NMR analysis of interaction of LqhalphaIT scorpion toxin with a peptide corresponding to the D4/S3-S4 loop of insect para voltage-gated sodium channel.

Voltage-gated sodium channels (Navs) are large transmembrane proteins that initiate action potential in electrically excitable cells. This central role in the nervous system has made them a primary target for a large number of neurotoxins. Scorpion alpha-neurotoxins bind to Navs with high affinity and slow their inactivation, causing a prolonged action potential. Despite the similarity in their mode of action and three-dimensional structure, alpha-toxins exhibit great variations in selectivity toward insect and mammalian Navs, suggesting differences in the binding surfaces of the toxins and th

Characterization of Mosquito Salivary Components.

Mosquito saliva is a powerful cocktail of bioactive molecules that help blood feeding and, in virtue of their immunomodulatory properties, may modify the local environment at the bite site facilitating transmission and establishment of mosquito-borne pathogens. We describe here some basic experimental procedures for studying and characterizing mosquito salivary components. These include mosquito salivary gland dissection, preparation of salivary gland protein extracts, and collection of mosquito saliva, both on a small scale from individual mosquitoes and on a larger scale by artificial membra

Sast1-mediated manifold effects inhibit Plutella xylostella fertility.

BACKGROUND: Plutella xylostella (Linnaeus) is a destructive pest of cruciferous crops due to its strong reproductive capacity and extensive resistance to pesticides. Seminal fluid proteins (SFPs) are the main effective factors that determine the reproductive physiology and behaviour of both sexes. Although an increasing number of SFPs have been identified, the effects of astacins in SFPs on agricultural pests have not yet been reported. Here, we elucidated the mechanisms by which Sast1 (seminal astacin 1) regulates the fertility of Plutella xylostella (L.). RESULTS: PxSast1 was specifically e

The molecular identification, odor binding characterization, and immunolocalization of odorant-binding proteins in Liriomyza trifolii.

The Liriomyza trifolii is a highly invasive polyphagia pest. Understanding the physiological functions of odorant binding proteins (OBPs) in the chemical communication of L. trifolii can lead to effective pest management strategies. Seven full-length OBPs were identified by transcriptome screening of L. trifolii adults. Bioinformatics analyses classified the seven OBPs into two subfamilies (six classic OBPs, one minus-C OBP). The analysis of their expression in different development stages revealed that LtriOBP5 was highly expressed in the larval stage, LtriOBP4 in the pupa stage, and LtriOBP1

THE SECRETION OF DIGESTIVE ENZYMES AND CAECAL SIZE ARE DETERMINED BY DIETARY PROTEIN IN THE CRICKET Gryllus bimaculatus.

In Gryllus bimaculatus, the size of the caecum decreases in the latter half of each instar to a stable minimal size with a steady minimal rate of digestive enzyme secretion until feeding resumes after ecdysis. The higher the percent protein in the newly ingested food, the faster and larger the caecum grows, and as a consequent the higher the secretion rate of trypsin and amylase. When hard boiled eggs (40% protein) are eaten the caecum is 2× larger, the trypsin secretion is almost 3× greater, and amylase 2.5× greater then when fed the same amount of apples (1.5% protein). Only dietary protein

SfRop is a potential target for controlling Spodoptera frugiperda through regulation of AcMNPV viral transportation.

Sec1/Munc18 (SM) proteins serve as core upstream regulators governing SNARE complex assembly. The SM protein Rop plays a critical role in SNARE-mediated membrane fusion, yet its function in baculovirus infection remains largely unexplored. Here, we demonstrate that SfRop, the Rop homolog in Spodoptera frugiperda, is essential for the infection cycle of Autographa californica multiple nucleopolyhedrovirus (AcMNPV). Knockdown of SfRop significantly reduced viral genome replication, early and late gene transcription, and intracellular viral genome copy numbers during the early infection stage. Co

Megalomyrmex milenae Transcriptome Reveals a Complex Venom Cocktail.

Megalomyrmex ant species have a rich natural history that provides an interesting backdrop to understanding how venom has been shaped by evolution. However, like many other species in the tribe Solenopsidini, alkaloid investigations have dominated, limiting our understanding of the diversity of venom components. Here we use transcriptomics to qualify and quantify the proteins and peptides within Megalomyrmex milenae, a species of ant native to the Panamanian rainforest along the Panama Canal. RNA transcripts associated with and over-expressed in the venom gland allow the description of putativ

Workers of honey bee (Apis mellifera L.) reared in small-cell combs in apiary conditions show higher activity of the proteolytic system and lower protein concentrations on the cuticle surface than workers reared in standard-cell combs.

This study is a continuation of innovative research on the impact of the simultaneous use of standard- and small-cell combs in bee colonies on the characteristics of worker bees and bee colonies. The nests of these colonies had two types of combs: small-cell (approximate cell width/size of 4.90 mm) and standard-cell (approximate cell width/size of 5.50 mm). The aim of the study was to compare the activities of proteases and their inhibitors present on the cuticle of workers reared in small-cell combs (SMC workers) and standard-cell combs (STC workers) in colonies kept simultaneously in standar

A potential link between aromatics-induced oviposition repellency behaviors and specific odorant receptor of Aedes albopictus.

BACKGROUND: The Asian tiger mosquito, Aedes albopictus, is a competent vector for the spread of several viral arboviruses including dengue, chikungunya, and Zika. Several vital mosquito behaviors linked to survival and reproduction are primarily dependent on a sophisticated olfactory system for semiochemical perception. However, a limited number of studies has hampered our understanding of the relationship between the A. albopictus acute olfactory system and the complex chemical world. RESULTS: Here, we performed a qRT-PCR assay on antennae from A. albopictus of differing sex, age and physiol

Thioester-containing proteins in the tsetse fly (Glossina) and their response to trypanosome infection.

Thioester-containing proteins (TEPs) are conserved proteins with a role in innate immune immunity. In the current study, we characterized the TEP family in the genome of six tsetse fly species (Glossina spp.). Tsetse flies are the biological vectors of several African trypanosomes, which cause sleeping sickness in humans or nagana in livestock. The analysis of the tsetse TEP sequences revealed information about their structure, evolutionary relationships and expression profiles under both normal and trypanosome infection conditions. Phylogenetic analysis of the family showed that tsetse flies

Lucilia cuprina genome unlocks parasitic fly biology to underpin future interventions.

Lucilia cuprina is a parasitic fly of major economic importance worldwide. Larvae of this fly invade their animal host, feed on tissues and excretions and progressively cause severe skin disease (myiasis). Here we report the sequence and annotation of the 458-megabase draft genome of Lucilia cuprina. Analyses of this genome and the 14,544 predicted protein-encoding genes provide unique insights into the fly's molecular biology, interactions with the host animal and insecticide resistance. These insights have broad implications for designing new methods for the prevention and control of myiasis

Mycoviruses and their role in fungal pathogenesis.

Nowadays, the focus of mycovirology research has expanded from plant pathogenic fungi and mycovirus mediated hypovirulence to include insect and human pathogenic fungi together with a range of mycovirus mediated phenotypes, such as hypervirulence, control of endophytic traits, regulation of metabolite production and drug resistance. In fungus-mycovirus-environmental interactions, the environment and both abiotic and biotic factors play crucial roles in whether and how mycovirus mediated phenotypes are manifest. Mycovirus infections result in alterations in the host transcriptome profile, via p

Expression of a highly antigenic and native-like folded extracellular domain of the human α1 subunit of muscle nicotinic acetylcholine receptor, suitable for use in antigen specific therapies for Myasthenia Gravis.

We describe the expression of the extracellular domain of the human α1 nicotinic acetylcholine receptor (nAChR) in lepidopteran insect cells (i-α1-ECD) and its suitability for use in antigen-specific therapies for Myasthenia Gravis (MG). Compared to the previously expressed protein in P. pastoris (y-α1-ECD), i-α1-ECD had a 2-fold increased expression yield, bound anti-nAChR monoclonal antibodies and autoantibodies from MG patients two to several-fold more efficiently and resulted in a secondary structure closer to that of the crystal structure of mouse α1-ECD. Our results indicate that i-α1-EC

Chemosensory System Decoding: Transcriptome-Wide Identification and Expression Profiling of Olfactory Genes in Lytta sifanica.

The insect olfactory system relies on a sophisticated repertoire of olfactory-related proteins to enable precise odor detection and subsequent signal transduction. Their expression and regulation are crucial for mediating communication between themselves and environments. Lytta sifanica (Coleoptera: Meloidae), a species of significant economic importance, represents a notable subject for olfactory studies, particularly as it is renowned for producing the medically relevant toxin cantharidin. However, the molecular basis of olfactory sensation in the Blister Beetle Lytta sifanica has not yet be

Novel expression of immunogenic foot-and-mouth disease virus-like particles in Nicotiana benthamiana.

Foot-and-mouth disease (FMD) is a highly contagious disease of cloven-hoofed animals and is endemic in Africa, parts of South America and southern Asia. The causative agent, FMD virus (FMDV) is a member of the genus Aphthovirus, family Picornaviridae. Vaccines currently used against FMDV are chemically inactivated virus strains which are produced under high-level biocontainment facilities, thus raising their cost. The development of recombinant FMDV vaccines has focused predominantly on FMDV virus-like particle (VLP) subunit vaccines for which promising results have been achieved. These VLPs a

Proteomic biomarkers for ageing the mosquito Aedes aegypti to determine risk of pathogen transmission.

Biomarkers of the age of mosquitoes are required to determine the risk of transmission of various pathogens as each pathogen undergoes a period of extrinsic incubation in the mosquito host. Using the 2-D Difference Gel Electrophoresis (2-D DIGE) procedure, we investigated the abundance of up to 898 proteins from the Yellow Fever and dengue virus vector, Aedes aegypti, during ageing. By applying a mixed-effects model of protein expression, we identified five common patterns of abundance change during ageing and demonstrated an age-related decrease in variance for four of these. This supported a

Leishmania donovani elongator protein Elp3a plays a crucial role in modulating the parasite response to genotoxic stress.

The eukaryotic elongator complex comprises six proteins Elp1-Elp6, with Elp3 being the catalytic subunit. In trypanosomatids, only Elp3 has been identifiable, and interestingly, these organisms have two Elp3 orthologs: Elp3a and Elp3b. This study was undertaken to examine the role of Elp3a in the trypanosomatid Leishmania donovani. The elp3a gene was successfully eliminated, with the insect form as well as mammalian form of the parasite surviving and propagating normally in its absence, signifying its non-essential nature under usual growth conditions. In examining the response of LdElp3a-depl

Molecular, enzymatic, and regulatory characterization of rat kidney cytochromes P450 4A2 and 4A3.

The cDNAs encoding cytochromes CYP 4A2 and 4A3 were cloned by RT-PCR amplification of male rat kidney and liver RNAs, respectively. Sequence analysis demonstrated that these cDNAs were nearly identical to the published sequences for CYPs 4A2 and 4A3. CYP 4A2 and 4A3 share extensive sequence homology that extends into their 3'- and 5'-untranslated segments ( approximately 97% overall nucleotide identity). Analysis of cDNA and genomic DNA sequences shows that a sequence of 123 bp, recognized as an intron during the processing of CYP 4A2 transcripts, is conserved in the 4A3 mRNAs and that these o

Methoprene-Tolerant (Met) Acts as Methyl Farnesoate Receptor to Regulate Larva Metamorphosis in Mud Crab, Scylla paramamosain.

The conserved role of juvenile hormone (JH) signals in preventing larvae from precocious metamorphosis has been confirmed in insects. Crustaceans have different metamorphosis types from insects; we previously proved that methyl farnesoate (MF) can prohibit larvae metamorphosis in mud crabs, but the molecular signal of this process still needs to be elucidated. In this study, methoprene-tolerant (Met) of Scylla paramamosain was obtained and characterized, which we named Sp-Met. Sp-Met contains a 3360 bp ORF that encodes 1119 amino acids; the predicted protein sequences of Sp-Met include one bHL
