Serum and Liver Lipidome Following Empagliflozin Administration for Six Months in a Fast Food Diet Mouse Model.

Empagliflozin is a sodium-glucose co-transporter inhibitor approved for the treatment of type 2 diabetes mellitus. The aim of this study was the 6-month effect of empagliflozin on serum and liver lipidome in C57BL/6J mice fed on a fast food diet (FFD). Three groups were studied; two of them fed on FFD, one with empagliflozin (EMPA group), and another without empagliflozin (FFD group); the third group fed on a chow diet and served as the control group (CD group). Following untargeted lipidomic analysis, the FFD and EMPA groups displayed largely similar serum lipid profiles, characterized by ele

A Comparative Bioinformatic Analysis of Optic Nerve Axon Regeneration Lipidomes Using the Xenopus laevis as a Model System.

Lipidomics is a rapidly growing branch of metabolomics that identifies lipid compositions of samples to learn more about disease and identify potential novel therapeutic targets. In the context of ophthalmology, lipidomic research has increased our understanding of optic nerve regeneration. The diversity of experimental designs for lipidomic research and the large datasets generated are two obstacles that must be addressed by bioinformatic tools to perform statistical analysis on lipidomics data. Our study provides an objective comparison of the features in two freely accessible web-based bioi…

Lipidomic Analysis Reveals Systemic Alterations in Servicemen Exposed to Repeated Occupational Low-Level Blast Waves.

INTRODUCTION: Occupational exposure to blast is a prevalent risk experienced by military personnel. While low-level exposure may not manifest immediate signs of illness, prolonged and repetitive exposure may result in neurophysiological dysfunction. Such repeated exposure to occupational blasts has been linked to structural and functional modifications in the brain, adversely affecting the performance of servicemen in the field. These neurological changes can give rise to symptoms resembling concussion and contribute to the development of post-traumatic stress disorder. MATERIALS AND METHODS:

Formation and lipidomic analysis of flavor substances in Tibetan dried pork slice with four different baking methods.

The research analyzed changes in lipids and volatile flavor substances in Tibetan dried pork slice (TDPS) under four baking methods: air fryer, oven baking, smoking and charcoal baking, using electronic nose (E-nose), gas phase ion mobility spectrometry (GC-IMS) and liquid chromatography-tandem mass spectrometry (LC-MS/MS). The study found that contents of alcohols (25.22 %), esters (42.66 %) and aldehydes (34.13 %) in raw meat were higher than that in TDPS. A total of 38 volatile flavor compounds were identified in TDPS, with alcohols (10), esters (9), ketones (8), and aldehydes (7) as primar

Plasma lipidomic signatures reveal age-associated patterns of septic shock risk and immune dysregulation in sepsis.

BACKGROUND: Sepsis is associated with significant lipidomic disturbances, but age-associated lipidomic patterns remain poorly characterized. Given the links between aging, immune dysfunction, and metabolic dysregulation, defining age-specific lipid profiles could improve sepsis risk assessment. This study investigates age-stratified lipidomic signatures in sepsis and identifies biomarkers for clinical severity. METHODS: We prospectively enrolled 62 sepsis patients (21 <65 years, 41 ≥65 years) and 40 healthy controls. Plasma lipidomics was performed via untargeted LC-MS/MS, identifying 1,277 …

Effects of Omega-3 PUFAs on lipid profiles and antioxidant response in depressed adolescents: A metabolomic and lipidomic study.

Adolescent depression is a significant global health challenge, with many patients responding inadequately to antidepressant treatments. Omega-3 polyunsaturated fatty acids (ω3 PUFAs) have been proposed as a potential adjunctive treatment, but their precise mechanisms remain poorly understood. This study aimed to explore the mechanisms through which ω3 PUFAs exert their antidepressant effects and to identify potential biomarkers for their therapeutic response. A comprehensive assessment of plasma metabolomic and erythrocyte membrane lipidomic was performed on 51 depressed adolescents who were

Maternal lipidomic signatures of preterm and small-for-gestational-age newborn infants in low- and middle-income countries.

Maternal lipid levels change dynamically during gestation to support normal fetal growth. To obtain a detailed footprint of these changes and their differences in pregnancies with preterm or small-for-gestational-age (SGA) neonates, we analyzed 641 lipids and 639 metabolites in plasma from women by 24 weeks of pregnancy from three cohorts from low- and middle-income countries: Bangladesh, Zimbabwe, and Kenya. We consistently found a significant lipid imbalance with increased lipid levels that preceded preterm birth and decreased levels that preceded SGA births. Changes were most pronounced in

Identification of gangliosides and ceramides as biomarkers for mucopolysaccharidosis type II (hunter syndrome) through untargeted lipidomic analysis.

INTRODUCTION: Mucopolysaccharidosis type II (Hunter syndrome) is an X-linked recessive disorder caused by iduronate-2-sulfatase deficiency, affecting mainly male patients. The lack of its enzyme activity causes the accumulation of the glycosaminoglycans heparan sulfate and dermatan sulfate in all body tissues and leads to a secondary accumulation of gangliosides and ceramides. OBJECTIVE AND METHODS: We conducted a lipidomic study to investigate the dysregulation of lipid pathways in neuronopathic MPS type II. A modified liquid extraction was performed for untargeted lipid analysis. A reverse

Deciphering ApoE Genotype-Driven Proteomic and Lipidomic Alterations in Alzheimer's Disease Across Distinct Brain Regions.

Alzheimer's disease (AD) is a neurodegenerative disease with a complex etiology influenced by confounding factors such as genetic polymorphisms, age, sex, and race. Traditionally, AD research has not prioritized these influences, resulting in dramatically skewed cohorts such as three times the number of Apolipoprotein E (APOE) ε4-allele carriers in AD relative to healthy cohorts. Thus, the resulting molecular changes in AD have previously been complicated by the influence of apolipoprotein E disparities. To explore how apolipoprotein E polymorphism influences AD progression, 62 post-mortem pat

Sexual dimorphism in lipidomic changes in maternal blood and placenta associated with obesity and gestational diabetes: A discovery study.

INTRODUCTION: The placenta uses lipids and other nutrients to support its own metabolism hence impacting the type and amount of these substrates available to the growing fetus. Maternal obesity and gestational diabetes (GDM) can disrupt placental lipid metabolism and thus lead to altered fetal growth contributing to adverse pregnancy outcomes and developmentally programing the offspring for disease in later life. Understanding obesity and GDM driven changes in placental lipid metabolism is thus important. METHODS: We collected maternal plasma and placental villous tissue following elective ce

Regiospecific analysis of lipidome in the brain from mammals of land and aquatic habitats-by liquid chromatography-mass spectrometry.

The brain is a complex organ demonstrated by the occurrence of specific types of functional lipids. Despite some studies focusing on providing the animal brain lipid signature, there are limited studies focusing on the comprehensive and regiospecific characterization of multiple animal brain lipidome. Herein we characterized about 294 lipid molecular species from six different lipid classes in different portions of the brain after fixation from mammals of different habitats, fully-aquatic (n = 6), semi-aquatic (n = 6), and terrestrial (n = 4), using liquid chromatography-mass spectrometry. High maternal adiposity during pregnancy programs an imbalance in the lipidome and predisposes to diet-induced hepatosteatosis in the offspring.

BACKGROUND: Exposure to high maternal adiposity in utero is a significant risk factor for the later-life development of metabolic syndrome (MetS), including non-alcoholic fatty liver disease (NAFLD). We have previously shown that high pre-pregnancy adiposity programs adipose tissue dysfunction in the offspring, leading to spillover of fatty acids into the circulation, a key pathogenic event in obesity-associated MetS. Herein, we hypothesized that programming of adipose tissue dysfunction in offspring born to overweight dams increases the risk for developing NAFLD. RESULTS: Females heterozygou
