Immunoexpression of androgen receptors and aromatase in testes of patient with Klinefelter's syndrome.

Klinefelter's syndrome (47, XXY) is the most common chromosome aneuploidy in men and is usually characterized by underdeveloped testes and s … histochemistry, nuclear AR staining was detected in Sertoli cells and peritubular cells, whereas in Leydig cells the staining was exclusively cytoplasmic. The immunostaining of aromatase was detected in the cytoplasm of Sertoli cells and Leydig cells. Increased levels of gonadotropins and decreased level of testosterone concomitantly with the cytoplasmic local …

Comparison of immunocytochemical and molecular features with the phenotype in a case of incomplete male pseudohermaphroditism associated with a mutation of the luteinizing hormone receptor.

We report the case of an infant who presented at birth with a hypoplastic phallus associated with hypospadias. Low testosterone production, normal serum levels of steroid precursors, and increased LH in response to LH-releasing hormone supported a defect in Leydig cell differentiation or function. Conventional microscopic study of the testes showed fibroblastic cells in the interstitium. However immunocytochemical analysis using anti-LH receptor and anti-P450c17 antibodies demonstrated that about one third of these cells were Leydig cells or precursors of Leydig cells. No histological feature …

Human testicular insulin-like factor 3 and endocrine disrupters.

The hormone insulin-like factor 3 (INSL3) is produced by testicular Leydig cells. Production of INSL3 is dependent on the state of Leydig cell differentiation and is stimulated by the long-term trophic effects of luteinizing hormone. INSL3 is, along with the other major Leydig cell hormone testosterone, essential for testicular descent, which in humans should be completed before birth. The incidence of cryptorchidism (incomplete descent of the testis) may have increased in some developed countries during recent decades. Experimental studies have shown that maternal exposure to endocrine-disrup…

The effects of ethane dimethane sulphonate (EDS) on bilaterally cryptorchid rat testes.

A single dose of ethane dimethane sulphonate (EDS) given to adult male rats has a specific destructive effects on Leydig cells, which are removed from the intertubular area by macrophages. This is associated with a decrease in testosterone (T) and a rise in serum FSH and LH for 21-28 days after EDS. Recovery of Leydig cells occurs from connective tissue precursors. This study uses the bilaterally cryptorchid rat to investigate the influence of local factors from the seminiferous tubules on the restoration of Leydig cell morphology and function. Morphometric data of intertubular tissue, hCG bin…

Testicular steroidogenesis in the testicular feminized (Tfm) mouse: loss of 17 alpha-hydroxylase activity.

Testicular feminized (Tfm) mice are totally insensitive to androgen and may be used to study the role of the androgen receptor in normal development and function. We have examined testicular and Leydig cell steroidogenesis in Tfm mice. Serum bioactive LH was high in Tfm mice but serum testosterone was low and this was associated with a severe reduction in testicular testosterone production in vitro. Examination of [3H]pregnenolone metabolism by testes of Tfm mice indicated that progesterone, rather than testosterone, was the major steroid produced. Leydig cells were isolated from normal and Tf…

Rotenone-induced cell apoptosis via endoplasmic reticulum stress and PERK-eIF2α-CHOP signalling pathways in TM3 cells.

Rotenone (ROT), a widely used natural pesticide, has an uncertain effect on reproductive toxicity. In this study, we used 20 mice distributed randomly into four groups, with each group receiving ROT doses of 0, 2, 4, and 8 mg/kg/day for 28 days. The results demonstrated that ROT induced significant testicular damage, including impaired spermatogenesis, inhibition of testosterone synthesis, and apoptosis of Leydig cells. Additionally, ROT disrupted the normal ultrastructure of the endoplasmic reticulum (ER) in testicular tissue, leading to ER stress in Leydig cells. To further explore whether R…

An oestrogen-producing seminoma responsible for gynaecomastia.

In feminising testicular tumours, oestrogens can be either secreted by the tumour itself or produced by normal Leydig cells in response to paracrine and/or endocrine stimulation by hCG. Typical hormonal Leydig cell tumour patterns include: plasma oestradiol levels > 300 pmol/l on day 3 following an hCG injection, reduced plasma testosterone, and normal plasma hCG and gonadotrophin levels. Except for elevated plasma oestradiol levels, opposite results are observed in seminomas. We report a case of oestrogen-secreting seminoma mimicking a Leydig cell tumour. A 24-year-old Caucasian patient had c…

Corticotropin-releasing factor: an antireproductive hormone of the testis.

Corticotropin-releasing factor (CRF), the key neuropeptide in the stress cascade, has major inhibitory actions on testicular function in addition to its known antireproductive effects at the central level (inhibition of sexual behavior and LH secretion). CRF is secreted by the Leydig cells of the testis and acts through high-affinity receptors at the Leydig cell membrane as a potent negative regulator of LH action, inhibiting gonadotropin-induced cAMP generation and androgen production. CRF is also a primary stimulus of beta-endorphin secretion by the Leydig cells, which in turn exerts paracri…

Human testicular insulin-like factor 3: in relation to development, reproductive hormones and andrological disorders.

Knockout of the gene encoding insulin-like factor 3 (INSL3) results in cryptorchidism in mice due to disruption of the transabdominal phase of testicular descent. This finding was essential for understanding the complete course of testis descensus, and wound up years of speculations regarding the endocrine regulation of this process. INSL3 is, along with testosterone, a major secretory product of testicular Leydig cells. In addition to its crucial function in testicular descent, INSL3 is suggested to play a paracrine role in germ cell survival and an endocrine role in bone metabolism. INSL3 is…

Macrophages in the interstitial tissue of the rat testis.

Macrophages were identified in the intertubular tissue of the rat testis by loading animals with a particulate vital dye (trypan blue or India ink) and by localizing immunocytochemically a macrophage membrane antigen (MRC W3/25). Leydig cells were identified by the histochemical staining reaction for 3 beta-hydroxysteroid dehydrogenase activity and by a monoclonal antibody. Macrophages were scattered in the interstitial tissue closely attached to and mixed with the Leydig cells. They were never found in the seminiferous tubules. The macrophages comprised about 25% of all the cells in the inter…

Immunodetection of inhibin in the human testis and epididymis during normal development and in non-tumoural testicular lesions.

Plasma concentrations of inhibin are correlated with spermatogenetic function. Inhibin is secreted mainly by the Sertoli and Leydig cells of the testis. In the human epididymis, the location and function of inhibin are contentious. Thus, the aim of the present study was to determine the location of inhibin in the human epididymis. Investigations were performed in samples with normal testicular function at different stages of development, as well as in samples in which testicular function or the testicular-epididymal connection were altered. In fetal, newborn and infant testes, Sertoli and Leyd…

[Klinefelter's disease and leydigiomas. A report of one case (author's transl)].

The authors present the case of a patient with Klinefelter syndrome who, at the age of 36 years, developed a multifocal Leydig cell tumour of the right testicle, presenting as hematuria. After orchidectomy, the patient developed an identical tumour in the remaining testes. This curious case of a patient in whom the clinical and laboratory features of Klinefelterer's syndrome, were masked by the Leydig cell tumour and vice versa, is analyzed. Study of this case, and a review of the literature of the three cases published in the past, offer no definitive solution to the problem of the possible l

Structural and functional abnormality of ectopic testes in rats.

Some males of a mutant strain of King-Holtzman rats exhibit an anomalous heritable defect manifested as either unilateral or bilateral ectopic testes. In the adult, these testes contain seemingly immature Sertoli and Leydig cells, seminiferous tubules greatly reduced in diameter, and exhibit arrested spermatogenesis. Thus, the affected testis is essentially sterile. An inability to produce normal amounts of testosterone and androstenedione by these gonads is probably a reflection of changes that have been effected in their Leydig cells. Thus, this study suggests that abnormal function of the L…

Adrenomedullin gene delivery reduces testicular Interstitial fibrosis via activation of the GSK-3β/β-catenin/TCF-4 signaling pathway.

This study was designed to assess whether adrenomedullin (ADM) can alleviate testicular interstitial fibrosis by triggering the GSK-3β/β-catenin/TCF-4 cascade, consequently reducing the levels of connective tissue growth factor (CTGF), fibronectin, and type I collagen. Primary Leydig cells underwent treatment with lipopolysaccharide (LPS) followed by transduction using an adeno-associated virus engineered to express ADM (Ad-ADM). Cell viability was subsequently measured employing the CCK-8 assay. Additionally, the gene and protein expression levels of fibronectin, CTGF, and collagen type I wer…

Expression of the insulin-like peptide 3 (INSL3) hormone-receptor (LGR8) system in the testis.

The new peptide hormone insulin-like peptide 3 (INSL3) is a member of the insulin-relaxin family, yet, unlike insulin, it signals through a new G-protein coupled receptor, LGR8, distantly related to the receptors for LH and FSH. INSL3 is produced in large amounts by the Leydig cells of the testis in both fetal and adult mammals. Using a combination of mRNA analysis by RT-PCR, immunohistochemistry, ligand-binding, and/or bioactivity assays, the distribution of LGR8 expression was assessed in testicular tissues and cells and in the epididymis. There was consistent agreement that LGR8 was express…

Toxic effect of heavy metals on cells isolated from the rat adrenal and testis.

Heavy metals including mercury, cadmium, cobalt, and copper (100 microM) exerted an adverse effect on the viability of isolated rat adrenal capsular (zona glomerulosa), adrenal decapsular (fasciculata and reticularis), and Leydig cells of the testis, with mercury being the most potent. Due to the decreased cell viability there was a parallel reduction in corticotropin-stimulated corticosterone production by adrenal decapsular cells and luteinizing hormone-stimulated testosterone production by Leydig cells. The results indicated a direct toxic action of these heavy metals on steroid-producing c…

Distribution of mercury and evaluation of testicular steroidogenesis in mercuric chloride and methylmercury administered rats.

Intraperitoneal administration of methylmercury chloride (MMC) and mercuric chloride (MC) to male rats in doses of 5, 10 micrograms MMC/kg or 50, 100 micrograms MC/kg for 90 days induced cellular disintegration of Leydig cells which was conspicuous on day 30 and onwards in the exposed groups. Progressive degeneration of Leydig cells and decrease in their nuclear diameter and population were associated with gradual increase in deposition of mercury. Gradual diminution of 3 beta-hydroxy-delta 5-steroid dehydrogenase activity in Leydig cells after MMC or MC treatment was correlated with different…

Branched chain amino acid sufficiency is necessary for proper luteinizing hormone response and testosterone synthesis.

Testosterone production by testicular Leydig cells (steroidogenesis) is vital to male fertility and overall male health. Information about how nutrition influences Leydig cell steroidogenesis is lacking. Branched chain amino acids (BCAAs - leucine, isoleucine, and valine) are essential amino acids and important regulators of protein synthesis and energy production. Circulating and tissue BCAA levels are tightly regulated by the enzyme branched chain a-keto acid dehydrogenase kinase (BCKDK), which inhibits their catabolism. This work explored how BCAAs, and especially leucine, modulate male fer…

Testicular gonadotrophins and their receptors in human cryptorchidism as revealed by immunohistochemistry and radioreceptor assay.

The localisation of endogenous FSH and LH was studied in 4 inguinal adult human testes by the immunoperoxidase technique utilising antisera against the beta-subunits of human FSH and LH. The content of available FSH and LH receptors was determined by radioreceptor assay. The Sertoli cells and about 10% of cells in the intersitium the Leydig cells, possibly the testicular macrophages, were similarly FSH-positive in cryptorchidism and control testes. The FSH receptor levels per testis were significantly lower in cryptorchidism than in control testes. Also the localisation of LH in Leydig cells i…
