Humoral immune responses in human onchocerciasis: detection of serum antibodies in early infections.

Sera of children (aged 1-15 years) living in a hyperendemic onchocerciasis area (Mali) were tested for antibodies by an enzyme-linked immunosorbent assay (ELISA) and a radioallergosorbent test (RAST), with an extract of adult Onchocerca volvulus as antigen. Serological results were related to age (equivalent to time of residence in the endemic area) and to parasitological findings (detection of skin microfilariae). This longitudinal study (over three years) allowed us to follow early antibody responses in exposed children with no parasitological or clinical evidence of infection and in childre…

[Invasive squamous cell carcinoma: about 2 cases].

Conjunctival tumors are common in tropical areas, where exposure to ultraviolet radiation is high and almost permanent. Malignant tumors are quite rare and the most represented is conjunctival squamous cell carcinoma. We report two cases of invasive squamous cell carcinoma of the conjunctiva received in the ophthalmology department of the Sominé Dolo hospital in Mopti, Mali. The patients were 25 and 51 years old, living in a rural desert area exposed to sunlight and dust. They presented with a mass developed in the area of the palpebral fissure, invading the cornea and preventing palpebral occ…

A novel variant in the GNE gene in a Malian patient presenting with distal myopathy.

GNE-myopathy (GNE-M) is a rare autosomal recessive disorder caused by variants in the GNE gene. We report a novel variant in GNE causing GNE-M in a Malian family. A 19-year-old male patient from consanguineous marriage was seen for progressive walking difficulty. Neurological examination found predominant distal muscle weakness and atrophy, decreased tendon reflexes, predominating in lower limbs. Electroneuromyography showed an axonal neuropathy pattern. However, whole exome sequencing (WES) revealed a novel biallelic variant in GNE c.1838G > A:p.Gly613Glu, segregating with the phenotype withi…

Dicrocoelium dendriticum: An Unusual Parasitological Diagnosis in a Reference International Health Unit.

Finding Dicrocoelium dendriticum eggs in human feces is exceptional and there are few prevalence data available. True infection occurs after accidental ingestion of ants containing metacercariae and spurious infection through the consumption of infected animal liver. Differential diagnosis between true and pseudo-infections is performed through stool examination after a diet free of liver. In addition, microscopy can help to differentiate the type of infection. We report six cases, all from sub-Saharan Africa, detection of this fluke at the Tropical Medicine Unit Vall d'Hebron-Drassanes (Barce

Multiple micronutrients including iron are not more effective than iron alone for improving hemoglobin and iron status of Malian school children.

Iron deficiency and anemia remain among the most important global public health problems facing school children. Helminth infections often peak at school age and aggravate nutritional risks. We conducted a 12-wk randomized controlled trial in 406 Malian anemic schoolchildren infected with Schistosoma hematobium to examine the effects of 2 doses of praziquantel (P) (40 mg/kg body weight), P + 60 mg/d iron (Fe), and/or a multiple micronutrient supplement (MM) that included 18 mg/d Fe. Supplements were administered to the children each school day (5 d/wk) throughout the study. Changes in hemoglob…

A simple kit system for rapid diagnosis of cerebrospinal meningitis in rural areas of developing countries.

A simple, easily operated, portable diagnostic kit, employing coagglutination reagents, has been developed for the rapid, bedside diagnosis of cerebrospinal meningitis. Field trials using this kit were conducted in a rural area of sub-Saharan Africa for identifying the etiological agents of meningitis outbreaks. West African village medical attendants were taught to use this kit and succeeded in making rapid specific diagnoses of meningitis cases. Other acute infections such as cholera and typhoid fever can also be rapidly diagnosed in a similar manner. This rapid diagnostic system offers appr

[Acute arthritis of knee with intra-articular presence of D.medinensis. Comparative clinical and immunologic evolution. 1 case].

The authors report a case of knee acute arthritis with intra-articular presence of microfilariae D. medinensis observed in a black African worker immigrated in France. It is a classical manifestion of dracunculosis articular complications which are however less frequent than intra-articular reaction arthritis without microfilariae, or than pyogenes arthritis due to neibourhood superinfections. The authors emphasize the significance of a double clinical and immunological supervision in the course of a convenient treatment including antibiotics, anti-inflammatory agents, local treatment, and reh

Transcriptomic analyses of differentially expressed human genes, micro RNAs and long-non-coding RNAs in severe, symptomatic and asymptomatic malaria infection.

Malaria transmission and endemicity in Africa remains hugely disproportionate compared to the rest of the world. The complex life cycle of P … analyses include individuals with severe (Gambia-20), symptomatic (Burkina Faso-15), asymptomatic (Mali-16) malaria as well as uninfected controls (Tanzania-20; Mali-36). Of the total 107 datasets retrieved, we identified 5534 differentially expressed genes (DEGs) among disease and control groups. A peculiar pattern of DEGs was observed, with individu …

Design, Synthesis, and Antifungal Activity of Novel Thiazole-2-carboxamide Derivatives Bearing Flexible Diaryl Linkers as Succinate Dehydrogenase Inhibitors.

To develop eco-friendly succinate dehydrogenase inhibitors with good fungicidal activity, various diaryl-substituted thiazole-2-carboxamide derivatives were designed and synthesized based on structure-activity relationships (SARs) and molecular docking. At 20 mg/L, most target compounds exhibited ≥80% inhibition against Valsa mali, Botrytis cinerea, Curvularia lunata, and Phytophthora capsici. Notably, compound G33 showed potent activity against B. cinerea (EC50 = 0.17 mg/L) and P. capsici (EC50 = 0.42 mg/L), outperforming commercial fungicides thifluzamide and boscalid. At 100 mg/L, G33 effec…

Vulnerability to climate-driven pathogen-specific diarrheal diseases: secondary analysis from the Global Enteric Multicenter Study and Vaccine Impact on Diarrhea in Africa follow-up.

The identification of climate-related risk factors for pathogen-specific diarrhea is critical for building resilience in high-risk and low-resource settings. Here we performed secondary analysis of published data from the Global Enteric Multicenter Study (GEMS) and Vaccine Impact on Diarrhea in Africa (VIDA) follow-up. We used daily counts of rotavirus, Shigella, Cryptosporidium, and enterotoxigenic E. coli producing heat stable toxin from two case-control studies of moderate-to-severe diarrhea (MSD) in The Gambia, Kenya and Mali (n = 9378). Using Random forests we identify influential climate…

Priming or Boosting Plasmodium falciparum Transmission-Blocking Responses With Recombinant Vaccines or Gametocyte Extract.

Plasmodium falciparum infection can induce antibody responses to the parasitic, gametocyte-expressed antigens Pfs48/45 and Pfs230. Retrospective analysis of healthy individuals from malaria-endemic Mali showed increasing prevalence of antibodies to these antigens from infancy to age 17 years, an expected response to malaria exposure. This supports examining the immune interplay between gametocyte exposure and vaccination with these antigens as transmission-blocking vaccine (TBV) candidates. Recombinant Pfs48/45 and Pfs230 were formulated with a liposome-display vaccine adjuvant system. In mice…

Mapping the Geographic Distribution of Tungiasis in Sub-Saharan Africa.

The geographic distribution of tungiasis is poorly understood, despite the frequent occurrence of the disease in marginalized populations of low socioeconomic status. To date, little work is available to define the geography of this neglected tropical disease (NTD). This exploratory study incorporated geostatistical modeling to map the suitability for tungiasis transmission in sub-Saharan Africa (SSA). In SSA, environmental suitability is predicted in 44 countries, including Angola, Nigeria, Ghana, Cameroon, Cote de Ivoire, Mali, Ethiopia, the Democratic Republic of the Congo, Kenya, Gabon, Ug…

Adaptive genetic diversification of Lassa virus associated with the epidemic split of north-central Nigerian and non-Nigerian lineages.

Lassa fever (LF) is a viral hemorrhagic fever that causes high morbidity and severe mortality annually. The disease is endemic to two geographically separate areas within tropical West Africa, one in Nigeria and the second predominantly in Sierra Leone-Guinea-Liberia-Mali. Lassa virus (LASV), the causative agent of the disease, exhibits clear delineation of phylogeography between the endemic areas. In order to characterize the genetic nature of Nigerian-non-Nigerian epidemic split, we performed molecular epidemiological analyses on non-Nigerian isolates (lineage IV as well as lineage V) and th…
