Candida albicans Sfl1/Sfl2 regulatory network drives the formation of pathogenic microcolonies.

Candida albicans is an opportunistic fungal pathogen that can infect oral mucosal surfaces while being under continuous flow from saliva. Under specific conditions, C. albicans will form microcolonies that more closely resemble the biofilms formed in vivo than standard in vitro biofilm models. However, very little is known about these microcolonies, particularly genomic differences between these specialized biofilm structures and the traditional in vitro biofilms. In this study, we used a novel flow system, in which C. albicans spontaneously forms microcolonies, to further characterize the arc

Advances in fungal sugar transporters: unlocking the potential of second-generation bioethanol production.

Second-generation (2G) bioethanol production, derived from lignocellulosic biomass, has emerged as a sustainable alternative to fossil fuels by addressing growing energy demands and environmental concerns. Fungal sugar transporters (STs) play a critical role in this process, enabling the uptake of monosaccharides such as glucose and xylose, which are released during the enzymatic hydrolysis of biomass. This mini-review explores recent advances in the structural and functional characterization of STs in filamentous fungi and yeasts, highlighting their roles in processes such as cellulase induct

Cholic Acid-Peptide Conjugates as Potent Antimicrobials against Interkingdom Polymicrobial Biofilms.

Interkingdom polymicrobial biofilms formed by Gram-positive Staphylococcus aureus and Candida albicans pose serious threats of chronic systemic infections due to the absence of any common therapeutic target for their elimination. Herein, we present the structure-activity relationship (SAR) of membrane-targeting cholic acid-peptide conjugates (CAPs) against Gram-positive bacterial and fungal strains. Structure-activity investigations validated by mechanistic studies revealed that valine-glycine dipeptide-derived CAP 3 was the most effective broad-spectrum antimicrobial against S. aureus and C.

Core N-Glycan Structures Are Critical for the Pathogenicity of Cryptococcus neoformans by Modulating Host Cell Death.

Cryptococcus neoformans is a human-pathogenic fungal pathogen that causes life-threatening meningoencephalitis in immunocompromised individuals. To investigate the roles of N-glycan core structure in cryptococcal pathogenicity, we constructed mutant strains of C. neoformans with defects in the assembly of lipid-linked N-glycans in the luminal side of the endoplasmic reticulum (ER). Deletion of ALG3 (alg3Δ), which encodes dolichyl-phosphate-mannose (Dol-P-Man)-dependent α-1,3-mannosyltransferase, resulted in the production of truncated neutral N-glycans carrying five mannose residues as a major

Systematic analysis of genome-wide fitness data in yeast reveals novel gene function and drug action.

We systematically analyzed the relationships between gene fitness profiles (co-fitness) and drug inhibition profiles (co-inhibition) from several hundred chemogenomic screens in yeast. Co-fitness predicted gene functions distinct from those derived from other assays and identified conditionally dependent protein complexes. Co-inhibitory compounds were weakly correlated by structure and therapeutic class. We developed an algorithm predicting protein targets of chemical compounds and verified its accuracy with experimental testing. Fitness data provide a novel, systems-level perspective on the c

Generalist core orchid mycorrhizal fungi structure symbiotic associations across multiple orchid species in the Ryukyu archipelago, an island biodiversity hotspot.

Plant-fungal symbioses influence plant community structure and dynamics, and orchids provide a model system for examining these interactions due to their strict dependence on orchid mycorrhizal fungi (OMF) throughout the life cycle. However, the organization of OMF communities across multiple orchid hosts in island ecosystems, and the identity of fungal lineages functioning as core symbionts, remain insufficiently resolved. We characterised OMF associated with 28 orchid species across the Ryukyu Archipelago, Japan using a two-step approach: preliminary Sanger sequencing followed by high-throug

Divergence in surface protein exposure between reference and clinical-derived Candida glabrata (Nakaseomyces glabratus) strains (CBS138 vs. BG2) - a preliminary proteomic perspective.

Candida glabrata (currently classified as Nakaseomyces glabratus) is an opportunistic fungal pathogen notable for its intrinsic antifungal tolerance and ability to persist in host environments. Although strain CBS138 has served as the principal model for genetic and functional studies, accumulating evidence indicates substantial intraspecies diversity that may shape virulence, immune interactions and stress adaptation. In particular, the widely used clinical isolate BG2 differs from CBS138 in genome structure, adhesin regulation and macrophage survival, yet the extent to which these difference

Antimicrobial Photodynamic Therapy Reveals Differential Species Susceptibility and Ecological Shifts in Candida albicans-Pseudomonas aeruginosa Mixed Model.

BACKGROUND: Candida albicans and Pseudomonas aeruginosa are frequently co-isolated from refractory endodontic infections. Antimicrobial photodynamic therapy (aPDT) is a promising adjunctive disinfection strategy; however, its efficacy against polymicrobial communities containing structurally distinct organisms remains unclear. OBJECTIVE: To evaluate methylene blue-mediated aPDT at three energy densities (15, 30, and 60 J/cm2) against a dual-species coculture model and to investigate the ecological consequences of differential species susceptibility. METHODS: Dual-species biofilms were establ

Targeting Sterylglucosidase A to Treat Aspergillus fumigatus Infections.

Invasive fungal infections are a leading cause of death in immunocompromised patients. Current therapies have several limitations, and innovative antifungal agents are critically needed. Previously, we identified the fungus-specific enzyme sterylglucosidase as essential for pathogenesis and virulence of Cryptococcus neoformans and Aspergillus fumigatus (Af) in murine models of mycoses. Here, we developed Af sterylglucosidase A (SglA) as a therapeutic target. We identified two selective inhibitors of SglA with distinct chemical scaffolds that bind in the active site of SglA. Both inhibitors ind

Comparison between human liver microsomes and the fungus Cunninghamella elegans for biotransformation of the synthetic cannabinoid JWH-424 having a bromo-naphthyl moiety analysed by high-resolution mass spectrometry.

PURPOSE: JWH-424, (8-bromo-1-naphthyl)(1-pentyl-1H-indol-3-yl)methanone, is a synthetic cannabinoid, which is a brominated analogue of JWH-018, one of the best-known synthetic cannabinoids. Despite the structural similarity to JWH-018, little is known about JWH-424 including its metabolism. The aim of the study was to compare human liver microsomes (HLM) and the fungus Cunninghamella elegans as the metabolism catalysts for JWH-424 to better understand the characteristic actions of the fungus in the synthetic cannabinoid metabolism. METHODS: JWH-424 was incubated with HLM for 1 h and Cunningha

A portable pathway specific transcription factor-promoter circuit enables heterologous production of fungal nonribosomal peptides.

Most fungal nonribosomal peptide synthetase (NRPS) biosynthetic gene clusters remain transcriptionally silent, severely restricting the discovery of bioactive peptide metabolites. Standard constitutive promoters fail to support robust expression of large multidomain NRPS enzymes. Here, we identify a conserved subgroup of Zn(ii)2Cys6 transcription factors (TFs) encoded within fungal NRPS clusters and functionally characterize TdaN, which triggers transcription from its cognate promoter P tdaA by recognizing paired CGG-type cis-regulatory motifs. The TdaN-P tdaA pair is repurposed as a portable

Transcriptomic and lipidomic analysis of the cryptococcal lung granuloma reveals macrophage boundary programs and sphingolipid remodeling.

Cryptococcosis is a life-threatening fungal disease caused by Cryptococcus neoformans. The immunocompetent response results in the formation of a lung granuloma, which contains the fungal cells. We have previously developed a mouse model of the cryptococcal granuloma that is histologically similar to the human lung granuloma. In this study, we characterize the lung granuloma in mice using transcriptomics and lipidomics, elucidating the immune cell types present and the transcriptional landscape of macrophages at the granuloma border. Macrophage subtypes are identified, which form a characteris

Atypical Presentation of Severe Fungal Necrotizing Fasciitis in a Patient with X-Linked Agammaglobulinemia.

X-linked agammaglobulinemia is a rare primary immunodeficiency due to a BTK mutation. The patients are characteristically deficient in peripheral B cells and serum immunoglobulins. While they are susceptible to infections caused by bacteria, enteroviruses, and parasites, fungal infections are uncommon in XLA patients. Here, we report a boy of Malay ethnicity who suffered from recurrent upper respiratory tract infections and severe progressive necrotizing fasciitis caused by Saksenaea erythrospora. Immunological tests showed a B cell deficiency and hypogammaglobulinemia. Whole-exome sequencing

Fungal sphingolipids: role in the regulation of virulence and potential as targets for future antifungal therapies.

INTRODUCTION: The antifungal therapy currently available includes three major classes of drugs: polyenes, azoles and echinocandins. However, the clinical use of these compounds faces several challenges: while polyenes are toxic to the host, antifungal resistance to azoles and echinocandins has been reported. AREAS COVERED: Fungal sphingolipids (SL) play a pivotal role in growth, morphogenesis and virulence. In addition, fungi possess unique enzymes involved in SL synthesis, leading to the production of lipids which are absent or differ structurally from the mammalian counterparts. In this rev

Zap1 regulates zinc homeostasis and modulates virulence in Cryptococcus gattii.

Zinc homeostasis is essential for fungal growth, as this metal is a critical structural component of several proteins, including transcription factors. The fungal pathogen Cryptococcus gattii obtains zinc from the stringent zinc-limiting milieu of the host during the infection process. To characterize the zinc metabolism in C. gattii and its relationship to fungal virulence, the zinc finger protein Zap1 was functionally characterized. The C. gattii ZAP1 gene is an ortholog of the master regulatory genes zafA and ZAP1 that are found in Aspergillus fumigatus and Saccharomyces cerevisiae, respect

Calcineurin target CrzA regulates conidial germination, hyphal growth, and pathogenesis of Aspergillus fumigatus.

The calcineurin pathway is a critical signal transduction pathway in fungi that mediates growth, morphology, stress responses, and pathogenicity. The importance of the calcineurin pathway in fungal physiology creates an opportunity for the development of new antifungal therapies that target this critical signaling pathway. In this study, we examined the role of the zinc finger transcription factor Crz1 homolog (CrzA) in the physiology and pathogenicity of the opportunistic human fungal pathogen Aspergillus fumigatus. Genetic replacement of the crzA locus in A. fumigatus resulted in a strain wi

Sulfonium Modification of Echinocandins: An Effective Strategy to Combat Intrinsically Resistant Cryptococcus via Membrane Disruption and Enhanced Cell Wall Biosynthesis Inhibition.

Fungal infections claim millions of lives annually and intrinsic resistance of Cryptococcus against echinocandins severely impedes clinical treatment. In this study, sulfonium covalent modification was performed based on natural pneumocandin B0 scaffold. The obtained derivatives exhibited potent activity against Candida species and reversed innate resistance of Cryptococcus neoformans, with MIC values reaching as low as 2 μg/mL. Structural modification elevated fungal membrane permeability and induced membrane depolarization, while enhancing the inhibitory effect on glucan synthase and disturb

Synthesis and Antifungal Activity of 4-Arylamino-1,2-Naphthoquinones-1,2,3-Triazoles Hybrids.

Quinones have been strategically combined with various heterocyclic scaffolds to enhance their biological properties. In this study, two new series of hybrid derivatives of 4-amino-1,2-naphthoquinones bearing a triazole ring were synthesized via a three-step procedure. These compounds were evaluated for antifungal activity and for their ability to generate reactive oxygen species (ROS). The compounds were active against both yeast and filamentous fungi. Notably, their antifungal effect was not linked to ROS generation or to interaction with lanosterol 14α-demethylase, the main accepted mechani
