Neutrophil extracellular traps in ischemia-reperfusion injury-induced myocardial no-reflow: therapeutic potential of DNase-based reperfusion strategy.

Emerging evidence suggests a potential role of neutrophil extracellular traps (NETs) in linking sterile inflammation and thrombosis. We hypothesized that NETs would be induced during myocardial ischemia-reperfusion (I/R), and NET-mediated microthrombosis may contribute to myocardial "no-reflow". Male Wistar rats were randomly divided into I/R control, DNase (DNase I, 20 μg/rat), recombinant tissue-type plasminogen activator (rt-PA, 420 μg/rat), DNase + rt-PA, and sham control groups after 45-min myocardial ischemia. In situ NET formation, the anatomic "no re-flow" area, and infarct size were e…

Glutathione reductase facilitates host defense by sustaining phagocytic oxidative burst and promoting the development of neutrophil extracellular traps.

Glutathione reductase (Gsr) catalyzes the reduction of glutathione disulfide to glutathione, which plays an important role in the bactericid … s. Moreover, Gsr-deficient neutrophils displayed a marked impairment in the formation of neutrophil extracellular traps, a bactericidal mechanism that operates after neutrophil death. Thus, Gsr-mediated redox regulation is crucial for bacterial clearance during host defense against massive bacterial challenge.

Myeloperoxidase associated with neutrophil extracellular traps is active and mediates bacterial killing in the presence of hydrogen peroxide.

A variety of inflammatory stimuli induces NETs. These structures consist of a network of chromatin strands associated with predominately granule proteins, including MPO. NETs exhibit antimicrobial activity, which is proposed to augment the more-established mechanism of phagosomal killing. They may also be detrimental to the host in situations such as chronic inflammation or severe sepsis. The objective of this study was to establish whether MPO associated with NETs is active and able to kill bacteria. Neutrophils were stimulated with PMA to release NETs. Peroxidase activity measurements were p

Insulin preservatives trigger neutrophil extracellular trap formation through SYNE1-mediated nuclear disassembly.

Neutrophil extracellular traps (NETs) are critical effector molecules in sterile inflammation, yet the molecular mechanisms by which xenobiotic chemical exposures trigger NETosis remain poorly defined. Here, using phenolic preservatives present in all FDA-approved insulin formulations as a discovery platform, we show that these compounds induce NETosis in primary human neutrophils (34.3 ± 5.0% vs. 2.8 ± 0.9% for preservative-free insulin; p < 0.001) via a mechanism distinct from canonical PKC- and calcium-dependent pathways. Data-independent acquisition mass spectrometry (n = 6 donors) reveals…

MIIP Inhibits Colorectal Cancer Progression by Modulating Neutrophils Infiltration and Neutrophil Extracellular Trap Formation.

Neutrophil extracellular traps (NETs) contribute to colorectal cancer (CRC) progression, but the underlying regulatory mechanism remains unknown. This study explored how migration and invasion inhibitory protein (MIIP) influences CRC progression by regulating tumor-associated neutrophils (TANs) infiltration and NETs formation. On the basis of bioinformatics analysis and clinical CRC samples, we observed that a high density of neutrophils and a high abundance of NETs were correlated with low MIIP expression in CRC tissue as well as distant metastasis and poor prognosis. In vitro co-culture assa…

Rapid trapping and label-free optical characterization of single nanoscale extracellular vesicles and nanoparticles in solution.

Achieving high-efficiency, comprehensive analysis of single nanoparticles to determine their size, shape, and composition is essential for understanding particle heterogeneity with applications ranging from drug delivery to environmental monitoring. Existing techniques are hindered by low throughput, lengthy trapping times, irreversible particle adsorption, or limited characterization capabilities. Here, we introduce Interferometric Electrohydrodynamic Tweezers (IET), an integrated platform that combines rapid molecular trapping, interferometric scattering imaging, and Raman scattering to rapi

Macropinocytosis of neutrophil extracellular trap by gastric cancer cells promotes immune escape and peritoneal metastasis.

BACKGROUND: Patients with gastric cancer (GC) and peritoneal metastasis (PM) have poor prognoses due to drug resistance and metastatic relapse. The mechanism underlying PM recurrence remains unclear. OBJECTIVE: This study investigates the mechanisms of ascitic neutrophil extracellular traps (NETs) in promoting GC immune escape and PM recurrence. DESIGN: Ascites from a patient with GC were analysed using flow cytometry, immunofluorescence and electron microscopy. In vitro and in vivo experiments demonstrated NETs promoted GC immune evasion. RNA sequencing and functional models identified the …

MyD88 promotes neutrophil extracellular trap formation and gut leakiness after ethanol and burn injury.

Myeloid differentiation primary-response protein 88 (MyD88), an adapter protein used by many Toll-like receptors (TLRs), plays a critical role in initiating inflammatory responses. We previously showed that alcohol intoxication combined with burn injury causes intestinal tissue damage and leakiness. This is accompanied by increased intestinal neutrophil infiltration along with bacterial abundance and translocation. An overgrowth of bacteria can activate TLRs on intestinal epithelial cells (IECs), promoting inflammation and subsequent gut-tissue damage. To examine the role of MyD88 in intestine

Nociceptive neurons inhibit neutrophil extracellular trap formation via MLKL-licensed histone release.

Neutrophil-involved neuroinflammation in dorsal root ganglion (DRG) plays double-sword roles in chronic pain. How DRG neuron-neutrophil interaction contributes to chronic pain remains unclear. Here, we report that MLKL, a key molecule in necroptosis, is constitutively expressed in the nucleus of nociceptive neurons and binds to histone H3. Periphery inflammation disrupted MLKL/H3 interaction, leading to cytoplasmic translocation of MLKL and release of histone H3. Extracellular histone H3 induces neuronal hyperactivity, neutrophil extracellular trap (NET), and hyperalgesia, possibly through P2X

Skin vasculitis and cardiac eosinophil extracellular trap formation in eosinophilic granulomatosis with polyangiitis: A case report.

The pathophysiology of organ involvement in eosinophilic granulomatosis with polyangiitis is characterised by vasculitis and eosinophilic inflammation, which can be accompanied by eosinophil extracellular trap cell death (EETosis). Clinically, eosinophilic inflammation is often observed in the lungs, gastrointestinal tract, and heart, whereas vasculitis more frequently affects the peripheral nerves, skin, and kidneys. However, histopathological confirmation of vasculitis and EETosis in different organs within the same patient has rarely been reported. We report a patient with EGPA presenting w

Cathepsin C-driven enhancement of neutrophil extracellular trap formation contributes to acute lung injury.

Acute lung injury (ALI) is a severe pulmonary inflammatory disease with high morbidity and mortality rates. The pulmonary inflammatory phenotype of ALI is driven by the excessive aggregation and activation of neutrophils, triggering a cytokine storm. Notably, the extensive formation of neutrophil extracellular traps (NETs) has been demonstrated to play a critical role in ALI. Our results revealed significant overexpression of Cathepsin C (CTSC) in macrophages after LPS stimulation, which subsequently promoted massive NET formation through a positive feedback mechanism. Specifically, LPS stimul…

NLRP3 Inflammasome Activation Modulates Neutrophil Extracellular Trap Formation and Aggravates Airway Inflammation in Bronchiectasis.

Neutrophilic inflammation and Pseudomonas aeruginosa infection have been implicated in the pathogenesis and progression of bronchiectasis. However, the mechanisms underlying the inflammasome activation and neutrophil extracellular traps (NETs) formation, which are triggered by lipopolysaccharide (LPS), thereby fostering relentless neutrophilic inflammation and airway destruction, remain poorly understood. Here, we investigated how nucleotide-binding oligomerization domain-like receptor protein 3 (NLRP3) inflammasome activation modulates NET formation, epithelial inflammation, and injury in bro…

Sepsis-induced coagulopathy: recent insights on the role and clinical application of neutrophil extracellular trap formation.

Sepsis is characterized by life-threatening organ dysfunction caused by an uncontrolled immune response to infection. Neutrophils play a vital role in this process, which can lead to immunothrombosis and disseminated intravascular coagulation (DIC) via the formation of neutrophil extracellular traps (NETs). This study aimed to validate the impact of NETs biomarkers in evaluating their potential as diagnostic, prognostic, and therapeutic indicators in critical care for patients with sepsis. We conducted a case-control study with a 7-day follow-up to assess mortality in 138 sepsis patients, focu…
