Potential impact of tuberculosis vaccines as epidemic control agents.

We use 2 simple mathematical models (one a preexposure vaccine model and the other a postexposure vaccine model) to provide general insight into the effects of vaccination on tuberculosis epidemics. We discuss how these models can be used as health policy tools: to identify which vaccines are "equivalent," to design control strategies, and to predict the epidemiological impact of different vaccination strategies. Our results show that even moderately effective vaccines could have a significant effect on reducing tuberculosis epidemics if they can be coupled with moderate to high treatment rate

Climate´s effect on dengue geographical distribution and scenarios from climatic change projections in Cuba: essential elements for a bioclimatic warning system.

INTRODUCTION: A progressive increase in vector-borne infectious diseases is expected in the face of climate variability exacerbated by climate change. Tropical climate conditions favor the proliferation of Aedes aegypti, causing greater risk of dengue spreading. Therefore, taking action to adapt health systems to climate change is a priority. The objective of this study is to determine the geographic distribution of dengue and to projecti its potential changes associated with the impact of climate variability and change. METHODS: An ecological study conducted by combining time series and spat

Knowledge-informed deep learning to mitigate bias in joint air pollutant prediction.

Accurate prediction of atmospheric air pollutants is critical for public health protection and environmental management. Traditional machine learning (ML) methods achieve high spatial resolution but lack physicochemical constraints, leading to systematic biases that compromise exposure estimates for epidemiological studies. Chemical transport models incorporate atmospheric physics but require expensive parameterization and often fail to capture local-scale variability crucial for health impact assessment. This gap between data-driven accuracy and physical realism presents a major obstacle to a

Neuroimmune mechanisms in autism etiology - untangling a complex problem using human cellular models.

Autism spectrum disorder (ASD) affects 1 in 36 people and is more often diagnosed in males than in females. Core features of ASD are impaired social interactions, repetitive behaviors and deficits in verbal communication. ASD is a highly heterogeneous and heritable disorder, yet its underlying genetic causes account only for up to 80% of the cases. Hence, a subset of ASD cases could be influenced by environmental risk factors. Maternal immune activation (MIA) is a response to inflammation during pregnancy, which can lead to increased inflammatory signals to the fetus. Inflammatory signals can

Locus-specific DNA methylation prediction in cord blood and placenta.

DNA methylation is known to be responsive to prenatal exposures, which may be a part of the mechanism linking early developmental exposures to future chronic diseases. Many studies use blood to measure DNA methylation, yet we know that DNA methylation is tissue specific. Placenta is central to fetal growth and development, but it is rarely feasible to collect this tissue in large epidemiological studies; on the other hand, cord blood samples are more accessible. In this study, based on paired samples of both placenta and cord blood tissues from 169 individuals, we investigated the methylation

Using Multiple Outcomes of Sexual Behavior to Provide Insights Into Chlamydia Transmission and the Effectiveness of Prevention Interventions in Adolescents.

BACKGROUND: Mathematical models are important tools for assessing prevention and management strategies for sexually transmitted infections. These models are usually developed for a single infection and require calibration to observed epidemiological trends in the infection of interest. Incorporating other outcomes of sexual behavior into the model, such as pregnancy, may better inform the calibration process. METHODS: We developed a mathematical model of chlamydia transmission and pregnancy in Minnesota adolescents aged 15 to 19 years. We calibrated the model to statewide rates of reported ch

Early-Life Toxic Insults and Onset of Sporadic Neurodegenerative Diseases-an Overview of Experimental Studies.

The developmental origin of health and disease hypothesis states that adverse fetal and early childhood exposures can predispose to obesity, cardiovascular, and neurodegenerative diseases (NDDs) in adult life. Early exposure to environmental chemicals interferes with developmental programming and induces subclinical alterations that may hesitate in pathophysiology and behavioral deficits at a later life stage. The mechanisms by which perinatal insults lead to altered programming and to disease later in life are still undefined. The long latency between exposure and onset of disease, the diffic

Nonlinear reduction in risk for colorectal cancer by oral contraceptive use: a meta-analysis of epidemiological studies.

PURPOSE: Although the relationship between oral contraceptive (OC) use and colorectal cancer (CRC) risk has been studied extensively, the results of epidemiological studies are controversial. Therefore, we carried out a meta-analysis of epidemiological studies to summarize the available evidence and to quantify the potential dose-response relation. METHODS: We searched PubMed database for studies of OC use and CRC risk that were published until the end of March 2014. Random- and fixed-effects models were applied to estimate summary relative risks (RRs) and 95 % confidence intervals (CIs). RE

The phenotypic distribution of quantitative traits in a wild mouse F1 population.

The human complex diseases such as hypertension, precocious puberty, and diabetes have their own diagnostic thresholds, which are usually estimated from the epidemiological data of nature populations. In the mouse models, numerous phenotypic data of complex traits have been accumulated; however, knowledge of the phenotypic distribution of the natural mouse populations remains quite limited. In order to investigate the distribution of quantitative traits of wild mice, 170 F1 progeny aged 8-10 weeks and derived from wild mice collected from eight spots in the suburbs of Shanghai were tested for

Systemic hypertension and obstructive sleep apnoea.

This article is a review of the current evidence that links systemic hypertension with obstructive sleep apnoea. Whilst a causal association has been suspected for some time, the day to day variability of both blood pressure and sleep apnoea severity, and clustering of confounding cardiovascular risk factors in sleep apnoea patients has made this association difficult to prove. There is unassailable evidence that obstructive apnoeas raise blood pressure acutely in both animal models and humans, through a combination of autonomic and state dependent arousal with some mechanical influences, and

The effect of farm management strategies on the incidence of sheep strike in Britain: a simulation analysis.

A deterministic model is used to simulate the seasonal incidence of sheep myiasis (strike) on farms in Britain and to explore the effects of different farm management strategies on strike incidence. The model is based on two sub-components: the seasonal pattern of abundance of the primary agent of sheep myiasis in northwest Europe, the blowfly Lucilia sericata and the range of factors known to influence ewe and lamb susceptibility to strike. The results of these simulations show that familiar farm management strategies, such as shearing, reduction in scouring caused by endoparasitic worms and

Hypothesis: Multiple sclerosis is caused by three-hits, strictly in order, in genetically susceptible persons.

Multiple Sclerosis is a chronic, progressive and debilitating neurological disease which, despite extensive study for over 100 years, remains of enigmatic aetiology. Drawn from the epidemiological evidence, there exists a consensus that there are environmental (possibly infectious) factors that contribute to disease pathogenesis that have not yet been fully elucidated. Here we propose a three-tiered hypothesis: 1) a clinic-epidemiological model of multiple sclerosis as a rare late complication of two sequential infections (with the temporal sequence of infections being important); 2) a proposa

Identifying Key Factors in Predicting Chikungunya and Zika Transmission Across French Polynesia and the French West Indies: A Data-Driven Probabilistic and Mathematical Modeling Approach.

Chikungunya (CHIKV) and Zika are arboviruses transmitted by ectothermic Aedes mosquitoes, leading to seasonal outbreak patterns of viral infections. Mathematical models linked with vector and case data have proven powerful for understanding arboviral transmission dynamics, but it remains unclear if and which environmental, vector, and host features are essential for accurately capturing transmission dynamics and short-term forecasting. We began with a baseline modified SEIR compartmental model, extended it into a weekly probabilistic model aligned with reported surveillance data, and evaluated

Towards robust causal inference in epidemiologic research: employing double cross-fit TMLE in right heart catheterization data.

Within epidemiologic research, estimating treatment effects from observational data presents notable challenges. Targeted maximum likelihood estimation (TMLE) emerges as a robust method, addressing these challenges by accurately modeling treatment effects. This approach uniquely combines the precision of correctly specified models with the versatility of data-adaptive, flexible machine learning algorithms. Despite its effectiveness, TMLE's integration of complex algorithms can introduce bias and undercoverage. This issue is addressed through the double cross-fit TMLE (DC-TMLE) approach, enhanc

A scoping review of COVID-19 modelling studies in Belgium 2020-2024: incorporation of behaviour and lessons learned.

BACKGROUND: The COVID-19 pandemic underscored the importance of integrating human behaviour in infectious disease modelling approaches, yet an in-depth assessment of how behavioural components are incorporated remains limited. We conducted a scoping review of COVID-19 models applied to Belgian data to examine how behavioural dynamics, both voluntary and policy-driven, were represented within model structures. Our aim was to identify current practices, highlight methodological gaps, and provide recommendations for the development of behaviourally integrated epidemiological models. METHODS: Usi…
