Manufacture of liposomes by isopropanol injection: characterization of the method.

Unilamellar liposomes are conventionally prepared by rapid injection of an ethanolic solution of lipids into an aqueous medium. The aim of the present study was to control, more efficiently, vesicle diameter by using an alternative solvent. The results show that isopropanol injection is a good alternative to ethanol injection for the manufacture of liposomes. Particle size can be controlled by the variation of process parameters, such as stirring speed of the aqueous phase and injection flow rate of lipid-isopropanol solution. Diameter of vesicles obtained by this method is less affected by th

[Resistance of various protein channels to proteolytic degradation].

The influence of some proteolytic enzymes on the properties of the ionic channels induced in the lipid bilayer by three different toxic proteins was studied. It was found that pronase and trypsin did not decrease the conductance of the membranes modified by melittin, alpha-staphylotoxin and latrotoxin and changed its cation-anion selectivity. The dependence of melittin-channels selectivity on the medium pH was investigated. It was suggested that the carbonyl groups of peptide bond take part in defining cation selectivity of the melittin channel.

Exploring the Mechanism of Alyteserin-1c in Gram-Positive and Gram-Negative Bacterial Membrane Models Using a Computational Approach.

Alyteserin-1c is a 23-amino-acid cationic antimicrobial peptide exhibiting greater activity against Gram-negative than Gram-positive bacteria (Conlon et al. 2009), yet the molecular basis of this differential membrane selectivity remains incompletely understood. Using all-atom molecular dynamics simulations (200 ns × 3 independent replicates per system), we examined the interactions of a pre-assembled Alyteserin-1c hexamer with simplified bilayer models representing Gram-positive (75% PG/25% PE) and Gram-negative (75% PE/25% PG) bacterial inner membranes. The hexameric assembly maintained stru

The location of fluorescence probes with charged groups in model membranes.

The location of commonly used charged fluorescent membrane probes in membranes was determined in order to: (1) investigate the relationship between the structure of hydrophobic molecules and their depth within membranes; and (2) aid interpretation of experiments in which these fluorescent probes are used to examine membrane structure. Membrane depth was calculated using parallax analysis, a method in which the quenching induced by lipids carrying a nitroxide group at different locations in the membrane is compared. Shallow locations were found for xanthene dyes (fluorescein, eosin, Texas Red a

Selection of Host Plasma Membrane Lipids by HIV-1 Gag Protein.

Human immunodeficiency virus type 1 (HIV-1) possesses an envelope enriched with a specific set of host plasma membrane (PM) lipids, a composition that is critical for viral infectivity. Virus budding is initiated by the binding of the viral Gag protein to phosphatidylinositol-4,5-bisphosphate (PI(4,5)P2) located in the inner leaflet of the PM. However, the mechanism by which inner leaflet-associated Gag protein contributes to the enrichment of specific outer leaflet lipids, such as sphingomyelin (SM) and cholesterol (Chol), remains poorly understood. Visualization of endogenous lipids using sp

Fluorescent peptides for membrane tension and domain structure reporting.

UNLABELLED: Cell mechanics play significant roles in all aspects of cell function. While many types of fluorescent linear force sensors inserted in cellular fibrillar elements have been developed, few tools are available to track two-dimensional tension in cell membranes. Here, we present a novel principle for tension detection using a fluorescent probe based on the scaffold of the GsMTx4 peptide from Grammostola venom. Previously, we have shown that amphipathic GsMTx4 binds to lipids and inhibits mechanosensitive channels by inserting more deeply into the membrane at tensions near activation

Characterization of lipid chain order and dynamics in asymmetric membranes by solid-state NMR spectroscopy.

We studied the structure and dynamics of asymmetric POPCout/(POPE/POPG)in and POPSout/(POPE/POPG)in lipid membranes. To this end, the outer layer of multilamellar POPE/POPG (molar ratio 9 : 1) vesicles was exchanged (using methyl-β-cyclodextrin) by either chain deuterated POPC-d31 or POPS-d31, for which 2H NMR order parameters were measured. As controls, we prepared symmetric POPC-d31/POPE/POPG and POPS-d31/POPE/POPG membranes of the composition of just the outer membrane of the asymmetric multilamellar vesicles and pure POPC-d31 or POPS-d31 multilamellar vesicles. Compared to symmetric membra

Molecular mechanisms for the destabilization of model membranes by islet amyloid polypeptide.

Misfolding of human islet amyloid polypeptide (hIAPP) into insoluble aggregates is associated with Type 2 diabetes. It has been suggested that hIAPP toxicity may be due to its accumulation in pancreatic islets, causing membrane disruption and cell permeabilization, however the molecular basis underlying its lipid association are still unclear. Here, we combine solid-state NMR, fluorescence and bright field microscopy to investigate hIAPP - lipid membrane interactions. Real-time microscopy highlights a time-dependent penetration of hIAPP oligomers toward the most buried layers of the lipid vesi

Evidence for a K+ channel in bovine chromaffin granule membranes: single-channel properties and possible bioenergetic significance.

A K+ channel was incorporated into voltage-clamped planar lipid bilayers from bovine chromaffin granules and resealed granule membranes ("ghosts"). It was not incorporated from plasma membrane-rich fractions from the adrenal medulla. The channel had a conductance of approximately 400 pS in symmetric 450 mM KCl, with the permeability sequence K+ > Rb+ > Cs+ > Na+ > Li+, and was insensitive to both Ca2+ and charybdotoxin. It exhibited complex gating kinetics, consistent with the presence of multiple open and closed states, and its gating was voltage-dependent. The channels appeared to incorporat…

Flavivirus NS1 structures reveal surfaces for associations with membranes and the immune system.

Flaviviruses, the human pathogens responsible for dengue fever, West Nile fever, tick-borne encephalitis, and yellow fever, are endemic in tropical and temperate parts of the world. The flavivirus nonstructural protein 1 (NS1) functions in genome replication as an intracellular dimer and in immune system evasion as a secreted hexamer. We report crystal structures for full-length, glycosylated NS1 from West Nile and dengue viruses. The NS1 hexamer in crystal structures is similar to a solution hexamer visualized by single-particle electron microscopy. Recombinant NS1 binds to lipid bilayers and…

Competition for block of a Ca2(+)-activated K+ channel by charybdotoxin and tetraethylammonium.

Single high-conductance Ca2(+)-activated K+ channels were incorporated into planar lipid bilayers, and the discrete block by charybdotoxin (CTX), a protein inhibitor of this channel, was studied. In particular, the effect of externally added tetraethylammonium (TEA) on CTX blocking kinetics was investigated. TEA decreases the on-rate of CTX in exact proportion to its blocking of the single-channel current. The CTX off-rate is unaffected by TEA. The results demonstrate that TEA and CTX are mutually exclusive in their binding to the channel. Since the site of TEA binding is known to be located o…

Scramblases and virus infection.

The asymmetric distribution of lipids, maintained by flippases/floppases and scramblases, plays a pivotal role in various physiologic processes. Scramblases are proteins that move phospholipids between the leaflets of the lipid bilayer of the cellular membrane in an energy-independent manner. Recent studies have indicated that viral infection is closely related to cellular lipid distribution. The level and distribution of phosphatidylserine (PtdSer) in cells have been demonstrated to be critical regulators of viral infections. Previous studies have supported that the infection of human immunod

Influence of Resveratrol on Oxidation Processes and Lipid Phase Characteristics in Damaged Somatic Nerves.

It has been shown that the intensification of oxidative processes is observed when somatic nerves of rats are damaged. Accumulation of malondialdehyde occurs, and the phase properties of the lipid bilayer change, especially in the distal part of the nerve. Under the same conditions, there are multidirectional changes in the activity of antioxidant enzymes, superoxide dismutase (SOD) activity decreases, and catalase (CAT) activity increases. Under the action of resveratrol, there is a decrease in the number of TBA-active products in both areas of the damaged nerve. Alongside resveratrol action,

Mechanosensitive ion channels of E. coli activated by amphipaths.

Mechanosensitive channels have been found in more than 30 cell types, including bacterial, yeast, plant and animal cells. Whether tension is transferred to the channel through the lipid bilayer and/or underlying cytoskeleton is not clear. Using the patch-clamp method, we found that amphipathic compounds, which are molecules having hydrophobic and hydrophilic character with positive, negative or no net electric charge at pH 7, could slowly activate the mechanosensitive channels of giant Escherichia coli spheroplasts, with effectiveness proportional to their lipid solubility. The cationic or ani

Nanoscale localisation of memantine governs structural modulation and loading in bicontinuous cubic phase systems.

Memantine is a primary pharmacological treatment for moderate-to-severe Alzheimer's disease but its efficacy and brain delivery remain limited. Lipid bicontinuous cubic phases and their nanoparticle dispersions (cubosomes) offer platforms for enhancing central nervous system drug delivery, yet the rigid adamantane scaffold of memantine can destabilise lipid organisation. This study resolves how an amphiphilic, adamantane-containing drug interacts with non-lamellar lipid bilayers using monoolein-based bulk cubic mesophases and cubosomes formulated with biomimetic lipid compositions and multiple…

Investigating Liposome Membrane Properties: Insights from Langmuir Monolayer Studies in the Corona Protein Environment.

Most nanoparticles approved for medical applications are liposome formulations. In the environment of biological fluids, the corona protein was found to alter both the surface of the liposome and its size and zeta potential. As a result, it can also affect the stability of the carrier and its ability to hold a drug and release it sustainably into the cell. The key element of novelty of this research is the introduction of the Langmuir-Blodgett technique together with imaging microscopy to study the interactions between the monolayer corresponding to one liposome leaflet and the corona protein.

Membrane-anchoring engineering mediated the self-assembly of multifunctional nanocarriers: An efficient platform for astaxanthin delivery.

To overcome the instability of astaxanthin (AST) during processing and gastrointestinal delivery, this study developed a multifunctional nanocarrier through the precise self-assembly of lipid bilayers that incorporated d-α-tocopherol polyethylene glycol 1000 succinate, a widely used biocompatible surfactant known as TPGS. Multidimensional characterization results demonstrated that the incorporation of TPGS formed a stable hydration layer, contributing to the distinctive mushroom-cloud-like architecture on the liposome surface. Spectroscopic analysis confirmed that the hydrophobic segment of TP…
