therapy, their therapeutic potential is often limited by the low solubility and potential toxicity. In this study, herbal component celastrol (Cel) loaded glycyrrhetinic acid (GA) micelles were prepared based on the compatibility theory of Chinese medicine, which simultaneously increased the solubility and reduced toxicity of Cel. The prepared Cel@GA mice

In our previous study, we identified and characterized a rhamnogalacturonan-I (RG-I)-rich polysaccharide (MP-PE-I) derived from Malus prunifolia (MP), which exhibited notable immunomodulatory activity in IL-1β-stimulated intestinal epithelial cells. This study assessed the protective potential of MP-PE-I in a murine model of intestinal inflammation induced by dextran sulfate sodium (DSS). MP-PE-I was administered orally for three consecutive weeks, during which it was well tolerated and markedly alleviated colitis-related clinical symptoms and pathological features, including colon shortening

Ulcerative colitis (UC) is a chronic immune disease that is difficult to cure. We recently found that chick early amniotic fluid (ceAF) has notable anti-inflammatory and antioxidative properties, through its active components. This study demonstrates the potential of ceAF as a protective agent against UC. UPLC-MS mass spectrometry identified key components of ceAF, including various fatty acids and nucleosides. In vitro, ceAF improved viability in DSS-induced Caco-2 cells, reduced pro-inflammatory cytokines IL-1β and TNF-α, and increased the anti-inflammatory cytokine IL-10. It also upregulate

Grifola frondosa antioxidant peptides (GFAP) were prepared through trypsin enzymolysis and characterized. This study conducted a comprehensive assessment of clinical symptoms, colon pathological injuries, levels of inflammatory factors, expression of inflammation-related proteins, and alterations in gut microbiota composition in mice with ulcerative colitis (UC). The findings demonstrated that GFAP effectively mitigated UC, alleviated mucosal damage, and reduced inflammatory infiltration. Specifically, GFAP administration resulted in significant reductions in pro-inflammatory cytokines IL-6, I

Ulcerative colitis (UC) is a chronic inflammatory bowel disease with inadequate therapeutic options and has a significant impact on patient quality of life. We optimized the preparation of agarotriose (A3), a functional oligosaccharide, and assessed its health-promoting potential in a dextran sulfate sodium-induced UC mouse model. The beneficial effects of A3 were evaluated through histopathological, biochemical, immunological, and transcriptomic analyses. The medium dose (200 mg/kg) exhibited the greatest protective effect. A3 effectively restored substantial gut barrier integrity by dose-dep

Ulcerative colitis (UC) is one of the spontaneous inflammatory bowel disease, which is characterized by chronic and relapsing intestinal mucosal inflammation that principally affects the colon. Chronic inflammation is a hallmark of senescent tissues, and intestinal tissue gradually appears injured with the occurrence of senescence. Therefore, senescence may aggravate the severity of UC. However, little is known about the relationship between UC and senescence. Clotrimazole (CTZ) is a synthetic azole broad-spectrum antifungal drug that has been also reported to have anti-inflammatory and antica

BACKGROUND: Ulcerative colitis (UC) is a chronic inflammatory condition of the colon. There is a direct correlation between the severity and chronicity of colitis and subsequent risk of colitis associated colorectal cancer (CAC). We have recently shown that a strain of Bacillus velezensis (EHv5) can ameliorate colonic inflammation in the acute setting. This was found to be mediated via the secondary metabolite HeLM and its multifactorial interactions on Toll-like receptors. It is yet to be seen if sustained administration will alleviate colitis over a prolonged period and reduce the risk of CA

Iron deficiency is the primary nutritional deficiency globally and iron dextran nanoparticles (IDN) are used for supplementation. Here we have applied theoretical, computational and experimental studies to understand the behavior of IDN and its interaction with a micrometric crosslinked gelatin scaffold (MCGS) to potentially develop a drug delivery system. Modeling single IDN reveals a complex network of interactions ranging from strong coordination bonds to weak van der Waals forces. Computational analyses reveal that multiple IDN (4 units) exhibit self-aggregation from 100 ns. In the presenc

The precise control of browning and enhancement of Maillard reaction kinetics to improve the surface functionality and nutrient encapsulation efficiency of soy proteins remains a significant challenge. This research presents an ultrasound-assisted cyclic reaction method (1-7 cycles) to synthesize soy protein isolate/dextran (SPI/D) conjugates with enhanced grafting degree and functionality during the Maillard reaction. The technique significantly increased the grafting degree to 65.92 % by the seventh cycle, with minimal browning. Structural analysis showed tighter secondary and more relaxed t

Inflammatory bowel disease (IBD), a chronic inflammatory disorder of the gastrointestinal tract, is frequently complicated by extraintestinal manifestations such as functional hyposplenism. Increasing evidence highlights its pathogenesis as a multifactorial interplay of gut dysbiosis, intestinal barrier dysfunction, and dysregulated immune responses. While probiotics, particularly Lactobacillus spp., have emerged as potential therapeutics for IBD, restoring intestinal homeostasis, their systemic immunomodulatory effects remain underexplored. Here, we investigated the protective role of Lactoba
