The transcription factor GNC optimizes nitrogen use efficiency and growth by up-regulating the expression of nitrate uptake and assimilation genes in poplar.

Plants have evolved complex mechanisms to cope with the fluctuating environmental availability of nitrogen. However, potential genes modulating plant responses to nitrate are yet to be characterized. Here, a poplar GATA transcription factor gene PdGNC (GATA nitrate-inducible carbon-metabolism-involved) was found to be strongly induced by low nitrate. Overexpressing PdGNC in poplar clone 717-1B4 (P. tremula × alba) significantly improved nitrate uptake, remobilization, and assimilation with higher nitrogen use efficiency (NUE) and faster growth, particularly under low nitrate conditions. Autophagy controls resource allocation and protein storage accumulation in Arabidopsis seeds.

Autophagy is essential for nutrient recycling and plays a fundamental role in seed production and grain filling in plants. Autophagy participates in nitrogen remobilization at the whole-plant level, and the seeds of autophagy mutants present abnormal C and N contents relative to wild-type (WT) plants. It is well known that autophagy (ATG) genes are induced in leaves during senescence; however, expression of such genes in seeds has not yet been reported. In this study we show that most of the ATG genes are induced during seed maturation in Arabidopsis siliques. Promoter-ATG8f::UIDA and promoter

Fruit shape diversity in the Brassicaceae is generated by varying patterns of anisotropy.

Fruits exhibit a vast array of different 3D shapes, from simple spheres and cylinders to more complex curved forms; however, the mechanism by which growth is oriented and coordinated to generate this diversity of forms is unclear. Here, we compare the growth patterns and orientations for two very different fruit shapes in the Brassicaceae: the heart-shaped Capsella rubella silicle and the near-cylindrical Arabidopsis thaliana silique. We show, through a combination of clonal and morphological analyses, that the different shapes involve different patterns of anisotropic growth during three phas

EXPSO: A PSO-based ensemble gene expression classifier for plant species using histone and chromatin accessiblity data.

Gene expression in plants is regulated by complex interactions among chromatin features, including histone modifications and chromatin accessibility. Accurate prediction of gene-expression states from epigenomic information can provide insights into transcriptional regulation and facilitate functional genomic studies. However, the nonlinear and high-dimensional nature of epigenomic data presents challenges for conventional statistical approaches. Here, we developed EXPSO, a PSO-assisted ensemble learning framework for classifying plant genes into high- and low-expression states using integrate

Lysine Biosynthesis Defines a Metabolic Checkpoint for Gibberellin-Mediated Growth in Arabidopsis thaliana.

Lysine represents a metabolic hub linking mitochondrial energy status with amino acid catabolism, yet its role in coordinating plant growth and development remains poorly characterised. The dapat mutant, which is deficient in L,L-diaminopimelate aminotransferase (DAPAT), exhibits severe growth inhibition associated with broad metabolic imbalance. This metabolic reprogramming is characterised by simultaneous depletion of carbohydrates (sugars and starch), and organic acids, accompanied by elevated amino acid pools, reflecting a profound disruption of carbon-nitrogen balance. These metabolic con

Plant Volatile Methyl Salicylate Primes Wheat Defense Against the Grain Aphid by Altering the Synthesis of Defense Metabolites.

Wheat (Triticum aestivum L.) is one of the most important staple crops all over the world. Its productivity is adversely affected by aphid infestation. Plant volatiles play a critical role in plant communication, inducing direct and indirect defenses against insect pests. However, little is known about the priming mechanism of key volatiles in wheat. To determine whether and how plant volatile induced defense priming in wheat against the grain aphid Sitobion avenae, a combination of insect bioassays, phytohormone and defense metabolite quantification, and transcriptome analyses were performed

Formation of embryogenic cell clumps from carrot epidermal cells is suppressed by 5-azacytidine, a DNA methylation inhibitor.

Using a direct somatic embryogenesis system in carrot, we examined the role of DNA methylation in the change of cellular differentiation state, from somatic to embryogenic. 5-Azacytidine (aza-C), an inhibitor of DNA methylation suppressed the formation of embryogenic cell clumps from epidermal carrot cells. Aza-C also downregulated the expression of DcLEC1c, a LEC1-like embryonic gene in carrot, during morphogenesis of embryos. A carrot DNA methyltransferase gene, Met1-5 was expressed transiently after the induction of somatic embryogenesis by 2,4-dichlorophenoxyacetic acid (2,4-D), before the

RNA-Seq and Electrical Penetration Graph Revealed the Role of Grh1-Mediated Activation of Defense Mechanisms towards Green Rice Leafhopper (Nephotettix cincticeps Uhler) Resistance in Rice (Oryza sativa L.).

The green rice leafhopper (GRH, Nephotettix cincticeps Uhler) is one of the most important insect pests causing serious damage to rice production and yield loss in East Asia. Prior to performing RNA-Seq analysis, we conducted an electrical penetration graph (EPG) test to investigate the feeding behavior of GRH on Ilpum (recurrent parent, GRH-susceptible cultivar), a near-isogenic line (NIL carrying Grh1) compared to the Grh1 donor parent (Shingwang). Then, we conducted a transcriptome-wide analysis of GRH-responsive genes in Ilpum and NIL, which was followed by the validation of RNA-Seq data b

A genome-wide association study reveals a novel regulator of ovule number and fertility in Arabidopsis thaliana.

Ovules contain the female gametophytes which are fertilized during pollination to initiate seed development. Thus, the number of ovules that are produced during flower development is an important determinant of seed crop yield and plant fitness. Mutants with pleiotropic effects on development often alter the number of ovules, but specific regulators of ovule number have been difficult to identify in traditional mutant screens. We used natural variation in Arabidopsis accessions to identify new genes involved in the regulation of ovule number. The ovule numbers per flower of 189 Arabidopsis acc

Identification of Freezing-Responsive microRNAs and Their Targets in Chinese Jujube by Small RNA and Degradome Sequencing.

The jujube tree fruit remains a primary fruit in northern China, yet its geographical distribution and yield are significantly constrained by freezing stress during winter. Numerous studies have highlighted the pivotal regulatory function of microRNAs (miRNAs) in plant responses to low-temperature stress. Nevertheless, the specific miRNAs involved in the response to low temperatures and their associated gene networks in Ziziphus jujuba Mill are not well understood. In this investigation, we utilized high-throughput sequencing to analyze small RNA libraries from branches subjected to temperatur

Overexpression of Arabidopsis ATX1 retards plant growth under severe copper deficiency.

The efficient delivery of CpG oligodeoxynucleotides (CpG ODN) adjuvants is constrained by the limited endosomal escape capability of lipid nanoparticles (LNPs). To evaluate this critical parameter, we constructed an in vitro escape evaluation platform that integrates serum protein corona simulation, a biomimetic phospholipid bilayer barrier, and pH regulation. This platform enables quantitative analysis of the transmembrane delivery mechanism of LNP-CpG ODN while overcoming limitations of cellular models for studying carrier-biointerface interactions. Through optimization of the biomimetic mem

Identification of ZmGLN5, a candidate gene for resistance to Gibberella ear rot in maize.

BACKGROUND: Gibberella ear rot (GER), caused by the fungal pathogen Fusarium graminearum (F. graminearum), severely affects maize yield and grain quality worldwide. Although numerous genetic loci associated with maize resistance to GER have been identified, the functional genes underlying resistance to GER remain largely uncharacterized. METHODS AND RESULTS: In this study, we performed comparative transcriptome analysis using two maize lines with contrasting resistance to GER. Differently expressed genes were mainly involved in defense-related pathway, including phenylpropanoid biosynthesis,

Ectopic expression of a Penicillium oxalicum cyclophilin gene in Arabidopsis thaliana imparts tolerance to cold stress.

Cyclophilins are ubiquitous proteins that are essential for cis-trans isomerisation of peptide bonds preceding the proline residue. Apart from catalysing the cis-trans isomerization, these proteins are also implicated in diverse cellular functions. We investigated the role of a cyclophilin gene PoxCYP62-2, isolated from a halotolerant strain of Penicillium oxalicum, in abiotic stress tolerance of plants. Ectopic expression of PoxCYP62-2 gene in transgenic Arabidopsis thaliana plants conferred tolerance to cold stress. The transgenic plants exhibited higher rosette fresh and dry mass, lower MDA

Genome-wide identification and cold-stress-responsive expression analysis of the NOX gene family in Cucumis melo.

NADPH oxidases (NOXs) are crucial enzymes for reactive oxygen species (ROS) generation in plants and play vital roles in growth, development, and stress responses. To elucidate the sequence characteristics of the NOX gene family and its low-temperature response patterns in melon (Cucumis melo L.), this study conducted genome-wide identification and expression profiling of NOX family members using bioinformatics analysis, RNA-seq transcriptome sequencing, and real-time quantitative PCR (RT-qPCR). The results revealed that eight NOX members were identified in the melon genome, distributed across

Silencing of RBOHF2 Causes Leaf Age-Dependent Accelerated Senescence, Salicylic Acid Accumulation, and Powdery Mildew Resistance in Barley.

Plant RBOH (RESPIRATORY BURST OXIDASE HOMOLOGS)-type NADPH oxidases produce superoxide radical anions and have a function in developmental processes and in response to environmental challenges. Barley RBOHF2 has diverse reported functions in interaction with the biotrophic powdery mildew fungus Blumeria graminis f. sp. hordei. Here, we analyzed, in detail, plant leaf level- and age-specific susceptibility of stably RBOHF2-silenced barley plants. This revealed enhanced susceptibility to fungal penetration of young RBOHF2-silenced leaf tissue but strongly reduced susceptibility of older leaves w

The soybean gene GmSP1L-8 improves salt tolerance through the enhancement of the ROS scavenging system.

Microtubule-associated proteins (MAPs) are key regulators of plant stress responses. Although salt stress-induced microtubule dynamics have been widely observed, how specific microtubule-associated factors coordinately regulate plant salt tolerance remains to be further elucidated. In this study, through genome-wide identification and expression profiling of the GmSP1L gene family in soybean (Glycine max), we screened and identified GmSP1L-8, a protein that is significantly induced by salt stress. Subcellular localization analysis revealed that GmSP1L-8 localizes to both the nucleus and cytopl

Generation and Characterization of Novel Low-Amylose Rice Germplasm via Prime Editing of the Wx Promoter.

Amylose content (AC) is primarily regulated by the Wx gene and acts as a key determinant of rice eating and cooking quality. Natural low-AC alleles are limited in fine-tuning effects across genetic backgrounds, and studies exploiting natural allelic variation are nearing saturation. Here, we employed prime editing (PE) to generate precise promoter deletions near the transcription start site (TSS) of Wx in japonica rice Huaidao 5, obtaining two homozygous mutant lines: Wx5d harboring the designed 5 bp deletion and Wx32d carrying an unintended 32 bp promoter deletion. Compared with the wild-type

The knockout of ClaCSLH1 induced dwarfing in watermelon.

In agriculture, selecting ideal plant types with desirable traits, such as dwarfing and upright stem structures, significantly enhances crop yield and quality by optimizing light absorption, spatial efficiency, and nutrient utilization. Developing new varieties of dwarf watermelon is a crucial objective in watermelon breeding. In this study, we constructed an F2 population using the wild-type V063 as the paternal parent and the dwarf variety dw-n as the maternal parent. The dwarfing trait was found to be governed by a pair of recessive alleles. Through bulk segregant analysis sequencing (BSA-s

Antisense Oligonucleotide as a New Technology Application for CsLOB1 Gene Silencing Aiming at Citrus Canker Resistance.

Citrus canker disease, caused by Xanthomonas citri subsp. citri, poses a significant threat to global citrus production. The control of the disease in the field relies mainly on the use of conventional tools such as copper compounds, which are harmful to the environment and could lead to bacterial resistance. This scenario stresses the need for new and sustainable technologies to control phytopathogens, representing a key challenge in developing studies that translate basic into applied knowledge. During infection, X. citri subsp. citri secretes a transcriptional activator-like effector that e
