Modulation of Ras signaling pathway by exosome miRNAs in T-2 toxin-induced chondrocyte injury.

This study aims to investigate the impact of T-2 toxin on the regulation of downstream target genes and signaling pathways through exosome-released miRNA in the development of cartilage damage in Kashin-Beck disease (KBD). Serum samples from KBD patients and supernatant from C28/I2 cells treated with T-2 toxin were collected for the purpose of comparing the differential expression of exosomal miRNA using absolute quantitative miRNA-seq. Target genes of differential exosomal miRNAs were identified using Targetscan and Miranda databases, followed by GO and KEGG enrichment analyses. Validation of

The International Consensus Classification (ICC) of hematologic neoplasms with germline predisposition, pediatric myelodysplastic syndrome, and juvenile myelomonocytic leukemia.

Updating the classification of hematologic neoplasia with germline predisposition, pediatric myelodysplastic syndrome (MDS), and juvenile myelomonocytic leukemia (JMML) is critical for diagnosis, therapy, research, and clinical trials. Advances in next-generation sequencing technology have led to the identification of an expanding group of genes that predispose to the development of hematolymphoid neoplasia when mutated in germline configuration and inherited. This review encompasses recent advances in the classification of myeloid and lymphoblastic neoplasia with germline predisposition summa

MicroRNA Expression Differences in Blood-Derived CD19+ B Cells of Methotrexate Treated Rheumatoid Arthritis Patients.

Rheumatoid arthritis (RA) is a complex disease with a wide range of underlying susceptibility factors. Recently, dysregulation of microRNAs (miRNAs) in RA have been reported in several immune cell types from blood. However, B cells have not been studied in detail yet. Given the autoimmune nature of RA with the presence of autoantibodies, CD19+ B cells are a key cell type in RA pathogenesis and alterations in CD19+ B cell subpopulations have been observed in patient blood. Therefore, we aimed to reveal the global miRNA repertoire and to analyze miRNA expression profile differences in homogenous

The clinical and genetic spectrum of pediatric hypertrophic cardiomyopathy manifesting before one year of age.

BACKGROUND: Hypertrophic cardiomyopathy (HCM) presents a wide range of clinical scenarios depending on the age of manifestation, with a less favorable prognosis in children. The genetic spectrum and clinical causes of HCM diagnosed before one year of age is rarely reported. METHODS: We analyzed the genetic causes and genotype-phenotype correlations in 68 children diagnosed with HCM during the first year of life. Genetic analysis was performed using targeted gene sequencing (39 HCM-related genes), followed by whole-exome sequencing for genotype-negative cases. The genetic data were correlated

Distinct sets of genetic alterations in melanoma.

BACKGROUND: Exposure to ultraviolet light is a major causative factor in melanoma, although the relationship between risk and exposure is complex. We hypothesized that the clinical heterogeneity is explained by genetically distinct types of melanoma with different susceptibility to ultraviolet light. METHODS: We compared genome-wide alterations in the number of copies of DNA and mutational status of BRAF and N-RAS in 126 melanomas from four groups in which the degree of exposure to ultraviolet light differs: 30 melanomas from skin with chronic sun-induced damage and 40 melanomas from skin wit

Clonality studies and N-ras and p53 mutation analysis of hematopoietic cells in Fanconi anemia.

Patients with Fanconi anemia (FA) have an extraordinary predisposition to acute myelogenous leukemia (AML). The genetic mechanisms underlying the neoplastic transformation of FA hematopoietic cells are unknown. In this study, we have investigated the molecular features of hematopoiesis in the course of FA at different stages of the disease, including aplastic anemia, myelodysplastic syndrome (MDS), and AML. The analysis focused on defining the clonality status of FA hematopoiesis as well as the putative involvement of N-ras, a dominantly acting oncogene, and p53, a tumor suppressor gene, which

Comprehensive bioinformatic and experimental approaches to analyze miR-200a, miR-1, and miR-548-3p expression and their targeted oxidative stress and glucose metabolic related hub genes in rheumatoid arthritis.

Rheumatoid arthritis is a chronic autoimmune disorder characterized by persistent synovial inflammation, oxidative stress, and metabolic reprogramming. Emerging evidence highlights miRNAs as key post-transcriptional regulators linking these pathogenic processes. Among them, miR-200a, miR-1, and miR-548-3p have been implicated in oxidative stress responses and glucose metabolism; however, their roles in RA remain largely unexplored. This study integrates bioinformatic prediction and experimental validation to examine the expression profiles of these miRNAs, identify their target genes related t

The association between inefficient repair of oxidative DNA lesions and common polymorphisms of the key base excision repair genes as well as their expression levels in patients with rheumatoid arthritis.

INTRODUCTION: Rheumatoid arthritis (RA) is a common autoimmune heterogeneous joint disease of still unknown etiology. The pathology of RA leads to chronic inflammation of the joint tissues, which causes joint cartilage and bone destruction. One of the characteristic features of RA is oxidative stress, most likely induced and stimulated by inappropriate B- and T-cell activity. This results in accumulation of oxidative DNA lesions in peripheral blood mononuclear cells (PBMCs) isolated from RA patients, as we have shown previously. We have hypothesized that oxidative stress together with an impai

Circulatory age-associated B cells: Their distinct transcriptomic characteristics and clinical significance in drug-naïve patients with rheumatoid arthritis.

Age-associated B cells (ABCs) have been implicated in the pathogenesis of autoimmune diseases. However, the global gene expression and clinical significance of circulatory ABCs in rheumatoid arthritis (RA) remain poorly understood. Here, single-cell RNA sequencing identified nine B cell subsets in peripheral blood of RA patients, including ABCs. Increased phagocytosis and antigen presentation were functionally enriched by the genes expressed differentially in ABCs. Network analysis and in vitro experiments demonstrated SYK as a key regulator defining the myeloid-like phenotypes in ABCs. Flow c

H-ras oncogene activation in invasive UVR-induced corneal sarcomas of the opossum Monodelphis domestica.

Chronic exposure to ultraviolet radiation (UVR) induces corneal sarcomas in the South American opossum Monodelphis domestica. Cell lines are readily established from these tumors. Northern blotting of mRNA from six such cell lines revealed high expression of the H-ras oncogene. H-ras cDNA from an eye tumor cell line was cloned and characterized; the germline sequence of codons 12, 13, and 61 was confirmed by examination of H-ras sequences amplified from liver DNA by the polymerase chain reaction. The Monodelphis H-ras coding sequence is 84-89% identical to that of other vertebrates at the nucl

T-cell receptor-major histocompatibility complex genetic interactions in rheumatoid arthritis.

Rheumatoid arthritis (RA) is a complex multifactorial illness. It is proposed that multiple genes involved in immune recognition interact to produce a state of genetic susceptibility for RA. The role of HLA and T-cell receptor and other background genes is discussed, with an emphasis on their role in shaping the T-cell repertoire.

Dissecting ARL15 Function in Rheumatoid Arthritis: Insights From Ex Vivo and in Vitro Synovial Fibroblast Models.

OBJECTIVES: ARL15, coding for a small GTPase, was identified as a non-HLA susceptibility gene in rheumatoid arthritis (RA) through a GWAS in a North Indian cohort, with serum adiponectin and ARL15 levels higher in RA patients with the associated genotype. This study aimed to delineate the functional role of ARL15 in RA pathobiology. METHODS: Differential transcriptomics in both ex vivo RA synovial fibroblasts and in vitro MH7A cells using a gene knockdown (KD) approach and standard analyses pipeline were performed to obtain insights into ARL15's role. RESULTS: In RASF, ARL15 KD led to downre

Integrating novel network pharmacology and experimental validation to study the mechanism of Wutou decoction against rheumatoid arthritis.

Wutou decoction (WTD) is a traditional remedy in Chinese medicine for rheumatoid arthritis (RA). However, its active ingredients are complex, and how it counteracts RA is still not clear. The objective of this research was to investigate the key components and targets of WTD action. RA-related genes were analyzed using the GSE55235 dataset. The active components of WTD were analyzed using online databases. Component-target-pathology (CTP) networks were built using the Cytoscape software. The functions and pathways of the key targets were analyzed using GO and KEGG. The component-target interac

Glycoprotein Acetyls Is a Novel Biomarker Predicting Cardiovascular Complications in Rheumatoid Arthritis.

The relationship between rheumatoid arthritis (RA) and early onset atherosclerosis is well depicted, each with an important inflammatory component. Glycoprotein acetyls (GlycA), a novel biomarker of inflammation, may play a role in the manifestation of these two inflammatory conditions. The present study examined a potential mediating role of GlycA within the RA-atherosclerosis relationship to determine whether it accounts for the excess risk of cardiovascular disease over that posed by lipid risk factors. The UK Biobank dataset was acquired to establish associations among RA, atherosclerosis,

Genetics of ACPA-positive rheumatoid arthritis: the beginning of the end?

Heritability is a measure for the contribution of genetic variation to the variation in liability to disease and for rheumatoid arthritis (RA) had previously been estimated to be about 60%. This has been recently confirmed and could show that the heritability of anti-citrullinated protein autoantibody (ACPA)-positive and ACPA-negative RA is similar. Apart from gender, the main known genetic factor is HLA, and its contribution to genetic variation has previously been estimated as 37% but recent studies indicate that this figure may be too high. HLA-linked genes, and in particular the HLA-DRB1 S

Local RAS and inflammatory factors are involved in cardiovascular hypertrophy in spontaneously hypertensive rats.

The renin-angiotensin system (RAS) and inflammation may play a decisive role in hypertensive end organ damage (EOD) including left ventricular (LV) hypertrophy and vascular remodeling. It is not clear whether the local RAS and inflammatory factors are involved in hypertensive EOD, and which RAS or inflammatory factors (systematic or local) is more important in EOD. Hemodynamics, cardiovascular hypertrophy, angiotensin II (Ang II) concentration and inflammatory factors in spontaneously hypertensive rats (SHR) were measured. mRNA expression of tissue RAS and inflammatory factors in SHR was assay
