Exploring the multilayered response of TB bacterium Mycobacterial tuberculosis to lysosomal injury.

Mtb subverts host immune surveillance by damaging phagolysosomal membranes, exploiting them as replication niches. In response, host cells initiate a coordinated LDR, integrating membrane repair, selective autophagy, and de novo biogenesis. This review delineates a systems-level model of lysosomal quality control governed by three critical regulatory axes: LGALS3/8/9, TRIM E3 ubiquitin ligases, and the AMPK-TFEB signaling pathway. LGALSs detect exposed glycans on ruptured membranes, triggering ESCRT-mediated repair and recruiting ARs. TRIM proteins mediate context-specific ubiquitination, enha

Understanding stress from a cultural perspective: the role of self-construal.

Stress is an adaptive psychophysiological process that plays an important role in human survival and adjustment. Increasing evidence suggests that stress responses are shaped not only by environmental demands but also by cultural and self-related psychological processes. Self-construal, which refers to how individuals define themselves in relation to others, has been widely regarded as an important mechanism linking culture to cognition, emotion, motivation, and behavior. However, its role in stress-related processes has not been systematically synthesized. The current narrative review integra

Neuroendocrine pharmacology of stress.

Exposure to hostile conditions initiates responses organized to enhance the probability of survival. These coordinated responses, known as stress responses, are composed of alterations in behavior, autonomic function and the secretion of multiple hormones. The activation of the renin-angiotensin system and the hypothalamic-pituitary-adrenocortical axis plays a pivotal role in the stress response. Neuroendocrine components activated by stressors include the increased secretion of epinephrine and norepinephrine from the sympathetic nervous system and adrenal medulla, the release of corticotropin

PP4 deficiency drives airway epithelial senescence via the PERK-eIF2α-ATF4-p21 axis in severe asthma.

BACKGROUND: Cellular senescence, defined by irreversible cell cycle arrest and the senescence-associated secretory phenotype (SASP), has emerged as a critical driver of airway inflammation and hyperresponsiveness in asthma. Despite well-established associations between senescence, aging, and chronic disease, its precise mechanistic role in asthma pathogenesis remains poorly understood. Protein phosphatase 4 (PP4), a serine/threonine phosphatase with broad physiological functions, is significantly downregulated in airway epithelial cells derived from patients with severe asthma, suggesting a po

Stress Urinary Incontinence as a Chronic Stress Model: Clinical Evidence and Emerging Epigenetic Implications for Mental Disorders.

Stress urinary incontinence (SUI) is conventionally viewed as a peripheral mechanical disorder, yet its psychological burden suggests that a broader biopsychosocial interpretation may be warranted. This perspective considers whether recurrent leakage, anticipatory anxiety, behavioural avoidance, and social evaluative threat may allow SUI to function as a chronic psychosocial stressor. It integrates clinical observations with established concepts in stress physiology, hypothalamic-pituitary-adrenal (HPA) axis regulation, allostatic load, inflammation, and stress-related epigenetic mechanisms, i

Activation of the cell integrity pathway is channelled through diverse signalling elements in fission yeast.

MAPK Pmk1p is the central element of a cascade involved in the maintenance of cell integrity and other functions in Schizosaccharomyces pombe. Pmk1p becomes activated by multiple stressing situations and also during cell separation. GTPase Rho2p acts upstream of the protein kinase C homolog Pck2p to activate the Pmk1 signalling pathway through direct interaction with MAPKKK Mkh1p. In this work we analyzed the functional significance of both Rho2p and Pck2p in the transduction of various stress signals by the cell integrity pathway. The results indicate that basal Pmk1p activity can be positive

Noise-induced inner hair cell ribbon loss disturbs central arc mobilization: a novel molecular paradigm for understanding tinnitus.

Increasing evidence shows that hearing loss is a risk factor for tinnitus and hyperacusis. Although both often coincide, a causal relationship between tinnitus and hyperacusis has not been shown. Currently, tinnitus and hyperacusis are assumed to be caused by elevated responsiveness in subcortical circuits. We examined both the impact of different degrees of cochlear damage and the influence of stress priming on tinnitus induction. We used (1) a behavioral animal model for tinnitus designed to minimize stress, (2) ribbon synapses in inner hair cells (IHCs) as a measure for deafferentation, (3)

Real-time Imaging of Response to Host-Induced Stress in Pseudomonas aeruginosa Aggregates.

Chronic infections such as those in cystic fibrosis (CF) are sustained by small, highly tolerant Pseudomonas aeruginosa aggregates that persist despite immune and therapeutic pressures. Unlike classical biofilms, these aggregates represent a distinct pathogenic unit - microscale, spatially organized communities that maintain structural integrity and physiological homeostasis under host-induced stress. However, the mechanisms that enable aggregates to remain intact under these conditions, and whether these homeostatic processes can be selectively disrupted, remain poorly defined. A key barrier

Network pharmacology and in vivo experimental studies reveal the protective effects of 6-hydroxygenistein against hypobaric hypoxia-induced brain injury.

Hypobaric hypoxia-induced brain injury (HHBI) is a progressive neurodegenerative disease that has still not been effectively treated. There are several different mechanisms involved in HHBI. Among them, oxidative stress and inflammation response predominate. 6-hydroxygenistein (4',5,6,7-tetrahydroxyisoflavone, 6-OHG) is a hydroxylated derivative of genistein with excellent antioxidant activity, however, the protective effects and underlying mechanisms against HHBI have not been clarified. In the present study, we aimed to explore the mechanisms of action of 6-OHG on HHBI using network pharmaco

Exercise, cognition, and affective disorders.

Anxiety and depressive disorders are major contributors to global disability and are frequently accompanied by cognitive impairment, stress-system dysregulation, and cardiometabolic comorbidity. Although pharmacological and psychotherapeutic treatments remain central, limited access, partial response, and persistent residual symptoms highlight the need for integrative and scalable interventions. This narrative review synthesizes evidence on physical activity and structured exercise as systemic biological interventions for depression and anxiety, integrating clinical findings with mechanistic p

Exertional Heat Illness in Active Populations.

AbstractExertional heat illness (EHI) is a leading yet preventable cause of morbidity and mortality in active populations. EHI develops when the body's integrated thermoregulatory responses, including cardiovascular adjustments and evaporative cooling, are unable to dissipate the combined burden of metabolic heat production, environmental heat stress, clothing or protective equipment, and other contributing factors. This review examines the physiological mechanisms that underlie EHI and the effects of heat stress on the exercising heart. We also summarize best practices for the prevention, rec

Translational insights from nonclinical studies of AAV gene therapies for hemophilia: mechanisms underpinning variability and durability of gene expression.

Adeno-associated virus (AAV) gene therapy is a promising approach for hemophilia, offering the potential for sustained therapeutic expression of coagulation factors. However, both variability and durability of transgene expression remain a challenge, limiting treatment predictability. Comparative preclinical and human liver biopsy studies suggest that transcriptional efficiency, rather than vector genome copy number (VCN), is a primary determinant of variability and durability in treatment response. Despite the presence of vector genomes in hepatocytes, transcriptional output varies significan

Chronic stress and cognitive dysfunction in myalgic encephalomyelitis/chronic fatigue syndrome: HPA axis dysregulation and hippocampal plasticity.

Cognitive dysfunction is a common and disabling clinical feature of myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS), often described by patients as "brain fog." These symptoms typically manifest as difficulties in attention, memory, and concentration. Chronic stress has been proposed as an important contributing factor in ME/CFS. The hypothalamic-pituitary-adrenal (HPA) axis plays a central role in the stress response, and prolonged adverse stress may contribute to HPA axis dysregulation, including altered cortisol rhythmicity and impaired negative feedback regulation. Such dysregu

Exploring the association between stress, coping, and sleep in a Midwestern College of Pharmacy Community: A mixed methods study.

INTRODUCTION: Stress, coping, and sleep quality are determinants of well-being in health professions education. Despite their importance, little is known about how these constructs interact among different academic roles. This study examined stress, coping, and sleep quality among members of a college of pharmacy, identified predictors of sleep quality, and explored the role of medication use. METHODS: A cross-sectional, mixedmethod survey was employed. Measures included self-reported medication use and validated Perceived Stress Scale, Toulouse Coping Scale, and Brief Pittsburgh Sleep Qualit

Developing a functional non-animal CNS stress model utilizing long-term potentiation with human iPSC-cortical neurons to evaluate therapeutics.

Cortisol, the main stress hormone of the hypothalamic-pituitary-adrenal (HPA) axis, has been hypothesized to cause considerable detriment to cognitive function in both a time and concentration-dependent manner. However, there is a current lack of functional in vitro models available to evaluate the stress condition. Long-term potentiation (LTP) has served as a quantitative correlate for memory and learning through in vitro neuronal network responses to electrical stimuli, where a high-frequency stimulation (HFS) protocol on microelectrode arrays (MEAs) evaluates synaptic integrity related to h

PD-L1-Targeted and ROS-Responsive Nanoplatform for Simultaneous Chemotherapy and Dual-Modal Imaging of Treatment Response in Triple-Negative Breast Cancer.

The evaluation of treatment response in triple-negative breast cancer (TNBC) still primarily relies on tumor size and ex vivo analyses, which provide very limited information on how tumors dynamically respond during therapy. In fact, oxidative stress rapidly emerges early after drug treatment, while PD-L1 reflects an adaptive signal associated with tumors. However, these treatment-associated features are commonly assessed using static detection methods, thereby limiting longitudinal in vivo evaluation. To address this, we developed a theranostic nanoprobe named RAPP, which integrates DOX deliv

Chronic Stress Segregates Mice into Distinct Behavioral Phenotypes Based on Glucocorticoid Sensitivity.

Chronic stress alters hypothalamic-pituitary-adrenal (HPA) axis function, affecting corticosterone regulation and adaptive responses. Understanding individual variability in stress adaptation requires identifying distinct HPA axis response patterns. Here, we assessed HPA axis sensitivity in male C57BL6 mice exposed to 30 days of chronic social defeat stress (CSDS). Negative feedback integrity was evaluated using the dexamethasone suppression test (DST), with corticosterone measured after saline or low-dose dexamethasone administration at days 10 and 30. Behavioral testing (open field, elevated
