Liposomes have a direct effect on multiple myeloma: a Mendelian randomization study.

BACKGROUND: Multiple myeloma (MM), a malignant disease of plasma cells originating in the bone marrow, is influenced significantly by genetic factors. Although plasma liposomes have been linked to MM, the nature of their potential causal relationship remains to be elucidated. This study aims to explore this relationship using Mendelian randomization (MR) analysis. METHODS: Liposome-associated genetic instrumental variables (IVs) were identified from plasma lipidomics data of 7,174 Finnish individuals within a Genome-Wide Association Study (GWAS) pooled database. A MM pooled dataset was source

Managing Trait Trade-Offs in Tomato Breeding Through Genome Editing.

Tomato improvement often involves trade-offs between fruit yield and quality, sweetness and fruit size, disease resistance and growth, reproductive performance and stress adaptation, and shelf life and flavour. Although CRISPR-Cas-mediated genome editing enables targeted modification of tomato genes, DNA-level precision does not guarantee an agronomically favourable phenotype because pleiotropy, functional redundancy, allele dosage, linkage, epistasis, genetic background, and environmental conditions can alter editing outcomes. This review evaluates genome editing as a strategy to manage, rath

Tourette syndrome.

Tourette syndrome is characterized by the presence of motor and phonic tics. Individuals with Tourette syndrome can have a spectrum of related behaviours including attention deficit hyperactivity features, obsessions, compulsions, impulsions and complex tics. The pathophysiology of Tourette syndrome is thought to reflect genetic influences on the development and function of cortical-striatal-thalamic-cortical and broader cortical-subcortical circuitry implicated in motor and inhibitory control and interoceptive processing, which may contribute to the characteristic 'urge-tic-relief' cycles. Early-life rotavirus infection susceptibility and later gastrointestinal cancer protection: Reverse antagonistic pleiotropy and potential vaccine benefits.

Rotavirus (RV) infection is a leading cause of gastroenteritis in children under five, often causing severe dehydration and hospitalization. The genetic factors influencing host susceptibility to severe RV infection remain poorly understood. We conducted whole-exome sequencing (WES) in children with severe RV infection and compared their profiles to those of healthy controls. Analyses identified seven candidate Single Nucleotide Polymorphisms (SNPs) across distinct genes, several of which are functionally linked to gastrointestinal cancer pathways. Transcriptomic data from acutely infected pat

Causal association between depression and myalgia-related disorders: A bidirectional mendelian randomization study.

OBJECTIVE: The connection between depression and myalgia-related disorders remains unclear. We investigated the bidirectional causal relationship between depression and five such disorders using Mendelian randomization (MR). METHOD: Genetic instrumental variables for depression were obtained from a GWAS of 170,756 individuals with depression and 329,443 controls. The primary two-sample MR analysis used the inverse variance-weighted (IVW) method. Robustness was tested with MR Egger, weighted median, and weighted mode approaches. We also conducted heterogeneity, pleiotropy, and sensitivity anal

Genetic evidence strengthens the connection between emotional disorders and TMD-related pain: A two-sample Mendelian randomization study.

OBJECTIVES: This study aims to evaluate the genetic association between emotional disorders and TMD-related pain through two-sample Mendelian randomization analysis. DESIGN: Single-nucleotide polymorphisms (SNPs) related to emotional disorders (worry, nerves, or depression) were selected from genome-wide association studies (GWAS) from UK Biobank consortia, and related these to SNPs from FinnGen consortia. The inverse-variance weighted (IVW) was used as the primary effect estimate between emotional disorders and TMD-related pain, and various methods were applied to test the reliability and st

The correlation between immune cells and endometriosis: a bidirectional two-sample mendelian randomization study.

OBJECTIVES: Endometriosis (EM), a prevalent estrogen-dependent inflammatory disorder affecting women of reproductive age, is characterized by the presence of endometrial-like tissue outside the uterus, resulting in pelvic scarring, pain, and infertility. Although the pathogenesis of EM remains poorly understood, there is growing evidence suggesting the involvement of the immune system in its etiology, pathophysiology, and associated morbidities such as pain, infertility, and adverse pregnancy outcomes. While previous studies have indicated a close relationship between the immune system and EM,

Dietary factors and rheumatoid arthritis: new perspectives from a Mendelian randomisation analysis.

Rheumatoid arthritis (RA) is a prevalent autoimmune disease, and there is growing evidence suggesting a potential correlation between dietary factors and the pathogenesis of this condition. In order to investigate the causal relationship between diet and RA, we conducted a two-sample Mendelian randomisation (MR) analysis to examine the causal associations between twenty-two dietary factors and RA. Summary data from genome-wide association studies (GWAS) of RA were obtained from large GWAS meta-analyses. GWAS summary data for twenty-two dietary factors were obtained from UK Biobank. Inflammatory bowel disease and bladder cancer risk: based on a Mendelian randomization study.

BACKGROUND: Prior epidemiological observational studies have duly documented a correlative link between inflammatory bowel disease (IBD) and bladder cancer (BC); however, the establishment of a definitive causal relationship has remained elusive. The principal objective of this meticulous investigation was to rigorously evaluate the causal nexus between IBD and BC, employing the robust methodology of Mendelian randomization (MR) analysis. METHODS: We meticulously performed both univariate and multivariate Mendelian randomization (MVMR) analyses employing publicly accessible genome-wide associ

Generation of a new mouse line with conditionally activated signaling through the BMP receptor, ACVR1: A tool to characterize pleiotropic roles of BMP functions.

BMP signaling plays pleiotropic roles in various tissues during embryogenesis and after birth. We have previously generated a constitutively activated Acvr1(ca-Acvr1) transgenic mouse line (line L35) through pronuclei injection to investigate impacts of enhanced BMP signaling in a tissue specific manner. However, line L35 shows a restricted expression pattern of the transgene. Here, we generated another ca-Acvr1 transgenic line, line A11, using embryonic stem (ES) transgenesis. The generated line A11 shows distinctive phenotypes from line L35, along with very limited expression levels of the t

Many faces of SMCHD1.

The chromatin scaffolding protein SMCHD1 (structural maintenance of chromosomes flexible hinge domain containing 1) was previously shown to have diverse roles in X-chromosome inactivation, imprinting and double-strand break repair, and mutations in SMCHD1 contribute to a type of muscular dystrophy. Now, development of the nose and eyes is added to its list of functions.

The Clinical and Genetic Landscape of Junctional Skin Disorder in Children: A Scoping Review.

Cellular junctions, including desmosomes, hemidesmosomes, adherens, tight, and gap junctions, are essential for epidermal integrity, barrier function, and cell signaling. Variants in genes encoding proteins involved in cellular junctions lead to a spectrum of skin disorders, commonly referred to as junctional genodermatoses, often presenting with phenotypic overlaps and multisystemic involvement in pediatric patients. This systematic scoping review aims to comprehensively map pediatric genodermatoses, correlating specific genetic variants with clinical manifestations to improve diagnostic and

Dissecting immune cell-specific genetics in migraine: a multi-omics framework for target discovery and therapeutic prioritization.

BACKGROUND: Migraine is a highly prevalent neurological disorder with complex pathophysiology and limited treatment efficacy. Emerging evidence suggests immune cell involvement in migraine, but the specific regulatory mechanisms remain unclear. This study aimed to identify immune cell-type-specific genetic drivers of migraine and prioritize potential therapeutic targets. METHODS: We conducted a Mendelian randomization (MR) analysis integrating single-cell expression quantitative trait loci (sc-eQTL) data from 14 immune cell types with large-scale genome-wide association studies (GWAS) of migr

Genetic risk factors in rheumatoid arthritis: A Mendelian randomization study of chronic kidney disease in European populations.

Rheumatoid arthritis (RA) is a heritable autoimmune disease linked to chronic kidney disease (CKD) in observational studies. However, whether this association is causal and driven by shared genetic risk remains unclear, warranting genetic investigation. To investigate possible causal relationships between RA and different CKD subtypes, we used Mendelian randomization (MR) analyses with data from genome-wide association studies. The inverse variance weighted (IVW) methodology was the main method, and sensitivity analyses were added to improve the validity of the causal estimations. Our analysis

Hypertension and frozen shoulder: A bidirectional 2-sample Mendelian randomization study.

Observational studies have demonstrated an association between hypertension and frozen shoulder (FS). However, there is still a lack of clarity about their possible causal relationship. This study aimed to explore whether a genetic relationship exists between hypertension and FS bidirectional through 2-sample Mendelian randomization (MR) analysis. In this study, we investigated the following using data from genome-wide association studies. Exposure-related genetic variants that are independent of each other, were rigorously screened for use as instrumental variables (IVs). To investigate the c

Is High-Density Lipoprotein Cholesterol Causally Related to Kidney Function? Evidence From Genetic Epidemiological Studies.

OBJECTIVE: A recent observational study with almost 2 million men reported an association between low high-density lipoprotein (HDL) cholesterol and worse kidney function. The causality of this association would be strongly supported if genetic variants associated with HDL cholesterol were also associated with kidney function. APPROACH AND RESULTS: We used 68 genetic variants (single-nucleotide polymorphisms [SNPs]) associated with HDL cholesterol in genome-wide association studies including >188 000 subjects and tested their association with estimated glomerular filtration rate (eGFR) using

A pleiotropic QTL on 2p influences serum Lp-PLA2 activity and LDL cholesterol concentration in a baboon model for the genetics of atherosclerosis risk factors.

Lipoprotein-associated phospholipase A(2) (Lp-PLA(2)), the major portion of which is bound to low-density lipoprotein, is an independent biomarker of cardiovascular disease risk. To search for common genetic determinants of variation in both Lp-PLA(2) activity and LDL cholesterol (LDL-C) concentration, we assayed these substances in serum from 679 pedigreed baboons. Using a maximum likelihood-based variance components approach, we detected significant evidence for a QTL affecting Lp-PLA(2) activity (LOD=2.79, genome-wide P=0.039) and suggestive evidence for a QTL affecting LDL-C levels (LOD=2.
