Influenza A virus (IAV) recognizes terminal sialic acid of sialoglyco-conjugates on host cells through the viral envelope glycoprotein hemagglutinin (HA), followed by initiation of entry into the cells. Molecular species of sialic acid are largely divided into two moieties: N-acetylneuraminic acid (Neu5Ac) and N-glycolylneuraminic acid (Neu5Gc). A receptor for IAV infection generally means Neu5Ac. Almost all equine IAVs and some human, swine, and duck IAVs bind not only to Neu5Ac but also to Neu5Gc. In nonhuman animals, Neu5Gc has been detected in swine and equine tracheas and the duck colon,

Mycobacterium avium complex (MAC) is a group of non-tuberculous mycobacteria that cause tuberculosis-like diseases in humans. Unlike Mycobacterium tuberculosis, MAC expresses high levels of glycopeptidolipids (GPLs) containing a well-defined tetrapeptide-amino alcohol core, composed of D-phenylalanine, D-allo-threonine, D-alanine, and L-alaninol, that is modified with a fatty acid and sugar residues. Surprisingly, however, a careful scrutiny of the mass spectrum of MAC GPLs revealed the presence of ions that could not readily accountable for the known GPL structure. The magnitude of the ions w

This study analyzed the structural differences of the carbohydrate chains of circulating free alpha-submit (alpha-SU) hypersecreted in various non-tumoral (primary hypothyroids, postmenopausal women, patients with chronic uremia, normal fetuses) and tumoral (gut carcinoids, TSH-, GH- and pure alpha-secreting pituitary adenomas) clinical conditions. Carbohydrate structures of free alpha-SU were investigated by means of lectin affinity chromatography using Concanavalin A (Con-A), which allows the separation of free alpha-SU in three different fractions (unbound = UB, weakly bound = WB and firmly

Carbohydrate structures constitute important factors in the pathogenesis of herpesvirus infections for three reasons: (i) Human herpesviruses specify several biologically active glycoproteins, where co-translational addition of carbohydrate units is essential for generation of functioning viral glycoproteins. (ii) Cellular carbohydrate structures are parts of the cellular receptor complex at least for some herpes-viruses. (iii) Peripheral carbohydrate structures are engaged in modulation of the immunological properties of herpesvirus glycoproteins. The present review will consider all these th

Polysaccharide-peptide conjugates constitute an important class of biomaterials for tissue engineering scaffolds and glycoconjugate vaccines. Yet, benign and chemoselective synthetic strategies that operate under aqueous, physiologically relevant conditions remain scarce. Herein, we present a proof-of-concept approach for the traceless conjugation of N-terminal l-cysteines to alginate. A glycine-derived thioester functionality was synthesized and covalently introduced onto the uronic acid residues of alginate (DS = 0.47), as confirmed by 1H and 13C NMR and ATR-IR spectroscopy. The resulting al

Accurate quantification of polysaccharide content is indispensable for the development and quality control of glycoconjugate vaccines. Conventional methods are often laborious, require large sample volumes, or are susceptible to interference. Ultraviolet-visible (UV-Vis) spectrophotometry presents an attractive alternative but suffers from an unexplained nonlinear concentration-absorbance relationship, precluding its reliable use. Here, we reveal that molecular behavior of pneumococcal polysaccharide conjugates including the solution conformation dynamics is the principal cause of this quantif

Sialic acid (Sia) is essential for human physiology and health, as emphasized by the range of human diseases that is linked to abnormalities in the Sia pathway. Sias are typically found at the outermost part of glycoconjugates that are involved in several biological processes, including cell adhesion and signaling. Sia metabolism is key to the production of cytidine 5'-monophosphate-Sia, the building block for sialylation, and is targeted as a therapeutic strategy to ameliorate the effects of abnormal sialylation in disease. Interestingly, patients with different genetic defects in Sia metabol…

Shigellosis is an intestinal infection causing severe and often life-threatening diarrheal disease, with high prevalence in children under five years of age in low- and middle-income countries. Given the rise of antimicrobial resistance to Shigella, the World Health Organization has included this pathogen among those for which the development of new interventions is a global health priority. No vaccines against Shigella are licensed yet, but several candidates based on the O-antigen portion of lipopolysaccharides are in clinical development, and combining a Shigella vaccine with another vaccin

The Tn antigen is an attractive target for cancer vaccine development. In this study, we report the efficacies of dendrimer-based multivalent Tn antigens to elicit profound antibody generation in mice, in a subtype-specific manner. Multivalency of the Tn antigen is achieved by utilizing poly(ether imine) (PETIM) dendrimers of first-, second-, and third-generations, leading to tetra-, octa-, and hexadecavalent Tn antigen moieties at their peripheries through squarate ester linkage. The in vivo toxicities of the multivalent Tn antigens on mice show that these antigens are well-tolerated, with no

The blood-brain barrier (BBB) glycocalyx is the dense layer of glycans and glycoconjugates that coats the luminal surface of the central nervous system (CNS) vasculature. Despite being the first point of contact between the blood and brain, not much is known about the BBB glycocalyx. Here, we performed a multi-omic investigation of the BBB glycocalyx which revealed a unique glycan landscape characterized by enrichment of sialic acid, chondroitin sulfate, and hyaluronan. We found that the BBB glycocalyx was thicker than glycocalyces in the peripheral vasculature and that hyaluronan was the majo…

GM1 gangliosidosis is an autosomal recessive neurodegenerative lysosomal storage disease caused by pathogenic variants in the GLB1 gene, limiting the production of active lysosomal β-galactosidase. Phenotypic heterogeneity is due in part to variant type, location within GLB1, and the amount of residual enzyme activity; in the most severe form, death occurs in infancy. With no FDA approved therapeutics, development of efficacious strategies for the disease is pivotal. CRISPR/Cas based approaches have revolutionized precision medicine and have been indispensable to the development of treatments

Francisella tularensis, a category A bioterrorism agent, causes tularemia in many animal species. F. tularensis subspecies tularensis (type A) and holarctica (type B) are mainly responsible for human tularemia. The high mortality rate of 30-60 % caused by F. tularensis subspecies tularensis if left untreated and the aerosol dispersal renders this pathogen a dangerous bioagent. While a live attenuated vaccine strain (LVS) of F. tularensis type B does not provide sufficient protection against all forms of tularemia infections, a significant level of protection against F. tularensis has been obse

Capsular polysaccharides (CPSs), with which most pathogenic bacterial surfaces are decorated, have been used as the main components of glycoconjugate vaccines against bacterial diseases in clinical practice worldwide. Pneumococcal conjugate vaccines (PCVs) are administered globally to prevent invasive pneumococcal disease (IPD). While PCVs have played important roles in controlling IPD in all age groups, their empirical, and labor-intensive chemical conjugation yield poorly characterized, heterogeneous, and variably immunogenic vaccines, with poor immune responses in high-risk populations such

In this paper, a new class of novel sulfonamides incorporating aminosaccharide tails were designed and synthesized based on the sugar-tail approach. Then, all the novel compounds were evaluated for their inhibitory activities against three carbonic anhydrase (CA, EC 4.2.1.1) isoenzymes (hCA I, hCA II and hCA IX). Interestingly, effective inhibition of these three CA isoforms were observed, especially the glaucoma associated isoform hCA II. It is worth noting that these glycoconjugated sulfonamide derivatives also showed better CA inhibitory effects compared to the initial segment carzenide. Am

OBJECTIVE: Neisseria meningitidis is a Gram-negative bacterium that causes meningitis. N. meningitidis serogroup W (NmW) capsule polymerase synthesizes capsular polysaccharide of this serogroup. This enzyme could be a tool for meningococcal glycoconjugate vaccine development. Our long-term goal is to control activity of the NmW capsule polymerase for production of defined carbohydrates for vaccines. The enzyme lacks a simple, high-throughput activity assay. Here, we describe the use of high-throughput bioluminescence assays (CMP-Glo and UDP-Glo by Promega) to investigate NmW capsule polymerase
