Mechanistic characterization of iron-catalyzed oxidation of polysorbate 80: The role of ferrous iron, hydrogen peroxide, and superoxide.

We investigated the role of individual radical species during Fe-catalyzed oxidation of PS80. Solutions containing 1 gL-1 PS80 (0.1 % w/v) in 10 mM acetate buffer (pH 6) were exposed to various amounts of either Fe(II) or Fe(III), hydrogen peroxide (H2O2), and various enzymes or antioxidants. PS80 oxidation was measured using a fluorescence micelle assay (FMA) alongside LC-MS. Hydrogen peroxide inhibited PS80 oxidation in the presence of Fe(II) but promoted oxidation in the presence of Fe(III). Furthermore, Ferrostatin-1 (Fer-1), an antioxidant which is known to preferentially react with alkox

Evaluation of ecological impacts with ferrous iron addition in constructed wetland under perfluorooctanoic acid stress.

In this study, ferrous ion (Fe(II)) had the potential to promote ecological functions in constructed wetlands (CWs) under perfluorooctanoic acid (PFOA) stress. Concretely, Fe(II) at 30 mg/L and 20-30 mg/L even led to 11.37% increase of urease and 93.15-243.61% increase of nitrite oxidoreductase respectively compared to the control. Fe(II) promotion was also observed on Nitrosomonas, Nitrospira, Azospira, and Zoogloea by 1.00-6.50 folds, which might result from higher expression of nitrogen fixation and nitrite redox genes. These findings could be explanation for increase of ammonium removal by

Iron(II)-α-keto acid complexes of tridentate ligands on gold nanoparticles: the effect of ligand geometry and immobilization on their dioxygen-dependent reactivity.

Two mononuclear nonheme iron(II)-benzoylformate (BF) complexes [(6Me2-Me-BPA)Fe(BF)](ClO4) (1a) and [(6Me3-TPMM)Fe(BF)](ClO4) (1b) of tridentate nitrogen donor ligands, bis((6-methylpyridin-2-yl)methyl)(N-methyl)amine (6Me2-Me-BPA) and tris(2-(6-methyl)pyridyl)methoxymethane (6Me3-TPMM), were isolated and characterized. The structural characterization of iron(II)-chloro complexes indicates that the ligand 6Me2-Me-BPA binds to the iron(II) centre in a meridional fashion, whereas 6Me3-TPMM behaves as a facial ligand. Both the ligands were functionalized with terminal thiol for immobilization on

Labile Iron Pool of Isolated Escherichia coli Cytosol Likely Includes Fe-ATP and Fe-Citrate but not Fe-Glutathione or Aqueous Fe.

The existence of labile iron pools (LFePs) in biological systems has been recognized for decades, but their chemical composition remains uncertain. Here, the LFeP in cytosol from Escherichia coli was investigated. Mössbauer spectra of whole vs lysed cells indicated significant degradation of iron-sulfur clusters (ISCs), even using an unusually gentle lysis procedure; this demonstrated the fragility of ISCs. Moreover, the released iron contributed to the non-heme high-spin Fe(II) species in the cell, which likely included the LFeP. Cytosol batches isolated from cells grown with different levels

Selective pressure of PFOA on microbial community: Enrichment of denitrifiers harboring ARGs and the transfer of ferric-electrons.

Perfluorooctanoic acid (PFOA), a class of permanent organic pollutants, is frequently detected in surface and ground water, with the latter made up primarily of porous media (such as soils, sediments, and aquifers) that harbor microbial communities. Therefore, we investigated the effects of PFOA on water ecosystems and found that, under stimulation by 2.4 μM PFOA, denitrifiers were significantly enriched due to their hosting antibiotic resistant genes (ARGs), which were 1.45 times more abundant than the control. Furthermore, denitrifying metabolism was enhanced by Fe(II) electron donation. Spe

NO3- Promotes Nitrogen-Containing Disinfection Byproduct Formation in Corroded Iron Drinking Water Pipes.

Nitrogen-containing disinfection byproducts (N-DBPs) are highly toxic DBPs in drinking water. Though, under normal conditions, NO3- could not directly participate in disinfection reactions to generate N-DBPs, here, we first found that NO3- could promote the formation of N-DBPs in corroded iron drinking water pipes. The coexistence of corrosion produced Fe(II) and iron oxides is a critical condition for the transformation of N species; meanwhile, most of the newly generated N-DBPs had aromatic fractions. The Fe-O-C bond formed between iron corrosion products and natural organic matter promoted

FeS as excellent co-activator driving nano calcium peroxide oxidation for contaminants degradation: Performance and mechanisms.

In this study, ferrous sulfide (FeS) was introduced to nano calcium peroxide (nCP)/Fe(III) system to facilitate the generation of Fe(II), more than 90% of naphthalene (NAP) could be removed at a wide pH range of 3-9. As a heterogeneous reductant, FeS could mitigate competitive reactions with reactive oxygen species (ROS), which favored the NAP degradation. As evidenced by scavenging experiments, HO• was the major ROS contributing to NAP degradation. The role of sulfur species (S2-, SO32-, and S2O32-) in nCP/Fe(III) system was investigated with S2O32- showing the preferable reactivity in Fe(III

Characterization of Oyster Protein Hydrolysate-Iron Complexes and their In Vivo Protective Effects against Iron Deficiency-Induced Symptoms in Mice.

Iron is one of the trace mineral elements, and iron deficiency is a common phenomenon that negatively influences human health. Food-derived iron supplements were considered excellent candidates for improving this syndrome. In this work, oyster-protein hydrolysates (OPH) and ferrous chloride successfully formed the OPH-Fe complex (6 mg/mL, 40 °C, 30 min), where the main binding sites involved were the carboxyl and amino groups. The OPH-Fe complex showed no obvious changes in the secondary structure, while the iron changed the morphological appearance and also showed fluorescence quenching, an u

Computational Study of Aromatic Hydroxylation Catalyzed by the Iron-Dependent Hydroxylase PqqB Involved in the Biosynthesis of Redox Cofactor Pyrroloquinoline Quinone.

PqqB from Methylobacterium extorquens is a unique nonheme iron-dependent hydroxylase involved in the biosynthesis of redox cofactor pyrroloquinoline quinone (PQQ). A series of recent experiments have demonstrated that PqqB catalyzes the stepwise insertions of two oxygen atoms into the tyrosine ring of the diamino acid substrate, generating the quinone moiety of PQQ; however, the reaction details have not been elucidated yet. In this paper, on the basis of the crystal structures, the enzyme-substrate complex models were constructed, and the catalytic mechanism of PqqB was explored by performing

Large Fractionation in Iron Isotopes Implicates Metabolic Pathways for Iron Cycling in Boreal Shield Lakes.

Stable Fe isotopes have only recently been measured in freshwater systems, mainly in meromictic lakes. Here we report the δ56Fe of dissolved, particulate, and sediment Fe in two small dimictic boreal shield headwater lakes: manipulated eutrophic Lake 227, with annual cyanobacterial blooms, and unmanipulated oligotrophic Lake 442. Within the lakes, the range in δ56Fe is large (ca. -0.9 to +1.8‰), spanning more than half the entire range of natural Earth surface samples. Two layers in the water column with distinctive δ56Fe of dissolved (dis) and particulate (spm) Fe were observed, despite diffe

Removal of tetracycline by biochar-supported biogenetic sulfidated zero valent iron: Kinetics, pathways and mechanism.

The application of zero-valent iron (ZVI) is limited due to passivation and agglomeration. Therefore, biochar loading (MB) and biogenetic sulfidation via sulfate-reducing bacteria (SRB) were used to improve the reactivity of ZVI (BS-ZVI@MB) towards tetracycline (TC) degradation. Biochar provided more attachment sites for ZVI and SRB, thus alleviating the agglomeration. Additionally, quinone groups on biochar enhanced the electrons transfer through the measurement of electron donating/accepting capacities, and biogenetic sulfidation could inhibit the surface passivation of ZVI. Fe(Ⅱ/Ⅲ) produced

Mechanistic studies of the cofactor assembly in class Ib ribonucleotide reductases and protein affinity for MnII and FeII.

Ribonucleotide reductase (RNR) is an essential enzyme found in all organisms. The function of RNR is to catalyze the conversion of nucleotides to deoxynucleotides. RNRs rely on metallocofactors to oxidize a conserved cysteine in the active site of the enzyme into a thiyl radical, which then initiates nucleotide reduction. The proteins required for MnIII2-Y• cluster formation in class Ib RNRs are NrdF (β-subunit) and NrdI (flavodoxin). An oxidant is channeled from the FMN cofactor in NrdI to the dimanganese center in NrdF, where it oxidizes the dimanganese center and a tyrosyl radical (Y•) is f

Phosphorus recovery as vivianite from waste activated sludge via optimizing iron source and pH value during anaerobic fermentation.

This study presented an innovative method for phosphorus (P) recovery as vivianite from waste activated sludge (WAS) via optimizing iron dosing and pH value during anaerobic fermentation (AF). The optimal conditions for vivianite formation were in the pH range of 6.0-9.0 with initial PO43- >5 mg/L and Fe/P molar ratio of 1.5. Notably, FeCl3 showed advantages over ZVI for the simultaneous release of Fe2+ and PO43- during WAS fermentation, especially in acidic conditions. The FeCl3 dosing at pH 3.0 could contribute to 78.81% Fe2+ release and 85.69% of total PO43- release from WAS. They were ulti

Proteome Response of a Metabolically Flexible Anoxygenic Phototroph to Fe(II) Oxidation.

The oxidation of Fe(II) by anoxygenic photosynthetic bacteria was likely a key contributor to Earth's biosphere prior to the evolution of oxygenic photosynthesis and is still found in a diverse range of modern environments. All known phototrophic Fe(II) oxidizers can utilize a wide range of substrates, thus making them very metabolically flexible. However, the underlying adaptations required to oxidize Fe(II), a potential stressor, are not completely understood. We used a combination of quantitative proteomics and cryogenic transmission electron microscopy (cryo-TEM) to compare cells of Rhodop

The interaction of isopenicillin N synthase with homologated substrate analogues δ-(L-α-aminoadipoyl)-L-homocysteinyl-D-Xaa characterised by protein crystallography.

Isopenicillin N synthase (IPNS) converts the linear tripeptide δ-(L-α-aminoadipoyl)-L-cysteinyl-D-valine (ACV) into bicyclic isopenicillin N (IPN) in the central step in the biosynthesis of penicillin and cephalosporin antibiotics. Solution-phase incubation experiments have shown that IPNS turns over analogues with a diverse range of side chains in the third (valinyl) position of the substrate, but copes less well with changes in the second (cysteinyl) residue. IPNS thus converts the homologated tripeptides δ-(L-α-aminoadipoyl)-L-homocysteinyl-D-valine (AhCV) and δ-(L-α-aminoadipoyl)-L-homocys

An autocatalytic radical chain pathway in formation of an iron(IV)-oxo complex by oxidation of an iron(II) complex with dioxygen and isopropanol.

Evidence of an autocatalytic radical chain pathway has been reported in formation of a non-heme iron(IV)-oxo complex by oxidation of an iron(II) complex with dioxygen and isopropanol in acetonitrile at 298 K. The radical chain reaction is initiated by hydrogen abstraction from isopropanol by the iron(IV)-oxo complex.
