Exploring alternative insecticide delivery options in a "lethal house lure" for malaria vector control.

The In2Care EaveTube is a house modification designed to block and kill malaria mosquitoes using an electrostatic netting treated with insecticide powder. A previous study demonstrated prolonged duration of effective action of insecticide-treated electrostatic netting in a semi-field setting. As part of a cluster randomized controlled trial (CRT) of the EaveTube intervention in Côte d'Ivoire, we investigated the residual efficacy of a pyrethroid insecticide deployed in EaveTubes under village conditions of use. We also explored the scope of using existing malaria control technologies including

Pharmacological and Genetic Evidence for Gap Junctions as Potential New Insecticide Targets in the Yellow Fever Mosquito, Aedes aegypti.

The yellow fever mosquito Aedes aegypti is an important vector of viral diseases that impact global health. Insecticides are typically used to manage mosquito populations, but the evolution of insecticide resistance is limiting their effectiveness. Thus, identifying new molecular and physiological targets in mosquitoes is needed to facilitate insecticide discovery and development. Here we test the hypothesis that gap junctions are valid molecular and physiological targets for new insecticides. Gap junctions are intercellular channels that mediate direct communication between neighboring cells …

A case of pubic lice resistant to pyrethrins.

BACKGROUND: Effective therapeutic protocols for the treatment of pubic lice include application of approved insecticidal products to all body and scalp hair, retreatment at seven days, and use of mechanical removal of lice and eggs. OBJECTIVE: We report here infection with pubic lice in a 43 year old male in Australia which failed to be eradicated by adequate therapy with a pyrethrin and piperonyl butoxide product. DISCUSSION: Pubic lice collected from the case survived after exposure to the same product in vitro. The infestation was cured by use of 5% permethrin. This appears to be the firs

The evolutionary origins of pesticide resistance.

Durable crop protection is an essential component of current and future food security. However, the effectiveness of pesticides is threatene … ance often evolves through selection of polygenic metabolic resistance from standing variation; and insecticide resistance evolves through a combination of standing variation and de novo mutations in the target site or major metabolic resistance genes. This has practical implications for resistance risk assessment and management, and lessons learnt from pesticide resist …

Tying pest insects in knots: the deployment of spider-venom-derived knottins as bioinsecticides.

Spider venoms are complex chemical arsenals that contain a rich variety of insecticidal toxins. However, the major toxin class in many spider venoms is disulfide-rich peptides known as knottins. The knotted three-dimensional fold of these mini-proteins provides them with exceptional chemical and thermal stability as well as resistance to proteases. In contrast with other bioinsecticides, which are often slow-acting, spider knottins are fast-acting neurotoxins. In addition to being potently insecticidal, some knottins have exceptional taxonomic selectivity, being lethal to key agricultural pest

Impact of ivermectin-blood meals from treated cattle on mortality and reproductive parameters of Aedes aegypti (Diptera: Culicidae) for vector control.

Growing of insecticide resistance in Aedes aegypti underscores the need for alternative vector control tools. Ivermectin, an endectocide with mosquitocidal properties, is currently evaluated against Anopheles for residual malaria control. Its potential use against Ae. aegypti within integrated strategies also warrants investigation. This study assessed the effects of ivermectin-treated cattle blood on the survival and reproductive parameters of two Ae. aegypti strains: Bora Bora (insecticide susceptible) and Bobo (insecticide resistant). Laboratory-reared females were fed on cattle treated wit…

Time-series analysis of transcriptomic changes due to permethrin exposure reveals that Aedes aegypti undergoes detoxification metabolism over 24 h.

Insecticide resistance is a multifaceted response and an issue across taxa. Aedes aegypti, the mosquito that vectors Zika, dengue, chikungunya, and yellow fever, demonstrates high levels of pyrethroid resistance across the globe, presenting a challenge to public health officials. To examine the transcriptomic shifts across time after exposure to permethrin, a 3'Tag-Seq analysis was employed on samples 6, 10, and 24 h after exposure along with controls. Differential expression analysis revealed significant shifts in detoxifying enzymes and various energy-producing metabolic processes. These fin…

Development of PCR and RPA-LFD molecular diagnostic assays for monitoring pyrethroid resistance in Chinese populations of Spodoptera exigua based on an intronic indel marker.

Widespread lambda-cyhalothrin resistance in Spodoptera exigua severely limits chemical control efficacy. Here, we assessed the frequency of resistance-associated CYP9A9a mutations across wild populations. Three mutations (V40L, Q160H and T208A) were highly prevalent in all six field populations tested (carrier frequencies: 70.8%-100%) but absent in the susceptible WH-S strain. Genetic analysis confirmed that a 55-bp intronic insertion in the resistant CYP9A9a allele is tightly linked to the resistant phenotype. Targeting this insertion, we developed a PCR genotyping assay that achieved 90.91%-

Nutrient and spinosad concentrations influence liquid bait consumption and mortality in spinosad-resistant melon flies (Diptera: Tephritidae).

BACKGROUND: The success of insecticide-laced liquid protein bait spray applications relies on attraction and consumption by the target pest. Commercial liquid baits often recommend a broad range of dilution rates, even though many insects adjust their intake of foods based on nutrient concentrations. This compensatory feeding response could influence the amount of toxin ingested, but this impact has not been thoroughly elucidated. We manipulated liquid diet and spinosad concentrations to reveal their effects on consumption and mortality in three lines of melon flies, Zeugodacus cucurbitae, tha

Insecticide Susceptibility of Phlebotomus argentipes and Residual Action of Alpha-Cypermethrin in India: A Multicenter Study.

Visceral leishmaniasis (VL), a potentially fatal disease caused by Leishmania donovani and transmitted by Phlebotomus argentipes sand flies, remains a public health issue in India, as evidenced by 385 cases reported in 2024. Despite low incidence, sporadic fluctuations necessitate vigilant vector control. Indoor residual spraying (IRS) is the principal strategy for controlling Ph. argentipes. After resistance to dichloro-diphenyl-trichloroethane (DDT) became widespread, alpha-cypermethrin IRS replaced it in 2016 for VL control in India. Although prior studies have assessed sand fly susceptibil

Cis-regulation of the CYP6CS1 gene and its role in mediating cross-resistance in a pymetrozine-resistant strain of Nilaparvata lugens.

Pymetrozine is currently one of the primary insecticides used to control the brown planthopper, Nilaparvata lugens Stål (Hemiptera: Delphacidae), but the long-term effectiveness of this chemical is threatened by growing issues of resistance. Previous studies in a laboratory selected strain of N. lugens, Pym-R, have shown that resistance to pymetrozine can evolve without target-site mutations. A key candidate gene identified is the cytochrome P450 gene CYP6CS1, which is overexpressed in the resistant Pym-R strain compared to the laboratory susceptible strain, Pym-S. In this study, we provide a

Bionomic responses of Spodoptera frugiperda (J. E. Smith) to lethal and sublethal concentrations of selected insecticides.

Since 2016, the invasive insect Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae) from the Americas has made maize production unattainable without pesticides in parts of Sub-Saharan Africa and Asia. To counteract this pest, farmers often resort to the use hazardous pesticides. This study aimed to investigate botanicals, microbials, and semi-synthetic insecticides in Ghana for pest control without harming local ecosystems. Under laboratory and on-station conditions, the present study evaluated the acute and sublethal responses of S. frugiperda to: (i) Pieris rapae Granulovirus (PrGV)

Treatment of pediculosis capitis: a critical appraisal of the current literature.

Pediculosis capitis is the most common ectoparasitic disease in children in industrialized countries and extremely common in resource-poor communities of the developing world. The extensive use of pediculicides with a neurotoxic mode of action has led to the development and spread of resistant head lice populations all over the world. This triggered the development of compounds with other modes of action. The current literature on treatment approaches of head lice infestation was searched, and published randomized controlled trials were critically analyzed. The following compounds/family of co

Therapy for head lice based on life cycle, resistance, and safety considerations.

The timing of head lice maturation most favorable to their survival in the presence of anti-lice agents is the maximum time as an ovum (12 days) and the shortest possible time of maturing from newly hatched nymph to egg-laying adult (8.5 days). Pediculicides that are not reliably ovicidal (pyrethroids and lindane) require 2 to 3 treatment cycles to eradicate lice. Ovicidal therapies (malathion) require 1 to 2 treatments. Treatment with an agent to which there is genetic resistance is unproductive. In the United States, lice have become increasingly resistant to pyrethroids and lindane but not
